본 연구에서는 전통주 제조시 부산물로 생산되는 주박과 누룩으로부터 추출물과 유기용매 분획물을 총 85종을 제조하고, 이들에 의한 항염증 활성을 연구하였다. 85종의 분획물 중 선별한 세 가지의 분획물(KSD-E1-3, KSD-E2-3, KSD-E4-3)에 의해서 LPS에 의해 염증이 유도된 RAW 264.7 세포주에서 nitric oxide 생산이 현저히 감소됨을 확인하였다. 또한, 세가지 분획물에 의해 염증유발 유전자인 COX-2, TNF-alpha, 그리고 iNOS 유전자의 발현이 감소되었다. 세 가지 분획물 중 KSD-E4-3에 의한 항염증 활성의 작용기전을 이해하기 위하여 oligo DNA microarray를 수행하였다. 마이크로어레이 결과 발현이 감소된 유전자 중 염증과 관련된 유전자 6개(IL-1F6, iNOS, IL-10, Fabp4, IL-1RN, CSF2)를 선택하여, RT-PCR과 정량적 real-time PCR을 수행하였다. 그 결과, 모든 유전자의 발현이 감소됨을 확인하였다. 결론적으로, 이러한 연구결과는 전통주 주박이 항염증 활성을 가지고 있는 식품이나 약품을 개발하는데 필요한 새로운 자원으로서 활용 가능함을 시사하는 것이다.
염증반응은 유해한 물질이나 병원체에 대항하여 활성화되는 생체 방어 기전이다. 그러나 과도한 염증반응은 그 자체가 생체에 좋지 않은 영향을 미칠 수 있다. 대식세포는 지질다당류와 같은 병원체를 인식한 후, NF-${\kappa}B$ 경로의 활성화를 포함한 다양한 경로를 통하여 산화질소와 같은 염증매개인자들을 분비하는 면역세포이다. 본 연구에서는 지질다당류로 활성화시킨 RAW 264.7 대식세포를 이용하여 등골나물(Eupatorium chinensis var. simplicifolium) 뿌리, 줄기 그리고 꽃 추출물들의 항염증 효과를 알아보았다. 그 중 등골나물 뿌리의 추출물은 농도의존적으로 산화질소의 생성을 감소시켰으며, 산화질소 합성유도효소(inducible nitric oxide synthase)와 고리형 산소화효소-2(cyclooxygenase-2)의 발현을 통계적으로 유의하게 감소시켰다. 또한 등골나물 뿌리의 추출물은 NF-${\kappa}B$ 경로에 있는 MAP (mitogen activated protein) 인산화효소와 단백질 인산화효소 B (protein kinase B)의 활성화를 감소시켰으며, 억제적 kappa B (inhibitory kappa B)의 분해 또한 감소시키는 것을 관찰하였다. 이러한 결과는 등골나물 뿌리의 추출물이 NF-${\kappa}B$ 경로와 산화질소 합성유도효소 발현의 억제를 통하여 항염증작용을 나타낼 수 있음을 제시한다.
파극천은 전통적으로 피부염증 치료에 사용되어온 약초이다. 본 연구에서는 파극천 뿌리를 고온($132^{\circ}C$) 및 고압($1.2kgf/cm^2$)에서 15분 동안 처리한 후 추출물을 분리하여 이들의 MMP-1 효소 억제 효능을 확인하였다. 고온고압 처리된 파극천 추출물의 성분변화를 확인한 결과, 고온고압 처리가 파극천 추출물의 페놀 및 플라보노이드 함량을 약 1.5배 이상 증가시키는 것으로 나타났다. DPPH 및 superoxide 라디칼 소거능을 확인해 본 결과 $500{\mu}g/mL$에서 각각 79.25%, 94.5%의 라디칼 소거 효과가 나타났으며, 기존 추출물 대비 농도 의존적으로 활성 증가를 확인하였다. 또한, 항염 효과를 5-LOX 및 COX-2 저해능을 통해 확인한 결과 고온고압 처리된 파극천 추출물에서 약 45% 증가된 저해능이 나타났다. 이에 자외선에 대한 고온고압 처리된 파극천 추출물의 피부 콜라겐 단백질 분해효소 발현 저해 양상도 농도 의존적으로 발현이 억제되었으며 약 16% 정도 저해 효과가 유의적으로 증가되었다. UVB의 세포보호 효과도 고온고압 처리된 파극천 추출물을 함께 처리하였을 때 세포생존율이 증가됨을 확인할 수 있었다. 결론적으로 파극천 추출물에 고온고압 처리를 통해 기존 추출물보다 뛰어난 항산화, 항염 및 MMP-1 발현 억제 효과의 증가를 확인하였으며, 앞으로 기능성 소재로서 화장품에 응용될 수 있을 것으로 사료된다.
