Effect of Zanthoxylum piperitum Extract on Human Skin Protection from UVB by Regulation of COP1 and PPAR-α (초피나무 열매 추출물의 COP1 및 PPAR-α 조절을 통한 자외선에 대한 피부 보호 효과)

  • Ultraviolet (UV) irradiation from the sun is the primary environmental factor that causes skin damages including skin cancer and premature skin aging. Because, even the most powerful sunscreen can't always afford enough protection, it is necessary to enhance the defensive power of skin against UV. Recently, constitutive photomorphogenic protein-1 (COP1) has shown to contribute to the regulation of UVB response of keratinocytes. In this study, we represent that COP1 and its associated protein, de-etiolated 1 (DET1), might participate in photoaging process in human skin as Arabidopsis COP1 does sun-protective function in plants. After UVB irradiation, the decrease of COP1 and DET1 mRNA expression was followed by the increase of c-Jun total protein. Moreover, transfection with DNA vectors expressing COP1 and DET1 down-regulated the c-Jun total protein. We found that Zanthoxylum piperitum extract (ZE) up-regulated the expression of COP1 and DET1 on human keratinocytes, and inhibited the expression of MMP1 which is one of the genes regulated by c-Jun signal. In addition, ZE has been reported to stimulate PPAR-${\alpha}$ and strengthen the skin barrier. We found that ZE decreased the UVB-induced IL-6 and IL-8 in NHEK cells. In human study, ZE protected skin against UV-B induced erythema and erythema-induced pigmentation. These results indicate that ZE could be useful for the protection against the adverse effects of UV irradiation through various mechanisms.

Immunomodulatory and Antigenotoxic Properties of Bacillus amyloliquefaciens KU801 (면역조절능과 유전독성 억제능을 가지는 Bacillus amyloliquefaciens KU801)

  • The Bacillus KU801 strain, due to its potential in the field of probiotics for animal use, was isolated from chicken feces. Strain KU801 was identified as Bacillus amyloliquefaciens KU801 based on the results of 16S rRNA sequencing. Vegetative and spore cells of B. amyloliquefaciens KU801 were resistant to artificial gastric juice and artificial bile acid. B. amyloliquefaciens KU801 was found to inhibit the production of nitric oxide (NO) and increase the production of Interleukin-1 alpha (IL-1${\alpha}$). DNA damage induced by N-methyl-Ntion of ninitroso-guanidine (MNNG) was significantly inhibited, in a dose dependent manner, by preincubating MNNG together with B. amyloliquefaciens KU801. These results demonstrate the potential use of B. amyloliquefaciens KU801 as a feed additive.

Inhibition Effects on Oxidative DNA Damage and Anti-inflammatory Effects of Nelumbinis Flos (연꽃의 산화적 DNA 손상 억제 활성 및 항염증 효과)

  • Objective : Nelumbo nucifera, its rhizome and semen have been used as a traditional medicine which was studied on antioxidant, hepatoprotective effect, anti-obesity and the others. However, Nelumbinis Flos have not studied. We investigated protective effects on oxidative DNA damage and anti-inflammatory effects of Nelumbinis Flos. Methods : The antioxidant activity was conducted by 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical, 2, 2'-Azino-bis (3-ethylbenzothiazoline-6 sulfonic acid) diammonium salt (ABTS) radical scavenging assay and reducing power assay. Total phenolic content was analyzed. Also, phenolic compounds were detected by HPLC/UV. The inhibitory effect on oxidative DNA damage was determined using ${\Phi}X-174$ RF I plasmid DNA cleavage assay. The anti-inflammatory effect of Nelumbinis Flos was measured by the amount of nitric-oxide (NO) produced and protein levels of iNOS, and COX-2 in LPS induced RAW 264.7 cells. Results : The results of DPPH and ABTS radical scavenging activity at $200{\mu}g/m{\ell}$ of extraction were $97.02{\pm}0.88%$ and $96.42{\pm}0.25%$. Reducing power (fold of L-ascorbic acid as control) was $100.14{\pm}0.31$ at $200{\mu}g/m{\ell}$. Total phenol content was $8.70{\pm}0.02mg/g$. Chlorogenic acid, catechin and epicatechin were found by HPLC. Nelumbinis Flos has inhibitory effect in dose-manner against oxidative DNA damage. In addition, it showed the anti-inflammatory effect by suppression of NO production as well as protein levels of iNOS, and COX-2. Conclusion : This study suggested that Nelumbinis Flos showed potential antioxidant and suppression activities of various factors were related in NO produced. Therefore, Nelumbinis Flos as natural plant resources that may help reduce inflammation and alleviate DNA damage.

