Induction and Proliferation of Adventitious Roots in Codonopsis spp. (더덕 속 식물의 부정근 유도와 증식)

  • We have established adventitious root culture systems of Codonopsis lanceolata, Codonopsis pilosula and Codonopsis ussuriensis. Root segments of C. lanceolata were the best explants for induction of adventitious roots and the number of adventitious root for explant was highest on solid medium with $0.5mg/{\ell}$ NAA and produced $18.8{\pm}1.9$ roots per explant. Root segments of C. pilosula were the best explants for induction of adventitious roots and the number of adventitious root for explant was highest on solid medium with $1.0mg/{\ell}$ NAA and produced $8.5{\pm}1.8$ roots per explant. Leaf segments of C. ussuriensis were the best explants for induction of adventitious roots and the number of adventitious root for explant was highest on solid medium with $0.5mg/{\ell}$ NAA and produced $7.8{\pm}0.4$ roots per explant. In liquid culture, the best production of adventitious root (fresh weight) was obtained in 1/2 MS medium with $1.0mg/{\ell}$ NAA. This study demonstrated for the first time to produce adventitious roots in C. pilosula and C. ussuriensis.

  • The effect of Codonopsis ussuriensis on red blood cell(RBC) count in rats was investigated. Among various fractions tested, a significant increase of RBC was observed in animal group treated with ether extract. From the ether soluble fraction, two compounds were isolated and their structures were elucidated as ${\alpha}-spinasterol$ and ${\beta}-sitosterol$ by chemical and spectral analyses.

Comparison of Growth Characteristics and Inorganic Components Between Korean and Japanese Codonopsis lanceolata

  • This study was performed to investigate the growth characteristics and inorganic components of Codonopsis lanceolata regarding regional differences. The plant height of Japanese Codonopsis lanceolata was 373.6 cm, so it's revealed that it has more vigorous growth than Korean won. The flowering time of Korean Codonopsis lanceolata was 2 weeks faster than Japanese one. Total fresh weight of root was 41.0 g and 39.0 g for Korean and Japanese respectively, thus, no significance difference was found. However, regarding fresh weight, Korean one had a more fresh weight (35.4 g) of main root parts, but Japanese one had a more fresh weight (9.6 g) of the lateral root part. Each inorganic component was found more in the aboveground parts, regardless of the region and the content of K was the largest. Regarding the content of macroelements for each part of Codonopsis lanceolata, the content of Na, Mg, P, S, and Ca in Korean Codonopsis lanceolata was found the highest on the leaf, followed by stem and root. In the case of Japanese Codonopsis lanceolata, same result was found on the content of Mg and Ca, however, the highest content of Na and P was found in the stem.

Effect of Extracts of Codonopsis lanceolata on Macrophage Activity and on the Growth of Lactic Starter Culture during Fermentation (더덕 추출물이 마크로파지 활성과 발효 중 젖산균 성장에 미치는 영향)

  • We examined the macrophage activity of Codonopsis lanceolata and its effect on the growth of lactic starter culture when it was added to fermented milk. Nitric oxide(NO) and interleukin-1${\alpha}(IL-1{\alpha})$ were increased significantly(p<0.05) by addition of Codonopsis lanceolata water extract at $1,000\;{\mu}g/mL$. Tumer necrosis factor-${\alpha}(TNF-{\alpha})$ was increased significantly (p<0.05) by addition of Codonopsis lanceolata water extract or 70% ethanol extract at $1,000\;{\mu}g/mL$. Water extract of Codonopsis lanceolata showed higher macrophage activity than 70% ethanol extract. Growth of lactic starter culture was inhibited by the increased addition of Codonopsis lanceolata water extract, resulting in less decrease in pH. A stirred type or drink type fermentation process seemed mote suitable than a set type in proper production of Codonopsis lanceolata extract added fermented milk.

Effect of Northern Root-knot Nematode, Meloidogyne hapla, on Growth of Codonopsis Zanceolata (당근뿌리 혹선충(Meloidogyne hapla)이 더덕(Codonopsis lanceolata)의 생육에 미치는 영향)

  • This study has been conducted to assess and analyze the damage of Codonopsis lanceolata by differential inoculation levels of Meloidogyne hapla in pot and plot. As the density of M. hapla increased, early growth of C. lanceolata was inhibited 60% in vine and 54% in root. However, growth of C. lanceolata was not remarkably different by density of M. hapla in the plot test. This result may be attribute to low density of M. hapla in the plot.

