Recently, it has been reported that some preservatives used in cosmetics lead to skin problems. Among the many cosmetic ingredients, 1, 2-hexanediol (HD) is used as both a preservative and humectant. In order to develop safer ingredients, we studied the synthesis of 1, 2-hexanediol galactoside (HD-G) by a transgalactosylation reaction using β-galactosidase (β-gal)-containing recombinant Escherichia coli cells. The transgalactosylation reaction was carried out under high-lactose conditions for 24 hr. After 12 hr had elapsed, a new spot was identified by thin-layer chromatography (TLC) analysis, and we presumptively designated this new spot as HD-G. Then, we carried out the purification of the presumptive HD-G spot from the reaction mixture by using silica gel chromatography, and its mass was measured by electrospray ionization-mass spectrometry. The purified new spot on the chromatograph was identified a sodium adduct ion ([M+Na]+, m/z = 303.1423) of HD-G. In addition, when purified HD-G was hydrolyzed using commercially available E. coli β-gal, it was observed that a galactose molecule was released from HD-G. That is, it was demonstrated that HD-G is a galactoside derivative of HD. Finally, we confirmed that HD-G was enzymatically synthesized by E. coli β -gal as a new molecular entity. In the future, we plan to determine the minimum inhibitory concentrations of HD-G against different bacterial species. The cytotoxicity of HD-G against human skin cells will also be examined. It is expected hopefully that the galactosylation of HD would improve the functionality of HD-G.
This study was conducted to confirm the application as ingredients of cosmetics through an examination of the function for anti-oxidant activity of the fraction isolated from Pyrus ussuriensis leaves. The dried leaf of Pyrus ussuriensis were extracted with acetone-$H_2O$ (6:4, v/v), concentrated and fractionated with the upper layer of acetone on a separatory funnel. Each fraction was freeze dried, then a portion of acetone soluble powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol as eluents and also used the MIC-gel using a series of aqueous methanol as developing solvent. The isolated compounds were identified by silica-gel TLC. The concentration of total phenolic compound of Pyrus ussuriensis acetate soluble fraction was high, 914 mg/g. The results obtained from the analyses of the anti-oxidanat effects of Pyrus ussuriensis acetate fraction can be summarized as follows: In the result of DPPH scavenging radical activity, Pyrus ussuriensis acetate soluble fraction showed more than 80% at 100 ppm. SOD-like activity of one of Pyrus ussuriensis acetate soluble fractions was 77% at 1000 ppm. Xanthine oxidase inhibition of Pyrus ussuriensis acetate soluble fraction was 38% at 100 ppm. From these results, we confirmed that acetate fraction of Pyrus ussuriensis has a great potential as a natural ingredients with a natural antioxidant and antimicrobial source.

In order to search for new whitening cosmetic ingredients from Chinese herbal extracts including Chinese herbs and complex prescriptions from TCM (Traditional Chinese Medicine), we screened about 47 TCM extracts collected from China. We tested their inhibitory effects on melanogenesis by using in vitro tyrosinase inhibition assay and B16 melanoma cells. We selected Siphonostegia chinensis and Salvianic miltiorrhiza Bunge. We isolated Danshensu ($\alpha$,3,4-trihydroxybenzenepropanoic acid sodium salt) from Salvianic miltiorrhiza Bunge extract and tested its inhibitory effect on melanin formation in B16-F10 melanoma cells. Danshensu suppressed melanin synthesis up to about 50% at a concentration of $100\;{\mu}g/mL$. Siphonostegia chinensis suppressed melanin synthesis up to about 60% at a concentration of $300\;{\mu}g/mL$. The results showed that these extracts could be used as new natural active ingredients for whitening cosmetics.

The harmful effects of ultraviolet (UV) radiation by increasing sun exposure are making people use sunscreens casually. To keep pace with this trend, many researches about mixing different ingredients or extracting effective ingredients from natural materials are conducted by cosmetics industry. In the present study, we evaluated the UV blocking effect of the sunscreen containing Scutellaria baicalensis Georgi extract. 10 volunteers were measured by minimal erythema dose (MED) and sun protection factor (SPF) of each product. The SPF results were $34.52{\pm}2.13$ and $32.67{\pm}1.44$ in the sunscreen containing Scutellaria baicalensis Georgi extract and that of not containing the substance, respectively. Although the difference of SPF between two products was statistically not significant, it is thought to be meaningful in evaluating clinical effects of the sunscreen using natural substance to humans without any adverse reaction.
This study was conducted to confirm the application as ingredients of cosmetics through an examination of the function for anti-cancer and anti-microbial of the fraction isolated from Pyrus ussuriensis leaves. The dried leaf of P. ussuriensis were extracted with acetone-$H_{2}O$ (6:4, v/v), concentrated and fractionated with the upper layer of acetone on a separatory funnel. The growth inhibition activity was measured using the MTT assay by the mouse meltioma (B16F10) cell. The cancer cell growth inhibition rate of fractions isolated from P. ussuriensis leaf was 80%. In anti-microbial activity test, the fraction of P. ussuriensis with 0.25 mg/disc resulted in the clear zone of 1.3 cm and 2 cm for Staphylococcus aureus and S. epidermidis of gram positive bacillus, respectively. In Escherichia coli of gram negative bacillus, the fraction with 0.5 mg/disc resulted in the clear zone of 1.1 cm~1.5 cm each fraction. From these results, we confirmed that acetate fraction of P. ussuriensis has a great potential as a natural ingredients with a anti-cancer and anti-microbial source.