Antioxidant and Antigenotoxic Effects of Sansuyu Fruit (Corni fructus) Extracted with Water at Different Temperatures (추출 온도에 따른 산수유의 항산화 활성 및 항유전독성 효과 비교)

  • The objective of this study was to evaluate the antioxidant and antigenotoxic activities of sansuyu fruit (Corni fructus, CF) at temperatures of $25^{\circ}C$, $50^{\circ}C$, and $90^{\circ}C$ using a water extraction method. Total phenolic content (TPC), DPPH radical-scavenging activity (RSA), and superoxide dismutase (SOD)-like activity, and ORAC (Oxygen radical absorbance capacity) values were determined. Also the antigenotoxicity of CF was determined by measuring inhibitory effects of $H_2O_2$ induced DNA damage in human leukocytes using the comet assay. The TPC in the CF extracts was 4.2, 4.6, and 5.5 g/100 g GAE in $25^{\circ}C$, $50^{\circ}C$, and $90^{\circ}C$, respectively. The DPPH RSA of the CF extracts increased in a dose-dependent manner over the range of $50\sim1000\;{\mu}g$/mL in all temperatures and the $SC_{50}$ of DPPH RSA of the CF extracts were not significantly different at different extraction temperatures. The $SC_{50}$ of SOD-like was the highest in CF extracted at $25^{\circ}C$ (1.1 mg/mL) followed by $90^{\circ}C$ (1.2 mg/mL) and $50^{\circ}C$ (1.3 mg/mL). The ORAC values of the CF extracts were not significantly different in low concentration ($10\;{\mu}M$/mL) and was in order of $25^{\circ}C$ ($5.7\;{\mu}M$ TE)< $90^{\circ}C$ ($6.2\;{\mu}M$ TE)< $50^{\circ}C$ ($8.5\;{\mu}M$ TE) in high concentration ($50\;{\mu}M$/mL). $200\;{\mu}M$ $H_2O_2$ induced DNA damages in human leukocytes were significantly reduced by the pretreatment with the CF extracts. These results suggest that sansuyu fruit (Corni fructus) can be used as a natural source for antioxidant activities and as antigenotoxic agents regardless of the water extraction temperature.

Effects of Fermentation on the Metabolic Activities of Pine Needle Juice (발효과정이 솔잎 착즙액의 항산화, alpha-Glucosidase 및 Angiotensin Converting Enzyme 저해 활성에 미치는 영향)

