Effect of $SO_2$ on Leaf Tip Blight of Ginkgo biloba caused by Coniothyrium sp. (은행나무의 잎끝마름병에 미치는 $SO_2$ 의 영향)

  • To examine the relationship between air pollution and occurrence of disease in plants, we investigated the alteration of physiology and biochemistry of Ginkgo biloba by inoculating with Coniothyrium sp. and fumigating with 0.1 ${\mu}l/l\;SO_{2-}$ Visual damage did not appear but photosynthesis, $CO_2$ use efficiency(CUE) and water use efficiency(WUE) were reduced when G. biloba was exposed to $SO_{2-}$for 7days (8 hours a day). When inoculated with Coniothyrium sp., the seventies of damage in G. biloba showed a threefold difference between $SO_{2-}$ treatment (SI) and $SO_{2-}$ free treatment (SFI) at day 42 from initial inoculation. Little difference was observed in sugar contents that may be used pathogens feed, among control, SFI and SI. In spite of the reduction in photosynthetic rate, sugar contents and CUE were maintained. WUE was enhanced 13% more at SI than SFI. The $CO_2$fixation boosted because of enhanced WUE, and thus sugar synthesis was not affected. In addition, sugar transport seems to be retarded for some internal alteration. Consequently, the severity of SI was more serious than that of SFI because Coniothyrium sp. easily invades into the physical texture of G. biloba weakened by $SO_2$ fumigation and because sugar was accumulated in leaves of G. biloba.

First Report of Impatiens necrotic spot virus in Hoya carnosa in Korea (호야에서 발생한 봉선화괴저반점바이러스 국내 첫 보고)

  • In 2016, less than 30% of virus-like symptoms such as chlorosis, necrosis and ringspots were observed in Hoya carnosa from commercial greenhouse in Eumseong, Korea. A total of 6 samples from Hoya carnosa were collected both symptomatic and asymptomatic plants and tested for virus infection by RT-PCR of 3 viruses known to infect Hoya spp. including Tomato spotted wilt virus (TSWV), Impatiens necrotic spot virus (INSV) and Tomato chlorotic spot virus (TCSV). Three symptomatic samples were positives for INSV. Also, it was not the virus detected in three asymptomatic samples. To further confirm the presence of INSV, complete nucleocapsid (N) gene of the virus were amplified and sequenced from two samples. BLAST analysis of the consensus sequence showed that two isolates (INSV-Hy1 and -Hy2) shared nucleotide sequence identities of 99% with each other and 97-99% with other INSV isolates available in the GenbBank. Phylogenetic analysis showed that these isolates closely related to the INSV isolates from ornamental from China. This is the first report of INSV on Hoya carnosa from Korea.

Chlorsulfuron-induced Phytotoxicity in Canola(Brassica napus L.) Seedlings (캐놀라 식물체내에서 클로르설푸론의 약해 유발 요인)

  • Chlorsulfuron, an acetolactate-synthase-inhibiting sulfonylurea herbicide, induces many metabolic and physiological changes in susceptible plants. The objective of this study was to determine to what extent chlorsulfuton-induced phytotoxicity was due to a shortage of final products(the branched-chain amino acids valine, leucine, and isoleucine) or to an accumulation of a toxic metabolite(2-ketobutyrate), or both, in a susceptible species. Chlorsulfuron-treated canola seedlings showed growth inhibition and injury symptoms that included chlorosis, downward leaf rolling, and accumulation of anthocyanins. Supplementation with valine, leucine, and isoleucine prevented the chlorsulfuron-induced growth inhibition and injury symptoms only partially, suggesting that factor(s) other than a shortage of the branched-chain amino acids also are involved in the phytotoxicity. Canola seedlings treated with 2-ketobutyrate showed reduced growth, but they showed different changes in metabolites than seedlings treated with chlorsulfuron. The results suggest that 2-ketobutyrate is not involved in chlorsulfuron-induced phytotoxicity. We conclude that chlorsulfuron-induced phytotoxicity is due at least in part to a shortage of branched-chain amino acids.

Influence of Humic or Fulvic Acid on Phytotoxicity of Bentazone (Bentazone의 약해(藥害)에 미치는 부식산(腐植酸) 및 훌브산(酸)의 영향(影響))

