Antidiabetic Effects of Herbal Medicine on db/db Mice (4종 한약 처방이 db/db 마우스의 혈당 개선에 미치는 영향)

  • The effects of herbal medicine on type 2 diabetic animal model were investigated. Herbal medicine were composed with the addition of Coicis Semen into Okchun-san (OCS). Commelinae Herba into Gangsim-tang (GST). Scrophulariae Radix into Hyunsamsunki-san (HSK). and Erythrinae Cortex into Yukmijihuang-hwan (YMH). We evaluated anti-hyperglycemic and body weight reduction activity in diabetic db/db mice. The experimental animals were divided into six groups. as control group and five sample groups. Each 200mg/kg/day of OCS, GST, HSK and YMH was administered with orally for 14days long. 5mg/kg/day of acarbose was administered with orally for 14days long. On day 14, OCS-treated db/db mice had significantly lower fasting blood glucose levels compared to control group(296±25.9 versus 593±16.4mg/dl. p<0.001). During the 2 h intraperitoneal glucose tolerance test (IPGTT), all the sample groups were improved compared to control group but insignificantly. After 14days of extract treatment. body weight in control. YMH and acarbose groups were increased. but OCS. GST and HSK groups reduced. However. it did not significantly lower hemoglobin Alc(HbAlc) in blood of db/db mice. These result suggest that OCS could be effective on insulin-independent type 2 diabetes.

An Experimental Study for Radiation Nephritis in Rabbits (가토(家兎)를 이용(利用)한 방사선신염(放射線腎炎)의 실험적(實驗的) 연구(硏究))

  • Experimental radiation nephritis was produced in 15 rabbits by X-irradiation. About $2,000{\gamma}$ (tissue doses) were given to both kidneys of a rabbit in 5 days. Other tissues and organs except both kidneys were protected with 2mm thickened lead plates. 5 weeks after the last irradiation, blood pictures, blood pressures, B.U.N., serum creatinine, Ca, Mg, Fe levels and serum erythropoietin activity of the irradiated rabbits were studied. After finishing above studies, rabbits were sacrificed and both kidneys were removed and examined histopathologically. Same laboratory and pathological studies were performed in 6 control rabbits. In this study, the author obtained following results. 1) Both kidneys of rabbitis with experimental raditation nephritis showed marked histopathological changes, i.e.: renal tubules showed diffuse cloudy swelling, impacted intraluminal hyaline casts and focal precipitations of lime salts on the tubular epithelium. Diffuse interstitial fatty necrosis and various degrees of fibrotic infiltrations on the interstitium were also seen in association with focal lymphocytic infiltrations. Hyaline degenerations were observed on the glomeruli and small vessels. 2) Experimental radiation nephritis rabbits showed marked lowering in R.B.C. counts, decreased hemoglobin levels, low hematocrit values and leucopenia in comparison with those of control rabbits. (P<0.01). (Table 1 & 2). 3) Mild proteinuria were observed in experimental radiation nephritis in rabbits. 4) The levels of B.U.N. and serum creatinine increased in experimental radiation nephritis. (P<0.01). (Table 1,3 & 4) 5) The levels of serum Ca and Mg Showed no statistical difference in comparison with those of control rabbits. (P>0.05) (Table 3 & 4). 6) No statistical correlations were observable between the levels of B.U.N. and Hb. values. (${\gamma}=-0.223$) No close correlations (${\gamma}=-0.328$) were noticed between the levels of B.U.N. and serum iron levels. 7) Erythropoietin activity (R.B.C. $^{59}Fe$ Incorporation) was measured by the modified Fried method. No change in its activity was noticed in radiation nephritis group comparing with that of the control group. (P>0.05). (Table 1,3 & 5). 8) Carotid artery blood pressures showed also no difference. Studies on Experimentally Induced Traumatic Hepatitis and Splenitis in Ruminants (반추수(反芻獸)의 창상성간염(創傷性肝炎) 및 비염(脾炎)에 관한 실험적(實驗的) 연구(硏究))