  • The objective of this study was to compare the content and metabolic activities between fresh pine needle juice (PNJ) and fermented pine needle juice (FPNJ). A variety of factors were measured, including total phenolic content (TPC), antioxidant activity [DPPH radical scavenging activity (RSA), total radical-trapping antioxidant potential (TRAP), oxygen radical absorbance capacity (ORAC), cellular antioxidant capacity (CAC)], anti-genotoxic activity, ${\alpha}$-glucosidase inhibitory activity, and angiotensin converting enzyme (ACE) inhibitory activity. The TPC was $17.3{\pm}0.2$ and $4.6{\pm}0.0$ mg GAE/g in PNJ and FPNJ, respectively. The DPPH RSA, TRAP, and ORAC values increased in a dose-dependent manner for both PNJ and FPNJ, with significantly higher activities in PNJ than FPNJ. The CAC against AAPH-induced oxidative stress in HepG2 cells was protected by both PNJ and FPNJ. Pretreatment with PNJ and FPNJ in human leukocytes produced significant reductions in $H_2O_2$-induced DNA damage at a concentration of $50{\mu}g/mL$. ${\alpha}$-Glucosidase inhibitory activity was significantly higher in FPNJ than PNJ. The ACE inhibitory activity was about 87.1% and 60.0% in 1:1 diluted PNJ and FPNJ, respectively. This study suggests that the fermentation of PNJ could enhance the regulation of blood glucose metabolism and both PNJ and FPNJ might be a new potential source of natural antioxidant, anti-diabetic, and anti-hypertensive agents applicable to food.

In vitro Antimutagenic Activity of Chitosan and Its Bio-antimutagenic Characteristics (Chitosan의 in vitro 돌연변이 억제효과 및 세포내 작용 특성)

  • The inhibitory effects of chitosan on mutagenicity induced by 3-amino-1-methyl-5H-pyrido [4,3-b] indole (Trp-P-2), sodium azide (SA), 2-nitrofluorene (2-NF), and 4-nitroquinoline oxide (4-NQO) were investigated using Salmonella typhimurium reversion assay and SOS Chromotest. In Salmonella typhimurium reversion assay. Chitosan showed 24-65% of inhibitory effect against the mutagenicity of an indirect-acting mutagen, Trp-P-2. On the other hand, no inhibitory effect was observed against the mutagenicity of direct-acting mutagens (2-NF, SA). In SOS chromotest. chitosan showed 46-49% effects on SOS function induced by 4-NQO. Chitosan inhibited the mutagenicity induced by Trp-P-2 with 9-39% of inhibition rate. It was also evaluated whether inhibitory effect of chitosan is due to its bio-antimutagenic or desmutagenic action. Chitosan at high concentrations showed a bio-antimutagenicity with dose-dependent manner, but it showed a desmutagenicity at low concentrations against the mutation induced by Trp-P-2.

