This study was conducted to find out some conditions for optimum cultivation of Angelica gigas Nakai by the investigation of root yield and decursin content from different seeding sizes and accumulative temperature. Accumulative temperature from April to October was $4,309^{\circ}C$ on altitude 100 m, $4,242^{\circ}C$ on 250 m, $3,662^{\circ}C$ on 530 m, $3,435^{\circ}C$ on 730 m, and altitude 530~730 m was less $650{\sim}870^{\circ}C$ than altitude 100 m cultivation areas of A. gigas. Seedling stand rate was increased from 86.4% to more than 90% as accumulative temperature decreases, and was increased in above 7 mm of seedling size, and 10% in non-mulching more than PE film mulching. Yield was increased as accumulative temperature decreases and in PE film mulching as 310.2 kg/10a. Also, Yield was increased in 7~9 mm than seedling diameter 5~7 mm as 313.0 kg/10a. Decursin content of primary roots was increased as altitude rises, ie, as 2.55% on altitude 100 m, 3.33% on 250 m, 5.51% on 530 m, and 6.24% on 730 m. Decursinol angelate content appeared little than decursin content as 1.08% on altitude 100 m, 1.37% on 250 m, 1.99% on 530 m, 2.38% on 730 m, and as altitude was heightened, content was increased.

It was investigated how the contents of four active ingredients, nodakenin, decursinol, decursin, and decursinol angerate, which are active ingredients of Angelica gigas Nakai, cause material changes depending on the type of bacteria. Fermentation experiments were conducted using 9 types of bacteria: 5 types of Bacillus EMD17, 9-3, HCD2, #8, 191 and 4 types of Lactobacillus KCTC 3320, WCP02, S65, P1201. 1. The contents of decursin and decursinol angerate, which are indicator substances, rapidly decreased after 2 days of fermentation by inoculating Bacillus bacteria in the extract of Angelica gigas Nakai. Even after 4 days of fermentation, the contents of decursin and decursinol angerate were the same as on the 2nd day. On the other hand, the content of nodakenin and decursinol increased after 4 days of fermentation. In addition, the content of decursin increased significantly after 6 days of fermentation. 2. Substance changes of nodakenin and decursinol after inoculation of Bacillus bacteria into the extract of Angelica gigas Nakai were almost non-existent regardless of the type of bacteria. The change in effective content of decursin and decursinol angerate was large in Bacillus EMD17 and 9-3. Changes in the contents of decursin and decursinol angerate were almost non-existent in Bacillus HCD2, #8, and 191 strains. 3. As a result of finding out the change in active ingredient after 8 days of fermentation using 4 types of Lactobacillus KCTC 3320, WCP02, S65, and P1201 extracts of Angelica gigas Nakai, there was almost no change in the contents of nodakenin and decursinol regardless of the type of bacteria. However, in the case of fermentation with Lactobacillus S65 and P1201, the contents of decursin and decursinol angerate were changed.
This experiment was conducted to understand the effects of climatic environment on the useful components in Angelicae gigantis radix. The variation of decursin contents in different seven cultivated regions and the relationship between decursin contents and meteorological factors were investigated. The appropriate analytical methods for decursin and decursinol angelate of Angelicae gigantis radix were HPLC method used normal phase column($\mu$-Porasil) and GC method. In different cultivated regions, the variation of decursin contents of Angelicae gigantis radix was relatively great, which were high with 4.86%, 4.75% in Bongwha, Yongcheon respectively and was low with 2.33% in Suwon. There was no significant relationship between decursin contents of Angelicae radix and meteorological factors during growing periods, but was negative correlation in average temperature and precipitation, and positive correlation in diurnal change of temperature, radiation and sunshine hours.

Two pyranocoumarin constituents have been isolated from Angelica gigas and were identified as decursinol angelate (1) and decursin (2) by means of NMR analysis, respectively. Human acyl-CoA:cholesterol acyltransferase (hACAT) inhibitory activity of decursinol angelate (1) and decursin (2) was evaluated. Decursin (2) showed significantly inhibitory activity against hACAT1 and hACAT2 with $IC_{50}$ value of 137 and $168\;{\mu}M$, respectively, whereas decursinol angelate (1) exhibited weak ACAT inhibitory activity. These results suggested that decusin from A. gigas might be effective for the prevention and the treatment of hypercholesterolemia or atherosclerosis by inhibitory effect on hACAT. The contents of decursinol angelate (1) and decursin (2) were analyzed in various parts of A. gigas including flower, seed, leaf and root using LC/MS/MS (ESI, positive ion mode, MRM mode). The content of decursinol angelate was increased in order of flower, seed, leaf, and root and decursin content was increased in order of flower, seed, leaf, and root. It was expected that unused parts including leaf and flower of A. gigas might be useful as new functional sources by their high contents of decursin and decursinol angelate.

