Comparison of process and cost of disposal methods for brucellosis infected bovine carcasses: burial, recycling, and incineration (브루셀라병 감염소 사체처리 방법별 절차 및 소요비용 비교: 매몰, 재활용, 소각)

  • In Korea, burial is the most common method of disposing animal carcasses culled due to brucellosis infection. However, burial has many disadvantages such as shortage of appropriate burial sites, possibile pollution of ground water supply, and negative view of the public. In this study, we have reviewed 3 legal methods for disposing bovine carcasses, which are burial, incineration, and rendering. We also described the overall process, advantages and disadvantages, and required costs for each method. About 75% of bovine brucellosis outbreak farms had less than 3 reactors, and in our study, rendering required the least amount of cost for farms with a small number of reactors (1-3 heads). Our findings suggest that the use of rendering should be encouraged for farms with bovine brucellosis and other methods considered only if rendering is inappropriate.

  • In the present study, the incidence of bovine tubeculosis (bTB) and brucellosis in Hanwoo (Korean native cattle) and dairy cow in Gyeongsangnam-do was investigated for three years from 2020 to 2022. The incidence bTB tested by tuberculin skin test with purified protein derivative (PPD) and gamma interferon (γ-INF) test with a commercial enzyme-linked immunosorbent assay. From 2020 to 2022, the incidence of bTB showed a decreasing trend in Hanwoo, while an increasing trend in dairy cow. In the case of Brucellosis, the positive rates for Hanwooe gradually increased. However, no brucellosis was found in dairy cow from 2020 to 2022. The increase in the incidence of these diseases is presumed to be related the small scale and poor sanitation facilities of livestock farms in Gyeongsangnam-do, and easy access of wild animals. Therefore, in order to suppress the incidence of the diseases, it is necessary to the farm scale from small to large and to strengthen sanitary facilities on farms.

The latent period and anti-epidemic measure of bovine brucellosis (소 부루세라병 잠복기간과 방역대책;-제주도 소 부루세라병 근절대책을 중심으로-)

  • The 37,000 holstein which had been raised in Jeju island were examined for the purpose of brucellosis detection and so on. We investigated the relationship between the farms showing positive outbreak and the number of examination, incubation period, and the outbreak rate of positive cattle. We endeavored to separate germs from brucellosis positive cattles blood and negative bulls semen against brucellosis serum test and concluded as follows. We intend to offer you institutional supplements and preventions of brucellosis for the future. 1. Outbreak of brucellosis could not lead the outbreak rate of positive cattle to decrease through 1-2 examination per year, For the past 4 years, the outbreak rate of brucellosis positive cattle had a tendency to show a gradual decrease through at least 5 examinations per year. 2. As we examined live-in cattle with positive cattle in mass outbreak farms for 9 months, we found out positive cattle every month. In a grazing land, the group of cattle producd enormous positive cattle in 9-12 months. 3. Annual brucellosis positive outbreak rate was 20-25% among live-in cattle with brucellosis positive cattle, but the rate might be 50-100% depending on extent of pollution. 4. 94's brucellosis examination showed that 200 positive cattle of 71,153 cattle and most of them were live-in cattle with confirmed brucellosis positive cattle. 5. 1 head of 200 positive cattle showed positivity against serum of bulls test and the rate of bulls was about 1%. 6. Brucellosis germ was separated from 2 cattle's semen among 52 negative bulls against brucellosis serum test and there was the one brucellosis positive cattle against tube agglutination of semen test, so the positive rate of brucellosis was 5.8%. 7. Brucellosis germ was not separated from 15 brucellosis positive cattle's blood.

A serological survey on large outbreak of bovine brucellosis in dairy farm (부루셀라병 다두이환 목장의 혈청항체가 조사)

  • This survey was conducted for the serological confirmation on large outbreak of bovine brucellosis in two dairy farms. Serological tests were performed by the plate agglutination test, tube agglutination test, enzyme linked immunosorbent assay(ELISA), complement fixation, ,test(CFT) and rose bengal plate test(RBPT). Total 200 heads(134 heads in farm A and 66 heads in farm B) were tested. The primigravida and positive group have been raised separately in the farm A and both group have been raised together in the farm B.. The result were summarized as follows ; 1. Positive ratios in positive herds of farms by the tube agglutination test were 68.3% in farm A and 53.2% in farm B. 2. Seroconversion to brucella was observed in the primigravidas group in farm B, but was not observed in the primigravidas group in farm A. 3. All calves born in positive herd were serologically negative at time of test. 4. Positive ratio of ELISA in farm A was higher than that of tube agglutination test. 5. Number of positive reactors by the CFT, RBPT in farm A were equal to those of tube agglutination test.