뇌신경 질환의 주요 원인이 되는 것으로 알려진 미세아교세포의 과도한 활성화에 의한 신경염증반응에서 풀무치 에탄올 추출물이 미세아교세포의 염증 반응에 미치는 영향을 검토하였다. 미세아교세포의 활성화를 유도하기 위해 LPS를 사용하였으며, LPS 처리에 의해 신경염증반응의 지표인 NO의 생성량과 이들을 조절하는 iNOS, COX-2의 발현이 증가됨을 확인 할 수 있었다. 그러나 풀무치 에탄올 추출물을 1시간 전처리 한 후 LPS를 처리한 경우 추출물의 농도에 의존적으로 이들의 발현량이 현저히 감소되는 것을 확인 하였다. 또한 LPS 처리로 인해 분비되는 염증성 cytokine들의 생성량도 풀무치 에탄올 추출물에 의해 현저히 억제 됨을 확인 할 수 있었다. 따라서 본 연구의 결과는 미세아교세포의 과도한 활성화로 인해 발생되는 뇌 신경질환의 치료 소재로서 풀무치 에탄올 추출물의 활성 가능성을 제시하였다.
Objectives : Hwanggeumjakyak-tang (huangqin shaoyao tang, HJT) has been used to treat acute enteritis in traditional oriental medicine. However, there has been a lack of studies regarding the effects of HJT on the inflammatory activities and effector inflammatory disease mechanism about macrophage before is not known. So we examined the effect of HJT water extract on pro-inflammatory mediators in lipopolysaccharide (LPS) - stimulated mouse macrophage, RAW 264.7 cells. Methods : Cells were treated with 2 ug/mL of LPS 1 h prior to the addition of HJT. Cell viability was measured by MTS assay. The production of nitric oxide (NO) was determined by reacting cultured medium with Griess reagent. The expression of cyclooxygenase-2 (COX-2), inducible NO synthase (iNOS) and mitogen-activated protein kinases (MAPKs) was investigated by Western blot, RT-PCR. The content of level of cytokines (prostaglandin (PG) $E_2$, interleukin (IL)-6, IL-12, Tumor necrosis factor-alpha (TNF-${\alpha}$) and monocyte chemoattractant protein-1 (MCP-1)) in media from LPS-stimulated Raw 264.7 cells was analyed by ELISA kit. Results : HJT inhibited the production of NO, $PGE_2$, IL-6 as well as the expressions of iNOS, COX-2 but did not inhibit the production of IL-12, TNF-${\alpha}$, MCP-1 in the murine macrophage, RAW 264.7 cells. HJT also had suppression effects of LPS-induced MAPKs activation Conclusion : These results suggest that HJT has an anti-inflammatory therapeutic potential, which may result from inhibition of MAPK phosphorylation, thereby decreasing the expression of pro-inflammatory genes.