  • The traumatic hepatitis and splenitis were experimentally induced in Korean native goats. General examinations, hematological and blood biochemical findings were investigated up to 10 days after induction of traumatic hepatitis and splenitis. Body weight of goats with traumatic hepatitis and splenitis showed no change or only a slight decrease. Changes in body temperature and respiratory rate were not noted in both experimental groups. Heart rate hart not been changed in traumatic splenitis group but were a little increased in traumatic hepatitis group. Erythrocytic series such as erythrocyte counts, hemoglobin concentration and packed cell volume showed same variation within normal range during the period of observation. Total leucocyte counts showed the considerable increase in both experimental groups. The increase of the percentage of neutrophil and the decrease of the percentage of lymphocyte were more remarkable in traumatic hepatitis group than that of traumatic splenitis group. Serum aspartate transferase activity was increased in both experimental groups after surgery, while serum alkaline phosphatase activity has been decreased conciderably up to 7 days after surgery. Serum lactic dehydrogenase activity has been increased up to 3 days after operation and thereafter the activity has returned to normal level in traumatic hepatitis group and the activity increased only the first day after surgery in traumatic splenitis group. There were no changes in total protein and albumin contents in both experimental groups during the period of observation. Serum total cholesterol contents showed a slight increase in both experimental groups and the range of increase in traumatic hepatitis group was more wide. Serum bilirubin contents were net changed in traumatic hepatitis group, but showed a slight increase in traumatic splenitis group.

Radiation Effect on Body Weight and Hematological Changes of Hybrid Mice by Conventional Fraction, Large Abdominal Field Irradiation (고식적 분할조사시 전복부 조사량에 따른 잡종 백서의 체중과 혈액상의 변화에 관한 연구)

  • Radiation effect on mammals, especially on hematologic changes, has been studied since discovery of x-ray. Various experimental animals were tried for radiobiological studies. 72hybrid mice with conventional faction $(15\times/week)$, large abdominal field $12\times3cm$, from symphysis pubic to xyphoid process) were used. Body weight was declined gradually by increasing irradiation doses, nadir was about $29.7\%$ in male; $30.4\%$ in female at 6000 rad irradiation group. Hemoglobin value was nearly normal throughout entire treatment. Significant dropping of WBC count was noted to $40-50\%$ of pretreatment values by only 1000rad irradiation. Change of differential count was interesting; : lymphocyte proportion showed gradual reduction, instead of gradual increasing of segmented neutrophil. Those proportion were reversed after 6000 rad irradiation. Urinary protein tests showed + - +++, showing no correlation with dosage. Application of our study in clinical combination therapy (radiation + chemotherapy) was discussed.

Reevaluation of the Necessity of Iron Injection to Newborn Piglets

  • The growth and immune responses to endotoxin lipopolysaccharide (LPS) challenge ( $20{\mu}g/kg$) of piglets with and without a iron dextran injection (Fe, 200 mg/head) two days after birth are compared. Sixty-four newborn piglets from eight litters were allocated randomly to one of four treatments. The control received no iron dextran and only saline (Sal) injection on the second and fifteenth day after birth (Sal-Sal). The remaining three groups received Fe-Sal, Sal-LPS, Fe-LPS treatments respectively. On fifteen days of age, blood samples of piglets were taken at 0 h, 1 h, 2 h and 4 d after saline or LPS injection to determine immune functions and blood characteristics. The trial terminated when the pig reached 56 days and the average daily gain of piglets was then measured. Daily gain, serum immunoglobulin G (IgG) concentration and red blood cell counts did not vary significantly among the four groups at any measuring times. Serum tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) concentration increased sharply 1 h after LPS injection. However, iron injection did not change TNF-${\alpha}$ concentration responds to LPS injection. White blood cell counts of two LPS injection groups were significantly lowered 1 h following the injection. In contrast, serum lactoferrin concentration had increased significantly 1 and 2 h postinjection. Furthermore, iron injection produced no further effects on these two criteria. Iron injection increased the hemoglobin (Hb) concentration of piglets at any measuring time, and LPS injection lowered Hb concentration. In conclusion, a 200 mg/head of iron dextran injection on the second day after birth increased Hb concentration, had no detrimental effect on the immune responses and growth of piglets. Moreover, if creep feed (175 mg Fe/kg feed) is provided from d 7 after birth, the Fe-injection does not contribute to overall performance of piglets and may not be a necessity in practice.