Centella asiatica (C. asiatica) has been widely used in food, cosmetics, and pharmaceutical industry as a functional material. In a previous study, we have investigated not only pharmacological effects such as antioxidative and anti-inflammatory effects, but also analyzed various functional ingredients. In this study, triterpenoids were analyzed using HPLC-DAD to determine marker compounds among functional ingredients. When triterpenoids were analyzed, asiaticoside from C. asiatica was determined as an optimal marker compound. Next, specificity, linearity, limited of detection (LOD), limited of quantification (LOQ), precision, accuracy, and range were evaluated using HPLC-DAD to determine asiaticoside contents in C. asiatica juice and extracts. The specificity was elucidated by chromatogram and retention time using an established analytical method. The coefficient of correlation obtained was 0.9996. LOD was 4.99 ㎍/mL and LOQ was 15.12 ㎍/mL. Intra- and inter-day precision of asiaticoside were determined to be 0.48~1.68% and 0.08~1.09%, respectively. Furthermore, the recovery rate of asiaticoside was 98.88% and the analytical range of Field-70E was determined to be 0.625~10 mg/mL. As a results of evaluating ABTS, DPPH, and FRAP antioxidative effect, Field-70E showed potent antioxidant activities. Results of this study could be used as basic data for quality standardization of C. astiatica juice and extracts.

In this study, we investigated the antioxidant effects and active ingredients of Gynura Procumbens extracts obtained through various extraction methods for the development of natural-based cosmetics and pharmaceutical materials. The contents of compounds, total flavonoids, chlorogenic acid, and caffeic acid were compared at different concentrations, revealing the highest content of active ingredients in the 100% ethanol extract. Antioxidant activity assessed by DPPH and ABTS radical scavenging assays showed a concentration-dependent increase in antioxidant activity with the ethanol concentration. Additionally, we validated the DNA damage inhibition and anti-inflammatory effects of Gynura Procumbens extracts through UVB irradiation on Hs68 cell models. The 100% ethanol extract demonstrated significant inhibition of the expression of p-p53, γ-H2AX, iNOS, and COX-2 induced by UVB, indicating its potential in alleviating photodamage effects. Consequently, the efficient extraction of Gynura Procumbens for skin functional material development was confirmed, suggesting the suitability of ethanol or alcohol-based solvents.
Properties of fucoidan used for functional cosmetic ingredients and the effect of fucoidan molecular weight on the cosmetic functions were studied. Fucoidan was extracted from Undaria pinnatifida sporophylls and molecular weight (35~160 kDa) of fucoidan was controlled by contact glow discharge electrolysis (CGDE). To test possibility of fucoidan as a cosmetics material, tyrosinase inhibition property, water-holding property, elastase activity inhibition property and DPPH free radical scavenging property were measured. Water-holding property of fucoidan was higher than that of hyaruronic acid, which is known as the one of the best water-holding material. The water-holding strength of fucoidan slightly increase as molecular weight of fucoidan decrease. Elastase activity inhibition (anti wrinkle effect) of fucoidan was higher than that of adenosine using standard material for anti wrinkle test. Optimum molecular weight of fucoidan to obtain highest tyrosin inhibition effect, elastase inhibition effect and radical scavenger effect is 100 kDa.

The experiments were carried out in vitro to determine anti-oxidant activities of Sanguisorba extracts on DPPH radical scavenging activity assay, Superoxidase scavenging activity assay, Elastase and collagenase activity assay. It show that the Sanguisorba extracts have the most significant anti-oxidant on free radical scavenging activity assay, and also inhibited significantly activities of elastase, collagenase. Further, Sanguisorba extracts are activated Type I collagen protein expression in CCD-986sk cells. These result suggest that the Sanguisorba extracts on DPPH radical scavenging activity assay, Superoxidase scavenging activity assay, elastase and collagenase activity assay, Type I collagen protein expression in CCD-986sk cells effected could be developed cosmetic ingredients for anti-aging.
Chitin deacetylase (CDA; EC 3.5.1.41) catalyzes the hydrolysis of N-acetamide bonds of chitin, converting it to chitosan. Chitosan has several applications in areas such as biomedicine, food ingredients, cosmetics, pharmaceuticals, and agriculture. In this paper, occurrence, assay and purification protocols, enzymatic characteristics, substrate specificity, and mode of action of microbial CDAs have been described. Several lines of evidence have substantiated the biological roles involved in cell wall formation and plant-pathogen interactions for fungal CDAs. The gene structure of CDAs has been compared with other family 4 carbohydrate esterases which deacetylate a wide variety of acetylated poly/oligo-saccharides. The use of CDAs for the conversion of chitin to chitosan, in contrast to the presently used chemical procedure, offers the possibility of a controlled, non-degradable process, resulting in the production of well-defined chitosan oligomers and polymers. Insect pathogen that can secrete high levels of chitin-metabolizing enzymes including CDA can be a possible alternative for new pest management tools.
