Stereoselective Synthesis of L-Deoxyaltronojirimycin from L-Serine

  • (2S,3R)-3-Hydroxy-2-(hydroxymethyl)-3,6-dihydro-2H-pyridine 8, an important precursor for the synthesis of polyhydroxylated piperidine azasugars, has been prepared from L-serine. Highly stereoselective nucleophilic addition to amino aldehyde 5 gave the corresponding allylic alcohol 6 which proceeded to give dihydro-2H-piridine 7a via a Grubbs II catalyzed RCM. Stereoselective H-bond directed epoxidation of allylic alcohol led to the oxiranyl alcohol 9 which was easily converted to L-deoxyaltronojirimycin by regioselective ring opening.

Optimal Pre-Plating Method of Chicken Satellite Cells for Cultured Meat Production

  • To establish a pre-plating method of chicken satellite cells with high purity, pre-plating was performed under culture conditions of 37℃ and 41℃, and the pre-plating time was set from a total of 3 hours to 6 hours in consideration of the cell attachment time. The purity of the cells was confirmed by staining paired box protein 7 (Pax7) after proliferation, and Pax7 expression was the highest in culture flasks shaken for 2 hours after incubation at 41℃ for 2 hours to prevent the attachment of satellite cells (p<0.05). Also, when pre-plating and proliferation were performed at 37℃ and 41℃, the Pax7 expression rate was higher at 41℃. The differentiation capabilities of the three groups (T3, T6, and T7) with high Pax7 expression were compared and the fusion index (%) and myotube formation area (%) determined by myosin heavy chain (MHC) staining was calculated. The T6 and T7 groups, which were cultured at 41℃, showed significantly higher values than the T3 group (p<0.05). There was no significant difference in the expression of Pax7 and MHC between the T6 and T7 groups (p>0.05). These results suggest that pre-plating at 41℃ for a total of 4 hours was the most efficient in terms of cost and time for purifying chicken satellite cells for cultured meat.

Proper Activity of Histone H3 Lysine 4 (H3K4) Methyltransferase Is Required for Morphogenesis during Zebrafish Cardiogenesis

  • While increasing evidence indicates the important function of histone methylation during development, how this process influences cardiac development in vertebrates has not been explored. Here, we elucidate the functions of two histone H3 lysine 4 (H3K4) methylation enzymes, SMYD3 and SETD7, during zebrafish heart morphogenesis using gene expression profiling by whole mount in situ hybridization and antisense morpholino oligonucleotide (MO)-based gene knockdown. We find both smyd3 and setd7 are highly expressed within developing zebrafish heart and knock-down of these genes led to severe defects in cardiac morphogenesis without altering the expressions pattern of heart markers, including cmlc2, vmhc, and amhc. Furthermore, double knock-down by coinjection of smyd3 and setd7 MOs caused the synergistic defects in heart development. As similar to knock-down effect, overexpression of these genes also caused the heart morphogenesis defect in zebrafish. These results indicate that histone modifying enzymes, SMYD3 and SETD7, appear to function synergistically during heart development and their proper functioning is essential for normal heart morphogenesis during development.

Expression Profiling of Lipopolysaccharide Target Genes in RAW264.7 Cells by Oligonucleotide Microarray Analyses

  • In inflammatory responses, induction of cytokines and other immune regulator genes in macrophages by pathogen-associated signal such as lipopolysaccharide (LPS) plays a crucial role. In this study, the gene expression profile changes by LPS treatment in the macrophage/monocyte lineage cell line RAW264.7 was investigated. A 60-mer oligonucleotide microarray of which probes target 32381 mouse genes was used. A reverse transcription-in vitro translation labeling protocol and a chemileuminescence detection system were employed. The mRNA expression levels in RAW264.7 cells treated for 6 h with LPS and the control vehicle were compared. 747 genes were up-regulated and 523 genes were down-regulated by more than 2 folds. 320 genes showing more than 4-fold change by LPS treatment were further classified for the biological process, molecular function, and signaling pathway. The biological process categories that showed high number of increased genes include the immunity and defense, the nucleic acid metabolism, the protein metabolism and modification, and the signal transduction process. The chemokine-cytokine signaling, interleukin signaling, Toll receptor signaling, and apoptosis signaling pathways involved high number of genes differentially expressed in response to LPS. These expression profile data provide more comprehensive information on LPS-target genes in RAW264.7 cells, which will be useful in comparing gene expression changes induced by extracts and compounds from anti-inflammatory medicinal herbs.

