Determination of Veterinary Antibiotic Residues: IV. Comparable Analytical Methods with EPA Methods 1694_A Review (시료 중 잔류 항생제 분석 방법: IV. EPA method 1694와 비교 가능한 기기 분석 방법)

  • In this study, 16 antibiotics were selected from among the top 30 veterinary antibiotics sold in South Korea in 2014, as well as from among the pharmaceuticals targeted by EPA method 1694, in order to review analytical methods for the detection of trace levels of antibiotics in environmental samples: surface water, soils, animal origin foods, and manures. LC-MS/MS was heavily used. In the chromatography for the detection of the selected antibiotics, the $C_{18}$ column was mostly used at the temperature of $30{\sim}40^{\circ}C$. Water and methanol/acetonitrile were commonly chosen as a nonpolar and a polar mobile phase, respectively. Gradient elution was applied to separate multiclass antibiotics. Volatile additives, such as formic acid, acetic acid, and ammonium acetate were mixed with the mobile phase to improve the ionization efficiency of analytes and the sensitivity in MS detection. Electrospray ionization (ESI) was widely used in the LC-MS/MS and positive ionization was preferred to determine the selected antibiotics. A protonated $[M+H]^+$ molecule was selected as a precursor ion, and its two transitions were analyzed, one for quantitative measurement and the other for confirmation. This study reviewed linearity of the calibration curve, recovery, repeatability, method detection limits (MDLs), and method quantification limits (MQLs) for each target compound used to validate the developed analytical methods.

  • This study explored the analytical conditions for 21 veterinary antibiotics which have been popularly sold in South Korea in 2014 but have not yet been targeted in EPA method 1694. Most of the selected antibiotics were separated by a reverse-phase C18 column with a combination of (buffered) water and organic polar solvent, which was commonly methanol and acetonitrile in the gradient elution mode. Volatile additives such as formic acid, ammonium acetate and ammonium formate were usually added to the mobile phases to minimize asymmetrical and tailing of antibiotics' peaks and to increase their ionization in mass spectrometry. The analytical methods of aminoglycoside antibiotics were distinct from those of the other antibiotics in terms of adoption of ion-pair chromatography (IPC) and hydrophilic interaction liquid chromatography (HILIC) capable of retaining and separating extremely polar compounds due to their hydrophilicity. Trifluoroacetic acid or heptafluorobutyric acid was frequently added to the mobile phase as an ion-pair reagent for the IPC. Tandem mass spectrometry was numerously applied to the detection of antibiotics using positive electrospray ionization (ESI) and the selected reaction monitoring (SRM) mode. All reviewed analytical methods had been/were validated by evaluating recovery, limits of detection and quantification, decision limit or detection capability of the methods.

  • Discovery's collection was originally paper documents, but with the advent of the digital age, its collection expanded. Resolving the issue of ESI has now become important in litigation. Therefore, this study analyzes the requirements of each domain for electronic records to be recognized as evidence. It also explained the factors that should be considered in record management. In addition, eDiscovery in the United States was selected as an advanced case to utilize records as evidence. This study investigated the Commentary on Legal Holdings: The Trigger & The Process and analyzed which functions must be considered in order for electronic records to be preserved as evidence. To this end, we analyze the functional requirements of the eDiscovery solution. Through this, necessary functional requirements are derived to help implement the system in preparation for eDiscovery.

A New Analytical Method for Erythromycin in Fish by Liquid Chromatography/Tandem Mass Spectrometry

