국내 인공수정센터의 웅돈에 대한 번식 관련 바이러스 조사 (A survey of viruses associated with reproductive failure in boar semen in Korean artificial insemination centers)

  • Artificial insemination (AI) of swine is a very useful reproductive tool and that offers convenience in the Korean swine industry. Since many viruses have been reported to be excreted through boar semen, we investigated the presence of antibodies and antigens against viruses causing reproductive failure in semen of boar in 349 semen samples collected from six Korean AI centers. Viral antigens were detected by polymerase chain reaction (PCR) or reverse transcription-PCR predominantly. The results was as follows. The major reproductive failure causing factor was porcine circovirus type 2 (PCV2), followed by porcine reproductive and respiratory syndrome virus (PRRSV) ($X^2$=166.64, P<0.001). PCV2 and PRRSV, Japanese encephalitis virus (JEV), encephalomyocarditis virus (EMCV) was detected in 73 samples (20.9%), 44 samples (12.6%), 4 samples (1.1%), 3 samples (0.9%), respectively and porcine parvovirus in one sample (0.3%) Classical swine fever virus (CSFV), bovine viral diarrhea virus and Aujeszky's disease virus (ADV) were not detected. Enzyme-linked immunosorbent assay was carried out in 111 serum samples from three AI centers. In most pigs, antibodies response was showed prominently in CSFV (105 sera, 94.6%) ($X^2$=82.580, P<0.001), followed by, in PRRSV (100 sera, 90.1%), PCV2 (92 sera, 90.1%), and PPV (8 sera, 82.9%). ADV antibody was not detected. Thus, the experimental results will be used for the base data, with respect to the state of viral stillbirth in general pig farms, as well as AI centers and breeding farms in Korea.

돼지에서 동결정액을 이용한 인공수정시 종모돈의 품종, 인공수정 횟수, 정자농도, 농장 및 연도가 번식성적에 미치는 영향 (Effects of Breed, Insemination Time, Sperm Concentration, Farm and Year on Reproductive Performance of Sows Inseminated by Frozen Boar Semen)

  • 돼지에서 동결정액을 이용한 인공수정이 번식 성적에 미치는 영향을 구명하고자 축산기술연구소 종축개량부 (충남, 성환)의 돼지 인공수정센터에서 사육중인 종모돈을 이용하여 1995년부터 2000년까지 인공수정을 실시하였다. 동결정액을 이용한 인공수정시 종모돈의 품종, 인공수정 횟수, 정자농도, 농장 및 연도가 번식성적에 미치는 영향을 조사하였다. 동결정액이 인공수정에 영향을 미치는 요인을 알아보기 위하여 5$m\ell$ maxi-straw에 포장된 동결정액을 이용한 인공수정시 번식성적을 조사한 결과 종모돈의 품종 (Landrace, Yorkshire, Duroc)간 차이가 없었다. 1회 발정당 수정횟수는 2회 수정과 3회 수정시 차이가 없었으며, 1회 주입정자 농도도 5.0, 4.0 및 3.0$\times$$10^{9}$ /dose 처리간에 차이가 없었다. 그러나 양돈장의 관리방법과 인공수정사의 기술은 수태율과 산자수에 영향을 주었다. 1995년부터 1999년까지 5년간 농장에서 매년 인공수정한 결과수태율은 68.3~74.6%, 분만율은 61.7~67.6%, 총산자수는 8.7~9.6두 그리고 생존산자수는 8.1~8.7두로 각각 나타났다.

임신전 불임 수퇘지 종부 및 무정자정액 주입에 의한 미경산돈의 산자수 증가에 관한 연구 (Studies on Increase of Litter Size by Infusion with Sterile Boar and Their Semen before Gestation in Gilts)