Effect of Rebamipide on nepato-Renal Dysfunction Caused by E.coli Lipopolysaccharide in Rat (E. coli Lipopolysaccharide 유발 간-신 기능장애에 있어서 Rebamipide의 효과)

  • The present study was aimed to investigate the preventive effects of rebamipide on the multiple organ dysfunction in a rat model of circulatory shock induced by bacterial endotoxin (E. coli lipopolysaccharide; LPS) in comparison with that of methotrexate. Endotoxemia for 6 hours resulted in little change in the levels of hemoglobin and neutrophils. However, treatment with methotrexate decreased significantly the numbers of circulating neutrophils. Significant increases in serum alanine aminotransferase (ALT,958 $\pm$ 250 lU/L, p<0.001) and aspartate aminotransferase (AST, 1350 $\pm$ 295 lU/L, p<0.001) levels induced by endotoxemia were significantly decreased by rebamipide and methotrexate. The increased level of lactic acid dehydrogenase (LDH) by LPS (2850 $\pm$ 467 lU/L, p<0.05) was significantly inhibited by rebamipide, but not by methot.elate. The elevated serum creatinine (1.2$\pm$0.1, p.0.05) and urea levels (55.3$\pm$6.5 mg/dL, p.0.01) by LPS were also decreased by rebamipide, but not by methotrexate. In line with these results, the plasma concentration of tumor necrosis factor-$\alpha$ (TNF-7,167 $\pm$ 20 pg/mL) was significantly increased upon injection of endotoxin at 1 hour by 1570$\pm$100 pg/mL, and declined to 312$\pm$35 pg/mL at 6 hours. The TNF-$\alpha$ level at 6 hours was significantly decreased by rebamipide to 207$\pm$8 pg/mL (P<0.05). Taken together, it is summarized that rebamipide inhibits the development of multiple ogran dysfunction by inhibition of neutrophil activation in association with inhibition of TNF-$\alpha$ formation in a murine model of endotoxemia.

AN EXPERIMENTAL STUDY ON THE EFFECT OF BLEACHING AGENTS (치아경조직(齒牙硬組織)에 있어 표백제(漂白劑)의 효과(效果)에 관(關)한 연구(硏究))

  • As widely known major cause of tooth discoloration is red blood cell in dentinal tubules after tooth trauma and extirpation. Hemoglobin in the red cells easily conbine with hydrogen sulfide and change natural tooth color into dark brown. To confirm and compare the effect of bleaching agents test material was made as fellows: No. 40 paper points were soaked in blood and put into the oven for 24 hours to dry. Dried paper points were again soaked in diluted yolk by distilled water for a minute and kept the points for a week to get fully sulfide. Hydrogen peroxide (25%), Sodium hypochlorite (2%), Sodium hyposulfate (2%), Sodium perborate and mixture of Hydrogen peroxide (25%) and Sodium perborate were employed as bleaching agents. About 0.2ml of each medicaments were put into the small test tubes and blood soaked paper points were gently placed vertically with the apex downward and kept 24 hours in the oven ($37^{\circ}C$). Every 24 hours the medicaments were renewed at 24 and 48 hours. Following are the results of the study: 1. Significant bleaching effect was found on the case of hydrogen peroxide (25%) after 24 hours. The area above 2mm from apex also fully bleached after 48 hours. 2. A mixture of hydrogen peroxide and sodium perborate treated case showed moderate effect at 24 hours and significant effect at 48 hours. High portion of the point (above 2mm) was bleached satisfactory at 72 hours. 3. The effect of sodium perborate at 48 hours showed moderate and significant at 72 hours. 4. Sodium hypochlorite and sodium hyposulfate were weak agent in bleaching. At 72 hours case revealed moderate effect and influenced on high portion of the point very weak effect.

Influence of Herbal Medicine and Acupuncture Treatment on the Pregnancy Rate in Infertile Women before In Vitro Fertilization-Embryo Transfer (체외수정 시술 전 한방치료가 여성 불임 환자의 임신성공율에 미치는 영향)