  • Erythromycin has been used to treat Streptococosis, Edwardsiel1osis, Vibriosis, Bacterial enteritis in the cultured fish. In this study, a rapid and effective erythromycin analysis method with new sample treatment protocol and liquid chromatography/tandem mass spectrometry (LC/MS/MS) system for fish products was developed. For the erythromycin extraction from fish muscle, the solvent mixture composed of 0.2% meta-phosphoric acid and methanol (6:4) showed good recovery rate, and the optimum extraction solvent volume was 20 mL. Erythromycin detection using LC/MS/MS were carried out under electro spray ionization (ESI) positive condition and erythromycin mass value 576.2 and 157.9. And the detection limit of the established method was 0.005 mg/kg in fish products. The recovery rate of the developed method applied to the fish species were as following, olive flounder, $87.6{\pm}5.0%$; black rockfish, $87.2{\pm}6.4%$; eel, $85.2{\pm}4.8%$; and rainbow trout, $86.0{\pm}6.2%$. In the established method in this study, the correlation of coefficient values ($R^2$) of erythromycin calibration curve (n=11) was 0.9998.

A Fast Determination of Globotriaosylsphingosine in Plasma for Screening Fabry Disease Using UPLC-ESI-MS/MS

  • Globotriaosylsphingosine (lyso-Gb3) is considered as one of the biological marker for Fabry disease. To date, a reliable biomarker that reflects disease severity and progression has not been discovered to guide the management of Fabry disease. A new method included a simple protein precipitation with acetonitrile in 100 μL of plasma following analyte separation on an Phenomenex Kintex- C18 column using a gradient elution (0.1% formic acid in 5-90% acetonitrile). Total run time was within 12 min including sample preparation and MS/MS analysis. The limit of detection and limit of quantitation were 1 ng/mL and 2 ng/mL, respectively. The calibration curve was linear over the concentration range of 2.0-200.0 ng/mL (r2 = 0.9999). Inter-day accuracy and precision at 7 level were 93.4-100.7% with RSD of 0.55-5.97%. Absolute recovery was 97.6-98.6%. The method was applied to human and mice plasma, proved the suitability for quantification of lyso-Gb3 for screening, diagnosis and therapeutic monitoring of Fabry disease patients.

Alternatives for Establishing Green Logistics System in Ulsan Port (울산항의 녹색물류체계 구축 방안)

  • After reviewing the concept and previous studies related to green ports, this study analyzes the implications of green port policy of advanced ports in foreign countries and analyzes problems in terms of environmentally-friendly green port policy for Ulsan port, and to present sustainable green logistics establishment measures. The literature survey and Benchmarking methods are adopted as research methodology and the results are as follows. First, the pan-government climate change response management system, legislation of relevant laws, implementation of fiscal support policies, and roadmaps should be established. Second, the foundation for eco-friendly green growth should be established through the discovery of business models in conjunction with leading industries in the Southeastern Metropolitan Economic Area. Third, the Ulsan Port Greenport, such as AMP, in-port LNG propulsion ship, and ESI vessel incentive, should be built. Fourth, a low-carbon, high-efficiency sea-shuttle service shall be established through the introduction of the sea-shuttle service along the sea route. Fifth, energy self-reliant ports, including all institutions in the metropolitan Ulsan port area that have exceeded the level of Ulsan port Authority, should be built. Finally, water-type ports need to be built through the creation of coastal forests, the purification of marine water quality, and the introduction of colors to port.

Anti-Cancer Effect of Gallic Acid in CT-26 Cells Inoculated Cancer Bearing Balb/C Mice (CT-26 세포 암 유발 마우스에서 Gallic acid의 항암 효과)

  • This study examined the anti-cancer activity of gallic acid(GA) isolated from P. suffruticosa. was analyzed by ESI-MS, $^1H$-NMR, and $^{13}C$-NMR. The anti-cancer activity was evaluated by measuring the cancer size in CT-26 cancer-allograft mice treated with GA(100 mg/kg p.o) for 14 days. The change in body weight, acute toxicity, weight change of the liver and spleen and biomaker of the liver were evaluated in the mice after the GA treatment. As a result, the cancer size of the CT-26 cancer-allograft mice treated with GA decreased significantly compared to that of the cancer mice without significant changes in weight loss (p<0.05) and acute toxicity. The weight of the liver and spleen and ALT, AST and LPO levels increased by cancer were decreased significantly after the GA treatment, and the GSH levels decreased by cancer were increased significantly with the GA treatment (p<0.05). Therefore, GA could be an attractive lead for the development of anticancer agents.

