This study was carried out to investigate the antioxidant activity of bamboo (Sasa borealis) leaf extract by measuring electron donating ability, superoxide dismutase (SOD)-like activity, reducing power, and lipid peroxidation inhibitory activity. Two crude extracts by water or 70% EtOH and five fractions of n-hexane, chloroform, ethyl acetate, n-butanol, and aqueous from the crude extract of 70% EtOH were prepared for this study. The crude extracts of water and 70% EtOH yielded 8.5% and 11.4%, respectively and the yields of n-hexane, chloroform, ethyl acetate, n-butanol, and aqueous fractions were 5.1% to 0.6%. Total polyphenol contents of the water and the 70% EtOH crude extracts were not significantly different; however, their total flavonoid contents were significantly greater in the 70% EtOH than in the water crude extract. Total polyphenol contents were the highest in chloroform fraction followed by ethyl acetate and n-butanol fractions and total flavonoid contents were the highest in ethyl acetate fraction followed by chloroform and n-hexane fractions. The two crude extracts as well as the five fractions showed election donating ability, SOD-like ability, reducing power, and lipid peroxidation inhibitory activity. Most of the antioxidant activities of each crude extract or fractions increased proportionally with the concentration. These results indicate that bamboo (Sasa borealis) leaf extracts show antioxidant activities due to its substantial content of polyphenol including flavonoid. Thus, it could be concluded that crude extracts by water or 70% EtOH and the fractions from the 70% EtOH extract, especially chloroform, ethyl acetate and n-butanol, would be useful as natural antioxidant substances.
The current study was conducted to confirm the anti-cancer effect of Sarijang, which is a mixture of extracts from purple bamboo salt, Rhynchosia nulubilis, garlic, and Ulmi cortex. Nude mice were injected with a human-derived colorectal cancer cell (HCT116 cell line) and subsequently administered Sarijang for 4 weeks, following which the body weight, organ weight, and tumor size were measured. To evaluate the anti-cancer mechanism of Sarijang, the levels of p16 and extracellular signal-regulated kinase (ERK), cell cycle regulators in colorectal cancer, were measured. To evaluate the toxicity of Sarijang on liver and kidney, aspartate transaminase, alanine transaminase, blood urea nitrogen, and creatinine were analyzed. Sarijang not only reduced the tumor size by enhancing p16 and suppressing ERK, but also showed no side-effect in the liver and kidneys. Taken together, Sarijang has the potential to inhibit tumor growth without side effects, and may be used as a useful functional food.
This study was performed to investigate the effects of Sineui-Dae (Sasa coreana Nakai, a kind of bamboo) extracts on the improvement of serum lipid composition by using rats fed a hypercholesterol diet for 4 weeks. The experiment animals were administered with the following diets; high cholesterol diet group (HC diet) as a control and three supplemented groups with high cholesterol diets (HCW, HCM or HCH diet). Three kinds of extracts were prepared by orderly extraction with hexane, methanol, and water. We measured free cholesterol, cholesteryl ester, total cholesterol LDL- and HDL-cholesterol triglyceride and phospholipid in the serum of rats in three experimental and control groups. There were no significant differences in body weights and feed intakes between the HC control group and the extract supplemented groups. The levels of total cholesterol and LDL-cholesterol in the serum of extract supplemented groups were lower than that of the group fed with HC diet only. Serum HDL-cholesterol levet which is known as an antiatherosclerosis factor, was higher in all groups supplemented with bamboo extract by 17.2-21.9% compared to the HC control group. All groups supplemented with bamboo extracts showed the lowering effect of atherogenic index compared to the HC control group (HC group: 2.96${\pm}$0.08, HCM group: 1.48${\pm}$0.02, HCW group: 1.69${\pm}$0.04, HCH group: 1.84${\pm}$0.01). Furthermore, serum triglyceride and phospholipid decreased significantly in the HCM diet compared to the HC control diet. These results suggest that Sineui-Dae bamboo extract, especially methanol extract, has improving effects on hyperlipidemia of rats fed a high cholesterol diet.
Glycosidase inhibitors are major targets in the treatment of type II diabetes, cancer and viral infections. This study was carried out to investigate the glycosidase inhibitory substances from bamboo (Phyllostachys bambusoides). Bamboo was extracted with methanol and then further fractionated with n-hexane, chloroform, n-BuOH and aqueous to get an active fraction. All extracts were evaluated for ${\alpha}$-glucosidase inhibitory activities to identify the n-hexane fraction with 33.5 ${\mu}$g/ml of IC50 value. Active compound 1 in the n-hexane fraction was identified as linoleic acid, which exhibited inhibitory activity with 12.4 ${\mu}$M of IC50 value. Mechanistic analysis showed that linoleic acid exhibited noncompective inhibition. This is the first study in which bamboo is reported to show ${\alpha}$-glucosidase inhibitory activity.