제2단계 BK21사업에 대한 학생들의 만족도 조사 - 인문·사회계열을 중심으로 - (A Survey of Students' Satisfaction on the 2nd Brain Korea (BK) 21 Project: a Case Study of the Humanities and Social Sciences at K University)

  • 본 연구의 목적은 BK21사업에 실질적으로 참여하는 대학원생들을 중심으로 교육과정지원, 취업지원, 학술활동지원, 교육 국제화지원 지원, 교육환경지원 측면 등에 대한 만족도 여부를 조사하여 대학원생들의 안정적인 연구여건 마련과 우수한 인력양성 촉진을 위한 방안들을 모색하는데 있다. 연구목적 달성을 위해 인문사회계열 BK21사업에 참여하는 학생들을 대상으로 설문조사를 실시하였고 그 결과를 분석하였다. 설문문항은 교육과정지원, 취업지원, 안정적 학술지원, 대외 활동지원, 교육 국제화지원, 영어 강의지원, 교육환경지원의 7가지 영역으로 구성되었으며 대체적으로 영역별로 긍정적인 반응이 나타났다. 그 중에서 높은 만족도를 나타낸 영역은 안정적 학술지원과 교육 국제화지원 지원에 관한 것이었으며 취업지원의 경우 만족도가 가장 낮게 나타났다. 연구결과를 통해 향후 발전적인 BK21사업을 위해 사업내용에 대한 인지도 확대, 취업준비 교육 및 관련 정보제공, 연구비 지원 영역 확대, 장단기 해외 연수프로그램 제고, 영어강의 지원체제 마련, 연구공간 확보방안 마련 등의 제언이 도출되었다.

Catabolic Plasmid-Mediated Heavy Metal Resistance in Herbicide Diuron-Degrading Pseudomonas species

  • Three Pseudomonas strains (Bk8, Bk9, Bk10) selected from soil for their ability to degrade herbicide diuron were tested for their heavy metal resistance. The growth of these catabolic strains on a minimal medium with various concentrations of $Cd^{2+},\;Zn^{2+},\;Ni^{2+}$, and $Hg^{2+}$ revealed a minimal effect on the carbon source for the inhibitory effect of the metals. One of these strains, namely, Bk8, exhibited a high resistance to the heavy metals as compared to the two other strains. This strain harbors plasmid pBk8 (110 kb) and contains at least fur determinants encoding heavy metal resistance. Nickel and zinc resistance are encoded by genes located on the chromosome, while cadmium and mercury resistance are on plasmid pBk8. Accordingly, the characteristics of strain Bk8 suggest that it would be useful in the bioremediation of aromatic compounds in the presence of toxic heavy metals as co-contaminants.

New 7-Hydroxycoumarin-Based Fluorescent Chemosensors for Zn(II) and Cd(II)

  • Five new 4- or 8-substituted-7-hydroxycoumarin derivatives (1-5) were synthesized as fluorescent sensors for metal ions. Fluorescent changes and selectivity for metal ions were compared based on the introduction of different ligands and/or testing with different substitution positions of 7-hydroxycoumarin in aqueous solution. Especially, probes 2, 3 and 5 displayed large fluorescence enhancements with $Zn^{2+}$ and $Cd^{2+}$. Probes 2 and 3 showed moderate selectivity for $Zn^{2+}$ over $Cd^{2+}$. On the other hand, probe 4 showed large fluorescence quenching effects upon the addition of $Ag^+$ and $Hg^{2+}$.