  • In order to prepare for the introduction of the E-Discovery system from the United States and to cope with some causable changes of legal systems, we propose a general E-Discovery process and essential tasks of the each phase. The proposed process model is designed by the analysis of well-known projects such as EDRM, The Sedona Conference, which are advanced research for the standardization of E-Discovery task procedures and for the supply of guidelines to hands-on workers. In addition, Machine Learning Algorithms, Open-source libraries for the Information Retrieval and Distributed Processing technologies based on the Hadoop for big data are introduced and its application methods on the E-Discovery work scenario are proposed. All this information will be useful to vendors or people willing to develop the E-Discovery service solution. Also, it is very helpful to company owners willing to rebuild their business process and it enables people who are about to face a major lawsuit to handle a situation effectively.

Effects of Qi Gong Exercise on the Immune Response, Pulse Wave Parameter and Heart Rate Variability(HRV) for Post Mastectomy Women (기공(氣功) 운동이 유방암 절제술 여성의 면역, 맥상파 및 심박변이에 미치는 영향)

  • Objectives The purposes of this study was to develope a Qi gong exercise that suits characteristics of post mastectomy women, and to evaluate the effect of Qi gong exercise on immune response, blood circulation index, pulse wave parameter and heart rate variability. Methods This study was applied to total 35 post mastectomy women, including 17 for experiment group and 18 for control group. The Qi gong exercise was composed of total 24 times of 90 minutes per each time, twice a week, and 12 weeks and it was conducted by the oriental medicine professor who was an expert of Qi gong exercise. Results 1. Two group comparison revealed that the experimental group had significantly improved immune response(p<.021), HR(beats/min)(p<.001), ESV(ml/beat)(p=.038), ESI($ml/beat/m^2$)(p=.040), ECO (L/min)(p=.019), ECI($L/min/m^2$)(p=.023), ECRI($dyne^*sec/cm$)(p=.015), Left Kwan($div^3$)(p=.021), Right Kwan($div^3$)(p=.038), Mean HRV(cycle/min)(p<.001), SDNN(ms)(p=.043), RMSSD(ms)(p=.040), and TP(log $ms^2$)(p=.039). 2. Two group comparison revealed that the experimental group had significantly decreased ECR ($dyne^*sec^*cm^{-5}$) (p=.034), Left RAI(p=.044), Right RAI(p=.042), and pNN50(%)(p=.038). Conclusions These results from Qi gong exercise program can be used as basic data for development of health promotion program for Post Mastectomy Women.

Botrytis cinerea hypovirulent strain △BcSpd1 induced Panax ginseng defense

  • Background: Gray mold, caused by Botrytis cinerea, is one of the major fungal diseases in agriculture. Biological methods are preferred over chemical fungicides to control gray mold since they are less toxic to the environment and could induce the resistance to pathogens in plants. In this work, we try to understand if ginseng defense to B. cinerea could be induced by fungal hypovirulent strain △BcSpd1. BcSpd1 encodes Zn(II)2Cys6 transcription factor which regulates fungal pathogenicity and we recently reported △BcSpd1 mutants reduced fungal virulence. Methods: We performed transcriptomic analysis of the host to investigate the induced defense response of ginseng treated by B. cinerea △BcSpd1. The metabolites in ginseng flavonoids pathway were determined by UPLC-ESI-MS/MS and the antifungal activates were then performed. Results: We found that △BcSpd1 enhanced the ginseng defense response when applied to healthy ginseng leaves and further changed the metabolism of flavonoids. Compared with untreated plants, the application of △BcSpd1 on ginseng leaves significantly increased the accumulation of p-coumaric acid and myricetin, which could inhibit the fungal growth. Conclusion: B. cinerea △BcSpd1 could effectively induce the medicinal plant defense and is referred to as the biological control agent in ginseng disease management.
