Phytoplankton flora and water quality in the stream of Taejon area (대전지역 하천의 수질과 식물성 플랑크톤군집)

  • This study was carried out to provide basic data for water environmental control and conservation, and to elucidate monthly water environmental factor, phytoplankton flora and dominant species that each stations was maked a selection of a sites by represent 3 branch-river in the Taejon area from Nov. 1997 to Dec. 1998. Range of water temperature was $2.4^{\circ}C{\sim}30.9^{\circ}C$(means of $16.02^{\circ}C{\sim}18.92^{\circ}C$), pH was 7.1~10.4(means of 7.40~8.68). BOD was generally under $5mg/{\ell}$ except Gap-chon kyo in Nov. and COD was measured highly by interfer of men at the each sites. Especially, occasion of Gap-chon kyo, data was estimated about 3~13 times higher than another sites. DO was showed 1.03~10.43ppm, SS was showed $1.0{\sim}120.0mg/{\ell}$ TN was that, upper stream was 0.63ppm, middle was 0.35ppm and then lower was 0.44ppm, upper stream was higher than lower. TP was at upper stream 0.03ppm, middle 0.05ppm, lower 0.06ppm, lower stream was higher than another. The phytoplankton flora was identified as total 230 taxa composed of 7 classes, 18 oders 35 families, 75 genara, 184 species, 42 virieties, 4 forms. There are composed of that, Chlorophyceae 94 taxa(40.9%), Bacillariophyceae 66 taxa(28.8%), Euglenophyceae 39 taxa(17.0%), Cyanophyceae 24 taxa(10.4%), Chrysophyceae 4 taxa(1.7%), Rhodophyceae 2 taxa(0.8%) Xanthophyceae 1 taxa(0.4%), composition of Spesies of each sites was Gapchon-kyo 119, Wonchon-kyo 94, Gasuwon-kyo and Mannyon-kyo 87, Ochung-kyo 78, Hyunam-kyo and Suchim-kyo 77, Moonchang-kyo 70 taxa and Sansung-kyo 58 taxa. Number of species was showed tendency of increase lower stream. Taejon-chon and youdung-chon and at the sites of confluence were presented Synedra ulna and Diatom vugare like bacillariophyceae, Gap-chon was dominated variously Oscillatoria princeps and chlorophyceae, confluence of 3 branch-river was presented Euglenophyceae.

Effect of Hot Water Extracts of Roasted Rhus vemiciflua Stokes on Antioxidant Activity and Cytotoxicity (볶음 처리한 옻나무 열수추출물의 항산화성 및 암세포 성장억제효과)

  • The inhibitory effects of hot water extracts of Rhus verniciflua Stokes pith and peel roasted at 170, 200 and $220^{\circ}C$ on lipid peroxidation, formation of DPPH free radicals and growth of four human cancer cells such as HepG2 (liver cancer), SNU-1 (stomach cancer), MCF-7 (breast cancer) and Widr (colon cancer) were examined. The antioxidant activities and growth inhibitory effects on cancer cells of hot water extracts of peel were higher than those of pith, and the activities were dose-dependent. The roasting temperature showing the highest antioxidant activities and growth inhibitory effects on cancer cells was in the range of $170\~200^{\circ}C$ The lipid peroxidation and formation of DPPH free radicals of hot water extracts of roasted pith and peel were inhibited to 50.9, $56.5\%\;and\;79.0,\;78.4\%$ at the concentration of $500\mu g/mL$, respectively. The growth inhibitory effects of roasted pith and peel on cancer cells were in the order of Widr (41.5, $36.0\%$) > HepG2 (61.5, $44.0\%$) > MCF-7 (92.0, $69.2\%$)> SNU-1 (100, $100\%$) cells at the concentration of $1,000\mu g/mL$ as compared with the control, respectively. These results suggest that roasted Rhus verniciflua Stokes could be an useful natural medicinal plant for colon cancer.

