수도 약배양의 효율을 높이기 위하여 callus 유기와 녹색체 출현율에 영향하는 화분의 발육단계, 화분의 저온처리와 기관 및 배지의 영향에 대한 일련의 실험을 수행하였으며 이에서 얻어진 결과를 요약하면 다음과 같다. 1. 단핵기의 화분에서 callus 유기율이 높았으며 이때는 대체로 엽신간장이 5~8cm에 해당되나 품종에 따라 차이가 있다. 2. 이삭을 상, 중, 하로 나누어 조사한 결과 엽이간장이 5~8cm의 것은 상부와 중간부분에 단핵기에 해당되는 것으로 나타났고 9~10cm 정도의 것은 중부와 하부 부분이 단핵기에 해당하는 것으로 나타났다. 3. 일반적으로 저온처리는 8$^{\circ}C$에서 12$^{\circ}C$가 callus 유기에 알맞는 것으로 나타났다. 그러나 온도에 대한 반응은 품종에 따라 차이가 있어 4$^{\circ}C$에서 callus 유기가 잘된 것도 있었다. 4. 저온 처리기간은 일반적으로 8~12일 정도에서 callus 유기가 잘 되었으나 품종간 차이가 있다. 5. 본 실험에서는 N6D 배지보다 N6 배지에서 callus 유기율이 높았다. 6. 대체로 녹색체 출현율은 4$^{\circ}C$와 8$^{\circ}C$로 처리한 것이 12$^{\circ}C$ 처리보다 높았다. 그러나 품종간 차이가 있었다. 7. 녹색체 출현에 좋은 결과를 주었던 온도 처리기간은 대체로 8~12일로 나타났다. 8. 본 실험에서 녹색체 출현율은 N6 배지보다 개량 MS 배지에서 2배 정도 높았다.
본 연구는 연속행동모니터링시스템(CBMS)을 사용하여 저온 충격에 노출된 참돔 Pagrus major 치어의 행동반응에 대한 임계 수온 및 행동 내성 반응을 규명하였다. 실험생물의 행동은 8.0~20.0℃ 수온구간에서 12시간과 24시간 마다 각각 2.0℃와 4.0℃씩 하강시키며 관찰하였다. 참돔 치어는 12.0℃ 이하로 수온이 하강한 경우 수온 노출시간과 변동 폭에 관계없이 유영활동이 감소하고 정지가 반복되는 불안정한 행동패턴이 관찰되었다. 또한 8.0~10.0℃ 수온에 노출된 생물의 유영능력은 급격히 저하되어 수조 바닥에서 머무르는 아치사 반응 행동을 보였다. 실험어류는 저 수온 스트레스 영향을 받아 생물의 50 %만 생존하였으며, 저온 충격(cold shock) 후 6시간 이내에 모든 개체가 사망하였다. 또한 행동지수(BI)는 급격히 감소하였으며 변이계수(CV)의 진폭변화는 타 수온보다 변동 폭이 큰 것으로 분석되었다(p<0.05). 참돔 치어의 저온 스트레스는 12.0℃를 경계로 촉진되며, 8.0~10.0℃의 저온 충격에 노출된 생물의 아치사 반응을 유발시키는 내성한계인 것으로 해석된다.
본 연구에서 L. crispatus KLB46을 저온 전처리함으로서 제제화 과정에 받게 되는 얼림과 녹임, 건조 스트레스뿐만 아니라, 여러 다른 환경 스트레스에 대한 내성이 증진된다는 것을 확인하였다. 그리고 내성 증진에 신규 단백질 합성이 필요함을 확인하였으며 나아가, 저온 충격 유전자 (csp)를 확인하였다. 따라서 이 균주를 제제화 하기 위한 방법으로 저온 전처리를 이용할 경우 생균력 유지에 큰 효과를 얻을 수 있다고 사료된다.
Blocking the first mitotic cleavage of the zygote is a key tool for chromosome-set manipulations in fish. We developed an improved method for inducing tetraploidy by blocking the mitosis with a combination of heat shock at 40.5$^{\circ}C$ for 1, 2 or 3 min followed by cold shock at $1.5^{\circ}C$ for 30, 45 or 60 min. When applied during the first cleavage metaphase of mud loach (Misgurnus mizolepis) zygotes, the optimal combination was heat for 2 min followed by cold for 45 min. At 1 month, the frequency of 4N survivors and the yield from total eggs fertilized was 55.7% and 14.4%, respectively, compared to heat shock alone with 20.0% efficiency and 3.6% yield. The effectiveness of the procedure was confirmed by diploid mitotic gynogenesis using transgenic markers. The overall yield of homozygous diploids, 34.0%, was better than that for single heat shock, 17.3%. The tetraploids and homozygous diploids had higher early mortality than normal diploid controls. However at 1 month, the viability of the tetraploids was the same as normal diploids. For gynogenetic diploids, the survival was similar to normal diploids after 3 months. The high efficiency of this new protocol extends the opportunity to study polyploidy in basic and applied research.
