매몰된 가축 사체의 부패 촉진 및 토양 비옥화를 위한 Corynebacterium glutamicum과 Bacillus licheniformis 처리 효과 (Effect of Corynebacterium glutamicum and Bacillus licheniformis on livestock material burial treatment)

  • Foot and mouth disease (FMD) is highly infectious disease of cloven-hoofed animals, particularly problematic in cattle, sheep, pigs and goats for economic reasons. Last FMD outbreak in February, 2017 caused tremendous social and economical impacts. The Korean FMD policy aims to vaccinate intact animals and euthanize and bury infected animals to prevent the disease spread. However, there was a problem that the buried livestock did not decompose after several years. Therefore, the study was purposed to investigate the effect of Corynebacterium glutamicum and Bacillus licheniformis on the degradation of buried cow carcasses and on the soil condition; such as temperature, decomposition course of carcasses, composition of amino acids in the soil around carcasses, and plant root elongation to measure soil conditions. As a result, the composition of amino acids in the soil treated with C. glutamicum and B. licheniformis was generally higher than those in the untreated soil. Plant roots in soil treated with C. glutamicum and B. licheniformis grew longer than in non-treated soil. The results suggested that the toxic effect on a grave land buried with FMD infected livestock is reduced when treated with C. glutamicum and B. licheniformis in regard of odor reduction, promoted decaying process, and soil fertilization.

  • Bacillus종은 그람양성 간균으로 포자를 형성하며 주변 환경 어디에나 존재하여 임상 가검물에서 분리되는 경우 대부분 오염균으로 생각된다. B. licheniformis에 의한 감염증인 경우 외상, 면역저하, 주사약물남용과 연관성을 가지며, 적절한 항생제 사용과 카테타 제거나 수술적 국소조직 제거에 의해 잘 치유되는 특징이 있다. 그러나 위험인자가 없는 상태에서도 B. licheniformis 균혈증은 생길 수 있으며 본 증례에서도 58세의 위험인자가 없던 여자에서 기관지경 후에 B. licheniformis 균혈증이 발생하였고 다른 증례들처럼 경험적 항생제 치료 후에 합병증 없이 치유되었다.

Bacillus licheniformis SCD121067 균체 생산성 증가를 위한 통계적 생산배지 및 발효조건 최적화 (Optimization of Medium and Fermentation Conditions for Mass Production of Bacillus licheniformis SCD121067 by Statistical Experimental Design)

  • In this work, mass production of Bacillus licheniformis SCD121067 through medium optimization by statistical experimental method was studied. First, galactose, yeast extract and potassium phosphate dibasic were selected as carbon, nitrogen and phosphate sources for mass production of B. licheniformis SCD121067 by using one factor at a time method. Second, according to the result of Plackett-Burman experimental design, key factors was yeast extract and $K_2HPO$. Finally, the response surface methodology was performed to obtain the optimum concentrations of two selected variables. The optimized medium composition consisted of 20 g/L galactose, 36 g/L yeast extract, 0.41 g/L $K_2HPO4$, 0.25 g/L $Na_2CO_3$, 0.4g/L $MgSO_4$ and 0.01g/L $CaCl_2$. Dry cell weight (15.4 g/L) by optimum production medium were increased 10 times, as compared to that determined with basic production medium (1.5 g/L). Fermentation conditions were examined for the mass production of B. licheniformis. The effect of temperature, agitation speed, pH and aeration rate on the mass production of B. licheniformis were also studied in a batch fermenter which was carried out in a 2.5 L bioreactor with a working volume of 1.5 L containing optimized production medium. As a result, dry cell weight of batch culture was 30.7 g/L at $42^{\circ}C$, 300 rpm, pH 8.0 and 2 vvm.