Seroprevalence of Brucellosis among Risk Population in Gyeongsangbuk-do, 2006 (경상북도 거주 고위험군에서 브루셀라 감염에 관한 혈청 역학적 연구)

  • Objectives : Cases of human brucellosis in Korea have recently increased due to the increasing incidence of bovine brucellosis. The authors conducted this study to elucidate the status of brucellosis through seroepidemiologic study. Methods : We selected our study population from a high risk group. We conducted a questionnaire survey and obtained blood samples to determine the seroprevalence of brucellosis antibodies for 10 days in February, 2005. The titers of brucellosis were measured by the combination of standard tube agglutination test (STA) and enzyme-linked immunosorbent assay (ELISA) test. Results : Our study subjects comprised 1,075 cases: 971 livestock workers, 51 veterinarians, and 53 artificial inseminators. In the STA test, 27 cases (2.5%) had titers of greater than or equal to 1:20. Of 1,068 cases (7 cases were excluded due to previous brucellosis), 7 cases of brucellosis were diagnosed with titers of 1:160, giving a seroprevalence of brucellosis of 0.66%. The seroprevalence in the male group was 0.95%, and that of livestock workers, veterinarians, and artificial inseminators was 0.52%, 4.17%, and 0.00%, respectively. The Spearman's correlation coefficient between the positive rate of bovine brucellosis per capita and household and human brucellosis was 0.806 and 0.744, respectively. The concordance rate between the Korea National Institute of Health and the Gyeongsangbuk-do Institute of Health and Environment by the STA and ELISA tests was 94.7% and 100.0%, respectively. Conclusions : The study results indicated in higher seroprevalence rate among veterinarians than among livestock workers and artificial inseminators. Because veterinarians may be exposed to this high risk, effective working guidelines for veterinarians to guard against brucellosis must be developed. Moreover, more extensive epidemiologic research for laboratory workers and meat handlers is needed.

Investigation of occurrence factors on brucellosis-outbreak farm in Korea (소 브루셀라병 동시 다두 발생 농장의 발생 요인 조사)

  • From the end of July 2012, several cases of abortion have been happened at the Korean indigenous cattle farm with 124 heads in Chungbuk province, Korea. Serological tests such as Rose-bengal test (RBT) and standard tube agglutination test (STAT) have been performed according to the standard official protocols of bovine brucellosis and 41 cattle turned out to be brucellosis-positive simultaneously. To find out the main factors of brucellosis outbreaks and spreads, additional serological, etiological and molecular investigation were applied. Totally, 11 B. abortus were isolated from 10 cattle's specimens including lymph-nodes and/or testis, and drinking water in cowhouse. In genotyping by multi-locus VNTR assay (MLVA) using 17 loci markers, the present B. abortus isolates were shown all the same pattern, D1 genotype, which has been reported in Gyeonggi and Gangwon province, Korea. These results suggest that the input of brucellosis might come from neighboring farms directly or indirectly, even if by unknown factor and expansion within farm would accelerate by materials related with aborting cows.

Characterization of Bovine Brucellosis in Korean Native Cattle by Means of Immunohistochemistry and Proteomics (면역조직 화학법 및 단백질체 변화 분석을 통한 한우에서 발생한 브루셀라증의 특성)

  • This study was conducted to examine the utilization of immunohistochemistry using the bovine anti-brucella immunoglobulin G (IgG) antibody in the diagnosis of brucellosis and to develop a functional biomarker relation for the progress of the disease. Anti-brucella IgG antibody was purified from the affected bovine serum using an affinity chromatography. We performed our investigation on 17 cases of brucellosis and 19 control cases with negative Rose-Bengal test results. Our purified anti-brucella IgG antibody showed a positive immunoreactivity in cytoplasmic hepatocytes of the centrilobular region, and glomeruli and tubular epithelium of the kidney. The protein pattern of the affected liver versus control was analyzed by two-dimensional electrophoresis, showing a different expression pattern of proteins between the two. Five protein spots were up-regulated and another were five down-regulated in the brucellosis liver. Significant upregulaton of catalase and 3-hydroxyacyl-CoA dehydrogenase might be due to a compensatory reaction in response to the endotoxic shock of brucella. In conclusion, the anti-brucella IgG antibody may be a good tool for discriminative diagnosis of the affected tissues and proteomics data suggest new target proteins underlying a possible pathogenic mechanism of brucellosis.

