본 연구의 목적은 누에형질전환 기술을 이용하여 녹색형광실크를 개발하는 것으로서, 본 실험에서는 피브로인 H-chain의 N-말단과 C-말단을 이용하여 피브로인 재조합 단백질 발현 시스템을 제작하였고, 종결코돈이 없는 EGFP 유전자를 위의 발현 시스템에 클로닝하여 녹색형광실크를 제작하였다. 누에형질전환체 선발을 위해서는 3xP3 promoter와 DsRed2를 이용하여 선발하였고, 1200 개의 누에알에 microinjection 하여 F1 세대에서 8 bloods의 형질전환체를 선발하였다. 선발된 누에형질전환체는 초기배 단계의 눈과 신경조직, 유충과 번데기 그리고 성충의 눈에서 DsRed2 형광단백질이 발현되는 것을 확인 할 수 있었다. 또한 실크의 피브로인에서 EGFP 단백질이 발현되는 것을 확인하기 위해, F2세대의 누에형질전환체 중에서 5령 3일 유충을 해부하여 견사선을 형광현미경으로 관찰하였고, 중부와 후부 견사선에서 형광단백질이 발현되는 것을 확인할 수 있었다. 그리고 F2 세대의 고치와 저온에서 정련한 실크에서도 녹색형광단백질의 발현을 확인할 수 있었고, Western blot 분석에서도 EGFP 재조합 단백질이 피브로인 H-chain과 융합된 형태로 존재하는 것이 확인되었다. 이상의 결과에서 녹색형광실크를 생산하는 누에형질전환체가 성공적으로 제작 되었음을 확인할 수 있었고 이러한 결과를 토대로 새로운 산업소재로서 실크를 활용할 수 있을 것으로 기대한다.
Silk has had a reputation as a luxurious and sensuous fabric but it is not popular due to the expensive price and poor durability. To develop the silk materials that apply the various industries, the artificially synthesized gene can be introduced into the silkworm and expressed in the silk gland. Transgenic silkworms for the mass production of green fluorescent silks are generated using a fibroin H-chain expression system. For commercial use, safety assessment of the transgenic silkworms is essential. The purpose of this study was to examine the potential acute oral toxicity of EGFP protein expressed in genetically modified (GM) fluorescence silkworm and to obtain the approximative lethal dose in the male and female at 6-weeks ICR mice. EGFP protein was fed at a dose of 2,000 mg/kg body weight in five male or five female mice. Mortalities, clinical findings and body weight changes were monitored for 1, 3, 7, 14 days after dosing. At the end of 14 day observation period, all mice were sacrificed, and the postmortem necropsy were performed. The test group was not observed death case. Also the effect was not admitted by test substance administration in common symptoms, the body weight and postmortem. The results of single-dose oral toxicity test showed that approximative lethal dose of EGFP protein expressed in fluorescence silkworm was considered to exceed the 2,000 mg/kg body weight in both sexes.

Background: The development of a vaccine for Jembrana disease is needed to prevent losses in Indonesia's Bali cattle industry. A DNA vaccine model (pEGFP-C1-tat) that requires a functional delivery system will be developed. Polylactic-co-glycolic acid (PLGA) may have potential as a delivery system for the vaccine model. Objectives: This study aims to evaluate the in vitro potential of PLGA as a delivery system for pEGFP-C1-tat. Methods: Consensus and codon optimization for the tat gene was completed using a bioinformatic method, and the product was inserted into a pEGFP-C1 vector. Cloning of the pEGFP-C1-tat was successfully performed, and polymerase chain reaction (PCR) and restriction analysis confirmed DNA isolation. PLGA-pEGFP-C1-tat solutions were prepared for encapsulated formulation testing, physicochemical characterization, stability testing with DNase I, and cytotoxicity testing. The PLGA-pEGFP-C1-tat solutions were transfected in HeLa cells, and gene expression was observed by fluorescent microscopy and real-time PCR. Results: The successful acquisition of transformant bacteria was confirmed by PCR. The PLGA:DNA:polyvinyl alcohol ratio formulation with optimal encapsulation was 4%:0.5%:2%, physicochemical characterization of PLGA revealed a polydispersity index value of 0.246, a particle size of 925 nm, and a zeta potential value of -2.31 mV. PLGA succeeded in protecting pEGFP-C1-tat from enzymatic degradation, and the percentage viability from the cytotoxicity test of PLGA-pEGFP-C1-tat was 98.03%. The PLGA-pEGFP-C1-tat demonstrated luminescence of the EGFP-tat fusion protein and mRNA transcription was detected. Conclusions: PLGA has good potential as a delivery system for pEGFP-C1-tat.

