Functional Analysis of the marB gene of Escherichia coli K-12

  • Antibiotic resistance is often associated with the production of inner membrane proteins (for example, AcrAB/TolC efflux pump) that are capable to extrude antibiotics, detergents, dyes and organic solvents. In order to evaluate the unknown MarB function of Escherichia coli, especially focused on the function of OmpF porin, several mutants were construted by T4GT7 transduction. MarA plays a major roles in mar (multiple antibiotic resistance) phenotype with AcrAB/TolC efflux pump in E. coli K-12. Futhermore, MarA decreases OmpF porin expression via micF antisense RNA. Expression of acrAB is increased in strains containing mutation in marR, and in those carrying multicopy plasmid expressing marA. MarB protein of E. coli K-12 showed its activity at OmpF porin & TolC protein as target molecule. Some paper reported MarB positively regulates OmpF function. MarA shows mar phenotype, and MarB along with MarA show decreased MIC through OmpF function. By this experiment, MarB could decrease MIC through the OmpF porin & TolC protein as target.

Phylogenetic Relationships in Korean Elaeagnus L. Based on nrDNA ITS Sequences

  • Molecular phylogenetic analyses of Korean Elaeagnus L. were conducted using seven species, one variety, one forma and four outgroups to evaluate their relationships and phylogeny. The sequences of internal transcribed spacer regions in nuclear ribosomal DNA were employed to construct phylogenetic relationships using maximum parsimony (MP) and Bayesian analysis. Molecular phylogenetic analysis revealed that Korean Elaeagnus was a polyphyly. E. umbellata var. coreana formed a subclade with E. umbellata. Additionally, the genetic difference between E. submacrophylla and E. macrophylla was very low. Moreover, E. submacrophylla formed a branch from E. macrophylla, indicating that E. submacrophylla can be regarded as a variety. However, several populations of this species were not clustered as a single clade; therefore, further study should be conducted using other molecular markers. Although E. glabra f. oxyphylla was distinct in morphological characters of leaf shape with E. glabra. But E. glabra f. oxyphylla was formed one clade by molecular phylogenetic with E. glabra. Additionally, this study clearly demonstrated that E. pungens occurs in Korea, although it was previously reported near South Korea in Japan and China. According to the results of ITS regions analyses, it showed a resolution and to verify the relationship between interspecies of Korean Elaeagnus.

Development of Finite Element Model of Hybrid III 5th Percentile Female Dummy (Hybrid III 5% 성인 여성 더미의 유한요소 모델 개발)

  • As the automobile industry is developing, the number of deaths and injuries has increased. To reduce the damages from automobile accidents, the government of each country proposes experimental conditions for reproducing the accident and establishes the vehicle safety regulations. Automotive manufacturers are trying to make safer vehicles by satisfying the requirements. The Hybrid III crash test dummy is a standard Anthropomorphic Test Device (ATD) used for measuring the occupant's injuries in a frontal impact test. Since a real crash test using a vehicle is fairly expensive, a computer simulation using the Finite Element Method (F.E.M.) is widely used. Therefore, a detailed and robust F.E. dummy model is needed to acquire more accurate occupant injury data and behavior during the crash test. To achieve this goal, a detailed F.E. model of the Hybrid III 5th percentile female dummy is constructed by using the reverse engineering technique in this research. A modeling process is proposed to construct the F.E. model. The proposed modeling process starts from disassembling the physical dummy. Computer Aided Design (CAD) geometry data is constructed by three-dimensional (3-D) scanning of the disassembled physical dummy model. Based on the geometry data, finite elements of each part are generated. After mesh generation, each part is assembled with other parts using the joints and rigid connection elements. The developed F.E. model of dummy is simulated based on the FMVSS 572 validation regulations. The results of simulation are compared with the results of physical tests.

Parametric study of laterally loaded pile groups using simplified F.E. models

  • The problem of laterally loaded piles is particularly a complex soil-structure interaction problem. The flexural stresses developed due to the combined action of axial load and bending moment must be evaluated in a realistic and rational manner for safe and economical design of pile foundation. The paper reports the finite element analysis of pile groups. For this purpose simplified models along the lines similar to that suggested by Desai et al. (1981) are used for idealizing various elements of the foundation system. The pile is idealized one dimensional beam element, pile cap as two dimensional plate element and the soil as independent closely spaced linearly elastic springs. The analysis takes into consideration the effect of interaction between pile cap and soil underlying it. The pile group is considered to have been embedded in cohesive soil. The parametric study is carried out to examine the effect of pile spacing, pile diameter, number of piles and arrangement of pile on the responses of pile group. The responses considered include the displacement at top of pile group and bending moment in piles. The results obtained using the simplified approach of the F.E. analysis are further compared with the results of the complete 3-D F.E. analysis published earlier and fair agreement is observed in the either result.

