Inhibitory Effect of Green Tea Extract on Collagenase Activity and Growth of Fish Pathogenic Bacteria (녹차 추출물을 이용한 어병세균의 collagen 분해효소 및 생육 억제)

  • Green tea extract has been suggested to possess properties of collagenase inhibition and antimicrobial effect to fish pathogenic bacteria. Most fish pathogenic bacteria showed collagenase activities of 0.08-0.7 unit/ml. Among them, Edwardsiella tarda produced large amount of collagenase in the ST medium at $25^{\circ}C$ after 16 hrs. Biosynthesis of the enzyme from E. tarda was decreased to 1/3 by addition of 0.08-0.8 mg/ml of tea extract in the culture medium. The collagenase activity was inhibited almost 100% in the reaction mixture by 0.2 mg/ml of the extract. Then, the minimal inhibition concentration against E. tarda growth appeared as 8 mg/ml of the tea extract in the culture medium.

Inhibition of Collagenase by Naturally-Occurring Flavonoids

  • We examined the inhibitory activities of various flavonoids, including the flavanones, flavones/isoflavones and flavonols, on collagenase from Clostridium histolyticum to establish their therapeutic potential against skin inflammation and photoaging. In general, the flavonols were stronger inhibitors than the flavones/isoflavones, and this indicated the importance of the C-3 hydroxyl substitution. Quercetin was the most active flavonoid among those tested, and it showed an $IC_{50}$ of 286 ${\mu}M$. These novel results suggest that certain flavonoids, particularly the flavonols, may prevent collagen breakdown by inhibiting collagenase in inflamed skin as well as photoaged skin.

Study on Biological Activities of Extracts for Cosmeceutical Development from Lagerstroemia indica L. Branch (화장품 개발을 위한 배롱나무(Lagerstroemia indica Linnaeus) 가지 추출물의 생리활성에 관한 연구)

  • The aim of the study was to examine the cosmetic and biological activity of Lagerstroemia indica L. and it is possible that can be used as a cosmetic ingredient for application of cosmetic industries. Lagerstroemia indica L. branch was extracted with 70% acetone in water. In the result of DPPH (1,1-diphenyl-2-picryl-hydrazyl) scavenging radical activity, acetone extract of Lagerstroemia indica L. branch were higher than 73% at the 50 ppm concentration. ABTS radical cation decolorization activity by acetone extract were higher than 78% at the 50 ppm. Both examine of DPPH and ABTS showed high antioxidative activities at the 50 ppm. In the result of nitrite scavenging ability, acetone extract were higher than 63% at the 50 ppm. Collagenase inhibition activity by extract were higher than 85% at the 50 ppm. Extract is showed high collagenase inhibition more than comparison group EGCG at all concentration. These results suggest that Lagerstroemia indica L. has a great potential as a cosmeceutical raw material as well as anti-oxidant and anti-inflammatory and collagenase inhibition activity.

Anti-oxidation and Anti-aging Effect of Mixed Extract from Korean Medicinal Herbs (한약재 복합 추출물이 항산화 및 항노화에 미치는 영향)

  • Objectives : The present study was designed to investigate effects of mixed extract from korean medicinal herbs (MIX) on oxidation/reduction reaction-related and aging-related enzyme in vitro. Methods : We performed MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity and xanthine oxidase inhibition assay. Results : Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. The MIX showed 97% inhibition of collagenase activity, and 64% inhibition of elastase activity at 1 mg/ml concentration of MIX, next only to positive control, which indicate good efficacy for anti-wrinkle ingredient. Also it's treatment showed 34% inhibition of tyrosinase activity, to relate whitening effect, at the same dose of MIX. Antioxidant activities were determined by DPPH radical scavenging, xanthine oxidase (XO) inhibiting activity and SOD-like activity. Also these scavenging, XO-inhibiting and SOD-like activities were measured in 91%, 80%, and 63% inhibition, respectively, at a treated dose of 1 mg/ml, compare to control. Conclusions : These results suggest that possibility of mixed korean medicinal herbs as a functional ingredient for anti-wrinkle and whitening, anti-oxidation and anti-aging cosmetic formula.