Cold shock proteins (CSPs) are a family of proteins induced at low temperatures. CSPs bind to single-stranded nucleic acids through the ribonucleoprotein 1 and 2 (RNP 1 and 2) binding motifs. CSPs play an essential role in cold adaptation by regulating transcription and translation via molecular chaperones. The solution nuclear magnetic resonance (NMR) or X-ray crystal structures of several CSPs from various microorganisms have been determined, but structural characteristics of psychrophilic CSPs have not been studied. Therefore, we optimized the purification process to obtain highly pure Lm-Csp and determined the three-dimensional structure model of Lm-Csp by comparative homology modeling using MODELLER on the basis of the solution NMR structure of Bs-CspB. Lm-Csp consists of a ${\beta}$-barrel structure, which includes antiparallel ${\beta}$ strands (G4-N10, F15-I18, V26-H29, A46-D50, and P58-Q64). The template protein, Bs-CspB, shares a similar ${\beta}$ sheet structure and an identical chain fold to Lm-Csp. However, the sheets in Lm-Csp were much shorter than those of Bs-CspB. The Lm-Csp side chains, E2 and R20 form a salt bridge, thus, stabilizing the Lm-Csp structure. To evaluate the contribution of this ionic interaction as well as that of the hydrophobic patch on protein stability, we investigated the secondary structures of wild type and mutant protein (W8, F15, and R20) of Lm-Csp using circular dichroism (CD) spectroscopy. The results showed that solvent-exposed aromatic side chains as well as residues participating in ionic interactions are very important for structural stability. Further studies on the three-dimensional structure and dynamics of Lm-Csp using NMR spectroscopy are required.
Antifreeze proteins (AFP) inhibit growth and recrystallization of ice, and permit organisms to survive in cold environments. The AFP from an Antarctic bacterium, Flavobacterium frigoris PS1, FfIBP (Flavobacterium frigoris icebinding protein), was produced in E. coli using a cold shock induction system. The culture temperature was shifted from $37^{\circ}C$ to $15^{\circ}C$ and a 20 L culture scale was used. The final weights of dried cell and FfIBP were estimated to be 126 g and 8.4 g, respectively. The thermal hysteresis (TH) activity ($1.53^{\circ}C$) of the produced FfIBP was 3.6-fold higher than that of the LeIBP (Leucosporidium ice-binding protein) produced in Picha. The current study demonstrates that large-scale production of FfIBP was successful and the result could be extended to further application studies using recombinant AFPs.
In spite of potential benefits of anther culture, low productivity of plant regeneration in some genotypes; e.g. tonsil and indica rice, is one of the major obstacles for practical use of anther culture. This study was conducted to improve cold shock method and carbohydrate source for increasing the efficiency of anther culture in rice. The most common carbon source, sucrose was replaced to maltose, which has two molecules of glucose. Maltose increased callus induction 1.4-to 1.8-fold higher in japonica rice, 3.2-to 11.6-fold in tongil types and 2.7-fold in indica rice IR50. Callus induction was increased from 0.2% to 12.5% in maltose medium compared to the medium supplemented with sucrose plus glucose in indica rice "Tetep". A simple procedure of vacuum packaging of panicles during cold shock treatment prolonged not only anther viability more than 15 days but also increased callus induction more than 2-fold compared to open-air storage (conventional method). Combining of above two methods, callus induction was increased 28 to 56% in japonica, 13 to 33% in tonsil type and 12 to 31% in indica rice. Plant regeneration was increased 14 to 35% in japonica, 10 to 20% in tonsil and 4 to 15% in indica rice, respectively.
춘채 휴면잠종을 산난후 약 50일째에 전기충격을 줌으로서 부화의 가능성여부를 실험한 결과 다음과 같은 결론을 얻었다. 1. 휴면란에 대한 전기충격은 배자의 발육을 촉진시켰다. 2. 전기처리시간은 5초에서 비교적 부화성적이 좋았다. 3. 침산처리일로부터 약 15~22일 사이에 산발적으로 부화가 시작되었다.
내한성이 약한 유채(B. napus)와 내한성이 강한 산동채(B. campestris)를 공시하여 발아초기단계에서의 장기저온순화처리가 peroxidase와 superoxide dismutase 활성도 및 동위효소 pattern에 미치는 영향을 분석하여 작물 내한성 기작을 생화학적으로 구명함과 아울러 내한성 관련 genetic marker를 탐색하고자 본 연구를 수행하였다. 장기저온순화시킨 유묘를 저온처리후 24시간 회복시켰을 경우, peroxidase 활성도는 유채와 산동채의 뿌리부위에서 각각 157%와 50%, 배축부위에서는 201%와 250% 정도 크게 증가하였고, superoxide dismutase 역시 배축 및 뿌리부위에서 크게 활성화가 일어남을 관찰할 수 있었다. 또한 배축부위에서의 단백질 함량도 유채는 약 11.4%, 산동채는 57.4% 증가하였다. 장기저온순화처리로 새로 생합성된 peroxidase 동위 효소들중 특히 5번 band(pl 6.4)는 유채와 산동채의 뿌리부위에서 모두 출현될 뿐만 아니라 내한성이 강한 산동채의 배축부위에서도 강하게 출현되어 내한성 관련 genetic marker로써의 이용 가능성을 높여 주었다. 이상의 결과와 전보의 결과들을 종합적으로 고찰하여 저온 shock에 따른 분자산소의 생화학적 상호전환 모델을 제시하여 작물 내한성 기작을 생화학적으로 추론하였다.