  • This research was conducted to assess the influence of humic or fulvic acid on Bentazone phytotoxicity using a bioassay with hydroponically grown cabbage (Brassica campestris subsp. napus var. pekinensis Makino). Concentrations of Bentazone in the water culture media were ranged from 0 to 32 ${\mu}M $ and those of the organic ligands were 1.0mM as a soluble carbon. Media were prepared in a complete factorial combination with pHs of 4.5, 6.5 and 8.5. The phytotoxicity indices on growth rate and dry weight decrement were employed to evaluate the effects of organic ligands on the Bentazone phytotoxicity. Humic or fulvic acid without Bentazone treatment enhanced the growth of cabbage and this effect was evident at low pH of 4.5. Bentazone led to chlorosis and necrosis on cabbage leaves resulting in the decreases of dry and fresh weights and growth rate. This phytotoxic effect was increased with Bentazone concentration and evident at low pH. At pH 4.5, dry weight was decreased about 63% with 8${\mu}$M of Bentazone treatment. Effective concentration of Bentazone causing 50% decreases in fresh weight as compared to the control was estimated to be 21${\mu}$M. Presence of organic ligand reduced the phytotoxicity of Bentazone to cabbage significantly by increasing yields and growth rates as compared to the treatment of Bentazone alone. At pH 4.5, fulvic acid reduced phytotoxicity of Bentazone upto 46%, and this efficiency of fulvic acid was better than that of humic acid under the same condition.

Action Characteristics of EK3143 and EK3150, New Cyclohexanedione Herbicides, and Their Effects on Acetyl CoA Carboxylase Activity (신규 cyclohexanedione계 화합물 EK3143과 EK3150의 제초작용 발현특성과 Acetyl CoA Carboxylase 활성에 미치는 효과)

  • This study was carried out to investigate the herbicidal action characteristics of EK3143 and EK3150, newly synthesized cyclohexanedione analogues, and their inhibitory effects on the acetyl-CoA carboxylase activity isolated from various plants. Sethoxydim, a typical cyclohexanedione herbicide, had a high herbicidal activity against only grasses with growth inhibition, leaf chlorosis and necrosis. EK3143 and EK3150 had a lower effect against grasses but a higher effect against broadleaf plants such as black nightshade and velvetleaf than sethoxydim had. Low rate application of EK3143 induced leaf bleaching(whitening), while high rate application induced the same symptoms as those of sethoxydim. EK3150 induced more significant whitening, additionally accompanying a curling of meristem part in broadleaf plants. A consistant tendency was observed between the growth inhibition against grasses and the senstivity of grass ACCase to these compounds, but not between herbicidal activities against broadleaf plants and the senstivity of dicotyledonous ACCase; i.e. EK3150, which had higher herbicidal activity against black nightshade and velvetleaf than EK3143 had in greenhouse, rather showed lower ACCase inhibition. In EK3150-treated barnyardgrass, growth inhibition occurred prior to decrease of photosynthetic pigments, in contrast to that of norflurazon and EK3140. These results suggest that EK3143 and EK3150 may have two target sites, ACCase and an unknown site on fatty acid synthesis related to growth inhibition and bleaching respectively, and their symptoms would be appeared in different degree by a differential binding affinity between two action sites.

Diagnosis and Sequence Analysis of Japanese yam mosaic virus from Yam (Dioscorea opposita) (마(Dioscorea opposita)에 발생한 Japanese yam mosaic virus 진단 및 염기서열 분석)

  • We surveyed the occurrence of Japanese yam mosaic virus (JYMV) on Yam in Gyeongsangbukdo pronvince from 2013 to 2015. The symptoms of JYMV were yellow stripes and chlorosis in yam leaves and the infection rate was ranged from 33.6% to 40.8%. We determined nucleotide sequence encoding the polyprotein of JYMV isolate BRI from yam leaves using next-generation sequencing (NGS) method. The partial nucleotide portion (7,736 nucleotides) of the genomic RNA of the JYMV isolate BRI has been sequenced (accession No. KU309315). The region sequenced includes a single open reading frame (ORF) encoding a polyprotein composed of 2,497 amino acids containing the coat protein (CP) and 3' untranslated region (UTR). The genomic organization of this isolate shows almost the same to that of other members of JYMV. The JYMV isolate BRI showed 77% to 79% nucleotide identity with the Japanese and Chinese strains and isolates. This is the first report of the genome nucleotide sequence of JYMV from Dioscorea opposita in Korea.

Characterization of Toxicity Symptom and Determination of Tissue Threshold Levels of Boron for Diagnostic Criteria in Domestically Bred Strawberries (국내육성 주요 딸기 품종에서 발생하는 붕소 과잉 증상 및 영양진단을 위한 식물체 내 한계농도)

  • Diagnosis of nutrient disorders in cultivating crops is based on the visual symptoms and results of soil and plant analysis. This study was carried out to investigate the influence of B concentrations in fertilizer solution on the growth of and nutrient uptake by domestically bred strawberries. Tissue analysis based on the dry weight were also conducted to determine the threshold levels in plants when toxicity developed in boron. The growth was seriously restricted in the three strawberry cultivars as the B concentrations in fertilizer solution were elevated. The fresh and dry weights were heavier and crown diameters were thicker in the 0.25 mM boron treatment than the other treatments tested. The toxicity symptoms of boron appeared on the older leaves of three strawberry cultivars while interveinal chlorosis symptoms appeared on the young leaves of 'Keumhyang' and 'Seolhyang' strawberries. The toxicity symptoms in lower leaves were developed when B concentrations in fertilizer solution were higher than 2 mM in 'Keumhyang' and 0.5 mM in 'Maehyang' and 'Seolhyang' strawberries. The elevated boron concentrations in fertilizer solution did not influence the tissue K, Ca, and Mg contents, but influenced the phosphorus contents with decreasing tendency. The tissue Fe and Zn contents decreased and increased, respectively, as the B concentrations in fertilizer solution were elevated. When the concentration of boron at which the growth of a plant is retarded by 10% is regarded as a upper threshold level, the boron contents based on dry weight of above ground plant tissue should be lower than 25.1, 44.2, and 62.5 $mg{\cdot}kg^{-1}$ in 'Keumhyang', 'Maehyang', and 'Seolhyang' strawberries, respectively.