Antibacterial Activity of Hippophae rhamnoides Leaf Extract and the Stability of a Cream with the Extract (비타민나무 잎 추출물의 항균 활성 및 크림의 안정성)

  • In this study, we investigated the antibacterial activity and stability of a cream containing Hippophae rhamnoides leaf extract. The MIC values of ethyl acetate fraction from an H. rhamnoides leaf on Escherichia coli, Pityrosporum ovale, Propionibacterium acnes and Staphylococcus aureus were 0.5%, 0.25%, 0.25% and 0.06%, respectively. Stability evaluations, pH, viscosity and absorbance of the cream containing 0.25% ethyl acetate fraction of H. rhamnoides, were performed. The cream was measured under 4 different temperature conditions under sunlight at 2-week intervals for 12 weeks. The viscosity and pH were measured by a comparison of the experimental cream with a similar control cream. The H. rhamnoides extract was found to have contributed to the stability of the emulsion product via a protective effect in maintaining the viscosity of the cream against sunlight. The absorbance variations of the experimental cream at 270 nm were, under sunlight; $45^{\circ}C$, $37^{\circ}C$, $25^{\circ}C$, and $4^{\circ}C$. In addition, any change in color or smell was not observed through the 12 weeks of the experimental period. These results indicated that the cream containing 0.25% ethyl acetate fraction of H. rhamnoides leaf extract was stable. Accordingly, this suggests that further study is needed to provide additional information for manufacturers, who are seeking the application of the extract to improve anti-oxidant and antibacterial activities and the stability of cosmetic products.

Taxonomical Classification and Genesis of Donggui Series in Jeju Island (제주도 토양인 동귀통의 분류 및 생성)

  • This study was conducted to reclassify Donggui series based on the second edition of Soil Taxonomy and to discuss the formation of Donggui series in Jeju Island. Morphological properties of typifying pedon of Donggui series were investigated and physico-chemical properties were analyzed according to Soil survey laboratory methods manual. The typifying pedon has very dark grayish brown (10YR 3/2) silt loam A horizon (0~17 cm), gravelly very dark grayish brown (10YR 3/2) silt loam BA horizon (17~42 cm), gravelly very dark grayish brown (10YR 3/2) silty clay loam Bt1 horizon (43~80 cm), brown (7.5YR 4/6) silty clay Bt2 horizon (80~105 cm), and brown (10YR 5/4) silty clay Bt3 horizon (105~150 cm). It is developed in lava plain and are derived from basalt and pyroclastic materials. The typifying pedon contains 1.3~2.1% oxalate extractable (Al + 1/2 Fe), less than 85% phosphate retention, and higher bulk density than 0.90 $Mg/m^3$. That can not be classified as Andisol. But it has an argillic horizon from a depth of 22 to 150 cm and a base saturation (sum of cations) of less than 35% at 125 cm below the upper boundary of the argillic horizon. That can be classified as Ultisol, not as Andisol and Inceptisol. It has udic soil moisture regime, and can be classified as Udalf. Also that meets the requirements of Typic Hapludalf. It has 18-35% clay at the particle-size control section, and have thermic soil temperature regime. Therefore Donggui series can be classified as fine loamy, mixed, thermic family of Typic Hapludalfs, not as fine silty, mixed, thermic family of Dystric Eutrudepts.

Enhancement of Immune Activity of Spirulina maxima by Low Temperature Ultrasonification Extraction (저온 초음파 추출에 의한 Spirulina maxima 면역활성 증진)

  • The marine microalga Spirulina maxima was extracted using water or ethanol at 100 or $80^{\circ}C$ and by ultrasonification in water at $60^{\circ}C$. The ultrasonification technique generated the highest yield (19.8%). To be therapeutically useful, the extraction should yield a product with low cytotoxicity and high immunity against skin infections. The cytotoxicity of all extracts (1.0 mg/mL) was below 25%. Moreover, the cytotoxicity of the extract generated by ultrasonification was 5%. Extracts prepared in the described manners could inhibit hyaluronidase activity by up to 40% compared to the control. Increased growth of human B, T and NK cells and an increase in cytokine secretion were observed, confirming the interrelationship between both human immune and skin immune activity. The extract prepared by ultrasonification increased the growth of human B, T and NK cells up to $10.3{\times}10^4$ cells/mL, $11.3{\times}10^4$ cells/mL and $19.1{\times}10^4$ cells/mL, respectively. The extract prepared by ultrasonification also greatly increased the secretion of both IL-6 and $TNF-{\alpha}$. Moreover, it was estimated that protein, Na and leucine occupy a high ratio. Accordingly, this study has confirmed that extracts prepared as described have the potential to effectively increase skin immunity.