배아줄기세포는 외부 기인 특정 요소를 이용한 세포 조절물질 활성을 통하여 원하는 세포형태로 분화될 수 있다. 배아줄기세포의 이용성세포침투성단백질(CPP)은 몇 개의 아미노산으로 구성된 작은 펩타이드로 유용한 물질 수송계 중의 하나로 인식되고 있다. 본 연구에서는 몇 종류의 CPP를 이용하여 특정 단백질을 생쥐배아줄기세포(R1)내로 트렌스펙션하는데 있어 그 정도를 결정할 수 있는 요소들의 영향을 분석하였다. CPP인 Buforin II, pEP-1을 강도를 강화한 녹색형광단백질(EGFP)에 부착되도록 하여 살아있는 세포에서 볼 수 있도록 하였다. 다양한 시간대별로 그리고 다양한 농도로 재조합 CPP-EGPF를 생쥐 배아줄기세포에 처리하였으며, 대조군으로 사용한 EGFP는 CPP를 갖고 있지 않았다. 재조합 CPP-EGFP들이 R1 배아줄기세포에 미치는 세포독성은 CPP 종류와 재조합 CPP-EGFP 농도에 의존적이었다. CPP-EGFP가 배아줄기세포의 세포질이나 핵질로 들어간 증거는 처리 후 30분 이내에서는 찾을 수 없었다. 1시간 후, pEP-1-EGFP-N을 처리한 일부 세포에서만 형광단백질이 세포안으로 들어간 것을 발견할 수 있었다. 처리를 24시간까지 하였으나 흥미롭게도 세포안으로 들어가는 정도와는 비례관계가 없었다. 또한, 배아줄기세포의 막을 통과할 수 있는 CPP의 경우, 처리 농도는 세포막을 침투하여 세포내로 이동하는 것과 비례적 관계는 없으나, 매우 높은 농도에서 약간 증가하는 경향을 보였다. 요약하면, CPP-EGFP의 트렌스펙션 효과는 전적으로 CPP에 의존적이며, CPP에 따라서는 His tag의 위치 또한 트렌스펙션에 영향을 줄 수 있으며, 처리시간은 CPP를 이용하여 배아줄기세포내로 단백질을 수송하는데 있어 중요 변수가 아니다. 또한, CPP-EGFP의 농도는 배아줄기세포막을 가로질러 수송되는데 큰 영향을 주지 않았다. 앞으로의 연구에서, 이들 CPP의 변형 또는 매개자 개발을 통한 배아줄기세포에서 좋은 단백질성 매개 수송자를 만드는 것이 필요하며 이를 이용한 분화 유도는 매우 유용할 것으로 사료된다.
Leishmaniasis is a worldwide uncontrolled parasitic disease due to the lack of effective drug and vaccine. To speed up effective drug development, we need powerful methods to rapidly assess drug effectiveness against the intracellular form of Leishmania in high throughput assays. Reporter gene technology has proven to be an excellent tool for drug screening in vitro. The effects of reporter proteins on parasite infectivity should be identified both in vitro and in vivo. In this research, we initially compared the infectivity rate of recombinant Leishmania major expressing stably enhanced green fluorescent protein (EGFP) alone or EGFP-luciferase (EGFP-LUC) with the wild-type strain. Next, we evaluated the sensitivity of these parasites to amphotericin B (AmB) as a standard drug in 2 parasitic phases, promastigote and amastigote. This comparison was made by MTT and nitric oxide (NO) assay and by quantifying the specific signals derived from reporter genes like EGFP intensity and luciferase activity. To study the amastigote form, both B10R and THP-1 macrophage cell lines were infected in the stationary phase and were exposed to AmB at different time points. Our results clearly revealed that the 3 parasite lines had similar in vitro infectivity rates with comparable parasite-induced levels of NO following interferon-${\gamma}$/lipopolysaccharide induction. Based on our results we proposed the more reporter gene, the faster and more sensitive evaluation of the drug efficiency.
polyhedron promoter, vesicular stomatitis virus G (VSVG), polyA, cytomegalovirus (CMV) promoter, enhanced green fluorescent protein (EGFP), protein transduction domain (PTD) 유전자가 포함된 재조합 베큘로바이러스를 구축하였다. 본 재조합 베큘로바이러스 시스템은 인간 섬유아세포와 여러 가지 조직에 감염하여 시험하였고 재조합된 유전자의 전달과 유전자 발현을 대조 벡터시스템과 비교하였다. 본 연구의 결과로 제작된 재조합 베큘로바이러스 시스템은 유전자의 전달과 발현에 있어서 대조 벡터시스템 보다 더욱 효과적이고 안전적이었다.

Pronuclear DNA microinjection has been the most universal method in transgenic animal production but its success rate of transgenesis in mammals are extremely low. To address this long-standing problem, we used retrovirus- and lentivirus-based vectors carrying the enhanced green fluorescent protein (EGFP) gene under the control of ubiquitously active cytomegalovirus (CMV) promoter to deliver transgenes to bovine embryos. The rate of transgenesis was evaluated by counting EGFP positive blastocysts after injection of concentrated virus stock into the perivitelline space of the bovine oocytes in metaphase II. Among two different types of lentivirus vectors derived from FIV (feline immunodeficiency virus) and HIV (human immunodeficiency virus), the former scored the higher gene transfer efficiency; almost 100% of the blastocysts developed from the oocytes infected with FIV-based vector were EGFP positive. As for the vectors derived Com HIV lentivirus, the transgenesis rate of the blastocysts was reduced to 39%.
