Genetic Analysis of Flower Color Traits in Calanthe discolor, C. sieboldii, and Variants Using Molecular Linkage Map (연관지도를 이용한 새우난초, 금새우난초, 변이종의 화색의 유전분석)

  • This study was conducted to clarify the genetic relationship between Calanthe discolor, C. sieboldii and variants, and the cause of flower color variations by using a molecular linkage map and a quantitative trait loci (QTL) analysis for flower and lip color in Calanthe species native to Korea. Twenty plants were included in three C. discolor and three C. sieboldii, and fourteen variants were obtained from their habitat, Jeju-do in Korea. The flowers of C. discolor were brownish red, the values of Commission Internationale de I'Eclairage (CIE) Lab were between 40 and 50. The flowers of C. sieboldii were yellowish, the values of CIE Lab were between 110 and 130. The variants had various mixed colors that were thought to have originated from natural hybridization between C. discolor and C. sieboldii, and the values of CIE Lab were between 50 and 70. The colors of the lips were usually divided into white and yellow. C. discolor had a white lip, C. sieboldii had a yellow one, and the variants had a white to yellow one. The CIE Lab value of each color was 90 in white and 110 to 120 in yellow lips. A molecular linkage mapping was constructed based on the segregation of 154 RAPD markers using a MAPL program. Sixteen linkage groups containing 66 markers were established. It covered a total map distance of 220.4 cM. The distance between adjacent markers ranged from 0 to 6.6 cM, with an average distance of 3.3 cM. These markers are thought to be closely associated with flower and lip color expression. Among the 16 molecular linkage groups, 3 QTLs had flower color trait loci and 1 QTL had lip color trait loci.

Effect of culture Media on Asymbiotic Seed Germination and Those Seedling Growth of Calanthe discolor and Habenaria radiata (새우난초와 해오라비난초 종자의 기내발아와 유묘생장에 미치는 배지의 영향)

  • The experiment was tried to identify culture media which was suitable for seed germination and seedling growth of wild orchids, Calanthe discolor and Habenaria radiata. When seeds of Calanthe discolor, which was treated with ultrasonics for 30 minutes, were sowed in Murashige and SKoog(MS) medium, germination was much more promoted than other treatments. Seedling of C. discolor grew more rapidly in 3g/L Hyponex and 2g/L tryptone(H$_3$T$_2$) medium and 3g/L Hyponex and 2g/L peptone(H$_3$P$_2$) medium, especially in H$_3$P$_4$ medium among those media, pseudobulb became more corpulent. Habenaria radiata, whose tubers were obtained from seedlings, were sprouted more vigorously in 3g/L Hyponex and 1g/L peptone(H$_3$P$_1$) medium and 1g/L Hyponex and 2g/L peptone(H$_1$P$_2$) medium; but multiplication of tubers, growth of daughter tubers and its corpulence were well established in 1g/L Hyponex and 2g/L peptone(H$_1$P$_2$) medium.

Selection of a New Calanthe discolor Lindle. Cultivar 'Narai' for fragrant plant. (방향성 자생새우란 "나래" 선발)

  • These studies were carried out to develop native Calanthe in Korea. Calanthe native to southern islands in Korea has beautiful flowers with various color and sweet fragrance, and it has been reported to have very good ornamental value. According to the morphological characteristics of leaves and flower color, 57 horticultural lines of Calanthe collected in natural crossing group were investigated. And the selected orchid was given a name of horticultural cultivar to ‘Narai’. In its color of flower, sepal was deep purplish red, petal was yellow and lip was white. And also, ‘Narai’ had strong fragrance and shape of blooming was flat-blooming type.

Selection of a New Calanthe discolor Lindle Cultivar 'Saegdong' for color variation by natural population (자생새우란 화색변이주 "색동" 선발)

  • These studies were carried out to develop native Calanthe in Korea. Calanthe native to southern islands in korea has beautiful flowers with various color and sweet fragrance, and it has been reported to have very good ornamental value. Concerning the classified 57 lines had surveyed their characteristics during the three years, and confirmed which that had manifestation stably. And then, a line was developed and given a name of horticultural cultivar to ‘Saegdong’. The major characteristics of the selected line, ‘Saegdong’, were as follows. In its color of flower, sepal was reddish orange, petal was yellow and lip was yellow. ‘Saegdong’ had a bended-inner blooming type.