Effects of Seed Storage Methods and Shading on Seed Germination and Seedling Growth of Endangered Species, Iris dichotoma and Iris setosa (종자저장방법 및 차광처리가 희귀식물 대청부채와 부채붓꽃의 발아와 유묘생육에 미치는 영향)

  • This study was conducted to determine the effects of seed storage method ($-20^{\circ}C$, $2^{\circ}C$ dry, $2^{\circ}C$ wet 30 days, $2^{\circ}C$ wet 60 days, stratification and room temperature) and shading treatment(control, 50%, 80%) on seed germination, seedling growth of endangered species, Iris dichotoma and Iris setosa. As a result, seed germination rate of I. dichotoma was the highest at 75% when seed were stored at $2^{\circ}C$ wet 60 days and then sown under non-shading condition. The seed of I. dichotoma belong to intermediate seed. Seed germination rate of I. setosa was the highest at 95% when seed were stored at $2^{\circ}C$ wet 60 days and then sown under 80% shading condition. The seed of I. setosa belong to recalcitrant seed. Seedlings of I. dichotoma and I. setosa showed not only the best seedling quality but also seedling vigor index in seed stored at $2^{\circ}C$ wet 60 days under non-shading condition, with the growth characteristics of plant height (6.4, 7.2 cm), number of leaves (3, 4), leaf width (4.6, 3.2 mm), leaf length (5.7, 6.8 cm), fresh weight (aerial/root part; 144/260, 97/153 mg), dry weight (aerial/root; 31/20, 17/17 mg) and seedling vigor index and modified seedling vigor index (13,895/9,479, 13,256/8,668). In this research, I. dichotoma and I. setosa seed were stored at $2^{\circ}C$ wet 60 days, and then sown in non-shading condition, seed germination rate was more than 75%, 90%, respectively, and production of superior quality seedlings.

Simulation of Detailed Wind Flow over a Locally Heated Mountain Area Using a Computational Fluid Dynamics Model, CFD_NIMR_SNU - a fire case at Mt. Hwawang - (계산유체역학모형 CFD_NIMR_SNU를 이용한 국지적으로 가열된 산악지역의 상세 바람 흐름 모사 - 화왕산 산불 사례 -)

  • The unexpected wind over the Mt. Hwawang on 9 February 2009 was deadly when many spectators were watching a traditional event to burn dried grasses and the fire went out of control due to the wind. We analyzed the fatal wind based on wind flow simulations over a digitized complex terrain of the mountain with a localized heating area using a three dimensional computational fluid dynamics model, CFD_NIMR_SNU (Computational Fluid Dynamics_National Institute of Meteorological Research_Seoul National University). Three levels of fire intensity were simulated: no fire, $300^{\circ}C$ and $600^{\circ}C$ of surface temperature at the site on fire. The surface heat accelerated vertical wind speed by as much as $0.7\;m\;s^{-1}$ (for $300^{\circ}C$) and $1.1\;m\;s^{-1}$ (for $600^{\circ}C$) at the center of the fire. Turbulent kinetic energy was increased by the heat itself and by the increased mechanical force, which in turn was generated by the thermal convection. The heating together with the complex terrain and strong boundary wind induced the unexpected high wind conditions with turbulence at the mountain. The CFD_NIMR_SNU model provided valuable analysis data to understand the consequences of the fatal mountain fire. It is suggested that the place of fire was calm at the time of the fire setting due to the elevated terrain of the windward side. The suppression of wind was easily reversed when there was fire, which caused updraft of hot air by the fire and the strong boundary wind. The strong boundary wind in conjunction with the fire event caused the strong turbulence, resulting in many fire casualties. The model can be utilized in turbulence forecasting over a small area due to surface fire in conjunction with a mesoscale weather model to help fire prevention at the field.

In Vitro Anticancer and Antioxidant Effect of Solvent Extracts from Tuna Dried at Low Temperature Vacuum. (저온진공건조 참치추출물의 in vitro 항암 및 항산화 효과)

  • We investigated the inhibitory effects of solvent extracts from dried tuna on the growth of cancer cell lines (HT1080 human fibrosarcoma and HT-29 human colon cancer cells) and $H_2O_2$-induced oxidative stress. Inhibitory effects of acetone with methylene chloride (A+M) and methanol (MeOH) extracts on the growth of HT1080 and HT-29 cancer cells increased in a dose dependent manner (p<0.05). The inhibitory effect was more significant on the growth of HT1080 cells, and A+M extracts had a higher inhibitory effect compared to MeOH extracts. The treatments of hexane, 85% aq. methanol, butanol and water fractions significantly inhibited the growth of both cancer cells (p<0.05). Among the fractions, hexane and 85% aq. methanol fractions showed higher inhibitory effects. In order to determine the protective effect on $H_2O_2$-induced oxidative stress, a DCHF-DA (dichlorodihydrofluorescin diacetate) assay was conducted. All fractions, including crude extracts of dried tuna, appeared to significantly reduce the levels of intracellular reactive oxygen species (ROS) with dose responses (p<0.05). Among the fractions, BuOH and 85% methanol fractions showed a higher protective effect on the production of lipid peroxides. These results indicate that the consumption of tuna may be recommended as a potent functional food for preventing cellular oxidation and cancer.