Purpose : Although human periodontal ligament stem cells (hPDLSCs) are a supportive factor for tissue engineering, oxidative stress during cell culture and transplantation has been shown to affect stem cell viability and mortality, leading to failed regeneration. The aim of this study was to evaluate the antioxidant and protective effects against cell damage of celecoxib, a selective cyclooxygenase-2 (COX-2) inhibitor, and the antioxidant signal of hPDLSCs in H2O2-induced oxidative stress. Methods : To induce oxidative stress in cultured hPDLSCs, H2O2 was used as an exogenous reactive oxygen species (ROS). Dose-dependent celecoxib (.1, 1, 10, or 100 µM) was administered after H2O2 treatment. WST-1 assay was used to assess cell damage and western blot was used to observe antioxidant activity of hPDLSCs in oxidative stress. Immunohistochemistry was performed for inverting the localization of the SOD and Nrf2 antibody. Results : We found that progressive cell death was induced in hPDLSCs by H2O2 treatment. However, low-dose celecoxib reduced H2O2-induced cellular damage and eventually enhanced the SOD activity and Nrf2 signal of hPDLSCs. Oxidative stress-induced morphological change in hPDLSCs included lowered the survival and number of spindle-shaped cells, and shrinkage and shortening of cell fibers. Notably, celecoxib promoted cell survival function and activated antioxidants such as SOD and Nrf2 by positively regulating the cell survival signal pathway, and also reduced the number of morphological changes in hPDLS. Immunohistochemistry results showed a greater number of SOD- and Nrf2-stained cells in the celecoxib-treated group following oxidative stress. Conclusion : By increasing SOD and Nrf2 expression at the antioxidant system, the findings suggest that celecoxib enhanced the antioxidative ability of hPDLSCs and protected cell viability against H2O2-induced oxidative stress by increasing SOD and Nrf2 expression in the antioxidant system.
Objectives : Citrus is the fruit that is readily available around us. Therefore, we investigated the anti-inflammatory effects of fraction isolated from the Citrus hassaku pericarp in RAW264.7 macrophage cells. Methods : The effects of fraction from Citrus hassaku pericarp on cell viability on RAW264.7 cells were measured by the MTT assay. The mRNA levels of iNOS and COX-2, its protein level by fraction of Citrus hassaku pericarp treatment in RAW264.7 macrophage cells were investigated by RT-PCR and immunoblots. Nitrite accumulation in the culture was measured colorimetrically by the Griess reaction using a Griess reagent. The amount of IL-6 and TNF-${\alpha}$ production was determined using an enzyme-linked immunosorbent assay (ELISA) kit. Results : The results indicated that the fraction of Citrus hassaku pericarp concentration highly suppressed lipopolysaccharide (LPS)-induced nitric oxide (NO) and IL-6 productions without a cytotoxic effect on RAW264.7 cells. fraction of Citrus hassaku pericarp inhibited the expressions of LPS-induced iNOS and COX-2 protein and their mRNA in a dose-dependent manner. Particularly, fraction of Citrus hassaku pericarp suppressed the level of nuclear factor-${\kappa}B$ (NF-${\kappa}B$) activity, which was linked with the suppression of LPS-induced phosphorylation of p65 at serine 276 and p65 translocation into nuclei, but not MAPK signaling. In addition, treatment with fraction of Citrus hassaku pericarp inhibited the production of IL-6 and TNF-${\alpha}$ in LPS-stimulated RAW264.7 cells. Conclusion : Our results indicate that fraction of Citrus hassaku pericarp potentially inhibits the biomarkers related to inflammation through the blocking of NF-${\kappa}B$ p65 activation, and it may be a potential therapeutic candidate for the treatment of inflammatory diseases.
본 연구는 제주산 자생 로즈마리 에센셜 오일의 항염 및 피부상재균에 대한 항균 활성을 확인하기 위한 것이다. 로즈마리 에센셜 오일은 물 증류법으로 추출하였다. 로즈마리 에센셜 오일의 항염 효능을 확인하기 위하여 RAW 264.7 세포에서 LPS에 의해 유도된 NO와 $PGE_2$의 생성을 농도 의존적으로 억제하는 것을 확인하였다. Western blot 실험을 통해 이들을 생합성하는 효소인 iNOS, COX-2 단백질의 발현이 농도 의존적으로 감소하는 것을 확인하였다. 또한 전염증성 cytokine인 TNF-${\alpha}$와 IL-6의 생성 억제 효능을 확인하였다. 항생제 내성균주 각 2종을 포함한 S. epidermidis 3종, P. acnes 3종에 대한 항균 활성을 실험한 결과, paper disc법에서 저해환이 관찰되었고, MIC, MBC 실험을 통하여 균생육 저해와 사멸를 확인하였다. 위의 실험 결과로부터 로즈마리 에센셜 오일의 항염증, 항균 효능을 확인 하였으며 향후 화장품 및 스킨케어 소재로서 활용 가능성을 확인하였다.