Archaeomagnetic Dating of Baked Earth Samples of Samheung-ri Kilns in Gangjin Region (강진 삼흥리 가마 소토시료의 고고지자기 연대측정)

  • Investigation on relics was performed by dividing the site of Samheung-ri, Gangjin-gun, Jeonranam-do into six districts, A to F, and as a result, total 16 kilns including Celadon kilns and pottery kilns were found. Among them, total 9 kilns found from A D E F districts including 3 Celadon kilns, 6 pottery kilns, went through archaeomagnetic analysis with baked earth samples collected from them. Since those baked earth samples showed good baking conditions as well as high soil quality, it was possible to gain archaeomagnetic data with high reliability. Through archaeomagnetic analysis, it was possible to obtain archaeomagnetic dating for each of the 9 kilns corresponding to the period between A.D. 990~1250, and the results also correspond to archaeological dating estimated to fall under the period of Goryeo. Also, it was examined if there was any difference in the operational periods of the kilns by districts, and there was difference of periods from archaeomagnetic dating in the order of district D(A.D.1100) ${\rightarrow}$ E F(A.D.1180) ${\rightarrow}$ A(A.D.1210). This result tells us that the kilns in district D stopped operating the earliest and were deserted, and next, those in districts E and F almost at the same time, and lastly, those in district A operated to the last and then were disposed.

  • Various approaches to the analysis of stiffened plating in ship structures have been proposed by a number of researchers. Among them, the finite element method is known to be the most powerful method. However, for many parts of ship's structure having simple geometry and boundary conditions, the F.E.M. is often extravagant and unnecessary. In this paper, the authors have attempted to introduce the finite strip method which was proposed by Y.K. Cheung to avoid the difficulties involved in F.E.M. The results of calculations on the displacements and stresses in various plates with or without stiffeners were satisfactory, which shows the F.S.M. is useful for structural analysis of ship's plating.

  • This paper presents a simple analysis method for inverter-fed induction motor using F.S.M and equivalent circuit. This analysis is carried out to analyze an induction motor driven by the sinusoidal voltage and the inverter. The results are verified by comparing with those of the time-step F.E.A and the experiment.

  • This paper presents an efficient algorithm to estimate the maximum load level for heavily loaded power systems with the load-generation vector obtained by ELD (Economic Load Dispach) and/or short term load forecasting while utilizing the elliptic pattern of the P-e curve. It is well known the power flow equation in the rectangular corrdinate is jully quadratic. However, the coupling between e and f makes it difficult to take advantage of this quadratic characteristic. In this paper, the elliptic characteristics of P-e curve are illustrated and a simple technique is proposed to reflect the e-f coupling effects on the estimation of maximum loadability with theoretical analysis. An efficient estimation algorithm has been developed with the use of the elliptic properties of the P-e curve. The proposed algorithm is tested on IEEE 14 bus system, New England 39 bus system and IEEE 118 bus system, which shows that the maximum load level can be efficiently estimated with remarkable improvement in accuracy.

An Analysis on the Over-Potentially Deposited Hydrogen at the Polycrystalline $Ir/H_2SO_4$ Aqueous Electrolyte Interface Using the Phase-Shift Method (위상이동 방법에 의한 다결정 $Ir/H_2SO_4$ 수성 전해질 계면에서 과전위 수소흡착에 관한 해석)

  • The relation between the phase-shift profile fur the intermediate frequencies and the Langmuir adsorption isotherm at the poly-Ir/0.1 M $H_2SO_4$ aqueous electrolyte interface has been studied using ac impedance measurements, i.e., the phase-shift methods. The simplified interfacial equivalent circuit consists of the serial connection of the electrolyte resistance $(R_s)$, the faradaic resistance $(R_F)$, and the equivalent circuit element $(C_P)$ of the adsorption pseudoca-pacitance $(C_\phi)$. The comparison of the change rates of the $\Delta(-\phi)/{\Delta}E\;and\;\Delta{\theta}/{\Delta}E$ are represented. The delayed phase shift $(\phi)$ depends on both the cathode potential (E) and frequency (f), and is given by $\phi=tan^{-1}[1/2{\pi}f(R_s+R_F)C_P]$. The phase-shift profile $(-\phi\;vs.\;E)$ for the intermediate frequency (ca. 1 Hz) can be used as an experimental method to determine the Langmuir adsorption isotherm $(\theta\;vs.\;E)$. The equilibrium constant (K) for H adsorption and the standard free energy $({\Delta}G_{ads})$ of H adsorption at the poly-Ir/0.1 M $H_2SO_4$ electrolyte interface are $2.0\times10^{-4}$ and 21.1kJ/mol, respectively. The H adsorption is attributed to the over-potentially deposited hydrogen (OPD H).

The Crack Analysis and Redesign of Horizontal Fin of F-5E/F's External Fuel Tank (F-5E/F 외부 연료탱크 수평 핀 균열 분석 및 재설계)

  • In this work the replacement material for magnesium alloy was investigated and an optimized design was suggested for the horizontal fin of a fighter's external fuel tank. For the replacement of magnesium alloy, Aluminum alloy, AL 2034-T351, was selected by considering material properties and its procurement. The strength and fracture toughness properties of AL 2034-T351 are stronger than those of magnesium alloy, but the specific weight of AL 2034-T351 is heavier than that of magnesium alloy by 65%. To meet the allowable limit of C.G. shift in the tank, the design of horizontal fin was optimized by reducing the original shape by 20% and resizing the maximum thickness to 7 mm. From the results of the static and dynamic stress analysis for improving the safety factor of the joint section and the joint hole, the radius of curvature in the aft joint section of the new fin was designed as 8.5mm.
