Ultrastructure of Germ Cell during Spermatogenesis and the Reproductive Cycle of the Hanging Cultured Male Scallop Patinopecten yessoensis (Pelecypoda:Pectinidae) on the East Coast of Korea

  • Ultrastructure of germ cell differentiation during supermatogenesis and the reproductive cycle in male Patinopecten yessoensis was studied by histological and cytological observations. The gonadosomatic index (GSI) in males rapidly increased and reached a maximum in April when seawater temperature gradually increased. Then the GSI gradually decreased from May through July when spawning occurred. Accordingly, monthly changes in the GSI in males coincided with testicular maturation and spawning periods. The sperm morphology of P. yessoensis belongs to the primitive type and showed general characteristics of external fertilization species. The head of the spermatozoon is approximately $3.50{\mu}m$ in length: the sperm nucleus and acrosome are approximately $2.90{\mu}m\;and\;0.60{\mu}m$ in length, respectively. The nuclear type of the spermatozoon is vase in shape, and the acrosome is cone type. The axoneme of the tail flagellum consists of nine pairs of microtubules at the periphery and a pair of central microtubules in the center The satellite body (which is formed by the centriole) and four mitochondria appear in the middle piece of the spermatozoon. The spawning period was from April through July and the main spawning occurred from May to June when seawater temperatures gradually increased. The reproductive cycle of this species can be classified into five successive stages; early active stage (September to November), late active stage (October to March), ripe stage (February to August), spawning stage (April to July), and spent/inactive stage (July to November).

Genetic and morphometric characteristics of Korean wild mice (KWM/Hym) captured at Chuncheon, South Korea

  • Laboratory inbred mice are used widely and commonly in biomedical research, but inbred mice do not have a big enough gene pool for the research. In this study, genetic and morphometric analyses were performed to obtain data on the characteristics of a newly developing inbred strain (KWM/Hym) captured from Chuncheon, Korea. All of five Korean wild male mice have the zinc-finger Y (ZfY) gene. Also, all of 19 Korean wild mice used in this analysis have the AKV-type murine leukemia virus gene, indicating that Korean wild mice might be Mus musculus musculus. To identify the genetic polymorphism in KWM/Hym, SNP analysis was performed. In a comparison with 28 SNP markers, there was a considerable difference between KWM/Hym and several inbred strains. The homogeneity between KWM/Hym and the inbred strains was as follows: C57BL/6J (39.3%), BALB/c AJic (42.9%), and DBA/2J (50%). KWM/Hym is most similar to the PWK/PhJ inbred strain (96.4%) derived from wild mice (Czech Republic). To identify the morphometric characteristics of KWM/Hym, the external morphology was measured. The tail ratio of male and female was $79.60{\pm}3.09$ and $73.55{\pm}6.14%$, respectively. KWM/Hym has short and agouticolored hairs and its belly is white with golden hair. Taking these results together, KWM/Hym, a newly developing inbred mouse originated from wild mouse, might be use as new genetic resources to overcome the limitations of the current laboratory mice.

Effect of Oxidation on Hot Ductility Behavior of Plain Carbon Steel (탄소강의 열간연성 거동에 미치는 산화의 영향)

  • The effects of oxidation behavior on the hot ductility of plain carbon steels were investigated at various temperatures in order to simulate the continuous casting process more precisely, in which the process undergoes in air atmosphere rather than Ar atmosphere. The high temperature oxidation behavior and scale morphology of the carbon steels exposed to the air and Ar atmosphere at various temperatures were also investigated in order to assess the mechanism of the RA value decreasing in an air atmosphere. The RA values obtained from the air atmosphere were marked below 45% by the test temperature, except for over 1000${^{\circ}C}$, with the RA values remaining in low values in both the low and high temperature region, at which the RA values generally recovered in the Ar atmosphere. The surface roughness of the specimen was developed by external and internal oxidation when the specimen was deformed in an air atmosphere at high temperature, with the result being the stress concentrated at the roughness of the specimen surface, resulting in low RA values. The hot ductility in the air atmosphere was found to be likely controlled by the oxidation rate instead of the microstructures corresponding to test temperatures.

