Acacia catechu의 플라보노이드 성분 (Flavonoid Constituents of Acacia catechu)

  • 전통생약으로서 다양한 효능과 염색제로서 사용되어온 A. catechu의 식물화학적 성분연구를 위해 열수추출물 중 ethyl acetate 분획으로부터 9종의 flavonoid와 1종의 tannin을 단리하였으며, 이들의 물리화학적 성상과 spectral data로부터 구조를 확인한 결과 (+)-catechin (1), (-)-epicatechin (2), (+)-afzelechin (3), (-)-epiafzelechin (4), (+)-mesquitol (5), kaempferol (6), quercetin (7), quercetin 3-methyl ether (8) caryatin (9) 및 ellagic acid (10)로 각각 확인 동정하였다. 이들 화합물 중 caryatin (9)은 Acacia 속으로부터 처음으로 분리되었다.

Changes in Bioactive Compounds Contents of 'Maehyang' and 'Seolhyang' Strawberry Fruits by UV Light Illumination

  • The net photosynthetic rate of 'Seolhyang' strawberry plants was measured daily for 7 days after treatment at three UV illumination dosages (0, 9.8, and 29.5 $kJ{\cdot}m^{-2}$). The net photosynthetic rates of the strawberry plants with 9.8 and 29.5 $kJ{\cdot}m^{-2}$ UV light illumination decreased by 20.2 and 61.4%, respectively, at 7 days after UV light treatments. UV treatments with two illumination dosages (7.9 and 15.7 $kJ{\cdot}m^{-2}$) altered the phenolic compounds contents during the cultivation period when compared to those in the control fruits. The anthocyanin content with 7.9 $kJ{\cdot}m^{-2}$ UV light illumination of 'Seolhyang' increased by 18.7% compared with those in control fruits at the 11 DAT. However, the anthocyanin content of 'Maehyang' was not significantly different among treatments during experiment period. The highest level of ellagic acid was found with 7.9 $kJ{\cdot}m^{-2}$ UV light illumination in both cultivars at the 11 DAT. Our results showed that strawberries illuminated with UV light during cultivation period had higher bioactive compounds contents than control fruits. These results suggest that UV light treatments may be a useful non-chemical way of promoting strawberry fruits quality.

밤 귀피의 용매분획별 항산화 활성과 항산화 물질의 분리 (Antioxidative Activity of Solvent Fraction and Isolation of ANtioxidative Compound from Chestnut Husk)

  • 밤 가공공장에서 폐기되고 있는 밤의 귀피를 항산화 기능성 자원으로 활용하고자 밤 귀치의 methanol 추출물 및 용매획분을 benzoic acid 수산화법으로 hydroxyl radical 소거능과 ferric thiocyanate법으로 과산화물생성 억제능을 시험하였다. 대부분의 획분에서 항산화성을 보였으며 특히 ethyl acetate 획분에서 BHA나 $\alpha$-tocopherol과 거의 유사한 rkdgkshydroxyl radical 소거능과 지질과산화물생성 억제능을 보였다. Ethyl acetate 획분으로부터 항산화성 물질을 분리하기 위하여 Sephadex LH-20 column chromatography 및 prep HPLC를 행하여 단일 물질로 정제하였다. 분리 정제된 물질을 $^{1}$H-NMR과 $^{13}$C-NMR로 구조 분석한 결과 gallic acid로 동정하였다. 페놀산을 GC로 분석한 결과 밤 귀피의 주요 페놀산으로 ellagic acid(172.22 mg%) 와 gallic acid(107.39 mg%)가 확인되었다.

Simultaneous Determination of Anthraquinone, Flavonoids, and Phenolic Antidiabetic Compounds from Cassia auriculata Seeds by Validated UHPLC Based MS/MS Method

  • A systematic isolation and characterization study for Cassia auriculata (CA) seeds resulted in identifying antidiabetic compounds 1,3,8-trihydroxyanthraquinone and quercetin, quercetin-3-O-rutinoside, gallic acid, caffeic acid, ferulic acid, and ellagic acid. The ultra-high-performance liquid chromatography based triple quadrupole mass spectrometry methodology was developed and validated for simultaneous identification and confirmation of these compounds from CA seeds. Multiple reaction monitoring (MRM) based quantification method was developed with MRM optimizer software for MS1 and MS2 mass analysis. The method was optimized on precursor ions and product ions with the ion ratio of each compound. The calibration curves of seven bioactive analytes showed excellent linearity (r2 ≥ 0.99). The quantitation results found precise (RSD, < 10 %) with good recoveries (84.58 to 101.42%). The matrix effect and extraction recoveries were found within the range (91.66 to 102.11%) for the CA seeds. This is the first MS/MS-based methodology applied to quantifying seven antidiabetic compounds in CA seeds and its extract for quality control purposes.

