Changes in quality characteristic of immature flat persimmon (Diospyros kaki Thunb) during heat treatment aging (열처리 숙성 중 반시 미숙과의 이화학적 특성)

  • The objective of this study was to investigate physicochemical and antioxidant characteristics of immature flat persimmon during 7 heat treatment aging steps. The pH decreased, and hence, acidity increased with aging. Hunter L value decreased with aging. The a value rapidly increased until step 3, whereas the b value decreased. Fructose content decreased while glucose contents slightly increased during aging. Hardness, chewiness and gumminess rapidly decreased, indicating softening in texture, but it was negligible after aging step 3. Phenolic compounds including gallic acid and homogentisic acid were detected at step 3 and then gradually increased with aging. However, contents of flavonoid and tannin were much higher in step 1 sample than others. The antioxidant activities observed by DPPH, ABTS, and FRAP assay were the highest at step 1 possibly due to the higher levels of flavonoids and tannin acid in the step 1 sample.

Functional Chemical Components and Their Biological Activities of Houttuynia cordata and Lespedeza cuneata (어성초와 야관문의 기능성 성분 분석과 항산화, 항고혈압, 및 항당뇨 활성)

  • For this study, we prepared organic solvent fractions from methanol extracts of Houttuynia cordata and Lespedeza cuneate, and analyzed their chemical components and various biological functions such as anti-oxidation, angiotensin-converting enzyme (ACE) inhibition, and α-glucosidase inhibitory activities. We found that DPPH radical scavenging activity was highest in the ethyl acetate fractions of Houttuynia cordata (90.8%) and Lespedeza cuneata (91.2%), whereas ABTS radical scavenging activity was highest in the ethyl acetate fractions of Houttuynia cordata (86.1%) and the chloroform fractions of Lespedeza cuneata (95.6%). FRAP activity was highest in the ethyl acetate fraction of Houttuynia cordata (360.1 mg TE/g) and Lespedeza cuneata (239.2 mg TE/g). ACE inhibitory activity was highest in the chloroform fraction of Houttuynia cordata (13.2%) and Lespedeza cuneata (35.2%). And, α-glucosidase inhibitory activity was highest in the ethyl acetate fraction of Houttuynia cordata (56.3%), and the water residue of Lespedeza cuneata (93.6%). Finally, we investigated the DPPH radical scavenging activity of 20 types of pure compounds identified in Houttuynia cordata and Lespedeza cuneate. The results show that quercetin demonstrates the highest DPPH radical scavenging activity. Overall, these results help us to understand the functional chemical components of Houttuynia cordata and Lespedeza cuneate and the biological effects of these components.

Antioxidant and Cell Proliferation Effects of Acanthopanax senticosus for Extract in Human Osteoblast-like MG-63 Cell Line (가시오가피 추출물의 항산화활성 및 MG-63 조골세포 증식과 alkaline phosphatase 활성에 미치는 효과)

  • Acanthopanax senticosus is a common Asian herb also known as "Siberian Ginseng". It is often used as a traditional herbal medicine for reducing damage in the liver, kidney, bone and muscle. In the present study we investigated the ferric reducing/antioxidant power and total phenolic contents of the ethanol-/water-extracts obtained from the stems and leaves of Acanthopanax senticosus. Osteoblast cellular proliferation was evaluated using the MTT and alkaline phosphatase activity assays in the human osteoblast-like MG-63 cell line. Acanthopanax senticosus extracts exerted remarkable ferric reducing/antioxidant power and contained high amount of phenolics. Among the extracts the stem-/ethanol-extract showed the highest antioxidant activity and total phenol content. Interestingly a highly positive correlation was found between antioxidant activity and total phenol content (p < 0.01). Proliferation of MG-63 osteoblast cells was highest in the stem-/ethanol-extract and alkaline phosphatase activity significantly increased in the water-extract of the stems (p < 0.05). These findings suggest that Acanthopanax senticosus extracts have antioxidant activity for preventing oxidative stress-related diseases and may have beneficial effects on bone health through the proliferation of osteoblast cells.

Antioxidant Activity of Greek-style Yogurt with Stevia Leaf Extracts (스테비아 잎 추출물을 첨가한 그릭 요거트의 항산화 활성 변화)

  • This study was carried out to evaluate the antioxidant activity and total polyphnol content of Greek-style yogurt in 12% solid content with added stevia leaf extracts. Stevia leaf extracts used as sweetener in preparing Greek-style yogurt were prepared in hot water ($100^{\circ}C$ for 6 h 3 times, and by 70% fermented ethanol for 24 h at room temperature 3 times). The antioxidant activities were measured by assessing the radical scavenging effect through DPPH, ABTS, and FRAP assays. To identify the compounds present, total polyphenol content was evaluated using a Folin-Dennis assay. The capability to scavenge free radicals and total polyphenolic content were the highest in Greek-style yogurt containing 1% stevia extract and fermented in 70% ethanol. According to the results, antioxidant activities significantly increased when high concentration of stevia extracts were added to the yogurt (p<0.05). Therefore, these results suggest that stevia leaf extracts can be used as a source of functional compounds in Greek-style yogurt. We also suggest that fermented ethanol extraction can be used to obtain stevia leaf extracts.

