When medicinal mushroom, Poria cocos, is cultured , inoculation method of spawn is cross slice inoculation of which the both sides of pine tree were peeled and spawn of P.cocos was inoculated. However, this method required lots of inoculation amount. This study was carried out to improve the culturing method of P. cocos. A good growth of P.cocos was observed in MCM(mushroom complete medium), showing proper mycelia growth and density. In inoculation amount, conventional method(cross slice inoculation) requires 20 bottles of spawn. In contrast, short log method required 8 bottles of spawn and drilling inoculation method 2~3 bottles, which could save by 60% and 85-90% respectively. In the selectrion of tree species, pine and larch had better condition for spawn culture and sclerotia formation condition.In terms of yield , pine was 33.7kg/3.3$m^2$. In the yield of pine, conventional method was 23.4kg/3.3$m^2$, drilling inoculation 29.4kg/3.3$m^2$, short log inoculation 31.7kg/3.3$m^2$, therefore drilling inoculation could increase by 25% and short log inoculation 35%, In addition, management cost was also saved.
Strain IMCC26207 was isolated from the surface layer of Lake Soyang in Korea by the dilutionto-extinction culturing method, using a liquid medium prepared with filtered and autoclaved lake water. The strain could neither be maintained in a synthetic medium other than natural freshwater medium nor grown on solid agar plates. Phylogenetic analysis of 16S rRNA gene sequences indicated that strain IMCC26207 formed a distinct lineage in the order Acidimicrobiales of the phylum Actinobacteria. The closest relative among the previously identified bacterial taxa was "Candidatus Microthrix parvicella" with 16S rRNA gene sequence similarity of 91.7%. Here, the draft genome sequence of strain IMCC26207, a freshwater actinobacterium, is reported with the description of the genome properties and annotation summary. The draft genome consisted of 10 contigs with a total size of 3,316,799 bp and an average G+C content of 57.3%. The IMCC26207 genome was predicted to contain 2,975 protein-coding genes and 51 non-coding RNA genes, including 45 tRNA genes. Approximately 76.8% of the protein coding genes could be assigned with a specific function. Annotation of the IMCC26207 genome showed several traits of adaptation to living in oligotrophic freshwater environments, such as phosphorus-limited condition. Comparative genomic analysis revealed that the genome of strain IMCC26207 was distinct from that of "Candidatus Microthrix" strains; therefore, we propose the name "Candidatus Limnosphaera aquatica" for this bacterium.

Many researchers have employed cryopreserved amniotic membrane (CAM) in the treatment of a severely damaged cornea, using corneal epithelial cells cultured on an amniotic membrane (AM). In this study, two Teflon rings were made for culturing the cells on the LAM and CAM, and were then used to support the AM, which is referred to in this paper as an Ahn's AM supporter. The primary corneal epithelial cells were obtained from the limbus, using an ex-plantation method. The corneal epithelium could be reconstructed by culturing the thirdpassage corneal epithelial cells on the AM. A lyophilized amniotic membrane (LAM) has a higher rate of graft take, a longer shelf life, is easier to store, and safer, due to gamma irradiation, than a (AM. The corneal epithelium reconstructed on the LAM and (AM, supported by the twoTeflon rings, was similar to normal corneal epithelium. However, the advantages of the LAM over that of the (AM make the former more useful. The reconstruction model of the corneal epithelium, using AM, is considered as a good in vitro model for transplantation of cornel epithelium into patients with a severely damaged cornea.