Morphological Characteristics and Molecular Identification of Calonectria ilicicola Causing Soybean Red Crown Rot (콩 검은뿌리썩음병균(Calonectria ilicicola)의 형태적 특징과 유전자 수준에서의 동정)

  • The 18 isolates of soybean red crown rot pathogen(Calonectria ilicicola) were isolated from infected soybean plants showing chlorosis symptom and red perithecia sign in 2002 and 2003 season in Korea. The morphological characteristics of the typical isolate SC03-15, such as macroconidia and macrovesicles in anamorph as well as perithecia, asci and ascospores in teleomorph, were observed from red crown rot pathogens. The perithecia were red-brown globose to ovoid, and $357.9(312.5{\sim}391.5.7){\mu}m$ high and $474.3(419{\sim}535){\mu}m$ diameter. Asci were hyaline and clavate and included 8 ascospores, which were straight to slightly curved, 1(-3)-septate, and $36.9(31.1{\sim}49.4){\times}5.2(4.5{\sim}6.1){\mu}m$. Macroconidia in anamorph were cylindrical, hyaline and in size $73.2(68.1{\sim}77.6){\times}5.8(4.9{\sim}6.8){\mu}m$. Further identification process was performed through molecular method, such as a phylogenetic analysis by rDNA sequencing of ITS and $\beta$-tubuline genes of all isolates tested in this study. All isolates were identified as Calonectria ilicicola by DNA sequencing of ITS and $\beta$-tubuline genes, and morphological characteristics.

Impacts of Air Pollution on Forests : A Summary of Current Situations (대기오염이 삼림에 미치는 영향 : 피해현황과 원인을 중심으로)

  • Issues of declining growth and vigor in forests are major concerns in many areas around the world, especially in response to predictions in the 1980s of widespread forest declines. This paper summarizes the current state of knowledge for forests in North America, Europe, and East Asia. Forest declines near point-sources of pollution(such as metal smelters) have been well recognized for a century, but evidence of widespread impacts away from point-sources remains relatively uncommon. In North America, significant forest decline has resulted from high concentrations of ozone near Los Angeles, California, and around Mexico City. Some high-elevation forests of red spruce in the eastern U.S. have declined in the past 20 years ; evidence indicates a role of low-pH fog in reducing they cold-tolerance of spruce. In Europe. most attention has focused on Norway spruce stands that developed yellow foliage, needle loss, and in some cases mortality. This syndrome appears to be related generally to an inadequate supply of magnesium, perhaps coupled with a very high supply of nitrogen. Despite localized areas that show declining trees, overall stand growth and standing biomass in Europe increased from. 1970 to 1990. Much less information is available for East Asia. Many industrialized regions in China have a pH of rain <4.5, and some connections between pollution and forest decline have been suggested. Pollution impacts on forests near cities in Korea include needle chlorosis, reduced needle retention, and declining species diversity. Overall, temperate forests show no widespread declines, and no evidence of substantial effects of pollutants on forest growth or vigor. Small areas showing declining forests may indeed demonstrate pollution impacts, and may provide cause for concerns about future impacts on larger areas.

Resistance Evaluation of Radish (Raphanus sativus L.) Inbred Lines against Turnip mosaic virus (순무모자이크바이러스에 대한 무 육종 계통 저항성 평가)

  • Leaves of twenties radish (Raphanus sativus L.) inbred lines were mechanically inoculated with Turnip mosaic virus (TuMV) strain HY to evaluate TuMV resistance of the radish inbred lines. The inoculated radish plants were incubated at $22^{\circ}C{\pm}3^{\circ}C$ and resistance assessment was examined using symptom development for 4 weeks. Based on the reactions of differential radish inbred lines, 16 radish lines were produced mild mosaic, mottling, mosaic and severe mosaic symptoms by TuMV infection. These results were confirmed by RT-PCR analysis of TuMV coat protein gene, suggesting that TuMV is responsible for the disease symptoms. Four resistant radish lines did not induce systemic mosaic symptoms on upper leaves and chlorosis in stem tissues for 4 weeks, showing they were symptomless by 8 weeks. Further examination of TuMV infection in the 4 radish lines showed no TuMV infection in all systemic leaves. These results suggest that the 4 radish lines are highly resistant to TuMV.