Quality characteristics and antioxidant capacities of oriental melon wine depending on pretreatments (전처리 방법에 따른 참외와인의 품질특성과 항산화능)

  • Oriental melon (Cucumis melo L. var. makuwa) has been widely consumed as various processed foods, such as dried products, jelly, wine, juice, and vinegar, in Asian countries. In fruit processing, blanching and pressure treatments affect its quality, such as antioxidant activities, sensory characteristics, and etc. This study was conducted to evaluate the effects of blanching and pressure pre-treatments on oriental melon wine-making (BP, blanching and pressure filtration; BNP, blanching and non-pressure filtration; NBP, non-blanching and pressure filtration; and NBNP, non-blanching and non-pressure filtration). Physicochemical properties and antioxidant capacities by ${\alpha}$, ${\alpha}$-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, ferric ion reducing antioxidant power (FRAP) assay, and total phenolic and total flavonoid contents were measured for comparison of the different pre-treatment methods. After the aging process, the alcohol contents and pH values showed no statistical differences, whereas the amount of soluble solids, reducing sugar, and titratable acidity were slightly different among the pre-treatments (p<0.05). The samples with blanching pre-treatment showed higher antioxidant capacities than those of other pre-treatments. In the sensory evaluation, the BNP was the best in overall acceptability. Thus, this study showed that the blanching treatment enhanced the antioxidant capacities and sensory qualities of the oriental melon wine.

Quality and antioxidant charactistics of Elaeagnus multiflora wine through the thermal processing of juice (과즙의 열처리에 따른 뜰보리수 과실주의 품질 및 항산화 특징)

  • In this study, the characteristics of alcohol fermentation using Elaeagnus multiflora juice were studied under static fermentation condition in an effort to develop new types of functional wine. After 9 days of fermentation at $25^{\circ}C$, the pH, soluble solids, reducing sugar, viable cell numbers, and alcohol contents were shown to be 3.32~3.33, $7.8{\sim}9.0^{\circ}C$, 29.84~31.05 g/L, 7.26~8.73 cfu/mL, and 11.0%, respectively. The heat treated juice exhibited significantly higher antioxidant activity than untreated juice while the soluble phenolic and flavonoid contents became higher. Also, the fermented wine after the heat treated at $120^{\circ}C$ for 30 min contained free sugar such as fructose (0.42 g/L) and glucose (0.09 g/L), major organic acids such as lactic acid (7.32 g/L), malic acid (2.59 g/L), succinic acid (2.16 g/L), and oxalic acid (3.08 g/L), and major flavanols and phenolic acids such as catechin (99.45 mg/L), epicatechin (264.55 mg/L), epigallocatechin (82.19 mg/L), gallic acid (6.44 mg/L), and salicylic acid (60.53 mg/L). In addition, DPPH radical and ABTS radical scavenging activities and FRAP assay results were 70.47%,, 65.93%, and 1.254, respectively. These results suggest that it is possible to produced a new type of wine using Elaeagnus multiflora fruits.

Antioxidant Effects of Picrasma quassioides and Chamaecyparis obtusa (S. et Z.) ENDL Extracts (소태나무 잎 및 편백나무 추출물의 항산화 효과)

  • In our study, we investigated the antioxidant effect of methanol extract from the leaves of Picrasma quassioides (PQ) and Chamaecyparis obtuse (S. et Z.) ENDL (CO). Total polyphenol contents of methanol extracts from PQ and CO varied from 138.3 to $367.52{\mu}g/mg$ and total flavonoid contents varied from 8.12 to $46.41{\mu}g/mg$. Contents of polyphenol and flavonoid in PQ were found to be extremely high. In addition, the methanol extract of PQ had a higher antioxidant activity in both DPPH ($4.79{\mu}g/ml$) and ABTS ($7.21{\mu}g/ml$) compared to other plants (CO). Based on the results of the FRAP assay, PQ showed a value of $8.52{\mu}mol/{\mu}g$ and CO exhibited a value of $1.77{\mu}mol/{\mu}g$. The methanol extracts from the leaves of PQ showed the highest radical-scavenging activity in various antioxidant systems.