  • Objectives: This study was performed to assess whether herbal medicine and acupuncture before in vitro fertilizationembryo transfer (IVF-ET) is effective on clinical pregnancy. Methods: From May 2010 to January 2011, a prospective analysis study was performed in 38 patients planning to undergo IVF-ET after taking herb medicine and acupuncture treatment. This study investigated the pregnancy rate and analyzed the change of dysmenorrhea by visual analog scale (VAS), body heat and condition of premenstrual syndrome (PMS), vaginal discharge and menstruation status. Results: 1. During herbal medicine and acupuncture treatment, five patients (13.16%) naturally became pregnant and six patients (15.79%) withdrew. After treatment, 15 patients (39.47%) received IVF-ET, 12 patients (31.58%) did not. 2. The biochemical pregnancy rate was 26.67%, the clinical pregnancy rate 26.67%, miscarriage rate 25% and ectopic pregnancy rate was 0%. 3. After treatment, PMS, dysmenorrhea and dysmenorrhea VAS was significantly decreased and the overall menstrual status improved. 4. After treatment, temperature difference of CV17-CV12 and CV4-CV12 increased, but it was not a statistically significant difference. 5. After treatment, decrease of hemoglobin and protein and increase of total bilirubin and creatinine were statistically significant. All the blood test results were within normal levels which proves safety of treatment. Conclusions: This study suggests that herbal medicine and acupuncture treatment before IVF-ET shows similar pregnancy rates with existing rates, but contributes to increasing the possibility of natural pregnancy.

A Study on Immune Response by Intoxication of Parathion, Chloroform and 2,6-Dichlorophenol in Rats (Parathion, Chloroform 및 2,6-Dichlorophenol의 중독에 의한 흰쥐의 혈액학적 소견과 면양적혈구에 대한 항체 생산세포수에 미치는 영향)

  • Among the environmental pollutants, parathion, chloroform and 2, 6-dichlorophenol may impair human health; they may inhibit or reduce the metabolic function of human body and may furthermore cause diseases directly or indirectly. This study was undertaken to investigate the effects on the immune response by intoxication of parathion, chloroform or 2, 6-dichlorophenol. Parathion(1.3mg/kg, olive oil 10ml), chloroform (100mg/kg, olive oil 10ml) were administered via intraperitoneal injection to rats. And 2, 6-dichlorophenol (13mg/kg, olive oil 10ml) was administered via oral injection. After 3 weeks, the rats were intoxicated with the above chemicals and immunized with sheep RBC. After 4 weeks the immune response of rat spleen cells was measured by the Jerne's technique. The results were obtained as follows. 1. There was no change of leukocyte counts by the intoxication of parathion, chloroform and 2, 6-dichlorophenol. 2. Parathion, chloroform and 2, 6-dichlorophenol reduced hemoglobin contents for most intoxicated and immunized groups. 3. Hematocrits were decreased by the intoxication of parathion, chloroform or 2, 6-dichlorophenol significantly. 4. It was determined that total protein, A/G (albumin/globulin), .alpha.-, .betha.-and .gamma.-globulins in rat serum were not changed. 5. Intoxication by parathion, chloroform or 2, 6-dichorophenol reduced the number of hemolytic plaque to the sheep RBC in rat spleen cells. Therefore, the capacity of erythrocyte production and the immune response of rat spleen cells were decreased by the intoxication of parathion, chloroform, or 2, 6-dichlorophenol.

Self-Management Knowledge, Self-Management, Physiological Indexes, and Symptoms Experience according to Dialysis Methods (투석방법에 따른 자가관리 지식, 자가관리, 생리적 지표와 증상경험)

  • Purpose: This study was a cross-sectional study comparing differences in self-management knowledge, self-management, physiologic indexes, and symptoms experience for two methods of dialysis. Methods: Participants were 90 patients on hemodialysis and 91 on peritoneal dialysis at A hospital. Results: There was no significant difference between the two groups for knowledge of self-management. A comparison of the categories in the evaluation of self-management showed that patients in the peritoneal dialysis group took better care of their dialysis access route (F=17.61, p<.001) and dialysis schedule (F=4.30, p=.040). The physiologic indexes between the two dialysis groups showed that hemoglobin levels were higher in the hemodialysis group (F=5.28, p=.023). The product of serum calcium and phosphate was higher in the peritoneal dialysis group (F=11.42, p=.001). Serum sodium level was also higher in the peritoneal dialysis group (t=5.36, p<.001) while serum albumin level (t=-3.36, p=.001) and mean arterial blood pressure (t=-2.50, p=.013) were higher in hemodialysis patients. There were no significant differences in the proportion of uncomfortable experiences between the two groups. Conclusion: Medical personnel should consider differences in self-management knowledge/self-management, physiologic indexes, and symptoms experience for hemodialysis and peritoneal dialysis populations, and should provide adequate education accordingly and promote behavioral change to improve physiologic indexes and reduce symptoms.