  • 피부는 항시 산소와 접촉하고 있고 자오선 조사에 크게 노출되어 있다. 따라서, 활성산소종으로 유도된 피부의 광산화적 손상 위험이 실질적으로 증가하고 있다. 활성산소종이란 한 무리로서 수퍼옥사이드 음이온과 히드록실 라디칼과 같은 산소 중심의 라디칼을 포함할 뿐만 아니라, 과산화수소나 싱글렛 옥시전과 같은 몇 종류의 비라디칼종들, 그 외의 다른 것으로, 활성산소종과 생체 성분과의 반응으로 유래된 과산화 라디칼, 알콕실 라디칼, 히드로과산화물 식세포에서 살균작용을 나타내는 HOCl등을 포함한다. 피부에는 복잡한 항산화 방어망이 발달되어 활성산소종에 대항하여 보호작용을 한다. SOD, 카탈라아제, 글루타치온퍼옥시다제 등의 항산화효소와 $\alpha$-토코페롤, 아스코르브산, 카로티노이드 등의 비효소적 항산화 물질들이 피부 항산화 방어망을 구축하고 있다. 그러나 계속된 자외선에의 노출로 생성된 과잉의 활성산소종은 실질직으로 피부의 효소적 그리고 비효소적 항산화 방어를 위태롭게 한다. 따라서 균형은 산화상태 쪽으로 유리하게 기울어진다 결과적으로 산화적 스트레스는 세포 성분들에 대한 손상을 야기시킨다. 지질 과산화, 단백질 산화, 탄력 섬유인 콜라겐과 엘라스틴의 사슬절단 밀 비장성적인 교차결합, 히아루론산 사슬의 절단, 멜라닌 생성반응 촉진, DNA 산화와 같은 생체 구성 성분들의 손상이 일어난다. 결국에는 탄력감소, 주름살 및 기미, 주근깨 등으로 특징 지워지는 피부노화가 가속화된다. 따라서 필요한 항상화제를 함유한 식품이나 화장품을 통한 계속적인 항산화제 보충으로 피부 항산화 방어망을 견고히 할 때 피부노화는 지연되고 억제될 것이다.주거를 구분하고 있는 것으로 조사되었으나, 아직까지 절반 가량(52.2%)의 상점들이 다른 사람 소유의 건물을 전 ·월세로 임대하여 사용하고 있음을 알 수 있었다. 이상에서 살펴본 바와 같이 고수동굴 주변의 상업적 특성은 90년대에 들어서면서 자기 고장에 있는 관광자원을 내 고향의 자랑거리로 생각하고 이를 지역발전의 밑거름으로 활용하고자 하는 인식이 늘어나고 있음을 알 수 있었다. 그러나, 관광지로서의 특성이라 할 수 있는 다른 관광동굴과의 차별성이 부족하다고 할 수 있다. 즉 다른 지역의 기념품점에서 구입할 수 없는 고수동굴만이 갖고 있는 특징적인 기념품을 판매함으로써 관광객의 구매욕구를 높인다거나 고수동굴의 일주관광 후 관광객을 좀 더 머무르게 할 수 있는 시설의 개발이 더 필요하다.유의한 차이를 나타내는 항목이 많았으며 12주에서 vehicle과 유의적인 차이를 나타내는 항목도 많으므로 3-APPA가 APSA 보다 광범위한 피부노화 억제 효과를 갖는 물질이라고 할 수 있다.주도적 역할을 수행해야 할 것이다. 넷째, 선박금융제도의 개선과 신금융상품의 개발이 요구된다. 내수 수요인 계획조선의 지원조건을 개선하고 연불수출자금을 BBC자금으로 활용토록하여 국내 선주들의 신조를 유도해야 할 것이다. 그 외에 향후 금융개방화에 맞추어 해외자금을 활용한 리스금융, 상사금융 등의 민간신용제도를 더욱 활성화하고 선진국의 선박금융기법에 대한 연구 및 도입 등 선주들에게 다양한 선박건조자금을 제공하여 내수기반 확충에도 노력해야 할 것 이다.있었다., 인삼이 성장될 때 부분적인 영양상태의 불충분이나 기후 등에 따른 영향을 받을 수 있기 때문에 앞으로 이에 대한 많은 연구가

Exploration of Beneficial Herbal Medicines to Attenuate Particulate Matter-induced Cellular Injury in Human Corneal Epithelial Cells (인간 각막상피세포에서 미세먼지로 인한 세포 손상을 완화할 수 있는 유익한 한약재의 탐색)

  • Particulate matter (PM) is known to be involved in the onset and progression of various diseases by promoting oxidative and inflammatory reactions as air pollutants containing various small particles that are harmful. In this study, the protective efficacy of herbal medicines was evaluated in human corneal epithelial cells (hCECs) to select natural products that can protect the eye, the primary organ directly exposed to external pollutants from PM. As a result, five candid ate herbal medicines [Cheonmundong, Asparagus Rhizome; Seokchangpo, Aciru Gramineri Rhizoma; Hwangryeon, Coptidis Rhizoma; Gamgug, Chrysanthemi Indici Flos; and Geumjanhwa (Marigold flower petals)] which showed inhibitory efficacy on PM2.5-induced cytotoxicity, were selected from among 12 candidate herbal medicines. To evaluate the antioxidant activity of these candidate substances, the reactive oxygen species (ROS) scavenging ability was investigated, and it was found that the extracts of Seokchangpo, Cheonmundong and Hwangryeon showed a significant inhibitory effect on PM2.5-induced ROS production, which was correlated with the preservation of mitochondrial activity. In addition, it was confirmed that they could block DNA damage caused by PM2.5 through analysis of 8-hydroxy-2'-deoxyguanosine generation and phosphorylated-H2A histone family member X (γ- H2AX) expression. Furthermore, the increase in inflammasome activity and inflammatory response in PM2.5-treated hCECs was also canceled in the presence of these extracts. Although additional studies are needed, the results of this study will be used as primary data to find novel natural compounds that protect hCECs from PM.

