  • 목재 섬유 기반의 셀룰로오스 나노피브릴(cellulose nanofibrils, CNF)은 생체적합특성이 우수하여 조직 공학용 스캐폴드, 약물 운반체, 상처 치유용 겔 등의 생체 의료 분야에서 많은 관심을 받고 있다. 하지만, 셀룰로오스 나노피브릴은 상대적으로 약한 기계적 강도를 나타내기 때문에 높은 기계적 특성을 요구하는 응용 분야에 사용되기 어렵다는 한계를 가지고 있다. 따라서 본 연구에서는 셀룰로오스 나노피브릴의 기계적 강도를 향상시키기 위해 TEMPO (2,2,6,6-tetramethylpiperidin-oxyl) 산화 처리된 셀룰로오스 나노피브릴에 금속 양이온을 도입하여 금속-카르복실레이트 배위 결합을 가지는 하이드로겔을 제조하였다. 또한, 큰 진폭 진동 전단(large amplitude oscillatory shear) 측정과 Live/Dead 세포 시험을 통해 하이드로겔의 비선형 점탄성 거동과 세포 생존 능력을 분석하였다. 특히, 첨가된 금속염의 종류에 따라 세포의 증식 및 생존 능력이 변화하였고, 이는 하이드로겔들의 유변 물성 특성에도 영향을 미쳤다.

미세가공 기술 기반의 마이크로니들 어레이 개발 및 패럴린 적용 가능성에 관한 연구 (Research on the Development of Microneedle Arrays Based on Micromachining Technology and the Applicability of Parylene-C)

  • In this research, we studied the development of a SUS304 microneedle array based on microfabrication technology and the applicability of Parylene-C thin film, a medical polymer material. First of all, four materials commonly used in the field of medical engineering (SUS304, Ti, PMMA, and PEEK) were selected and a 5 ㎛ Parylene-C thin film was deposited. The applicability of Parylene-C coating to each material was confirmed through SEM analysis, contact angle measurement, surface roughness(Ra) measurement, and adhesion test according to ASTM standards for each specimen. Parylene-C thin film was deposited based on chemical vapor deposition (CVD), and a 5 ㎛ Parylene-C deposition process was established through trial and error. Through characteristic experiments to confirm the applicability of Parylene-C, SUS304 material, which is the easiest to apply Parylene-C coating without pretreatment was selected to develop a microneedle array based on CNC micromachining technology. The CNC micromachining process was divided into a total of 5 steps, and a microneedle array consisting of 19 needles with an inner diameter of 200 ㎛, an outer diameter of 400 ㎛, and a height of 1.4 mm was designed and manufactured. Finally, a 5 ㎛ Parylene-C coated microneedle array was developed, which presented future research directions in the field of microneedle-based drug delivery systems.

Comparative in vivo biodistributions of nanoparticles and polymers of 177lutetium-labeled hyaluronic acids in mice during 28 days

  • Hyaluronic acid (HA) has been investigated for biomedical and pharmaceutical applications. This study was conducted to determine the distributions of HA nanoparticles (NPs; size 350-400 nm) and larger HA polymers in mice at intervals after application. $^{177}Lutetium$ (Lu)-labeled HA-NPs or HA polymers were intravenously injected (5 mg/kg) into male ICR mice, and radioactivity levels in blood and target organs were measured from 0.25 h to 28 days post-injection. In blood, the radioactivities of HA-NPs and HA polymer peaked at 0.5 h after injection but were remarkably decreased at 2 h; subsequently, they maintained a constant level until 6 days post-injection. HA-NPs and HA polymers were observed in the liver, spleen, lung, kidney, and heart (in ascending order) but were seldom observed in other organs. After 3 days, both the HA-NP and HA polymer levels showed similar steady decreases in lung, kidney, and heart. However, in liver and spleen, the HA-NP levels tended to decrease gradually after 1 day and both were very low after 14 days, whereas the HA polymer level accumulated for 28 days. The results indicate that HA-NPs, with their faster clearance pattern, may act as a better drug delivery system than HA polymers, especially in the liver and spleen.

