Correlative Effect of Adenine Sulphate and Benzylaminopurine on the Regeneration Potentialily in Cotyledonary Explants of Groundnut (Arachis hypogaea L.)

  • An efficient method of shoot regeneration of peanut is described. In vitro shoot organogenesis from the callus of cotyledon explants of Arachis hypogaea L. was stimulated by addition of Adenine sulphate (Ads) along with 6 - benzylaminopurine (BAP) and - napthalene acetic acid (NAA). Ads (13 ${\mu}{\textrm}{m}$) had a stimulatory effect on shoot bud differentiation when combined with BAP (13 ${\mu}{\textrm}{m}$) and NAA (2 ${\mu}{\textrm}{m}$). Shoot organogenesis was markedly higher (92%) from callus induced on Ads, BAP and NAA combined media than from those formed by the individual supplementation of Ads or BAP with NAA. The shoots elongated on the media with GA$_3$ (1 ${\mu}{\textrm}{m}$). Elongated plantlets rooted with MS media containing IBA (9 ${\mu}{\textrm}{m}$).

  • In days to calyx-flex stage test, flower development affected by light intensity, light duration and the prevailing temperature. As, 'Jacaranda' variety shorter in under 46 days of flower bud formation and 13 days of calyx-flex stage than the other variety with 'Queen Elizabeth' control variety. About cut flower only 'Grand Vicki Brown' (as within 46 days of flower formation, 13 days of calyx stage and 'Jacaranda', both with shorter plants than the other varieties produced cut flower classified as short and medium-stemmed cut flowers. 'Jacaranda', 'Vicki Brown', 'Golden Times', 'Champagne', 'Osiana', and 'Queen Elizabeth (control variety)' flowered earlier from 46.7 to 50.8 days from pruning. Flower development was likewise faster in all the above mentioned cultivars; while 'Tineke' had significantly delayed flower development reaching calyx-flex stage from 0.5 cm bud size after 51.0 days. The top ten high yielding varieties were 'American Orange', with a net income of P3,081,676.00; 'Yellow Island' with P1,572,874.00; 'Frosty Pink' with P1,085,183.00; 'Champagne' with P980,620.00; 'Coral Pink' with P959,669.00; 'Lara Pink' with P918,444.00; 'Lara Orange' with P714,252.00; 'Grand Gala' with P705,787.00; 'Golden Times' with P806,489.00; and 'Lady X' with P778,060.00 net returns.

  • The following is a taxonomic list of the specific features of external and internal shape and pattern analysis of Paeonia lactiflora PALL. and P. obovata MAX as the standard of herbal medicine. 1. External shape of original plant P. lactiflora has lancelate and elliptical leaves with no hair on both sides. It bears a flower bud at the shoot apex or leaf axil. On the other hand, P. obovata has obovate leaves with hair on the back side. It has a single flower bud at the shoot apex. 2. Physical characteristics of herbal states P. lactiflora has a bright and smooth external surface whose color is light red brown, light gray on its section and it is not easy to cut. On the other hand, P. obovata has a coarse external surface whose color is dark brown, pinkish on its section and it is easy to cut. 3. Physical characteristics of herbal state in currents Paeonia Radix Alba is brown on the external surface, and whitish on its section. On the other hand, Paeonia Radix Rubra is dark brown on the external surface, and yellowish-white on its section. 4. Internal characteristics Internal shape is correspond to that of Paeonia Radix described in literatures. Only P. lactoflora has a concentrated vascular bundle toward centeral cylinder. On the other hand, P. obovata has a scattered vascular bundle at the cortex. 5. Physicochemical pattern by HPLC Both P. lactoflora and P. obovata showed the same pattern. Paeoniforin, a main component, appears upon 13.68 in retention time (Rt) in both. In addition, both contain a large amount of paeoniflorin. Such results will, in the future, be used as basic source for the additional research, such as biological reactions and genetic discrimination.

