The inhibitory effect of egg white lysosome extract (LOE) on melanogenesis through ERK and MITF regulation

  • Lysosome organelle extract (LOE) was derived from egg whites. The lysosome is an intracellular organelle that contains several hydrolysis enzymes. Previous studies have reported that LOE performs important functions, such as melanin de-colorization and anti-melanin production in B16F10 melanoma cells. However, its principal molecular and cellular mechanisms have not been elucidated till date. In non-cytotoxic conditions, LOE significantly inhibited α-MSH induced melanin synthesis of murine B16F10 cells. The anti-melanogenic activity of LOE was mediated by suppressing the mRNA expression of tyrosinase enzyme, tyrosinase related protein-1/2 (TRP-1/2), and microphthalmia-associated transcription factor (MITF) genes. By performing western blot analysis, we found that LOE significantly attenuated melanogenesis. In this case, LOE helped in increasing extracellular receptor kinase (ERK) phosphorylation in α-MSH induced B16F10 cells. Furthermore, MITF is found to be a key regulatory transcription factor in melanin synthesis; it was down-regulated by LOE through ERK phosphorylation. In this experiment, PD98059 (MEK inhibitor) was used to check whether LOE directly regulated the activity of ERK. Although LOE exerted inhibitory effect on melanin synthesis, we could not observe this effect in PD98059-treated α-MSH induced B16F10. These results strongly indicate that LOE is related to ERK activation and MITF degradation in anti-skin pigmentation. Hence, LOE should be utilized as a whitening agent of skin in the near future.

Inhibitory Effects of 6,8-diprenylorobol on Melanin Synthesis (6,8-Diprenylorobol의 멜라닌 합성 억제 효능)

  • This study was performed to elucidated the inhibitory effects of 6,8-diprenylorobol on melanin synthesis by measuring the levels of cell viability, mRNA expression, tyrosinase activity, and melanin production in the B16F10 cell line. The effects of 6,8-diprenylorobol on tyrosinase-related protein 1 (TYRP1), TYRP2, tyrosinase (TYR), and microphthalmia-associated transcription factor (MITF) mRNA expression levels and melanin content were determined. Quantitative real-time RT-PCR shows that 6,8-diprenylorobol decreases the mRNA expression levels of TYRP1, TYRP2, TYR, and MITF in B16F10 cell line, resulting in lower levels of melanin production compared to α-MSH-treated B16F10 cells. Tyrosinase activity assays reveal that 6,8-diprenylorobol decreases melanin production in B16F10 cells. These results demonstrate the whitening effects of 6,8-diprenylorobol on B16F10 cells; thus, 6,8-diprenylorobol is a potent ingredient for skin whitening. Further research is needed on the mechanism of action of 6,8-diprenylorobol. Such research will benefit not only cosmetics, but also the health food and medical industries.

The Branch Extracts of Vaccinium oldhamii Stimulate Melanin Synthesis Through Activation of Tyrosinase Activity in B16F10 Melanoma Cells

  • In this study, we investigated the effect of branch extracts from Vaccinium oldhamii (VOB) on melanin synthesis in B16F10 cells. VOB promoted melanin production in absence or presence of ${\alpha}$-melanocyte-stimulating hormone (${\alpha}-MSH$) in B16F10 cells. However, VOB did not affect the expression of tyrosinase and TRP-1 associated with melanin synthesis at the mRNA and protein level in B16F10. But, VOB decreased TRP-2 protein level and induced tyrosinase activation in B16F10 cells. Inhibition of tyrosinase activity and tyrosinase knockdown attenuated VOB-mediated melanin synthesis. In conclusion, it is thought that VOB may stimulate melanin synthesis through activating tyrosinase activity.

The Antimelanogenic Effect of Inularin Isolated from Flowers of Inula britannica on B16F10 Melanoma Cells and Zebrafish Embryos

  • In the search for novel, natural melanogenesis inhibitors, a new sesquiterpene, inularin, was isolated from the flowers of Inula britannica, and the structure was determined using spectroscopic and chemical methods. The antimelanogenic effects of inularin on B16F10 melanoma cells and zebrafish embryos were evaluated. Inularin dose-dependently reduced melanocyte-stimulating hormone-induced melanin production and L-DOPA oxidation in B16F10 cells. Zebrafish embryos were used to confirm the antimelanogenic activity. Inularin significantly decreased the pigmentation of embryos compared with untreated controls.

Inhibitory Effects on Melanin Production in B16 Melanoma Cells of Fallen Pear (B16F10 Melanoma 세포에서 낙과 배 물 추출물의 멜라닌 생성 저해 효과)

  • This study investigated the water extracts of fallen pear (FPWE) on tyrosinase activity and melanogenesis. In the present study, we examined the effects of FPWE on mushroom tyrosinase activity in vitro, B16F10 melanoma cell tyrosinase activity, melanin contents, and expression of melanogenic enzyme proteins such as tyrosinase. An apparent down-regulatory effect on tyrosinase activity was observed when B16F10 cells were incubated with FPWE. Results of melanin assay using B16F10 cells treated with different concentrations (50, 125, and $250{\mu}g/mL$) of FPWE showed a dose-dependent decrease in melanin content. To determine whether or not FPWE indirectly affects tyrosinase activity, we assessed mushroom tyrosinase activity upon treatment with various concentrations (125, 250, 500, and $1,000{\mu}g/mL$) of FPWE. In addition, we investigated changes in the protein level of tyrosinase by using Western blotting. Tyrosinase and microphthalmia-associated transcription factor expression levels in B16F10 melanoma cells were reduced in a dose-dependent manner by FPWE. These results suggest that FPWE reduced melanin formation by inhibition of tyrosinase activity. Therefore, we suggest that FPWE could be used an effective whitening agent for skin.