Embedding Cobalt Into ZIF-67 to Obtain Cobalt-Nanoporous Carbon Composites as Electrode Materials for Lithium ion Battery

  • Lithium ion batteries (LIBs) is a kind of rechargeable secondary battery, developed from lithium battery, lithium ions move between the positive and negative electrodes to realize the charging and discharging of external circuits. Zeolitic imidazolate frameworks (ZIFs) are porous crystalline materials in which organic imidazole esters are cross-linked to transition metals to form a framework structure. In this article, ZIF-67 is used as a sacrificial template to prepare nano porous carbon (NPC) coated cobalt nanoparticles. The final product Co/NPC composites with complete structure, regular morphology and uniform size were obtained by this method. The conductive network of cobalt and nitrogen doped carbon can shorten the lithium ion transport path and present high conductivity. In addition, amorphous carbon has more pores that can be fully in contact with the electrolyte during charging and discharging. At the same time, it also reduces the volume expansion during the cycle and slows down the rate of capacity attenuation caused by structure collapse. Co/NPC composites first discharge specific capacity up to 3115 mA h/g, under the current density of 200 mA/g, circular 200 reversible capacity as high as 751.1 mA h/g, and the excellent rate and resistance performance. The experimental results show that the Co/NPC composite material improves the electrical conductivity and electrochemical properties of the electrode. The cobalt based ZIF-67 as the precursor has opened the way for the design of highly performance electrodes for energy storage and electrochemical catalysis.

The Anatomical Identification-key of Ephedrae Herba and its adulterant (마황(麻黃)과 그 위품의 내부형태감별)

  • Objectives : Ephedra regeliana, although similar to the other E. sinica, E. intermedia, and E. equisetina, is not authorized by The Korean Pharmacopoeia and yet has also been imported under the name of 'Ma-Hwang'. The aim of this study was to perform a comparative anatomical evaluation of Ephedrae Sinicae Herba (ESH), Ephedrae Intermedicae Herba (EIH), Ephedrae Equisetinae Herba (EEH) and Ephedrae Regelianae Herba (ERH). Methods : Permanent paraffin-embedded specimens were produced using the paraffin-tertiary butyl alcohol (TBA) method, to visualize their anatomical features via light microscopy. Results : ESH and ERH transverse sections were in the shape of a long ellipse, while those of EIH and EEH were in the shape of a circle. These anatomical characteristics substantiated the results of external morphological examination. The sections revealed that each of the four samples showed significantly different cortexes. The cortex of ESH was wider than two thirds of the section, unlike that of the other samples which was relatively narrow. The xylems of ERH, EIH, and EEH demonstrated interconnectivity with other adjacent xylems, contrary to those in ESH. Therefore, the characteristic development of the xylems over time was used to distinguish ERH from the other. Furthermore, parenchymal morphology in the central cylinder was also a good criterion to differentiate between EIH and EEH, since EIH demonstrated parenchymal cells that were shaped like a rounded-triangle, whereas EEH parenchymal cells were chrysanthemum-shaped. Conclusions : The identification-keys established in this study would facilitate the accurate identification of microscopic features of the four varieties of 'Ma-Hwang'.