  • For centuries, Panax ginseng Meyer (Korean ginseng) has been widely used as a medicinal herb in Korea, China, and Japan. Ginsenosides are a class of triterpene saponins and recognized as the bioactive components in Korean ginseng. Ginsenosides, which can be classified broadly as protopanaxadiols (PPD), protopanaxatriols (PPT), and oleanolic acids, have been shown to flaunt a vast array of pharmacological activities such as immune-modulatory, anti-inflammatory, anti-tumor, anti-diabetic, and antioxidant effects. In recent years, a number of ginseng and ginsenoside researches have increasingly gained wide attention owing to its unique pharmacological properties. Although good efficacies of ginsenosides have been reported, lack of target specific delivery into tumor sites, low solubility, and low bioavailability due to modifications in gastro-intestinal environments limit their biomedical application in clinical trials. As a result to this major challenge, nanotechnology and drug delivery techniques play a significant role to solve this problematic issue. Thus, we reported the preparation of poly-ethylene glycol (PEG) and glycol chitosan (GC) functionalized to ginsenoside (Compound K and PPD) conjugates via hydrolysable ester bonds with improved aqueous solubility and pH-dependent drug release. In vitro cytotoxicity assays revealed that PEG-CK, and PPD-CK conjugates exhibited lower cytotoxicity compared to bare CK and PPD in HT29 cells. However, GC-CK conjugates exhibited higher and similar cytotoxicity in HT29 and HepG2 cells. Furthermore, GC-CK-treated RAW264.7 cells did not exhibit significant cell death at higher concentration of treatment which supports the biocompatibility of the polymer conjugates. They also inhibited nitric oxide production in lipopolysaccharide (LPS)-induced RAW64.7 cells. In addition to polymer-ginsenoside conjugates, silver (AgNps) and gold nanoparticles (AuNps) have been successfully synthesized by green chemistry using different m. The biosynthesized nanoparticles demonstrated antimicrobial efficacy, anticancer, anti-inflammatory, antioxidant activity, biofilm inhibition, and anticoagulant effect. Special interest on the effective delivery methods of ginsenoside to treatment sites is the focus of metal nanoparticle research.In short, nano-sizing of ginsenoside results in an increased water solubility and bioavailability. The use of nano-sized ginsenoside and P. ginseng mediated metallic nanoparticles is expected to be effective on medical platform against various diseases in the future.

설포베타인 키토산의 실크 블렌드 필름의 제조 및 그들의 성질 (Preparation of Sulfobetaine Chitosan, Silk Blended Films, and Their Properties)

  • Bombyx mori silk fibroin(SF)과 블렌드 필름을 만들기 위하여 키토산에 1,3-propanesultone을 반응시켜 수용성 sulfobetaine chitosan(SCs)을 제조하였다. 여러 가지 비율의 SF/SCs 블렌드 필름을 B. mori SF와 SCs의 수용액을 혼합하여 제조하였다. 수용액으로부터 얻어진 SF/SCs 블렌드 필름의 구조와 형태 변화는 분광학적 및 열적 분석을 통해 규명하였다. SF와 SCs의 혼합 비율에 따른 인공 피부나 화상치료 목적의 비이오재료로서의 물리적 및 기계적 성질에 미치는 영향을 조사하였다. X-선 분석으로 두 생체고분자 사이에 좋은 친화성을 보여주고 있음을 알 수 있었으며 기계적 성질도 SCs의 함량이 증가하면 크게 증가하였다. $37^{\circ}C$에서 phosphate buffered saline solution 용액 중에서 in vitro 분해 실험을 8주 동안 시행한 결과 46.4%가 분해됨을 알 수 있었다. MC3T3-E1 세포에 의한 독성 실험 결과 무독성을 나타내 주었으며, 3일의 배양 후 SF/SCs 필름의 상대 세포 수는 최적화된 tissue culture plastic보다 약간 낮게 나타남을 알 수 있었다.

Highly Stretchable and Sensitive Strain Sensors Fabricated by Coating Nylon Textile with Single Walled Carbon Nanotubes

  • Stretchable strain sensors are becoming essential in diverse future applications, such as human motion detection, soft robotics, and various biomedical devices. One of the well-known approaches for fabricating stretchable strain sensors is to embed conductive nanomaterials such as metal nanowires/nanoparticles, graphene, conducting polymer and carbon nanotubes (CNTs) within an elastomeric substrate. Among various conducting nanomaterials, CNTs have been considered as important and promising candidate materials for stretchable strain sensors owing to their high electrical conductivity and excellent mechanical properties. In the past decades, CNT-based strain sensors with high stretchability or sensitivity have been developed. However, CNT-based strain sensors which show both high stretchability and sensitivity have not been reported. Herein, highly stretchable and sensitive strain sensors were fabricated by integrating single-walled carbon nanotubes (SWNTs) and nylon textiles via vacuum-assisted spray-layer-by-layer process. Our strain sensors had high sensitivity with 100 % tensile strain (gauge factor ~ 100). Cyclic tests confirmed that our strain sensors showed very robust and reliable characteristic. Moreover, our SWNTs-based strain sensors were easily and successfully integrated on human finger and knee to detect bending and walking motion. Our approach presented here might be route to preparing highly stretchable and sensitive strain sensors with providing new opportunity to realize practical wearable devices.