현재까지 외래 유전자를 도입하여 형질전환 동물을 생산하는 방법이 다방면으로 연구되어 왔다. 그 중에서 본 연구에서는 정자를 EGFP 유전자와 공배양한 후 이를 난모 세포내에 미세 주입한 다음, 수정란의 발달과 ECFP 유전자의 발현을 조사하였다. 즉, 동결후 융해나 Triton X-100 처리 등으로 세포막을 파괴하여, 이들 정자를 EGFP 유전자와 다분간 공배양함으로써 정자와 EGFP 유전자와의 결합을 유도하였다. 정자나 정자두부의 미세주입에 의해 수정된 난자는 0.3%의 BSA가 첨가된 CR1aa 배양액에서 배양하였으며, EGFP 유전자의 발현은 형광현미경 하에서 관찰하였다. 통결 후 융해로 처리된 정자와 Triton X-100 처리 한 정자를 미세주입한 결과 난할율은 85.7과 80.1%였고, 배반포 발생율은 32.4 과 35.0%로서 유의차가 없었다. 동결 후 융해와 Triton X-100 으로 처리된 정자를 각각 미세주입한 수정란의 EGFP 유전자 발현율은 각각 19.1과 13.9%로서 전자가 유의하게 높았다. 또 정자 배양액에 첨가된 EGFP유전자의 농도가 54 ng/${\mu}\ell$일 때 EGFP 발현율은 15.4% 로서, 27 ng/${\mu}\ell$일 때의 9.0%와 63.5 ng/${\mu}\ell$일 때의 5.1% 보다 유의하게 높았다. 발현율을 높히기 위한 방법중 하나로써 electric shock의 방법을 이용해 보았으나 기존의 공배양 방법으로 얻은 최고 발현율인 19.1%에 못 미치는 2%를 보였다. EGFP 유전자가 발현된 수정란의 배반포 발생율은 0%로서 비발현 수정란의 29.5%보다 유의하게 낮았으며, EGFP 유전자의 발현은 mosaicism 형태를 보였다. 본 연구에서는 비록 낮은 외래 유전자 도입율을 보이기는 하나 (19.0%), 정자를 매개로 한 형질전환 동물의 생산은 그 방법이 간단하고 비용이 적게 든다는 장점이 있다. 기존 보고들의 효율성을 재고하여 볼 때, 난자내 정자 직접 주입술에 의한 형질전환 동물 생산의 연구는 향후 밝은 전망을 시사하고 있다.
진핵세포의 원형질막은 외부환경으로부터 세포를 격리하는 물리적인 장벽뿐만 아니라, 선택적 물질수송, 신호전달등 중요한 기능을 수행한다. 원형질막의 세포질쪽 지질층은 주로 Phosphatidylethanolamine (PE), Phosphatidylserine (PS), Phosphatidylinositides (PIs) 등의 인지질로 구성되어 있는데, 다양한 단백질들이 이들과 직접 결합하거나 이들의 성질을 이용해 원형질막으로 위치된다. 본 연구에서는 원형질막의 안쪽 지질층에 존재하는 특정 인지질과 결합하여, 원형질막에 위치되는 단백질을 과발현시켰을 때 나타나는 세포의 모양변화를 분석하였다. 이를 위해 PS와 PI등과의 선택적 결합으로 원형질막에 위치하는 단백질들과 소수성 또는 정전기적 상호작용으로 원형질막으로 위치되는 단백질들을 HEK293T세포에 과발현시켜 보았다. 그 결과, 대조군으로 사용한 EGFP 단백질과 PI4P에 선택적으로 결합하는 EGFP-P4M-SidM단백질의 발현은 세포의 모양변화를 유도하지 않았다. 반면, PI(4,5)P2에 결합하는 EGFP-PLCδ1(PH), PI(3,4,5)P3에 결합하는 EGFP-AKT1(PH), PI4P와 PI(4,5)P2에 결합하는 OSH2(PH)x2-EGFP의 발현은 세포의 크기가 줄어드는 수축현상이 일어나면서, Lamilapodia나 Filopodia가 형성되는 것을 확인할 수 있었다. 반면에, PS결합을 통해 원형질막에 위치되는 Lact-C2-EGFP과 소수성결합에 의해 원형질막에 위치되는 ApPDE4 long-form인 L(N20)-EGFP이나, 정전기적인 결합을 통해 원형질막으로 위치되는 ApPDE4 short-form인 S(N-UCR1-2)-EGFP단백질의 경우는 세포의 크기가 줄어드는 수축현상은 일어나지만 Lamilapodia나 Filopodia와 같은 모양변화는 나타나지 않는 것을 확인할 수 있었다. 본 연구를 통해, 특정 인지질에 결합하여 원형질막에 위치되는 단백질들이 각각의 인지질 결합 특성에 따라 분류 가능한 세포의 모양변화가 일어남을 확인할 수 있었다.