Soil Acclimatization of Calanthe discolor through Multiple Shoot Formation from Tissue Culture (새우난초(Calanthe discolor)의 조직배양으로부터 다신초형성을 통한 토양순화)

  • This experiment was conducted to establish the micropropagation of Calanthe discolor through multiple shoot formation from the culture of leaf, corm and root explants. Frequency of adventitious shoot formation from leaf explants was higher than those of corms and root explants. Frequency of adventitious shoot formation on medium with various concentrations of BA (0. 1.0, 3.0, and 5.0 mg/L) and NAA (0, 0.1, 0.5, and 1.0 mg/L) was tested. The maximun induction of adventitious shoot was obtained on half strength Murashige and Skoog (MS) medium supplemented with 3.0 mg/L BA and 1.0 mg/L NAA after 6 weeks of culture. Multiple shoots were transferred onto half strength MS medium with various concentrations of GA3 (0, 0.1, 0.5, 1.0, 3.0, and 5.0 mg/L). The number and length of multiple shoots on medium were highest on medium with 3.0 mg/L GA3. All the adventitious shoot grew well and rooted on half strength MS medium with 3.0 mg/L NAA. The plantlets were acclimatized up to 100% on sand with TKS-II or pearlite with TKS-II.

Studies on the Growth Environment of Calanthe Species Native to Cheju Province (제주(濟州) 자생(自生) 새우난초의 생육환경(生育環境))

  • Natural habitats for Calanthe species were generally established in forests of deciduous broad-leaved trees. The habitat areas had lower summer temperatures, more precipitation, and harrow humidity ranging 75~90% throughout the year than adjacent non-habitat areas. The most influential factor on the habitat establishment was light intensity. The light levels in the habitats are cut down by tree canopies, which also reduce temperature before the defoliation in autumn. The most frequent distribution of the habitat was in the areas with light intensity of 400~1,500 lux under tree canopies and of 30,000~50,000 lux in winter after defoliation. Calanthe was characteristically distributed in the broad-leaved forests where high levels of light are reduced in summer, and cold temperatures in winter are insulated by mulched layers of fallen leaves.

Morphological characteristics and genetic diversity of Calanthe species native to Korea (한국 자생 새우난초의 형태적 특성 및 유전적 다양성)

  • This study was conducted to research the morphological characteristics and analyze the genetic diversity by using RAPD in Calanthe species native to Korea. Nine samples were selected by flower color and 19 morphological characteristics. In the length and width of leaf, dorsal sepal, the lateral sepal, the petal, the central lip, and the lateral lip, C. discolor was the shortest and narrowest, respectively, but C. sieboldii was the longest and the widest, respectively. The flower stalk length was the shortest in C. discolor, and the longest in C. sieboldii. Three variants were the intermediate between C. discolor and C. sieboldii in the above morphological characteristics, but spur length was the longest in C. discolor, the shortest in C. sieboldii, and intermediate in the variants. The ovary length in C. discolor was shortest and C. sieboldii and variants were similar with each other. The flower color of C. discolor were brownish red, the value of CIE Lab was between 40 and 50. The flower color of C. sieboldii was yellowish, the value of CIE Lab was between 110 and 130. And variants had various colors between 50 to 70 in the value of CIE Lab. By analyzing multiple band patterns of PCR products, 154 bands were selected as polymorphic RAPD markers. The analysis of genetic similarity of Calanther species using RAPD showed that C. discolor and C. sieboldii are more distant from each other than variants, and these results demonstrated that genetic position of variants located between C. discolor and C. sieboldii.

  • According to the morphological characteristics of leaves, and flowers, including leaf and flower color, 24 horticultural cultivars of Calanthe discolor and C. striata which were collected from Jaeju island of Korea and cultivated at similar environmental conditions in Taegu from 1995 to 1996 were classified newly. Variation ranges were relatively narrow flower shape and leaf. but wide in other characteristics among cultivars.

Micropropagation of Calanthe discolor Lindl. through Induction of Multiple Shoots from Axillary Bud Culture (새우난초(Calanthe discolor Lindl.)의 액아배양으로부터 다신초 형성을 통한 대량증식)

  • This study was conducted to establishment of in vitro micropropagation through induction of multiple shoots from axillary bud culture in Calanthe discolor Lindl. Shoots initiation from axillary bud was the most effective on half strength MS medium supplemented with $0.5mg{\cdot}L^{-1}$ NAA and $2.0mg{\cdot}L^{-1}$ TDZ during four weeks of dark culture followed by culture under a 16-h photoperiod. Multiple shoots (12.5 shoots per explant) were proliferated on half strength MS medium containing $4.0mg{\cdot}L^{-1}$ TDZ and $2.0mg{\cdot}L^{-1}$ IBA. On the other hands, the abnormally emerged shoots during the multiple shoot proliferation stage were recovered to normal shoots on half strength MS hormone free medium. Multiple shoots were well elongated and rooted on half strength MS medium with $0.1mg{\cdot}L^{-1}$ NAA. The plantlets were acclimatized up to 100% on TKS substrate after pretreating with $10mg{\cdot}L^{-1}$ NAA for 30 min. and these plantlets showed good growth as well.