Exploring the Metabolomic Responses of Bacillus licheniformis to Temperature Stress by Gas Chromatography/Mass Spectrometry

  • Owing to its high protein secretion capacity, simple nutritional requirements, and GRAS (generally regarded as safe) status, Bacillus licheniformis is widely used as a host for the industrial production of enzymes, antibiotics, and peptides. However, as compared with its close relative Bacillus subtilis, little is known about the physiology and stress responses of B. licheniformis. To explore its temperature-stress metabolome, B. licheniformis strains ATCC 14580 and B186, with respective optimal growth temperatures of $42^{\circ}C$ and $50^{\circ}C$, were cultured at $42^{\circ}C$, $50^{\circ}C$, and $60^{\circ}C$ and their corresponding metabolic profiles were determined by gas chromatography/mass spectrometry and multivariate statistical analyses. It was found that with increased growth temperatures, the two B. licheniformis strains displayed elevated cellular levels of proline, glutamate, lysine, pentadecanoic acid, hexadecanoic acid, heptadecanoic acid, and octadecanoic acid, and decreased levels of glutamine and octadecenoic acid. Regulation of amino acid and fatty acid metabolism is likely to be associated with the evolution of protective biochemical mechanisms of B. licheniformis. Our results will help to optimize the industrial use of B. licheniformis and other important Bacillus species.

Bacillus licheniformis KJ-9를 이용한 항균발효퇴비의 생산 (Production of Antifungal Compost by Using Bacillus licheniformis KJ-9)

  • 환경친화적인 항균발효퇴비를 생산할 목적으로 식물병원성 균주에 대하여 항균활성이 우수한 B. licheniformis KJ-9를 톱밥우분에 접종하여 고품질의 항균발효퇴비(B. licheniformis KJ 9 fermented compost, BLC)의 개발을 행하였다. B. licheniformis KJ-9가 생산한 물질의 온도 안정성은 $100^{\circ}C$, 10분간 열처리하여도 약 60% 이상의 항균활성을 유지하였으며, pH 안정성은 산성보다는 pH 7.0 이상의 중성 및 알칼리성 영역에서 높은 항균활성을 유지하였다. 퇴비를 제조하는 과정에서 BLC의 경우는 우분 냄새가 확실히 줄어들었으며 또한 퇴비의 발효시간도 3일 정도 단축되었다. 고추의 pot실험 결과 BLC를 첨가한 시험구 이외의 다른 시험구에서는 고추 잎이 마르고 약간의 황색반점이 발생하였다. 그리고 BLC를 이용한 시험구는 밭 흙만을 사용한 시험구 및 밭 흙에 기존의 판매퇴비(commercial available compost, CC)를 혼합한 시험구보다 잎의 수가 많고(1.5~2배), 줄기 및 뿌리의 성장이 빠르며, 줄기가 굵은(1.5~3배) 것으로 나타났다. 마늘 및 들깨를 재배한 현장적용 실험결과 두 시험구 모두 자연발생적인 병충해의 피해는 없었지만 BLC를 사용한 밭에서 마늘 및 들깨의 생육이 현저하게 빠른 것을 관찰할 수 있었다.

Bacillus licheniformis K12 균주 분자 선발 마커 개발 (Development of a Molecular Selection Marker for Bacillus licheniformis K12)

  • 본 연구에서는 미생물균총에서 B. licheniformis K12 균주의 양상을 확인하기 위한 선발마커 개발을 시도하였다. Bacillus licheniformis는 바위게 내장에서 유산생산균주로써 분리되었다. 본 균주는 중온에서 성장이 양호할 뿐만 아니라 유전체 분석결과 protease, amylase, cellulase, lipase, protease, xylanase 등 다양한 고분자 물질들을 분해할 수 있는 효소류들을 보유하고 있는 것으로 나타났다. 선발마커 발굴을 위해 recombinase, integrase, transposase, phage-related genes, bacteriocin 등 유전자 변이 유발이 쉬운 게놈상의 영역에 대해 탐색을 실시하였다. 그 결과, 선발마커로써 가능성이 높은 5개 부위를 확보하였다. 후보마커는 recombinase 부위 3개(BLK1, 2, 3) integrase 부위 1개(BLK4), phase 관련 1개(BLK5)로 나타났다. 후보 선발마커로써 Bacillus species가 다른 B. licheniformis, B. velezensis, B. subtilis, B. cereus 등에 대해 PCR 분석을 실시하였다. 그 결과 BLK1 recombinase, BLK2 recombinase family protein, BLK4 site-specific integrase 등에서 B. licheniformis에서 특이적인 위치에 PCR 산물이 나타나는 것을 확인하였다. 또한, B. licheniformis 표준균주로써 subspecies에 대한 PCR을 수행한 결과 BLK1 및 3이 선발마커로써 양호한 것으로 나타났다. 따라서 BLK1이 미생물균총에서 종(species) 및 아종(subspecies) 선발마커로써 활용 가능할 것으로 판단된다.