Neuronal Cell Protection and Antioxidant Activities of Hot Water Extract from Commercial Buckwheat Tea (시판 메밀차 열수 추출물의 항산화 및 신경세포 보호효과)

  • The antioxidant and neuronal cell-protective effects of hot water extract from commercial buckwheat tea (CBTE) were evaluated. The 2,2'-azino-bis(3-ethyl-benzthiazoline-6-sulfonic acid) (ABTS) radical scavenging activity, ferric reducing antioxidant power (FRAP), and malondialdehyde (MDA) inhibitory effect of the CBTE increased in a dose-dependent manner. The Intracellular reactive oxygen species (ROS) accumulation that resulted from hydrogen peroxide ($H_2O_2$) treatment more significantly decreased when CBTE was present in the media than when the PC12 cells were treated only with $H_2O_2$. In the neuronal cell viability assay using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazoliumbromide (MTT), the aqueous extracts showed a protective effect against $H_2O_2$-induced neurotoxicity, and the lactate dehydrogenase (LDH) release into the medium was also inhibited by CBTE. The total phenolics of CBTE was 9,608.10 mg/100 g, and the major phenolic compounds were rutin (13.42 mg/100 g) and quercitrin (0.90 mg/100 g). These data suggested that CBTE, including the aforementioned phenolics, may be useful in reducing the risk of neurodegenerative disease.

Antioxidant and anti-inflammatory effects of seed ethanol extracts of Rubus coreanus miquel (복분자 종자 추출물의 항산화 및 항염증 효과)

  • This study investigated the antioxidant and anti-inflammatory effects of seed ethanol extracts from Rubus coreanus Miquel (SERC). To investigate the antioxidant activity, total polyphenol and flavonoid content, ABTS and DPPH radical scavenging activity, and reducing power were measured. The total polyphenol and flavonoid contents in seed ethanol extracts of R. coreanus Miq. were 4.09 ㎍ gallic acid equivalents (GAE)/mg and 16.25 ㎍ quercetin equivalents (QE)/mg, respectively. DPPH and ABTS radical scavenging activity showed concentration-dependent scavenging activity, and the RC50 values of SERC were 26.68 ㎍/mL and 39.30 ㎍/mL, respectively. Moreover, the ferric reducing antioxidant power (FRAP) assay was performed to assess the reducing power, and SERC showed 0.61 ± 0.01 mM FeSO4 E/mg. To measure the anti-inflammatory effect, the cytotoxicity and nitric oxide (NO) production inhibitory efficacy in RAW 264.7 cells was confirmed. SERC showed a NO production inhibitory effect at 500 ㎍/mL without cytotoxicity. As a result of verifying the antioxidant and anti-inflammatory activity using SERC, its potential as an antioxidant and anti-inflammatory material was confirmed.

Effects of steamed Polygonatum odoratum extract on inhibition of adipocyte differentiation and lowing lipid in 3T3-L1 adipocytes (증자 둥굴레 추출물의 3T3-L1 지방세포에서 분화억제 및 지질강하 효과)

  • Objectives : The purpose of this study was to investigate inhibitory effects of steamed Polygonatum odoratum extract (POE) on differentiation and adipogenesis in 3T3-L1 adipocytes. Methods : Polygonatum odoratum (P. odoratum) extract was extracted with ethyl acetate. Total phenolic and flavonoid contents in POE were measured for antioxidant activity. The spectrophotometric method was used to determine the DPPH and ABTS radical scavenging activity and ferric-reducing antioxidant potential (FRAP). MTT assay was examined for cell toxicity, oil red O staining was performed for intracelluar adipogenesis in differentiated 3T3-L1 adipocytes. Western blot analysis for measurement of CCAAT/enhancer-binding protein ${\alpha}$ ($C/EBP{\alpha}$), peroxisome proliferator-activated receptor${\gamma}$ ($PPAR{\gamma}$) and AMP-activated protein kinase (AMPK) expressions were performed. Results : The results revealed that POE has antioxidant activities. Contents of total polyphenolics and flavonoids were $50.83{\pm}1.52$ GAE mg/100g dry weight of POE and $17.05{\pm}2.47$ RE mg/100g dry weight of POE, respectively. DPPH radical scavenging activity, and FRAP in 10 mg/ml concentration were $92.1{\pm}0.6%$, $244.8{\pm}9.0{\mu}M$ Fe(II) and ABTS inhibition in 5 mg/ml concentration was $84.8{\pm}4.1%$. Treatment of POE in adipocytes inhibited the differentiation and adipogenesis of 3T3-L1 adipocytes compared to those of vehicle control. Additionally, protein expressions of $C/EBP{\alpha}$ and $PPAR{\gamma}$, major transcription factor for the adipogenic genes, were significantly decreased compared to those of vehicle control (p<0.05). Futhermore, phosphorylation of AMPK was increased in 3T3-L1 adipocytes treated with POE compared to that of vehicle control (p<0.05). Conclusions : we demonstrate that steamed P. odoratum extract (POE) has potentiating antioxidant activities, inhibits differentiation and lipid accumulation and also induces energy expenditure in adipocytes, which may contribute to antiobesity property.