The Morphological Characteristics of Morus tiliaefolia Makino Growing Wild in the Korean Peninsula (한반도에 자생하는 돌뽕나무(Morus tiliaefolia Makino)의 형태학적 특성)

  • The morphorogical characteristics of the Morus tiliaefolia growing wild in the Korean peninsula are as follows. The chromosome number of somatic cell is 2n=84,that is hexaploid. The shape of male inflorescence is a long cylinder shape of 3.6∼6.0 cm. The length of female inflorescence is 1.8 cm, red violet or violet and the weight of mulberry fruits is 1.39∼1.65 g. The winter bud is very big in comparison with the branch and the accessory bud does not exist. The leaf is big and lobed leaf or entire leaf, the density of lenticell is 9.42$\pm$1.29 ㎠. The length of leaf trichome is 236∼438 ㎛ and the distribution density is 16∼37/25 ㎟ and heigh density of leaf back. The trichome shape of the leaf surface is cylinder shaped, in the leaf back it is thin and long corn shaped.

Effects of Removing Time of Flower Buds on Root Yield and Paeoniflorin Content in Paeonia lactiflora Pallas (작약(芍藥) 화뇌제거시기에 따른 근수량(根收量)과 Paeoniflorin함량(含量) 변화(變化))

  • This experiment was carried out to determine the proper time for flower bud removing to improve growth, yield and paeoniflorin content of root in peony(Paeonia lactiflora Pallas). The flower buds removement caused short stem length by $7{\sim}9cm$ compared with control, but number of stem and stem diameter were similar to that of control. The number of root, fresh root weight and paeoniflorin content in peony increased when their flower buds were removed at earlier stages. The root dry weight was highest of 1.647kg/10a, and increased by 27% compared with control when their flower buds were removed at early stage (bud diameter was less than 10mm). The root dry weight increased by 13% and 10%, respectively. when their flower buds were removed prior to flowering and at flowering stage. Paeoniflorin content in leaf was higher than that in stem, and was higher at early stage. Leaf and stem showed higher paeonif1orin content when their flower buds were removed.

Antifungal and Antioxidant Activities of Extracts from Leaves and Flowers of Camellia japonica L. (동백나무 잎과 꽃 추출물의 항미생물 활성 및 항산화 효과)

  • This research was performed to investigate the possibilities of industrial usage of camellia (Camellia japonica L.) by examining the antioxidant and antimicrobial effects of methanol extract with different sections. Content of total phenolics, DPPH radical scavenging activities and antibacterial activity of young leaf, mature leaf, flower bud, flower, bark, and seed of camellia were compared in vitro experimental models. Total phenolics was contained the higher in young leaf (74.62 mg), flower bud (65.02 mg) and flower (62.42 mg) but less than 20.95 mg per 100 g of dry weight in other parts of Camellia japonica L. And effects of antioxidant measured by DPPH radical scavenger activity ($RC_{50}$, reduce concentration 50%), was shown higher $7.16{\sim}18.14\;{\mu}g/m{\ell}$ in methanol extract of young leaf, flower bud and flower than $61.23\;{\mu}g/m{\ell}$ of BHT as a chemical oxidant. Also, the antimicrobial activity of Camellia japonica L. extracts determined using a paper disc method against food-borne pathogen and food spoilage bacteria, the young leaves extracts showed the most active antimicrobial activity against 7 kinds of harmful microorganisms. Flower bud extracts showed the highest antibacterial activity against P. aeruginosa and Enterobacter spp. C1036. In addition, the minimum inhibitory concentration (MIC) of young leaf extract against B. subtillis,S. fradiae,S. aureus,E. coli,P. aeruginosa, Enterobacter spp. C1036, and S. typhimurium were revealed 1 to 15 ${\mu}g/m{\ell}$. As a result, antimicrobial activity of camellia extracts was shown higher gram positive bacteria than gram negative bacteria.