A Study on Skin Care Effects of Rapeseed Meal Extract (유채박 추출물의 피부미용 효과에 관한 연구)

  • This research was to investigate the inhibitory effects of tyrosinase, elastase and collagenase using rapeseed meal extract for the functional cosmetics. Gamma-tocopherol and alpha-tocopherol contents were 304.9 and 212.2 mg/kg, respectively. In the case of delta-tocopherol and plastochromanol-8, they were 12.1 and 35.7 mg/kg, respectively. The total phenol content of methanol extract was the highest (49.6 mg/g) which was about 4.96 fold higher than that of water extract. The maximum nitrite scavenging activities of methanol and acetone extract at pH 1.2 were 85.2 and 80.1%, respectively, at 8.0 mg/mL. When the extract concentration of rapeseed meal increased upto 2.0 mg/mL, cell viabilities did not appear to have any significant cytotoxic effect, irrespective of extracts. Tyrosinase and elastase inhibition activities increased from 25.2 to 42.5% and 25.3 to 48.0%, respectively, as methanol extract concentration increased from 0.5 to 1.0 mg/mL. The collagenase inhibition activities of methanol and acetone extracts at 1.0 mg/mL were 67.2 and 68.0%, respectively. These results showed that the methanol and acetone extract of rapeseed meal can be used as a new source of functional cosmetic agent.

Study of Cosmeceutical Activities of Hovenia dulcis var. koreana Nakai Extracts (헛개나무 추출물의 화장품 생리활성에 관한 연구)

  • Hovenia dulcis var. koreana Nakai has been reported to liver function improvement effect as functional materials for food and medicine. On these facts, biological activity and safety test were conducted to evaluate biological activities of the fruit petiole and root extracts of H. dulcis as a potential cosmeceutical ingredient. Cosmeceutica activities of different extracts were examined by l.l-diphenyl-2-picrylhydrazyl (DPPH) radical generation, the ABTS+ cation decolorization, tyrosinase activity, collagenase activity and elastase activity compared with the properties of the commercial antioxidant butylated hydroxytoluene (BHT) and L-ascorbic acid (AA). The antioxidant activities HDFW, HDFE, HDRW and HDRE were 83.6%, 39.6%, 85.9% and 74.5% in DPPH assay, 99.5%, 13.7%, 96.4% and 88.6% in ABTS assay. Tyrosinase inhibitiory activities HDFW were 56.0% at 1,000 ppm. Measured the inhibition effect of the H. dulcis about collagenase and elastase where break the peptide bonds in collagen and enzyme from the class of proteases where exists in the dermis. The H. dulcis was inhibition the two kind enzymesm, collagenase activities being on a high scale inhibition, was same concentration. Uses the anti oxidation effect and a anti-wrinkle effect of this resultant H. dulcis and with the functional cosmetics use is thought with the fact that will be possible.

Comparative evaluation on biological activities of Ziingiber officinale Roscoe extracted from different solvents (추출 용매를 달리한 생강 추출물에 대한 생리활성의 비교 평가 연구)

  • Objective : This study was designed to investigate anti-oxidative, anti-aging and anti-inflammatory activities of Zingiber officinale Roscoe water (ZD) and 50% ethanol extracts (ZE). Method : The antioxidant capacity of ZD or ZE was analyzed by DPPH, ABTS scavenging activities and reducing power, respectively. The anti-aging efficacy was performed with tyrosinase and collagenase inhibition activities. The anti-inflammatory activities were confirmed through inhibition of nitric oxide production and TNF-�� expression in LPS-treated Raw 264.7 cells. Result : As a result of measuring DPPH and ABTS radical scavenging activities, both ZD and ZE extracts increased in concentration-dependent manners. The ABTS radical scavenging activities of water and 50% ethanol extract were similar to that of positive control at high concentration. On reducing power, both ZD and ZE showed higher activities than the positive control at high concentration. On inhibition activities of tyrosinase and collagenase both ZD and ZE showed the large inhibition rates at low concentration. Also, the inhibitory efficacy of ZD on NO and TNF-�� production was better than ZE in RAW 264.7 cells. Conclusion : These results suggest that ZD and ZE have anti-oxidative, anti-aging and anti-inflammatory effects in vitro. Especially, ZD was more effective than ZE. Thus, Ginger extract is considered to be highly useful as a cosmetic or food material with anti-oxidative, anti-aging (prevention of whitening and wrinkle) and anti-inflammatory effects. Also these effects may vary depending on the extraction solvent.