  • 본 연구는 초임전 임신항원인자로 정소상체제거 및 정관절제 수퇘지를 종부하거나 무정자 정액을 사용하였을 때 미경산돈의 번식향상 효과를 조사하기 위하여 수행하였다. 임신항원인자의 주입에 따른 번식성적을 조사하기 위하여 7∼8개월령의 교잡종 미경산돈을 160두 공시하였다. 시험돈은 4개의 시험군으로 나누었으며, 처리는 A처리구는 정소상체제거 교배, B처리구는 무정자 정액, C 처리구는 정관절제 교배, D처리구는 무처리하여 각구당 40두를 배 치 하여 완전임의배치법으로 하였다. 미경산돈의 초임시 총산자수는 A, B, C 및 D 처리구가 각각 10.05, 10.44, 11.63 및 9.97두로 가장 많은 처리구는 정관절제교배구인 C처리구였다(P<0.05). 비경산돈의 생존산자수는 총산자수와 유사하였다. 특히 C처리구는 10.70두로 9.12 및 9.11 두인 A군과 무처리보다 많았다(P<0.05). 반면 정관절제군과 무정자 처리구간에는 총산자수 및 생존산자수에서 차이가 없었다. 한편 미경돈의 혈중 progesterone 농도는 교배후 6일에서 C처리구가 다른 처리구보다 유의성 있게 높았다. 그러나 8일 이후에는 다른 처 리구보나 낮았다. 한편 혈중 cortisol농도는 처리간에 통계적인 유의성이 없었다. 이상의 시험결과를 볼 때 임신전 정관절제 수퇘지나 무정자정액을 항원으로 주입으로 미경산돈의 산자수 증가에 의한 번식효율을 증진을 확인할 수 있었다. 미경산돈의 산자수 증가를 위한 항원으로 정관절제 수퇘지나 무정자정액을 자궁에 주입하는 방법이 효과가 있다고 할 수 있다.

Effect of L-carnitine on sperm quality during liquid storage of boar semen

  • Objective: This study was conducted to investigate the effect of L-carnitine on the pig semen characteristics during storage. Methods: Spermatozoa samples were examined for spermatozoa quality and then randomly divided into 5 groups: 0 (control), 12.5, 25, 50, and 100 mM L-carnitine. Sperm motility, plasma membrane integrity and antioxidant parameters (total reactive oxygen species, total antioxidant capacity, and malondialdehyde) were evaluated after 0, 3, 5, and 10 day cooled-storage at 17℃. Moreover, ATP content, mitochondria activity as well as sperm-binding and in vitro fertilizing ability of preserved boar sperm were also investigated. Results: Supplementation with 50 mM L-carnitine could effectively maintain boar sperm quality parameters such as sperm motility and membrane integrity. Besides, we found that L-carnitine had positive effects on boar sperm quality mainly through improving antioxidant capacities and enhancing ATP content and mitochondria activity. Interestingly, by assessing the effect of L-carnitine on sperm fertility and developmental potential, we discovered that the extender containing L-carnitine could improve sperm quality and increase the number of sperms bounding to zona pellucida, without improving in vitro fertility and development potential. Conclusion: These findings suggested that the proper addition of L-carnitine to the semen extender improved boar sperm quality during liquid storage at 17℃.

Effects of Storage in Different Commercial Semen Extenders on Sperm Motility, Viability and Membrane Integrity of Korean Native Boar Spermatozoa

  • The objective of this study was to compare the effect of semen extenders on the sperm motility, viability, acrosome integrity and functional integrity of plasma membrane (HOST: hypo-osmotic swelling test) during liquid preservation of Korean Native boar semen. In this experiment, semen was diluted in Androhep plus, Beltsville Thawing Solution (BTS), ModenaTM, Seminark and Vitasem LD. Sperm-rich fractions were collected from three Korean Native boars and sub-samples were diluted ($30{\times}10^6$ spermatozoa/ml) in different semen extenders. Semen samples were stored at $17^{\circ}C$ for 96 hours. On everyday (0, 24, 48, 72, 96 h) after storage, the sperm characteristics relevant for fertility, such as sperm motility, viability, acrosome integrity and HOST positive were evaluated. The motility of spermatozoa stored in different extenders was no significantly different among other extenders (P>0.05). Also, no difference was observed among samples processed with different extenders in the percentage of sperm viability, acrosome integrity and HOST positive. All extenders maintained a high percentage (70%) of sperm motility, viability and acrosome integrity through 96 h of storage. The result of this study show that there was no significant differences among extenders in their capacity to preserve motility, viability and membrane integrity of spermatozoa from normal, fertile Korean Native boars for 96 h of liquid preservation at $17^{\circ}C$.