  • A rapid micropropagation protocol was established for Exacum travancoricum Bedd. The effect of two cytokinins viz. BA and kinetin were studied to evaluate the propagation of plants through nodal explants. MS medium supplemented with 13.32 ${\mu}M$ BA induced early bud break and subsequent production of multiple shoots. Rooting of shoots occurred when cultured on 1/2 strength MS medium supplemented with 14.7 ${\mu}M$ IBA. Rooted plants were acclimatized to greenhouse conditions. The propagated plants were transferred successfully to field with 65% success. As the plant was amenable to propagation in vitro, this can be employed as a tool for conservation of this critically endangered and endemic ornamental herb.

  • Several plant growth regulators were examined for on their effect on the in vitro propagation of Kalopanax pictus$(T_{HUNB}.)$ $(N_{AKAI}.)$ on WPM medium. Among the cytokinins tested, BA at $13.32\;{\mu}M$ appeared to be the most effective for multiple axillary shoot formation. Although the addition of $2.89\;{\mu}M\;GA_3$ promoted stem elongation, it produced morphologically abnormal leaves and stems. For rooting of the shoots, $4.9\;{\mu}M$ IBA seemed to be more effective than $2.69\;{\mu}M$ NAA. When the regenerated plants were transferred on artificial mixture containing vermiculite and peat-moss (1 : 1, v/v), 81% of them survived and grew normally.

Characterization of an Easter Lily Calmodulin cDNA Clone (백합실물에서 하나으 Calmodulin cDNA 클론 연구)

  • A clone, LCM1, which encodes calmodulin (CaM) was isolated and characterized from monocot lily (Lilium longiflorum Thunb.) plants. The clone is 681 bps and contains the 447 bp coding region, 8 bp leader sequence, 210 bp 3'-untraslated region, and a poly(A) tail. The coding region of 149 amino acids encodes a protein of predicted Mr 17 kD. Comparison of the LCM1 amino acid sequence with other CaMs revealed that the protein is highly conserved among various living organisms. The expression level of calmodulin gene in lily was studied by RNA blot analysis. The LCM1 mRNA was present in all tissues tested. However, a higher level of calmodulin was observed in anther and floral bud. The level of calmodulin mRNA in anther was about 10 times higher than that in anther was about 10 times higher than that in vegetative tissues. The anther preferential expression of CaM in lily is currently investigated in dicot plants.

  • Rice is sensitive to day-length and short-day plants. It has a characteristic that the photosensitivity response required for flower bud differentiation decreases under long-day conditions. To identify critical photoperiod required for flower bud differentiation of major cultivation rice varieties, the average temperature was fixed at 28 ℃, and the day length was set at 12 hours and 10 minutes intervals from 13 hours to 14 hours 30 minutes. The critical photoperiod for each cultivar was set to day-length, where the daily cumulative response [(X(Critical Photoperiod)-Y(Set day-length))/(X(Critical Photoperiod)-12:00(Optimal Day-length)) × (28.0(Set Temperature)-10(Minimum Temperature))/(29.2(Maximum Temperature)-10(Minimum Temperature))] was the same for each day-length conditions. The flower bud differentiation time of all varieties was 32 days before heading at the average temperature of 28 ℃ conditions. The critical photoperiod of the early maturing type, such as Woonkwang, Odae, Koshihikari, Jopyeong, were 19:20, 18:14, 18:58, 17:30, respectively. Medium maturing type, such as Daebo, Haiami, Samdeok, were 16:08, 16:15, 16:55, respectively. Mid-late maturing type, such as Saenuri, Sindongjin, Chucheong, Samkwang, Ilpum, Saeilmi, Hwangkeumnuri, Dongjinchal, Ilmi, Hopum, Yeonghojinmi, were 15:58, 15:56, 16:36, 16:44, 15:35, 16:26, 15:33, 16:20, 16:29, 16:13, 15:41.