Inhibitory Effect on Melanin Formation, Collagenase and Elastase Activity by synthesized Coenzyme $Q_{10}$ Derivatives (세포내 멜라닌 생성 및 Collagenase와 Elastase에 대한 Coenzyme $Q_{10}$ 유도체들의 억제활성)

  • Coenzyme $Q_{10}$ and six derivatives of coenzyme Qn were synthesized and tested for their inhibitory effects on melanogenesis occurred in murine melanoma (B16/F1) cells and on collagenase/elastase activities as well. As the result, synthetic coenzyme Qn showed a potent inhibitory effect on melanin formation, collagenase and elastase activities in all tested concentrations. Among these synthetic compounds, coenzyme $Q_1$ and coenzyme $Q_2$ potentially inhibited melanin formation and elastase activity when compared to other coenzyme Qn derivatives. For the collagenase activities, all coenzyme Qn derivatives inhibited 80-85% of controls. As compared, coenzyme Qn derivatives exhibited strong inhibitory activities with the decrease of isoprenoid unit number of coenzyme Qn derivatives except for collagenase activity. For the inhibition of collagenase activity, moiety of benzoquinone might be considered as the active functional group. Taken together, coenzyme $Q_1$ and coenzyme $Q_2$ might be used for functional cosmetics.

Cosmeceutical Activities of Doinseunguitang and Its Composition (도인승기탕 및 그 구성약재의 화장품약리활성)

  • Objectives : Cosmeceutical activities such as anti-oxidative and anti-aging effects of the Doinseunguitang and its composition, a traditional prescription, were evaluated. Methods : We performed MTT assay, melanin synthesis inhibition assay, DPPH free radical scavenging assay, SOD-like activity, xanthine oxidase inhibition assay, astringent activity assay, tyrosinase inhibition assay, elastase and collagenase inhibition assay. Results : The results were obtained as follows : DPPH free radical scavenging of water extract Doinseunguitang (DISG) and ethanol extract DISG was 60% and 50% at 1,000 ppm. Xanthine oxidase inhibition effect of ethanol extract showed more than 80% at 500 ppm. Tyrosinase inhibition and inhibition melanin synthesis activities were measured in 40% and 50% at 1,000 ppm and 100 ppm. Elastase and collagenase inhibition rate of ethanol extracts DISG 40% and 80% at 1,000 ppm. It was concluded that compositive ingredients (Persicae Seman, Glycyrrhizae Radix, Cinnamomi Ramulus, Rhei Rhizoma) influenced the most results. Conclusions : The results indicated that, ethanol extract which is superior in its anti-oxidative and anti-aging effects is useful to be applied in herbal cosmetic industry.

Mass Production of Chaff-vinegar and Its Effect of Anti-Aging and Whitening (왕겨초액의 대량생산과 항노화 및 미백 효과)

  • Chaff-vinegar is known for having a variety of useful purposes in the fields of health and lifestyles. In a previous study we isolated and identified the active fractions of the polyphenol compound 7 species as a potential biomaterial for cosmeceuticals. To further test for its potential use as a functional material, we carried out an MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity assay and a xanthine oxidase inhibition assay. Chaff-vinegar exhibited potent collagenase and elastase inhibitory activities in a concentration dependent manner, indicating that the agent has the potential to alleviate the skin wrinkling process. Chaff-vinegar also showed 80% tyrosinase inhibition at a concentration of $100{\mu}L/mL$. DPPH radical scavenging, xanthine oxidase inhibition, and SOD-like activity results for each activity were 80%, 80%, and 100%, respectively. Taken together, the present study suggests that chaff-vinegar is a good candidate for use as an anti-wrinkling and/or whitening agent.