Studies on Plant Parasitic Nematodes in the Fields of Codonopsis lanceolata (더덕(Codonopsis lanceolata) 재배지 기생선충의 발생상황)

  • This study were conducted to assess plant­parasitic nematodes infesting Codonopsis lanceolata. Ten kinds of plant­parasitic nematode gem including Meloidogyne and Pratylenchus were identified in 34 localities of C. lanceolata fields in three provinces in Korea. Meloidogyne, a dominant genus, was found in 97% localities and showed high average density as 1,700 nematodes per 300$m\ell$ soil. Its average density was 10 times higher in upland field than in paddy field.

  • Codonopsis lanceolata belongs to the Campanulaceae family, which is a perennial herb. It is widely used as an antitussive agent and an expectorant in oriental medicine. Codonopsis lanceolata contains large amounts of polysaccharide and phenolic compounds and is known to have anti-oxidative effect and improve immunity. We evaluated the efficacy and the skin safety and stability of cosmetic that is applied Codonopsis lanceolata extract fermented by lactic acid bacteria. As result, formulation of Codonopsis lanceolata extract improved the human skin moisturizing effect, increased TEWL 196%, skin hydration 15%, and the sebum rate 131%.

  • Codonopsis lanceolata has long been used as a seasonal food and as a traditional tonic medicine with anti-inflammatory and anti-oxidation properties. The present study investigated the in vitro effect of Codonopsis lanceolata extracts on immune function in mice. After preparing a single cell suspension splenocyte proliferation was determined by the MTT(3-[4,5-dimethylthiazol-2-y]-2,5-diphenyl terazolium bromide) assay. The cytokines IL-1${\beta}$, IL-6, and TNF-$\alpha$ were not secreted by macrophages stimulated with or without LPS as determined by an ELISA cytokine kit assay. After a 48-hr incubation with the mitogens ConA or LPS there was an increase in splenocytes proliferation and in the production of IL-1${\beta}$, IL-6, and TNF-$\alpha$ in the suspensions supplemented with 50, 100, 250, 500 ${\mu}g/m{\ell}$ Codonopsis lanceolata water extract. The results suggest Codonopsis lanceolata water extract may enhance immune function by regulating splenocyte proliferation and stimulating cytokine production.

Effects of Pinching times on Root Yield and Quality of Codonopsis lanceolata (S.et Z.) Trautv. (더덕순 채취가 수량 및 품질에 미치는 영향)

  • To produce edible Codonopsis shoots in spring, the effect of shoot pinching times on root yield and quality of Codonopsis was evaluated during 3 years from 1992 to 1994. Shoot growth decreased by increasing picking times and one time pinching of Codonopsis shoot was optimal for seed production. In case of 3 times shoots harvest per year. yield of edible Codonopsis shoots amounted to 29.5MT/ha for 3 years. Increase of shoot picking times exerted harmful effect on root growth and decreased marketable value of harvested shoots. It was recommended from the results. One-time pinching of shoot a year for the production of marketable shoots and roots after 3-year cultivation.

Introduction of RAG25gene into Codonopsis lanceolata by Agrobacterium tumefaciens (Agrobacterium tumefaciens에 의한 RAG25유전자의 더덕 (Codonopsis lanceolata)으로의 도입)

  • A RAG25 gene regulating flowering time was introduced into Codonopsis lanceolata through high efficiencies (ca. 90%) of plant regeneration. The leaf explants were immersed in YEP media containing Agrobacterium tumefaciens (pGA 1209) harboring RAG25 gene, and cocultivated for 3 days. After cocultivation, they were cultured in shoot inducing media (SIM), N2B2 (NAA 2 mg/L, BA 2 mg/L and kanamycin 20 mg/L) and N2B4 (NAA 2 mg/L, BA 4 mg/L and kanamycin 20 mg/L), and the putative transformants were regenerated. The introduction of nptII and RAG25 gene into Codonopsis lanceolata was confirmed by 0.7 kb and 0.6 kb bands from polymerase chain reaction and reconfirmed by Southern hybridization using PCR product of RAG25 gene.