Extraction and verification of highly immunologenic antigen for diagnosis of bovine brucellosis (소 브루셀라병 진단용 고면역원성 항원의 추출과 검증)

  • Bovine brucellosis, an important zoonosis, is diagnosed with serological tests such as the RBT, TAT using inactivated whole bacterial cells or bacterial lipopolysaccharide (LPS) antigen in Korea. However, a strong cross-reaction between Brucella spp. and Yersinia enterocolitica O9 in these tests has seriously complicated the diagnosis of animal brucellosis because Brucella spp. shares common antigenic determinants with Y. enterocolitica O9 in the smooth LPS region. In this study, Brucella-field strains were isolated from Brucellapositive Hanwoo in Kimhae, Korea and outer membrane protein (omp) which has low cross-reaction with Y. enterocolitica O9 and high immunogenicity was extracted from the field strains Then we compared ELISA using the extract with RBT-TAT. Fifteen field strains were isolated from 47 supra-mammary-lymph nodes, which were collected from 18 farms. Isolation rate was 32%. Brucella-specific antigen was identified by performing SDS-PAGE or Western blotting on extracted omp with at 0.5% n-lauroylsarcosine One hundred and ninety-two serum-samples were used in the experiment: 142 negative and 50 positive samples verified by RBT-TAT. According to ELISA results, 127 samples were negative and 15 appeared positive among 142 negatives by RBT-TAT, while 42 samples were positive and 8 were negative among 50 positives by RBT-TAT. Therefore, it showed 89.4% of specificity and 84% of sensi-tivity. Through the current experiments, we could set up an ELISA based on the omp which has low cross-reaction and high immunogenicity and concluded that the omp could be a good material for accurate diagnosis of bovine brucellosis.

Genotyping of Brucella abortus isolated from cattle in Gyeongbuk province by MLVA (MLVA를 이용한 경북지역 소에서 분리된 Brucella abortus의 유전형별)

  • Brucella (B.) abortus is a facultative intracellular pathogen that infects a wide variety of animal species and human. Brucellosis is the zoonosis and an extremely important disease around the world. Although the eight species can be differentiated by conventional phenotypic tests, these species display a high degree of DNA homology in DNA-DNA hybridization assay (>90%). Various methods have been established for genotyping of Brucella species, but most of analytical methods are lack reproducibility and limited capability to differentiate them. The attempt of this study was to evaluate multiple-locus VNTR analysis (MLVA) for use of epidemiological trace-back analysis in bovine brucellosis. Ninety-four B. abortus isolates from Gyeongbuk province during 2006~2010 were analyzed using 16 VNTR locus. High resolution automatic capillary electrophoresis system was used for more throughput, simpleer, faster, and better discriminable than conventional gel electrophoresis. As a result, 13 different genotypes were identified from 94 B. abortus isolates. MLVA could contribute to epidemiological trace-back analysis of bovine brucellosis.

Prevalence of bovine tuberculosis, brucellosis and Q fever in Korean black goats (국내 흑염소에서의 소결핵, 브루셀라, Q fever 유병률 연구)

  • As the meat of black goats has become popular as a healthy food, domestic goat meat-related industries are steadily growing. However, previous studies are scarce of informations about the zoonotic disease originated from the black goat in Korea. In this study, we investigated Korean black goat's infectious diseases representing bovine tuberculosis, brucellosis, and Q fever. One hundred and eighty samples were collected from a local slaughter house located in Jeollanam-do. Three typical zoonotic diseases were separately examined by carrying out enzyme linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR). Histopathological test was additionally performed in tuberculosis. In case of tuberculosis, results of the PCR and histopathological test were negative but the ELISA results were positive in eight samples. In case of brucellosis, one out of the total samples was shown to be positive in the ELISA and none in the PCR. In case of Q fever, there were forty one positive in the ELISA and twenty positive in the real-time PCR. Those results indicate that the Korean black goat could be a natural reservoir in the possible chain-infections among human, cows and goats. Thus, further study needs in order to improve productivity as well as to prevent the zoonosis spreading and circulation of other livestock with the black goat in this country.