Effect of Natural Plant Mixtures on Behavioral Profiles and Antioxidants Status in SD Rats (자생식물 혼합 추출물이 SD 흰쥐에서의 행동양상 및 항산화 체계에 미치는 영향)

  • Caffeine, a psychoactive stimulant, has been implicated in the modulation of learning and memory functions due to its action as a non-selective adenosine receptors antagonist. On the contrary, some side effects of caffeine have been reported, such as an increased energy loss and metabolic rate, decrease DNA synthesis in the spleen, and increased oxidative damage to exerted on LDL particles. Therefore, the aim of this study was to develop a safe stimulant from natural plants mixture (Aralia elata, Acori graminei Rhizoma, Chrysanthemum, Dandleion, Guarana, Shepherd's purse) that can be used as a substitute for caffeine. Thirty SD rats were divided into three groups; control group, caffeine group (15.0 mg/kg, i.p.), and natural plants mixture group (NP, 1 mL/kg, p.o.). The effect of NP extract on stimulant activity was evaluated with open-field test (OFT) and plus maze test for measurement of behavioral profiles. Plasma lipid profiles, lipid peroxidation in LDL (conjugated dienes), total antioxidant capacity (TRAP) and DNA damage in white blood, liver, and brain cells were measured. In the OFT, immobility time was increased significantly by acute (once) and chronic (3 weeks) supplementation of NP and showed a similar effect to caffeine treatment. Three weeks of caffeine treatment caused plasma lipid peroxidation and DNA damage in liver cells, whereas there were no changes in the NP group. NP group showed a higher plasma HDL cholesterol concentration compared to the caffeine group. The results indicate that the natural plants mixture had a stimulant effect without inducing oxidative stress.

Cytoprotective Effect of Ethanol Extract from Maesil (Prunus mume Sieb. et Zucc.) on Alloxan-induced Oxidative Damage in Pancreatic-cell, HIT-T15 (Alloxan에 의한 HIT-T15 세포의 산화적 손상에 대한 매실(Prunus mume Sieb. et Zucc.) 주정추출물의 세포보호효과)

  • The present study was designed to examine the potential antidiabetic and antioxidant effect of ethanol extract from $Prunus$ $mume$ fruit (PME) against alloxan-induced oxidative stress in pancreatic ${\beta}$-cells, HIT-T15. To evaluate the antidiabetic effect of PME, 3-(4,5-dimethylthiazolyl-2)-2,5-diphenyltetrazoliu bromide (MTT) cell proliferation assay, lactate dehydrogenase (LDH) release assay, $NAD^+$/NADH ratio and insulin secretion were assessed. We also measured its antioxidant effect against alloxan-induced oxidative stress in the cells by assessing the levels of the antioxidant enzymes including superoxide dismutase (SOD), glutathione S-transferase (GST), glutathione reductase (GR) and glutathione peroxidase (GPx). The results of this analysis showed that alloxan significantly decreased cell viability, increased LDH leakage, and lowered $NAD^+$ /NADH ratio and insulin secretion in HIT-T15 cells. However, PME significantly increased the viability of alloxan-treated cells and lowered LDH leakage. The intracellular $NAD^+$ /NADH ratio and insulin secretion were also increased by 1.5~1.9-fold and 1.4-fold, respectively, after treatment with the PME. The HIT-T15 cells treated with alloxan showed significant decreases in the activities of antioxidant enzymes, while PME significantly elevated the levels of antioxidant enzymes. Based on these results, we suggest that PME could have a protective effect against the cytotoxicity and dysfunction of pancreatic ${\beta}$-cells in the presence of alloxan-induced oxidative stress.