두채생산에서 관수는 상면살수 방식과 하면담수 방식으로 대별된다. 본 연구는 관수방식에 따른 숙주나물의 생산과 상품성에 관한 정보를 제공하고자 중록1호를 공시재료로 상면살수와 하면담수 방식으로 대별되는 관수방식의 차이가 숙주나 물의 생장, 형태, 색도, 전단력과 재배용기내의 온도변화를 조사하고자 실시되었으며, 그 결과를 요약하면 다음과 같다. 1. 품질과 밀접히 관련된 세근은 하면담수 방법으로 재배할 경우 전혀 형성되지 않았던 반면, 상면살수 방법으로 재배할 경우 일부 개체에서 발생되는 것으로 조사되었다. 2. 하배축은 상면살수 방식에서, 뿌리는 하면담수 방식에서 길었으나, 하배축과 뿌리 길이를 합한 전체길이와 하배축 중간 및 자엽 바로 아래의 직경은 관수방법간에 차이가 없었다. 3. 개체당 전체건물중은 관수방법간에 차이가 없었던 반면, 전체생체중은 하면담수 방식보다 상면살수 방식에서 많았다. 이러한 차이는 주로 하배축 생체중의 차이에 기인되는 것으로 나타났다. 4. 두 관수방법 모두 재배중 플라스틱통 및 재배기내의 온도가 상승되었으나, 상면살수 방식에 이용되는 플라스틱 재배 통 내의 온도 상승이 큰 것으로 측정되었다. 이러한 온도 상승이 하면담수 방식보다 상면살수 방식에서 하배축 및 개체당 전체 생체중을 증가시킨 원인으로 해석되었다.
In order to clarify the effects of acid rain on soil microorganisms, the inpact of acid to soil microorganisms was survyed for 14 weeks using soil microcosms from industrial site A and B, Gaejok mountain, and Daechong lake in Taejeon area. The acid tolerant-microorganisms in natural soil, using culturing method were counted to be 5.8 - $8.0{\times}10^6$CFU/g soil. The number of microorganisms using ATP-biomass analysis for natural soil samples were also analyzed and 2.2 - $2.6{\times}10^9$ cell/g soil in industrial site A and B, Gaejok mountain, and Daechong lake were determined. In soil samples, which were treated with artificial acid rain, the number of acid tolerant microorganisms were counted 2.9 - $5.8{\times}10^5$ and 2.8 - $7.5{\times}10^8$, respectively. Therefore, we conformed that the numver of soil microorganisms were influenced by acid rain. Also, long term acid tolerant microorganisms were identified as Rhodotorula sp. and Pseudomonas sp.
We carried out an image analysis of living cells forming their contacts at the bottom of the cell culturing substrate. In order to visualize the contact area selectively, we adopted total-internal-reflection-fluorescence (TIRF) method, which can illuminate the specimen volume within only several hundred nano-meters above the substrate. From the fluorescent intensity of the TRF image, we could calculate the distance of the cell surface from the substrate. As a result, we visualized the origin of cell contacts, their movements, and the change of cell-contact type from the close-contact into focal-contact with information of its vertical displacement representing the temporal evolution process of the three-dimensional cell-surface-profile near the contact area during this metamorphosis.
In the conventional biology, the most of cell studies was carried out by culturing cells in the Petri dish and by investigating cellular behavior under the diverse bio-molecule (cell signalling materials, drugs or etc.) conditions. However, in vivo environments, diverse stimulations including chemical, mechanical and topological environments involved in the proliferation, differentiation and migration of cells and it is almost impossible to provide these conditions with traditional method. We have developed the methods to provide the well defined chemical and mechanical stimulations using microfluidic devices and applied these approaches to the study of environmental effect on cells. In this paper, we will introduce our microfluidic chips to provide microenvironment and its applications using several cells.
This paper describes a monitoring system for fish farm formation effect based on personal computer by using an image processing technique. This method is based on image processing technique incorporating concept of window and threshold processing to track the target object and to distinguish it from background. The image processing program runs in the veal time so that all program modules are able to process multi-task. The effectiveness is evaluated through the comparative study on the motion of lantern net for the scallop culturing by wave action in an experimental wave tank.
In this work, imaging and study of elastic property of the living cell was performed. The motivation of this work was to seek the possibility of exploiting Young's modulus as a disease indicator using Atomic Force Microscope (AFM) and also to gain fundamental understanding of cell mechanics for applications in medical nanorobots of the future. L-929 fibroblast adherent cell was used as the sample. Imaging condition in cell culturing media environment was done in very low speed ($20{\mu}m/ s$) compared to that in the ambient environment. For measuring the Young's modulus of the living cell, AFM indentation method was used. From the force-distance curve obtained from the indentation experiment the Young's modulus could be derived using the Hertz model. The Young's modulus of living L-929 fibroblast cell was $1.29{\pm}0.2$ kPa.
