Two fungal strains were isolated from rods of Quercus sp. (NAAS02335) and Pinus densiflora (NAAS05299) in Korea. These strains were identified as Gyrodontium sacchari by their morphological and mycological characteristics. The optimal growth temperature of NAAS02335 and NAAS05299 are $25^{\circ}C$ and $30^{\circ}C$, respectively. Production of cellulase, xylanase, and ligninase was tested on agar media supplemented dyes or substrates. Production of cellulase and xylanase of NAAS05299 was higher than those of NAAS02335, however ligninase activity of NAAS02335 was higher than that of NAAS05299. The activities of cellulase, xylanase, and amylase of strain NAAS05299 were estimated at 6.7~10.2 times higher than that of NAAS02335. Laccase activity was only estimated by strain NAAS02335. The lignocellulytic enzymes are induced by substrates such as rice straw, wooden chips of pine, oak, and poplar. The NAAS05299 was able to degrade filter paper completely after 4 weeks of culturing in liquid media containing a piece of filter paper at $28^{\circ}C$ with continuous shaking. NAAS05299 was able to degrade rice straw, pine chips, and oak chips after 4 months in solid culture, however NAAS02335 decomposed only rice straw among tested 4 kinds of biomass.
The influence of urushiol, as an allergen on laccase property of Fomitella fraxinea was investigated. The enzyme production was reached to the highest level after 10 days, cultivation and the activity and mycelial biomass were increased by 2.5 and 1.5 folds, respectively, by adding urushiol in the culture medium. In liquid cultures using a Cu Mn-free medium, laccase lactivity was decreased by 3.8-9.2 folds, with similar dry cell weight. Two isoenzymes, were purified using anion exchange, hydrophobic interaction and size-exclusion chromatographies. Both isoenzymes are monomeric proteins, with $M_W$ around 67 kDa(Lac1) and 66 kDa(Lac2), and isoelectric points of 3.67 and 3.81. The optimal conditions for purified isoenzymes were found to be pH 4.5-5.0 and $30-35^{\circ}C$. Activity decreased by the addition of $Fe^{2+}$, $Mg^{2+}$, $Na^+$, and strongly inhibited by EDTA and sodium azide.

In this study, we investigated the potential of producing bioethanol from Liriodendron tulipifera by using oxalic acid pretreatment. Amounts of fermentable sugars, mostly xylose and glucose, in the liquid fraction (hydrolysate) was $40.22g/{\ell}$ after the biomass was pretreated with 0.037 g/g of oxalic acid for 20 minutes at $160^{\circ}C$. Production amounts of ethanol was $8.6g/{\ell}$ from the 72 hours of simultaneous saccharification and fermentation (SSF) on solid fraction of the pretreated sample. At the same condition, when the reaction time increased to 40 minutes, $32.66g/{\ell}$ of fermentable sugars in the hydrolysate and $9.5g/{\ell}$ of ethanol was produced from the process of pretreatment and SSF. As a result of analyzing the fermentation inhibitors, such as acetic acid, 5-HMF, furfural and total phenolic compounds, as the reaction time increased, the amount of the fermentation inhibitors in the hydrolysate increased. Production of the fermentation inhibitors was more affected by initial concentration of oxalic acid rather than reaction time. $3.39{\sim}5.78g/{\ell}$ of acetic acid was produced by pretreatment with 0.013 g/g of oxalic acid, and the amount of furfural produced by decomposition of xylose was 2~3 times higher than the amount of 5-HMF produced by decomposition of glucose. All the hydrolysates contained more than $5g/{\ell}$ of total phenols considered as the degradation product of lignin. Therefore, by analyzing the amount of fermentable sugars and fermentation inhibitors in the hydrolysate, and producing ethanol from SSF of solid fraction of the pretreated sample, the biomass pretreated with 0.037 g/g of oxalic acid for 20 minutes at $160^{\circ}C$ can be expected to produce the most ethanol.