  • Silk fibroin (SF) is one of the typical protein polymer produced by silkworm, Bombyx mori. SF has been used as textile fiber and surgical suture fur thousands of years due to its unique gloss, handle, and mechanical properties. Recently, SF has been intensively studied to diverse usage for biotechlological and biomedical fields because of their reproducibility, environmental compatibility, non-toxicity, and biological compatibility. Based on its biocompatibility, the possible uses of regenerated SF have been proposed including substrate for cell culture[1], enzyme immobilization[2], and matrix for drug release[3]. (omitted)

Preparation of a Porous Chitosan/Fibroin-Hydroxyapatite Composite Matrix for Tissue Engineering

  • Chitosan, fibroin, and hydroxyapatite are natural biopolymers and bioceramics that are biocompatible, biodegradable, and resorb able for biomedical applications. The highly porous, chitosan-based, bioceramic hybrid composite, chitosanlfibroin-hydroxyapatite composite, was prepared by a novel method using thermally induced phase separation. The composite had a porosity of more than 94% and exhibited two continuous and different morphologies: an irregularly isotropic pore structure on the surface and a regularly anisotropic multilayered structure in the interior. In addition, the composite was composed of an interconnected open pore structure with a pore size below a few hundred microns. The chemical composition, pore morphology, microstructure, fluid absorptivity, protein permeability, and mechanical strength were investigated according to the composition rate of bioceramics to biopolymers for use in tissue engineering. The incorporation of hydroxyapatite improved the fluid absorptivity, protein permeability, and tenacity of the composite while maintaining high porosity and a suitable microstructure.

Electrochemical Immunosensor Using a Gas Diffusion Layer as an Immobilization Matrix

  • The modification of a gas diffusion layer (GDL), a vital component in polymer electrolyte fuel cells, is described here for use in the electrochemical detection of antibody-antigen biosensors. Compared to other substrates (gold foil and graphite), mouse anti-rHBsAg monoclonal antibody immobilized on gold-coated GDL (G-GDL) detected analytes of goat anti-mouse IgG antibody-ALP using a relatively low potential (-0.0021 V vs. Ag/AgCl 3 M NaCl), indicating that undesired by-reactions during electrochemical sensing should be avoided with G-GDL. The dependency of the signal against the concentration of analytes was observed, demonstrating the possibility of quantitative electrochemical biosensors based on G-GDL substrates. When a sandwich method was employed, target antigens of rHBsAg with a concentration as low as 500 ng/mL were clearly measured. The detection limit of rHBsAg was significantly improved to 10 ng/mL when higher concentrations of the 4-aminophenylphosphate monosodium salt (APP) acting on substrates were used for generating a redox-active product. Additionally, it was shown that a BSA blocking layer was essential in improving the detection limit in the G-GDL biosensor.

Hydroxyapatite-Based Biomaterials for Hard Tissue Applications

  • Over the past few decades, much effort has been made to improve the mechanical and biological performance of HA, in order to extend its range of applications. As a major inorganic component of human hard tissues, hydroxyapatite bioceramic is regarded as being one of the most biocompatible materials. Numerous in vitro and in vivo studies have confirmed its excellent bioactivity, osteoconductivity and bone forming ability. However, because of its poor mechanical properties, its use in hard tissue applications has been restricted to those areas in which it can be used in the form of small sized powders/granules or in the non-load bearing sites. A number of researchers have focused on improving the mechanical and biological performance of HA, as well as on the formulation of hybrid and composite systems in order to extend its range of applications. In this article, we reviewed our recent works on HA-based biomaterials; i) the strengthening of HA with ceramic oxides, ii) HA-based bioactive coatings on metallic implants, iii) HA-based porous scaffolds and iv) HA-polymer hybrids/composites.