재배종 및 야생 산딸기의 기능성 성분 함량과 항산화 활성 (Contents of Bioactive Constituents and Antioxidant Activities of Cultivated and Wild Raspberries)

  • 유용성분 함량이 많고 생리기능성이 우수한 산딸기 품종의 육성을 위해 국내 농가에서 재배되고 있는 7종의 산딸기와 야생에서 자생하는 2종의 산딸기를 복분자 연구소에서 재배한 후 이들을 대상으로 과실의 품질특성, 유용성분의 함량, 항산화능을 측정하였다. 충남 아산의 야생 산딸기가 크기는 작았지만 당도는 16.20%로 시료 산딸기 중 가장 높고 산도는 가장 낮았다. 경북 울릉도의 야생 산딸기는 작은 크기에 당도 및 산도가 매우 낮아 과실로서의 이용가치는 매우 낮았으나, 총 페놀성 화합물과 ellagic acid 함량은 각각 182.97, $55.25mg{\cdot}100g^{-1}FW$로 시료 중 가장 높았다. 경남 김해에서 재배되는 산딸기 중 '왕딸'은 크기가 크고 당도는 12.80%로 다소 낮은 반면, 품종이 불명확한 다른 산딸기는 야생 산딸기 정도로 크기는 작지만 당도는 14.4%로 '왕딸' 보다 높았다. 김해 지역에서 재배되는 '왕딸'과 품종이 불명확한 산딸기의 총 페놀성 화합물(159.62, $165.94mg{\cdot}100g^{-1}FW$) 및 ellagic acid(45.7, $52.1mg{\cdot}100g^{-1}FW$) 함량은 울릉도 야생 산딸기보다 낮았으나, 총 플라보노이드 화합물(14.28, $14.90mg{\cdot}100g^{-1}FW$)과 총 안토시아닌(28.69, $30.48mg{\cdot}100g^{-1}FW$) 함량은 이보다 높았다. 경북 울릉도 야생 산딸기, 김해 지역의 '왕딸' 및 품종이 불명확한 산딸기는 FRAP법에 의한 항산화능과 DPPH 및 ABTS radical 소거능도 다른 품종의 산딸기보다 현저하게 높아 육종 소재 및 기능성 성분 소재로서의 이용 가치가 높은 것으로 사료된다.

Comparison of Chemical Composition and Radical Scavenging Activity of Pomegranate Extracts from Different Growing Areas

  • The pomegranate (Punica granatum L.) is a promising source of functional food, which contains several phytochemicals that perform important roles in reducing the risk of pathological diseases. Chemical compositions, such as the total sugar, uronic acid, polyphenols, and anthocyanin contents, and radical scavenging activity were determined and compared among PEs from six different cultivation areas. Total anthocyanin contents (TAC) and total polyphenol contents (TPC) from various growing areas were detected in the following order, respectively: Spain (19.08 ${\mu}g$/mL)> Turkey (12.91 ${\mu}g$/mL)> Iran-A (6.67 ${\mu}g$/mL)> Taiwan (4.77 ${\mu}g$/mL)> Uzbekistan (1.88 ${\mu}g$/mL)> Iran-B (0.76 ${\mu}g$/mL) and Turkey (639.52 ${\mu}g$/mL)> Uzbekistan (502.19 ${\mu}g$/mL)> Spain (306.40 ${\mu}g$/mL)> Iran-B (249.20 ${\mu}g$/mL)> Taiwan (162.78 ${\mu}g$/mL)> Iran-A (143.93 ${\mu}g$/mL). PEs from six different cultivation areas were determined to vary in ellagic acid content from 8.90 ${\mu}g$/mL to 332.52 ${\mu}g$/mL. The amounts of total sugars in PE from Iran-A evidenced relatively high total sugar contents, but low uronic acid levels (11.92 mg/mL). DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activities were as follows: Turkey> Uzbekistan $\gg$ Spain> Iran-B> Iran-A> Taiwan. ABTS [2,20-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)] radical scavenging activities were detected in the following order: Turkey$\geq$ Uzbekistan$\gg$ Spain> Iran-B> Iran-A> Taiwan. These results indicate that the PEs from Turkey with higher levels of TPC, TAC, ellagic acid, and higher radical scavenging activity may constitute a promising functional source for the prevention of several degenerative diseases.

  • The methylated compound of ellagic acid was isolated from Potentilla chinensis (Rosaceae). The isolated compound was 3, 3', 4-tri-O-methylellagic acid, [2, 3, 7-trimethoxy-8-hydroxy[1] benzopyrano-[5, 4, 3, cde][1] benzopyran-5, 10-dione], $C_{17}H_{12}O_8$, m.p. $293-295^{\circ}C$. The isolation of trimethylellagic acid was conducted by the column chromatography and the identification of the compound was carried out by the methods of IR, NMR and MS spectroscopy.

복분자 딸기 잎의 페놀성 물질 (Phenolic Compounds from the Leaves of Rubus coreanum)

  • Chemical examination of the leaves of Rubus coreanum has led to the isolation and characterization of six phenolic compounds. The structures of these compounds were elucidated as kaempferol(l), quercetin(2), sodium salt of quercetin 3-O-$\beta$-D-glucuronide(3), sodium carboxylate of quercetin 3-O-$\beta$-D-glucuronide(4), ellagic acid(5) and sanguiin H-5(6) on the basis of physicochemical and spectroscopic evidences.

  • A phenolic compound isolated from the ethylacetate extract of Duchesnea indica (Andr.) Focke (Rosaceae) showed antimicrobial activities against Staphylococcus aureus, Shigella dysentriae and Pseudomonas aeruginosa. This compound was identified as ellagic acid by spectral analysis.