Degradation of Bradykinin, a Cardioprotective Substance, during a Single Passage through Isolated Rat-Heart

  • Angiotensin converting enzyme (ACE) inhibitors have cardioprotective effects in different species including human. This cardioprotective effect is mainly due to the inhibition of bradykinin (BK) degradation rather than inhibition of the conversion of angiotensin I to angiotensir. II. Bradykinin, a nonapeptide, has been considered to be the potential target for various enzymes including ACE, neutral endopeptidase 24.11, carboxypeptidase M, carboxypeptidase N, proline aminopeptidase, endopeptidase 24.15, and meprin. In the present study, the coronary vascular beds of Sprague Dawley rat isolated hearts were perfused (single passage) with Krebs solution alone or with different concentrations of BK i.e. $2.75{\times}10^{-10},\;10^{-7},\;10^{-6}\;and\;10^{-5}M$ solution. Percent degradation of BK was determined by radioimmunoassay. The degradation products of BK after passing through the isolated rat-hearts were determined using RP-HPLC and mass spectroscopy. All the four doses of BK significantly decreased the perfusion pressure during their passage through the hearts. The percentage degradation of all four doses was decreased as the concentration of drug was increased, implying saturation of a fixed number of active sites involved in BK degradation. Bradykinin during a single passage through the hearts degraded to give [1-7]-BK as the major metabolite, and [1-8]-BK as a minor metabolite, detected on HPLC. Mass spectroscopy not only confirmed the presence of these two metabolites but also detected traces of [1-5]-BK and arginine. These findings showed that primarily ACE is the major cardiac enzyme involved in the degradation of bradykinin during a single passage through the coronary vascular of bed the healthy rat heart, while carboxypeptidase M may have a minor role.

BK Channel Deficiency in Osteoblasts Reduces Bone Formation via the Wnt/β-Catenin Pathway

  • Global knockout of the BK channel has been proven to affect bone formation; however, whether it directly affects osteoblast differentiation and the mechanism are elusive. In the current study, we further investigated the role of BK channels in bone development and explored whether BK channels impacted the differentiation and proliferation of osteoblasts via the canonical Wnt signaling pathway. Our findings demonstrated that knockout of Kcnma1 disrupted the osteogenesis of osteoblasts and inhibited the stabilization of β-catenin. Western blot analysis showed that the protein levels of Axin1 and USP7 increased when Kcnma1 was deficient. Together, this study confirmed that BK ablation decreased bone mass via the Wnt/β-catenin signaling pathway. Our findings also showed that USP7 might have the ability to stabilize the activity of Axin1, which would increase the degradation of β-catenin in osteoblasts.

Analysis of Aluminum Stress-induced Differentially Expressed Proteins in Alfalfa Roots Using Proteomic Approach

  • Aluminum (Al) is one of the major factors adversely affects crop growth and productivity in acidic soils. In this study, the effect of Al on plants in soil was investigated by comparing the protein expression profiles of alfalfa roots exposed to Al stress treatment. Two-week-old alfalfa seedlings were exposed to Al stress treatment at pH 4.0. Total protein was extracted from alfalfa root tissue and analyzed by two-dimensional gel electrophoresis combined with MALDI-TOF/TOF mass spectrometry. A total of 45 proteins differentially expressed in Al stress-treated alfalfa root tissues were identified, of which 28 were up-regulated and 17 were down-regulated. Of the differentially expressed proteins, 7 representative proteins were further confirmed for transcript accumulation by RT-PCR analysis. The identified proteins were involved in several functional categories including disease/defense (24%), energy (22%), protein destination (9%), metabolism (7%), transcription (5%), secondary metabolism (4%), and ambiguous classification (29%). The identification of key candidate genes induced by Al in alfalfa roots will be useful to elucidate the molecular mechanisms of Al stress tolerance in alfalfa plants.