Objective : Nelumbo nucifera, its rhizome and semen have been used as a traditional medicine which was studied on antioxidant, hepatoprotective effect, anti-obesity and the others. However, Nelumbinis Flos have not studied. We investigated protective effects on oxidative DNA damage and anti-inflammatory effects of Nelumbinis Flos. Methods : The antioxidant activity was conducted by 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical, 2, 2'-Azino-bis (3-ethylbenzothiazoline-6 sulfonic acid) diammonium salt (ABTS) radical scavenging assay and reducing power assay. Total phenolic content was analyzed. Also, phenolic compounds were detected by HPLC/UV. The inhibitory effect on oxidative DNA damage was determined using ${\Phi}X-174$ RF I plasmid DNA cleavage assay. The anti-inflammatory effect of Nelumbinis Flos was measured by the amount of nitric-oxide (NO) produced and protein levels of iNOS, and COX-2 in LPS induced RAW 264.7 cells. Results : The results of DPPH and ABTS radical scavenging activity at $200{\mu}g/m{\ell}$ of extraction were $97.02{\pm}0.88%$ and $96.42{\pm}0.25%$. Reducing power (fold of L-ascorbic acid as control) was $100.14{\pm}0.31$ at $200{\mu}g/m{\ell}$. Total phenol content was $8.70{\pm}0.02mg/g$. Chlorogenic acid, catechin and epicatechin were found by HPLC. Nelumbinis Flos has inhibitory effect in dose-manner against oxidative DNA damage. In addition, it showed the anti-inflammatory effect by suppression of NO production as well as protein levels of iNOS, and COX-2. Conclusion : This study suggested that Nelumbinis Flos showed potential antioxidant and suppression activities of various factors were related in NO produced. Therefore, Nelumbinis Flos as natural plant resources that may help reduce inflammation and alleviate DNA damage.
들깨(Perilla frutescents (L.) Britton var.) 새싹은 꿀풀과에 속하는 1년생 초본이다. 본 연구의 목적은 들깨 새싹 에탄올 추출물이 사이토카인으로 유도된 췌장 베타 세포 손상에 대한 예방 효과를 평가하기 위함이다. 췌장 소도 주위에 염증 세포 침습으로 의해 분비되는 사이토카인은 1형 당뇨병의 발병원인에 해당된다. 인터루킨-$1{\beta}$ (IL-$1{\beta}$), 인터페론-${\gamma}$ (IFN-${\gamma}$), 종양괴사인자-${\alpha}$ (TNF-${\alpha}$) 등의 사이토카인은 활성산소 형성을 유도한다. 세포 내 활성산소 축적은 췌장 베타 세포 기능장애와 세포사멸을 이끈다. 들깨 새싹 추출물은 항산화 효과를 증가 시켰으며 활성산소 생성을 억제하였다. 사이토카인은 세포생존율을 감소시켰고, iNOS와 COX-2의 발현을 증가시키고 산화질소 생성을 유도하였다. 들깨 새싹 추출물은 사이토카인으로 유도된 세포생존을 농도 의존적으로 예방하였다. 또한, 사이토카인에 의한 산화질소 생성과 iNOS와 COX-2의 단백질 발현 증가를 억제하였다. 더 나아가 들깨 새싹 추출물은 췌장 베타 세포주(RIN-m5F)에서 $I{\kappa}B{\alpha}$ 인산화 억제를 통해서 NF-${\kappa}B$의 활성화를 상당히 감소시켰다. 요약하자면, 본 연구 결과는 들깨 새싹 추출물이 사이토카인으로 유도된 췌장 베타 세포 손상에 대한 보호 효과를 가지고 있다는 것이 확인되었다. 결과적으로 들깨 새싹은 혈당 증가에 의한 산화 스트레스와 염증성 사이토카인에 의한 베타 세포 손상을 완화하여 당뇨에 유익할 것으로 사료된다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