Electrical and Optical Properties of Fluorine-Doped Tin Oxide Films Fabricated at Different Substrate Rotating Speeds during Ultrasonic Spray Pyrolysis Deposition (초음파 분무 열분해 증착 중 기판 회전 속도에 따른 플루오린 도핑 된 주석산화물 막의 전기적 및 광학적 특성)

  • Fluorine-doped tin oxide (FTO) has been used as a representative transparent conductive oxide (TCO) in various optoelectronic applications, including light emitting diodes, solar cells, photo-detectors, and electrochromic devices. The FTO plays an important role in providing electron transfer between active layers and external circuits while maintaining high transmittance in the devices. Herein, we report the effects of substrate rotation speed on the electrical and optical properties of FTO films during ultrasonic spray pyrolysis deposition (USPD). The substrate rotation speeds were adjusted to 2, 6, 10, and 14 rpm. As the substrate rotation speed increased from 2 to 14 rpm, the FTO films exhibited different film morphologies, including crystallite size, surface roughness, crystal texture, and film thickness. This FTO film engineering can be attributed to the variable nucleation and growth behaviors of FTO crystallites according to substrate rotation speeds during USPD. Among the FTO films with different substrate rotation speeds, the FTO film fabricated at 6 rpm showed the best optimized TCO characteristics when considering both electrical (sheet resistance of 13.73 Ω/□) and optical (average transmittance of 86.76 % at 400~700 nm) properties with a figure of merit (0.018 Ω-1).

Comparison study between recovered carbon black and commercial carbon black filled epoxy conductive materials

  • Waste tire management and recycling have grown to be significant issues because they bring up a global environmental concern. Thus, turning recycled waste tires into useful products may help tackle the environmental issue. This research aims to study and compare the effect of recycled carbon black (rCB) and commercial carbon black (CB) at certain 15 vol. % of filler loading on the mechanical, thermal, morphology and electrical properties of epoxy/CB composites. For this project, epoxy resin, diethyltoluenediamine (DETDA), recovered carbon black (rCB) and commercial carbon black (CB) graded N330, N550, N660 and N774 were mixed and compared accordingly to the formulation determined. The CB content was dispersed in the epoxy matrix using the mechanical mixing technique. The distribution and dispersion of CB in the epoxy matrix affect the characteristics of the conductive composites. rCB content at 15 vol% was selected at fixed content for comparison purposes due to the optimum value in electrical conductivity results. The flexural strength results followed the sequence of rCB>N774>N660>N550>N330. As for electrical conductivity results, epoxy/N330 exhibited the highest conductivity value, while the others achieved a magnitude of X10-3 due to the highest external surface area of N330. In terms of thermal stability, epoxy/N330 and epoxy/N774 were slightly more stable than epoxy/rCB.

Deep Learning-Based Lumen and Vessel Segmentation of Intravascular Ultrasound Images in Coronary Artery Disease

  • Background and Objectives: Intravascular ultrasound (IVUS) evaluation of coronary artery morphology is based on the lumen and vessel segmentation. This study aimed to develop an automatic segmentation algorithm and validate the performances for measuring quantitative IVUS parameters. Methods: A total of 1,063 patients were randomly assigned, with a ratio of 4:1 to the training and test sets. The independent data set of 111 IVUS pullbacks was obtained to assess the vessel-level performance. The lumen and external elastic membrane (EEM) boundaries were labeled manually in every IVUS frame with a 0.2-mm interval. The Efficient-UNet was utilized for the automatic segmentation of IVUS images. Results: At the frame-level, Efficient-UNet showed a high dice similarity coefficient (DSC, 0.93±0.05) and Jaccard index (JI, 0.87±0.08) for lumen segmentation, and demonstrated a high DSC (0.97±0.03) and JI (0.94±0.04) for EEM segmentation. At the vessel-level, there were close correlations between model-derived vs. experts-measured IVUS parameters; minimal lumen image area (r=0.92), EEM area (r=0.88), lumen volume (r=0.99) and plaque volume (r=0.95). The agreement between model-derived vs. expert-measured minimal lumen area was similarly excellent compared to the experts' agreement. The model-based lumen and EEM segmentation for a 20-mm lesion segment required 13.2 seconds, whereas manual segmentation with a 0.2-mm interval by an expert took 187.5 minutes on average. Conclusions: The deep learning models can accurately and quickly delineate vascular geometry. The artificial intelligence-based methodology may support clinicians' decision-making by real-time application in the catheterization laboratory.