Washing solution and centrifugation affect kinematics of cryopreserved boar semen

  • Cryopreservation is a widely-used efficient means of long-term sperm preservation. However, unlike other types of semen, cryopreserved boar semen has reduced fertility and the efforts continue to optimize post-thawing sperm recovery. In this study, we evaluated the effects of various washing solutions (Hulsen solution, lab-made DPBS and commercial DPBS) on post-thawing porcine sperm kinematics (CASA system), viability (SYBR-14/PI) and acrosome integrity (PSA/FITC). We also examined the effect of washing-centrifugation on frozen-thawed semen kinematics. The results indicate that type of washing solution and post-thawing centrifugation alters parameters linked to sperm quality (total motility, progressive motility, viability and acrosome integrity). Significantly higher (p < 0.05) motility and progressive motility were obtained when cryopreserved semen was processed with Hulsen solution. The post-thaw percentage of live and intact acrosomal sperm was significantly higher in group 1 (Hulsen solution) as compared to other groups. Following thawing-centrifugation, the results showed significantly higher motility and progressive motility in group 1 than other groups. However, the latter two DPBS groups did not differ statistically. Taken together, Frozen-thawed spermatozoa motility, acrosome integrity and viability can be affected by the type of washing solution used. Moreover, centrifugation of frozen-thawed semen has an unfavorable effect on total motility and progressive motility.

돼지 난모세포의 체외성숙 및 체외수정시 배양액과 액상정액의 효과 (Effect of Matruation Media and Liquid Boar Semen on Maturation and Fertilization of Pig Oocytes In Vitro)

  • This present study was carried out to examine the effect of maturation media and liquid boar semen on in vitro maturation and feritilization of pig oocytes. The results obtained were as follows : When the oocytes were cultured for 36∼42 hours in mTCM-199, Waymouth MB 725/1 and mTLP-PVA medium, the maturation rates were 90%, 92% and 88%, respectively. The sperm penetration rates of pig oocyte matured in vitro were 87%(mTCM-199), 90%(Waymouth MB 725/1) and 86%(mTLP-PVA), respectively. The rates of nuclear maturation and fertilization of pig oocytes among three different media did not differ. However, the rate of male pronucleus formation of pig oocytes was significantly higher in pig oocytes matured in Waymouth MB 725/1(91%) than oocytes matured in mTCM-199(66%) and mTLP-PVA(62%) medium (P<0.05). When the collected sperm-rich fraction without diluent was used fro in vitro fertilization in mTCM-199 fertilization medium, the fertilization rate was 87.9%. However, when the liquid boar semen diluted with B tschwiler diluent was used at day 3 and 5 after dilution, the fertilization rate was 40.8% and 0.0%, respectively.

돼지 액상정액의 정자농도가 번식성적에 미치는 영향 (Effect on Fertilizing Capacity According to Sperm Concentration of Liquid Boar Semen)

  • 본 연구는 돼지 액상정액의 주입농도가 번식성적에 미치는 영향을 구명하기 위하여 실시하였다. 1회 주입 정자수를 80$m\ell$ 병당 3.0$\times$$10^{9}$ , 2.5$\times$$10^{9}$ 2.0$\times$$10^{9}$$1.5\times$$10^{9}$ 으로 구분하여 인공 수정한 결과 주입 정자 농도가 $1.5\times$$10^{9}$ 일 경우 분만율 82.2%, 총산자수 10.9두로 나타났으며 처리별 농도간에 통계적인 유의차가 없었다. 또한 1회 적정 주입 정자농도를 추정한 결과 2.0~2.3$\times$$10^{9}$ 으로 나타났다.

Effect of Different Inoculation Concentration of Escherichia coli on Boar Sperm Quality and Reproductive Performance in Sow

  • The objective of this study was to determine the effects of E. coli on boar sperm quality and reproductive performance in sows after artificial insemination. Three different levels of E. coli were artificially inoculated to semen with following concentrations; Control, 500, 5,000 and 50,000 colony forming unit (cfu)/ml. Semen samples were preserved at $17^{\circ}C$ for 5 days. Sperm motility was significantly decreased (p<0.05) on day 3 in the group inoculated with 5,000 cfu/ml compared to control groups. In all treatment groups, sperm motility was gradually decreased as storage time increased, but the decline pattern was more drastic in the groups inoculated with 5,000 and 50,000 cfu/ml groups from day 3 (p<0.05) compared to control group. After 3 day of storage at $17^{\circ}C$, sperm viability in sample inoculated with the highest concentration (50,000 cfu/ml) of bacteria was less (p<0.05) than that of control group. The pH of semen sample pH was maintained 7.2~7.5 in all groups during the experimental period. No differences (p>0.05) were found for both storage time and bacterial concentration. The pregnancy rate and live born piglets tend to decrease by increasing the concentration of E. coli in semen. In particular, the rate of pregnancy was lower in the group inoculated with 50,000 cfu/ml (58.3%) compare to the other groups (81.8, 75.0, 76.5%). These results suggest that the contamination of E. coli in boar semen negatively affects fertilizing ability of boar sperm and the reproductive performance obtained from sows after artificial insemination.