This study aimed to maximize the efficacy of Angelica gigas by finding a more effective way to extract active compounds from Angelica gigas. After extracting Angelica gigas by hydrothermal, ethanol, and supercritical carbon dioxide extraction methods, analysis of decursin and decursinol angelate content, quantification of total polyphenol content, and evaluation of efficacy of antioxidant, whitening, and antibacterial were conducted. The content of decursin and decursinol angelate was very high at 38.65% of the supercritical carbon dioxide extract, and the total polyphenol content was high in the order of hydrothermal extract, ethanol extract and supercritical carbon dioxide extract, but the difference was relatively small. The antioxidant effects were consistent with the total polyphenol content, and the antibacterial effects were consistent with the decursin and decursinol angelate content. In other words, through this study, we found that the optimal method for extracting active compounds from Angelica gigas is the supercritical carbon dioxide extraction method.

Background: Angelica gigas Nakai is a perennial herb belonging to the family Umbelliferae. Its roots are utilized in traditional medicine. The aim of this study was to increase the yield of and the content of two indicator components (decursin and decursinol angelate) in A. gigas Nakai. Methods and Results: The roots of A. gigas Nakai were harvested 4-times from late August to late October in 2017. Two agents (trace element-TE, and plant hormone-HM) were applied 4 times at intervals of 2 weeks. The content of the two indicator components were analyzed by high performance liquid chromatography. The HM treatment showed the greatest increase in underground part yield and root diameter. The content of the two indicator components in the control (non-treatment) group was the highest in the underground part, but was higher in the aerial parts in the agent treatment group. After treatment with the agents, the content of the indicator components tended to decrease in the underground part. However, the total content of the indicator components in the two agent treatment groups exceeded the level of 6% set by the Korean Pharmacopoeia. Conclusions: The highest underground part yield was found in the HM treatment group, while the highest content of decursin and decursinol angelate were found in the control group. This study provides basic information for yield improvement in A. gigas.
The effects of decursinol and decursin on chloroplast-mediated electron transport and phosphorylation in barley seedlings were investigated in comparison with coumarin in the dark or light. The changes of CP-complexes were also studied. Decursinol, decursin and coumarin caused marked inhibitory effects on germination of seed and electron transport and phosphorylation activity of seedlings. The following order of inhibitory effectiveness was exhibited; decursinol>coumarin>decursin. Loss of chlorophyll and decrease of electron transport activity were retarded in the dark, but were reversely accelerated in the light by these three chemicals. The changes of CP-complex patterns were also similar to effects on chlorophyll content and the electron transport activity. These opposite effect in the dark and light suggest that these three chemicals act as natural growth retardants rather than cytokinins or growth inhibitors.
The purpose of this study was investigation of quantitative analysis of marker substances in solid fermented Angelicae Gigantis Radix by High performance liquid chromatography(HPLC). HPLC was performed for determination of nodakenin and decursin in solid fermented Angelicae Gigantis Radix extract, the separation method was performed on C18 column ($250\;mm\;{\times}\;4.6\;mm$, $5\;{\mu}m$, RS tech) using gradient solvent mixtures of water-acetonitrile with photodiode array detector (330 nm). The flow rate was 1.0 ml/min. Retention time of nodakenin and decursin was about 11.47, 46.79 min and linearity of calibration was showed good result(r2=0.9999, 0.9999), respectively. Content of nodakenin was $0.76\;{\pm}\;0.02%$ in control, $0.31\;{\pm}\;0.00%$ in Angelicae Gigantis Radix extract fermented with Paecilomyces japonica(SDT)(p<0.01), $0.51\;{\pm}\;0.02%$ in Angelicae Gigantis Radix extract fermented with Ganoderma lucidum(SYT)(p<0.01), $0.82\;{\pm}\;0.03%$ in Angelicae Gigantis Radix extract fermented with honey(SST)(p<0.05) and $0.88\;{\pm}\;0.01%$ in Angelicae Gigantis Radix extract fermented with Nuruk(SNT)(p<0.01). Content of decursin was $4.50\;{\pm}\;0.08%$ in control, $2.90\;{\pm}\;0.05%$ in Angelicae Gigantis Radix extract fermented with Paecilomyces japonica(SDT)(p<0.01), $2.65\;{\pm}\;0.08%$ in Angelicae Gigantis Radix extract fermented with Ganoderma lucidum(SYT)(p<0.01), $4.46\;{\pm}\;0.11%$ in Angelicae Gigantis Radix extract fermented with honey(SST) and $4.73\;{\pm}\;0.04%$ in Angelicae Gigantis Radix extract fermented with Nuruk(SNT)(p<0.05), respectively.

Angelica gigas Nakai (Korean danggui), a member of the Umbelliferae family, is a Korean traditional medicinal plant whose roots have been used for treating gynecological diseases. In this study, the growth characteristics and decursin contents of Angelica gigas Nakai were compared according to the difference of cultivation area. As a result, the root diameter of A. gigas Nakai was the highest in the Duil-ri, and the number of the root of A. gigas was the highest in Topdong-ri. The fresh weight and dry weight of Angelica gigas Nakai was the highest in the Topdong-ri. The soil properties of cultivation area was Sandy Clay Loam and the content of organic matter was the highest in Topdong-ri. The total content of decursin was 5.31% the highest in Duil-ri. And 4.96% in Namhoeryong-ri, 4.41% in Kosun-ri, 4.25% in topdon-ri, 3.96% in Gugok-ri, and 3.64% in Gongjeon-ri was accumulated.