Many studies of the phytoplankton community structure have been conducted using the CHEMTAX program on the basis of the photosynthetic pigment concentrations measured by a HPLC (High-Performance Liquid Chromatography) technique. The CHEMTAX program determines the contribution of each phytoplankton class to total phytoplankton biomass (chlorophyll a) based on the ratios of marker pigment to chlorophyll a of phytoplankton group. In this study, the marker pigment/chlorophyll a ratios were investigated in phytoplankton species isolated from marine waters around the Korean peninsula. These results were used as the input pigment ratios of the CHEMTAX program to investigate phytoplankton community structure in Korean coastal waters (Yeoja and Gamak Bay). There were significant differences in the ratios of marker pigment to chlorophyll a among the different species within the same algal class. There was a significant difference between the values of our ratios and the previously used ratios in other regions of the world. When phytoplankton community composition was calculated using our initial ratios in Yeoja and Gamak Bay, our results were significantly different from the results calculated on the basis of initial ratios of marker pigment in phytoplankton suggested in other marine waters. The estimates of the contributions of the major algal groups (bacillariophyceae and dinophytes) to total chlorophyll a varied within 5% depending on the initial ratios chosen. The variations of estimates for the pico- and nanoplankton (cyanophytes and prasinophytes), which have relatively low contributions to total chlorophyll a, were higher than those for major algal group. Although the HPLC-pigment measurements combined with CHEMTAX analysis are useful for identifying and qualifying phytoplankton community structure, further researches for the pigment ratios of the dominant phytoplankton species presenting in a given area are also needed.
To establish a optimum culture condition in vitro for production of seedlings in Wasabia japonica, the effects of salt strength, sucrose concentration, and $NH_4/NO_3$ ratio on the shoot growth in MS medium were investigated. The full strength of MS medium was the best condition for shoot growth, and in $1/4{\sim}1/2x$ or 2x MS medium the growth was significantly suppressed. Growth of shoots was enhanced when sucrose was added in MS medium after 2 weeks of culture, and the best result was found at 3% sucrose addition. ${NH_4}^+/{NO_3}^-$ ratio of 10:50 gave the highest biomass of shoot in the liquid culture with MS medium, and increasing the ${NH_4}^+$ ratio in the medium significantly reduced the growth of shoot.

Chlorophyll a in seawater which is an indicator of phytoplankton biomass and primary production is determined by High Performance Liquid Chromatography (HPLC), Fluorometry and Spectrophotometry. In this study, various methods for chlorophyll a determination in seawater are compared using in situ seawater samples from Korean seas. Three inter-laboratory comparison campaigns were carried out using chlorophyll a standard samples (R0) and in situ seawater samples, collected from the East China Sea (R1) and the East Sea (R2). 6 laboratories by HPLC methods, 4 laboratories by fluorometry, and 3 laboratories by spectrophotometry participated. Precisions, defined as the coefficient of variation (CV) were within 9% in standard samples, 0.8~20% (average: 6.1%) in R1, 4~21% (average: 13.2%) in R2. Discrepancy in three methods was approximately 20% within the range of the sample homogeneity intended the laboratory precision (R1: 8%, R2: 15%). The discrepancy in laboratories was greater than the discrepancy in methods. The chlorophyll a concentrations can be produced within 20% discrepancy in spite of using different methods. It is recommended to consider this 20% discrepancy when using the chlorophyll a data produced by different laboratories and different methods.

This study aimed to isolate and identify freshwater algae from the organic agricultural ecosystems and investigate its biological characteristics to study the possibility of utilizing a biomass freshwater algae in organic farming. In the survey area, average water temperature was $12.4{\sim}28.2^{\circ}C$ and the pH ranges were from 6.1 to 8.5. The solid culture method is more suitable than liquid culture method for isolation of freshwater algae with lower contamination level and higher isolation frequency. A total of 115 strains were isolated from six freshwater algae habitats in nine regions in Korea. BGMM (BG11 Modified Medium) amended with NaNO3 and $KNO_3$ as a nitrogen, and $Na_2CO_3$ as carbon source was designed to isolate and culture freshwater algae. Absorbance of freshwater algae culture has increased dramatically to four days and decreased after eight days after inoculation. CHK008 of the seven isolates showed the highest absorbance in seven days after culturing in BGMM. The optimal pH of BGMM for culturing freshwater algae was pH 6-7. As light intensity increased, growth of freshwater algae increased. Among the five kinds of carbon sources, glucose and galactose promoted good growth of freshwater algae in BGMM. The colony color of purified 16 green algae isolates showed a separation of green, dark and light green, and of them, eleven algae strains showed a strong fluorescent light under fluorescence microscopy. Cell size of the green algae showed a wide range of variation depending on the species. General morphology of the green algae strains was spherical. Chlamydomonas sp. was elliptical, and Chlorella sorokiniana was ellipsoidal and cylindrical. All strains of the green algae except for Chlamydomonas sp. did not have flagella. One isolate of Chlamydomonas sp. and five isolates of C. sorokiniana secreted mucus. Sixteen isolates of 16 green algae were identified as two family and six species, Chlorella vulgalis, C. sorokiniana, C. pyrenoidosa, C. kessleri, C. emersonii, and Chlamydomonas sp. based on their morphological characteristics.