Nitrogen Balance in Paddy Soil of Control-Release Fertilizer Application (완효성비료 시용 논 토양중의 질소행동에 관한 연구)

  • The increasing of greenhouse gases may change agricultural environment. The agronomic productivity will depend upon change of temperature, precipitation, solar radiation and fertilization. Particularly, nitrogen fertilization considerably influences rice productivity and agricultural environments. This experiment was conducted to study transformation of nitrogen and to determine the primary yield components responsible for yield differences in paddy soil. $NH_4-N$ concentration of NPK plot in surface water of paddy soil was $2.07mg\;L^{-1}$ at 5 days after transplanting, and then was decreased sharply due to rice absorption and loss to environment. $NO_3-N$ concentration of NPK plot in surface water was $3.97mg\;L^{-1}$ at 10 days after transplanting. $NO_3-N$ concentration range of CRF plot in surface water was $3-5mg\;L^{-1}$ at 30th after transplanting. The accumulation of $NH_3$ volatilization in NPK plot was $22.39kg\;ha^{-1}$, which accounted for 20% of N fertilizer applied but using of CRF fertilizer can reduce $NH_3$ volatilization by 67% in paddy soil. Use efficiency of N fertilizer was not different between CRF70% and CRF100% plot. Rate of N use efficiency were 27.4%, 51.2%, 49.0% in paddy field NPK, CRF70% and CRF100% plots respectively. The yield of CRF70% showed the best effect with 9.3% increase production ratio, compare with NPK plot.

The Effects of Anoxic Treatments on Color and Mechanical Property in Fabrics, Natural Dyed Fabrics, Papers, Natural Dyed Papers and Paints (저산소 농도 살충처리가 직물, 염색 직물, 종이, 염색지 및 채색편의 색상 및 기계적 성질에 미치는 영향)

  • Fabrics, natural dyed fabrics, papers, natural dyed papers and paints were examined effects of colors and mechanical properties for materials of museum collections under anoxic treatment. Anoxic conditions using nitrogen and argon were oxygen concentration 0.01%, temperature($20^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$), 50% RH and exposure time 30 days. Examined fabrics were raw silk fabric, UV irradiated raw silk fabric, degummed silk fabric, UV irradiated degummed silk fabric, cotton fabric, and UV irradiated cotton fabric. Natural dyed silk and cotton fabrics were dyed with fresh indigo, indigo, safflower, gromwell, madder sappanwood, amur cork tree, turmeric, gardenia, barberry root, pagoda tree flower, cochineal, lac, alnus japonica, gallnut, chestnut shell, and combination(indigo and safflower, indigo and amur cork tree, indigo and pagoda tree flower, indigo and sappanwood). Papers were Korean papers(mulberry paper, mulberry(70%) and rice straw(30%) mixed paper), Japanese paper(gampi paper), cotton paper, refined linen paper, cotton, linen & manila mixed fibre furnish, copy paper, news print, and alum sized mulberry paper. Natural dyed papers were dyed with indigo, sappanwood, madder, safflower, gardenia, amur cork tree, and pagoda tree flower. Paints were painted on alum-sized papers and silk fabrics using glue and pigments(azurite, malachite, cinnabar, vermilion, orpiment, gamboge, red lead, haematite, iron oxide red, indigo(lake), lac, cochineal, safflower, madder root lake, celadonite, smalt, ultramarine blue, lapis lazuli, prussian blue, kaolin, lead white, oyster-shell white, and clam-shell white). The color differences(${\Delta}E^*$) of all examined materials were below 1.5 or lowered than control samples after anoxic treatment. The variations of tenacity of yarns of fabrics and natural dyed fabrics after anoxic treatment were within that of standard silk and cotton fabrics. Gases(nitrogen and argon) and temperatures of anoxic treatment did not also affected color differences and variations of tenacity of materials.
