Electrochemical Biosensors for Biomedical and Clinical Applications: A Review

  • There are strong demands for accurate, fast, and inexpensive devices in the medical diagnostic laboratories, such as biosensors and chemical sensors. Biosensors can provide the reliable and accurate informations on the desired biochemical parameters, which is an essential prerequisite for a patient before going for a treatment. They can be used for continuous measurements of metabolites, blood cations, gases, etc. Of these, electrochemical biosensors play an important role in the improvement of public health, because rapid detection, high sensitivity, small size, and specificity are achievable for clinical diagnostics. In this paper, the clinical applications with electrochemical biosensors are reviewed. An attempt is also made to highlight some of the trends that govern the research and developments of the important biosensors that are associated to clinical diagnosis.

  • To date, many researchers have developed a variety of biosensors to detect the biomolecular interactions. Recently, electrochemical biosensors have been attracting great interest as one of key technologies in a ubiquitous healthcare (U-healthcare) system since they are highly sensitive and feasible to miniaturize. Here we overview the current electrochemical biosensors based on strip-type, nanowire/nanotube, field effect transistor (FET), and nanogap electrode.

  • Over the past few decades, considerable research has been conducted on field-effect transistor (FET) biosensors; however, other than electrochemical sensors for pH, they have not reached the commercialization stage and still remain at the basic research level. Although several reports have been published on experiments with real biological samples, no reports exist of developments that have reached commercialization or finalized approval for use. In this paper, we explain the reason for the experiments of FET biosensors to induce spurious signals in an experimental setup and explain the existence of misunderstandings regarding the operating principle of FET biosensors owing to the spurious signals. Based on the thoughtful review of the results of previously published papers, we show that the electrochemical read-out principle of FET biosensors requires our intensive understanding of the interfacial potential between the solution and the sensor electrode for further progress in the FET biosensor research.

  • This article reviews the recent progress in nanowell array biosensors that use the label-free detection protocol, and are detected in their natural forms. These nanowell array biosensors are fabricated by nanofabrication technologies that should be useful for developing highly sensitive and selective also reproducible biosensors. Moreover, electrochemical method was selected as analysis method that has high sensitivity compared with other analysis. Finally, highly sensitive nanobiosensor was achieved by combining nanofabrication technologies and classical electrochemical method. Many examples are mentioned about the sensing performance of nanowell array biosensors will be evaluated in terms of sensitivity and detection limit compared with other micro-sized electrode without nanowell array.

  • Colorectal cancer (CRC) is one of the most prevalent diseases in modern society, constituting a serious threat to global health. Currently, routine clinical screening and early removal of precancerous polyps are the most successful methods for reducing CRC incidence and mortality. However, the high cost and invasive detection of sigmoidoscopy and colonoscopy limited the CRC-screening participation and prevention. The emergence of biosensors provides an inexpensive, sensitive, less invasive tool for detecting CRC disease biomarkers. This review highlights some of recent efforts made on developing biosensors with electrochemical and optical techniques targeting CRC specific protein biomarkers for early diagnosis and prognosis, potential applications, and future perspectives.

  • Among the many studies of carbon-based nanomaterials, carbon-based dots (CDs) have attracted considerable interest owing to their large surface area, intrinsic low-toxicity, excellent biocompatibility, high solubility, and low-cost with environmentally friendly routes, as well as their ability for modification with other nanomaterials. CDs have several applications in biosensing, photocatalysis, bioimaging, and nanomedicine. In addition, the fascinating electrochemical properties of CDs, including high active surface area, excellent electrical conductivity, electrocatalytic activity, high porosity, and adsorption capability, make them potential candidates for electrochemical sensing materials. This paper reviews the recent developments and synthesis of CDs and their composites for the proposed electrochemical sensing platforms. The electrochemical principles and future perspective and challenges of electrochemical biosensors are also discussed based on CDs-nanocomposites.

Electrochemical Immunosensor Using a Gas Diffusion Layer as an Immobilization Matrix

  • The modification of a gas diffusion layer (GDL), a vital component in polymer electrolyte fuel cells, is described here for use in the electrochemical detection of antibody-antigen biosensors. Compared to other substrates (gold foil and graphite), mouse anti-rHBsAg monoclonal antibody immobilized on gold-coated GDL (G-GDL) detected analytes of goat anti-mouse IgG antibody-ALP using a relatively low potential (-0.0021 V vs. Ag/AgCl 3 M NaCl), indicating that undesired by-reactions during electrochemical sensing should be avoided with G-GDL. The dependency of the signal against the concentration of analytes was observed, demonstrating the possibility of quantitative electrochemical biosensors based on G-GDL substrates. When a sandwich method was employed, target antigens of rHBsAg with a concentration as low as 500 ng/mL were clearly measured. The detection limit of rHBsAg was significantly improved to 10 ng/mL when higher concentrations of the 4-aminophenylphosphate monosodium salt (APP) acting on substrates were used for generating a redox-active product. Additionally, it was shown that a BSA blocking layer was essential in improving the detection limit in the G-GDL biosensor.

  • 바이오센서의 구조와 기능 그리고 종류에 대해 소개하였다. 이 분야의 연구가 과거 15년간 각국에서 경쟁적으로 연 구하였으나 glucose sensor가 상품화되 었을 뿐 연구결과를 상 품화한 것은 극히 적은 수였다. 그러나 최근 몇 년간 특히 포켓용 i-STAT point-of-care system의 도입 그리고 surface plasmon resonance와 evanescent wave 측정 장치 의 출현으로 상품화된 바이오샌서의 수가 크게 증가하는 추세에 었다. 이들 중 의료임상과 생물공정 그리고 환경오염 측정용으로 응 용되고 있는 몇 가지 상품에 대하여 약술하였다.

웰니스 의류에 적용 가능한 바이오센서 동향 연구 (A Review Study of Biosensors applicable to Wellness Wear)

  • 본 논문은 전기적 감지 방식 바이오센서의 개념을 리뷰하고, 의류 및 텍스트 기반의 바이오센서의 연구 사례를 조사하였다. 생체 신호를 측정 할 수 있는 바이오센서는 생물학적 감지 물질을 이용하여 생물학적 물질의 물리적, 화학적 특성을 감지하는 장치이다. 따라서 바이오센서를 사용하여 생체신호를 측정할 수 있는 웰니스 의류는 U-Health 서비스를 제공하는데 중요한 역할을 한다. 기존 센서와 다르게 바이오센서의 차별화된 특징은 선택적 반응과 생물학적 물질의 결합을 사용한다는 점이다. 이러한 바이오센서 중 전기적 감지 바이오센서는 전기 신호의 처리로 인해 크기가 매우 작아 유비쿼터스 환경을 조성하는데 이용될 수 있다. 따라서 웰니스 의류를 개발하기 위해 소형화가 쉬운 전기적 감지 바이오센서를 연구할 필요가 있다. 본 논문에서는 전기적 감지 바이오센서(전기화학적 방식, 나노와이어/탄소나노튜브 기반 FET 방식)에 대해 자세히 기술하였다. 마지막으로, 이러한 고찰을 통해 향후 웰니스 의류에 적용 가능할 바이오센서의 기술개발 방향을 제언하였다.