Sweetpotato shoot tops (leaves, tips and petioles) are known to be very useful parts as vegetables because of their high nutritive values and great biomass yield. In this study, the phenolic compound contents, antibacterial activity, mutagenic activity, and antimutagenic activity were investigated in sweetpotato tips that were 10-15cm of shoot top including stems, petioles and tender leaves after sprout of storage roots. The study was done by extracting sweetpotato tips with 80% ethanol and the ethanol fraction was re-extracted with hexane, chloroform, ethyl acetate, butanol and water. In ethyl acetate and butanol fractions, total phenolic compounds contained 95. 6mg/g extract and 69.3 mg/g extract, respectively, The antibacterial activity was measured using the paper disk method with concentrations of 1, 2, 5 and 10 mg/disk of butanol and ethyl acetate fractions against L. monocytogenes and S. Typhimurium strains. Higher doses of solvent extracts showed the higher antibacterial activities. In addition, 5, 10 and 20 mg/mL of the extracts were tested to determine the antibacterial activity in liquid culture. The sweetpotato leaf extract by ethyl acetate showed 1 log reduction compared to control after 24 hrs on Listeria monocytogenes, but 20 mg/ml of butanol extract completely inhibited the growth of the pathogen after 12 hrs. The extracts from ethyl acetate or butanol on Salmonella Typhimurium did less than 1 log reduction during cultivation compared to control. The numbers of S. Typhimirium TA98 and TA100 revertant colonies were 29-33 and 159-188 CFU/plate, respectively, indicating that solvent extracts were no mutagenic activity. The antimutagenic test was performed by adding direct mutagen 2-NF and MMS, and butanol and ethyl acetate showed antimutagenic effect. Thus, this study showed that sweetpotato tips had high phenolic contents and both antimicrobiol and antimutagenic properties. Sweetpotato tips would be good nutritive source because of their high nutrient content without any toxicity in consuming.
Objective of this research was to investigate the influence of $NH_4^+$ and $NO_3^-$ ratios in liquid feeding on the growth of snapdragon 'Potomac Red' and changes in medium chemical properties. The seeds were sown into 200 plug trays and fertigated once a week with nutrient solution containing various ratios of $NH_4^+$ and $NO_3^-$ such as 0 : 100, 27 : 73, 50 : 50, 73 : 27, and 100 : 0. The total N concentrations were adjusted to 50, 100 and $150\;mg{\cdot}L^{-1}$ in plug stages of 2, 3, and 4, respectively. Determination of seedling growth and analysis of plant tissue and root medum were conducted at 56 days after sowing. The treatment of 27 : 73 ($NH_4^+:NO_3^-$) had the greatest plant height, fresh weight, and dry weight. The N and P contents in 27 : 73 ($NH_4^+:NO_3^-$) treatment based on the above ground plant tissues were 2.39 and 0.39%, respectively, which were the greatest among treatments. The elevation of $NH_4^+$ ratio in fertigation solution decreased tissue Ca and Mg contents, but that did not influence tissue K content. The variations in $NH_4^+:NO_3^-$ ratios impacted the soil solution pH and the difference among treatments had been severe since three weeks after sowing. Elevation of $NH_4^+$ ratios in fertigation solution increased electrical conductivity and concentrations of K, Ca, and Mg in soil solution of root medum. The $NH_4^+$ and $NO_3^-$ concentrations in the soil solution were high in weeks 2, 3, and 4, then decreased gradually as the biomass of seedlings increased. Medium P concentration decreased gradually as seedlings grew, but statistical differences were not observed among treatments.