Gonadal Maturation and Spawning of River Puffer Takifugu obscurus Indoor Cultured in Low Salinity (저염분에서 사육한 황복 Takifugu obscurus 생식소의 성숙과 산란)

  • Monthly changes in the gonadosomatic index (GSI) and hepatosomatic index (HSI) of wild river puffer Takifugu obscurus, and water quality environment in spawning area during breeding season were investigated from March 1995 to February 1996. Monthly changes in GSI and HSI of T. obscurus, that was cultured in low salinity, were calculated. The external morphology of the gonads, germ cell differentiation during gametogenesis and the reproductive cycle with the gonad developmental phases were investigated by histological analysis. The optimum water quality environment in Ganggyung, Choongcheongnam-do, where is spawning ground of wild T. obscurus, was $15-20^{\circ}C$ (water temperature) and 0 psu (salinity). Monthly changes in the GSI in females and males reached a maximum in May, and then rapidly decreased. Therefore, it is assumed that in the natural condition the spawning period of wild T. obscurus is May to June. In females and males, it showed a negative correlationship between the GSI and HSI. The external morphology of the gonads in female and male T. obscurus, that was cultured in low salinity, is composed of a pair of saccular structure. Based on monthly changes in the GSI, it is assumed that in female T. obscurus, that was cultured in low salinity, spawn from March through May. Therefore, it showed a negative correlationship between changes in the GSI and HSI. On the whole, in females and males, it showed a similar pattern between wild and cultured T. obscurus. The reproductive cycle with the gonad developmental phases can be classified into successive five stages in females: the early growing stage, late growing stage, mature stage, ripe and spent stage, and recovery and resting stage. In males, that can be divided into successive four stages: the growing stage, mature stage, ripe and spent stage, and recovery and resting stage. In case of wild T. obscurus, the spawning period has once a year, however, those cultured in the high water temperature ($20-27^{\circ}C$) - low salinity (under 3.3 psu) condition have reproductive characteristics having possibilities of discharge of eggs and sperms year-round as a multiple spawner.

Phylogeny and Conservation of the Genus Bupleurum in Northeast Asia with Special Reference to B. latissimum, Endemic to Ulleung Island in Korea (울릉도 고유종인 섬시호를 중심으로 동북아시아 시호속 식물의 계통과 보전생물학)

  • Based on external morphology, each of five species can be classified into three groups: 1) B. falcatum group (B. falcatum, B. scorzonerifolium), 2) B. euphorbioides group (B. euphorbioides) and 3) B. longiradiatum group (B. longiradiatum, B. latissimum). B. falcatum group has cauline leaves linear or lanceolate in shape and attenuate at base and not surrounding the stem. In contrast, B. longiradiatum group and B. euphorbioides group have cauline leaves ovate, lanceolate or panduriform in shape and auriculate or cordate at base and completely surrounding the stem. The inflorescence is basically compound umbels terminated at the apex of stem. But B. euphorbioides group is small in size and pedicels are rather short and the number of the pedicel is ca. 20. On the other hand, B. longiradiatum and B. falcatum groups are large in size and their pedicels are long and the number of the pedicel is around 10. The pore of pollen aperture of B. longiradiatum and B. latissimum is partially projected or not while those of B. falcatum group and B. euphorbioides is usually remarkably projected. The number of somatic chromosomes was counted as 2n=20 in B. falcatum, 2n=12 in B. scorzonerifolium and B. longiradiatum, and 2n=16 in B. euphorbioides and B. latissimum. Although chromosome numbers of B. euphorbioides and B. latissimum are the same, the two species are not likely to relate because the karyotypes of the two species are different from each other. Based on these observations, it can be concluded that B. latissimum is most closely related to B. longiradiatum. However, molecular data indicated that the species is probably related to B. bicaule distributed in central Siberia. In terms of conservation of B. latissimum, overexploitation by human and grazing by goat are most threatened factors.