Effect of Proliferation Inhibition on B16/F10 Melanoma Cell by Chloroform Extract from Cornis fructus (산수유 클로로포름 추출물에 의한 B16/F10 melanoma세포의 증식억제효과)

  • Cornis fructus have various biological effects and major chemical components have been tannins, saponins, ursolic acids, gallic acids, linoleic acids, morronisides, cornins and loganins. The main aim of the present study is measurment the effect of chloroform extract from Cornis fructus on proliferation inhibition and Cell death. Cells were cultured in the presence of chloroform extracts from Cornis fructus for 48 h. after 48h treatment of B16/F10 melanoma cells with chloroform extracts, the cells were observed a dose-dependent inhibitions of cell viability with cell death in their proliferation. the cells were estimated cell viability, cell number, total DNA fragmentation and chromatin condensation in a dose-dependent manner. It also caused cell death as measured by cell morphology, DNA fragmentation and nucleus chromatin condensation. therefore, these results suggest that chloroform extracts from C. fructus is inhibitory proliferation and is related to cell death in this cells.

Antioxidant Activity and Whitening activity of Psidium guajava leaf extract (구아바 잎 추출물의 항산화 및 미백 활성 효과)

  • The purpose of this study was to investigate the applicability of the Psidium guajava leaf extract as a whitening functional cosmetic material. We measured DPPH radical scavenging activity, intracellular ROS, cytotoxicity in B16F10 melanoma cells and cytoprotective effect on ultraviolet A, in vitro tyrosinase inhibitory effect and melanin biosynthesis inhibitory effect. The antioxidative effect was confirmed through high DPPH radical scavenging activity and intracellular ROS activity inhibition measurement of the Psidium guajava leaf extract. The survival rate of B16F10 melanoma cells was more than 98% at all concentrations, and the cytoprotective effect from ultraviolet ray A was found to increase in a concentration-dependent manner. In addition, in vitro tyrosinase activity inhibitory effect of 10% and melanin biosynthesis inhibitory effect of 20% were observed. Through less toxicity for B16F10 melanoma cell, high antioxidant activity, inhibition of tyrosinase activity and melanin biosynthesis inhibitory effect, we confirmed the possibility of developing the Psidium guajava leaf extract as a whitening functional cosmetic material with a safe and excellent whitening effect.

Anti-melanogenesis in B16F0 Melanoma Cells by Extract of Fermented Cordyceps militaris Containing High Cordycepin (Cordycepin-고함유 동충하초(Cordyceps militaris) 발효 추출물의 미백효과)

  • To find a novel skin whitening agent, the effect of cordycepin-enriched Cordyceps militaris (CM${\alpha}$) extract fermented by fungi on anti-melanogenesis in B16F0 mouse melanoma cells was investigated. Fermented CM${\alpha}$ was prepared with fungi, including Monascus purpureus (Mp), Aspergillus oryzae (Ao), Aspergillus kawachii (Ak), and Rhizopus oryzae (Ro), respectively. When the content of the phenolics and the flavonoids and the activities of the antioxidant and the mushroom tyrosinase inhibition were measured in the CM fermented by Ak (AkF-CM), the highest content of the phenolics was 46 mg/g dry weight and the highest content of the flavonoids was 0.93 mg/g; the highest activity of the DPPH radical scavenging was 62.74% and the highest activity of the mushroom tyrosinase inhibition was 79.97% CM${\alpha}$CM${\alpha}$. From this result, AkF-CM${\alpha}$ exhibited the highest mushroom tyrosinase inhibitory activity and so it was used in subsequent anti-melanogenesis. B16F0 melanoma cells were treated with 1-10 mg/ml concentrations of AkF-CM${\alpha}$ and 200 ${\mu}M$ arbutin as the positive control. The melanin content and cell viability of the melanoma cells by arbutin treatment decreased to 43% and 92% of the control, respectively. AkF-CM${\alpha}$ treatment at 1, 3, and 5 mg/ml concentrations decreased the extracellular melanin release induced by IBMX treatment by 35%, 45%, and 53%, respectively. AkF-CM${\alpha}$ showed inhibitory activity against both intracellular tyrosinase in melanoma cells and mushroom tyrosinase. AkF-CM${\alpha}$ reduced the protein level of tyrosinase in the IBMX-stimulated cells. These results indicate that AkF-CM${\alpha}$ suppressed the activity and protein content of cellular tyrosinase and decreased the total melanin content in cultured B16F0 melanoma cells.

A Study on the Depigmenting Effect of Carthamus tinctorius Seed, Cyperus rotundus and Schizonepeta tenuifolia Extracts (홍화자, 향부자, 형개 추출물의 미백효과에 관한 연구)

  • The objective of the present study was to evaluate the skin depigmentation effect of the extracts of three herbs, Carthamus tinctorius seed, Cyperus rotundus and Schizonepeta tenuifolia. Their effects on tyrosinase and melanin synthesis inhibitory action were assessed. We found that the C. tinctorius seed ethanol extracts reduced the tyrosinase activity and melanin formation of B16F10 melanoma cells. The C. tinctorius seed suppressed the expression in microphthalmia associated transcription factor (MITF), tyrosinase, tyrosinase related protein 1 (TRP-1), and tyrosinase related protein 2 (TRP-2) in B16F10 melanoma cells. These results show that C. tinctorius seed inhibited melanogenesis on the B16F10 melanoma cell. The underlying mechanism of C. tinctorius seed whitening activity may be the inhibition of tyrisinase, MITF, tyrosinase, TRP-1, and TRP-2 expression. The results suggested that C. tinctorius seed has considerable potential as a natural functional ingredient with a depigmentation effect.