Certain parts of 36 kinds of plant were extracted by 75% ethanol and water. The extracts were tested their microbial inhibition activities against several food spoilage microorganisms, Bacillus subtilis, Lactobacillus plantarum, Leuconostoc mesenteroides, Pseudomonas fluorescens, Bacillus cereus and Saccharomyces cerevisiae. The ethanol extract of amur cork was shown inhibitory effect on all 6 species of the microorganisms tested. Chinese pepper, sesame cake, gromwell and oak were on 5 species except S. cerevisiae or P. fluorescens and bamboo leaves, lycopi herba, paulownia and rigida were on 4 species. In general amur cork exhibited the strongest inhibition with a few exceptions on certain species. By disc diffusion method, the ethanol extract of leaf mustard showed the highest inhibition effect on B. subtilis, amur cork on L.plantarum, L. mesenteroides and B. cereus, and amur cork and gallnut on P. fluorescens. Mostly the ethanol extracts in comparison with water extracts showed higher inhibition in most of plants but a few exhibited higher in water extracts.
Plants are an invaluable source of potential new anti-cancer drugs. Sasa quelpaertensis Nakai (Korean name, Jeju-Joritdae) is one of these plants with medical value, which is a bamboo grass widely distributed in Mt. Halla on Jeju Island, Korea. Here, we investigated the apoptotic effects of S. quelpaertensis leaf extracts in six human cancer cell lines (A549, MCF-7, HepG-2, Hela, HCT116 and A375). MTT assay signified the antiproliferative nature of S. quelpaertensis extracts against all tested cancer cells: S. quelpaertensis displayed slight cytotoxicity against A549, MCF-7 and HepG-2 cells, whereas it was exclusively cytotoxic to Hela, HCT116 and A375 cells. Apoptotic cells were evaluated using PI staining of DNA fragmentation by flow cytometry (sub-G1 peak). PI staining indicated increasing accumulation of Hela, HCT116 and A375 cells at sub-G1 phase. Further events like generation of nitric oxide ($NO^{\bullet}$) were accompanied in the S. quelpaertensis Nakai-induced apoptosis. Augmented $NO^{\bullet}$ generation resulted in the DNA fragmentation of Hela, HCT116 and A375 cells by treatment with S. quelpaertensis leaf extracts. These results suggest that S. quelpaertensis may be a potential natural resource for treating cancer cell. To identify the exact mechanisms of molecular mechanism of S. quelpaertensis induced apoptosis awaits further investigation.
The study was carried out to evaluate the anticancer effects in Korean soybean paste(doenjang) added with sun-dried salt(S-D), roasted salt(R-D), bamboo salt roasted once(B1-D) or bamboo salt roasted nine times(B9-D). In MTT assay, S-D, R-D, B1-D and B9-D exhibited a significant inhibitory activity(64-87% and 68-92%) against the proliferation of AGS human gastric adenocarcinoma cells by adding 0.05% and 0.1% of methanol extract. Among Balb/c mice injected with sarcoma-180 cells, the bodies, livers, spleens, kidneys and heart weight of the mice administered with 4 kinds of doenjang extracts were heavier than the non-administered ones of the group. However, no difference was found between the control and doenjang administered groups. Four kinds of doenjang inhibited significantly the tumor growth, especially R-D and B1-D showing an inhibition of tumor cell growth up to 97% by the administration of 1.0 mg/kg methanol extracts of doenjang. The activity of natural killer(NK) cells was relatively high in mice administered with four kinds of doenjang. Particularly, mice administered with R-D methanol extract showed a stronger activity of 88.2%. The activity of glutathione S-transferase(GST) in mice administered with four kinds of doenjang was higher than that of the non-administered group. In particular, the GST activity was the strongest in the group with B1-D. As these results indicate the various kinds of salt have different effects on the anticancer activity of doenjang.
In east Asia, the leaves of various Sasa species have been used in folk medicine for centuries. The effects of the methanolic extract and its subsequent fractions derived from the leaves of Sasa quelpaertensis Nakai on cell proliferation in human leukemia HL-60 cells were evaluated. The ethyl acetate fraction of this extract (ESQL) significantly reduced cell viability in a dose-dependent manner ($0-250\;{\mu}g/mL$). ESQL ($IC_{50}=24.8\;{\mu}g/mL$) exhibited growth inhibition comparable to the main constituent of green tea, epigallocatechin ($IC_{50}=26.2\;{\mu}g/mL$), which was used as a positive control. ESQL treatment induced apoptosis, which was confirmed by the presence of nuclear condensation and annexin V-staining. These results demonstrate that ESQL contains chemopreventive phytochemicals that may be useful in neutraceutical applications.

Among extracts prepared from Alstonia scholaris, Phyllostachys pubescens and Bambusa veitchii, methanol fraction of Alstonia scholaris was found to have antiplasmodial effect by inhibiting growth of the chloroquine-resistant Plasmodium falciparum strain FCR-3 with less than 14 μg/ml of EC50 values. Methanol fraction 2 of Alstonia scholaris revealed the strongest anti plasmodial effect with 40 μg/ml of EC50 value. Especially, this fraction showed higher than 3-folds selective toxicity on a Plasmodium as the EC value was 116 μg/ml on the host FM3A cell. This is the first report on which an extract compound from Alstonia scholaris showed antimalarial effect.