Inoculation with Bacillus licheniformis MH48 Promotes Nutrient Uptake in Seedlings of the Ornamental Plant Camellia japonica grown in Korean Reclaimed Coastal Lands

  • The objective of this study was to determine whether inoculation with Bacillus licheniformis MH48 as a plant growth-promoting rhizobacterium (PGPR) could promote nutrient uptake of seedlings of the ornamental plant Camellia japonica in the Saemangeum reclaimed coastal land in Korea. B. licheniformis MH48 inoculation increased total nitrogen and phosphorus content in soils by 2.2 and 20.0 fold, respectively, compared to those without bacterial inoculation. In addition, B. licheniformis MH48 produced auxin, which promoted the formation of lateral roots and root hairs, decreased production of growth-inhibiting ethylene, and alleviated salt stress. Total nitrogen and phosphorus uptake of seedlings subjected to bacterial inoculation was 2.3 and 3.6 fold higher, respectively, than the control. However, B. licheniformis MH48 inoculation had no significant effect on the growth of seedlings. Our results suggest that inoculation with B. licheniformis MH48 can be used as a PGPR bio - enhancer to stimulate fine root development, promote nutrient uptake and alleviate salt stress in ornamental plant seedlings grown in the high-salinity conditions of reclaimed coastal land.

Bacillus spp. 및 Serratia marcescens에 의한 사과 푸른곰팡이병의 생물적 방제 (Biological Control of Blue Mold of Apples by Bacillus spp. and Serratia marcescens)

  • 사과 푸른곰팡이병을 방제하기 위하여 사과, 배, 복숭아 등 370점의 과일을 채취하여 과일표면으로부터 세균 1,080균주를 분리하였다. 분리한 1,080균주를 공시 하여 사과 저장병을 일으키는 Penicillium expansum, Alternaria alternata 그리고 Botrytis cinerea을 대상으로 항균활성 및 병 진전억제효과를 검정하여 8종의 길항세균을 선발하였다. 이 들 길항세균에 대한 세균학적 특성과 Biolog system에 의한 영양원 이용성을 조사한 결과 Bacillus amyloliquefaciens가 2균주, B. megaterium, B. subtilis var. gladioli, B. licheniformis, B. pumilus 및 Serratia marcescens로 동정하였다. Glucose, mannose, starch 등 8종의 탄소원을 공시하여 병원균의 생장 및 길항균의 중식에 미치는 영향을 조사한 결과, 병원균의 증식을 억제하고 길항균의 증식을 촉진하는 영양원으로 glucose가 선발되었다. 길항균의 처리효과를 높이기 위하여 길항균 현탁액에 glucose를 1% 첨가하여 사과에 처리한 다음 푸른곰팡이 병균을 $10^5$포자/ml 농도로 처리하였을 때, 병 진전억제효과가 S. marsecens P76-9처리에서 48%, B. amyloliquefaciens P43-2 처리에서 46% 그리고 B. licheniformis P94-1 처리에서 14% 증가되었다. 사과표면의 상처부위에서 처리한 길항균의 밀도변동에 있어서는 Bacillus 속 세균인 B. amyloliquefaciens P43-2과 B. licheniformis P94-1는 처리 후 7일까지 밀도가 증가하다가 처리 후 10일부터는 감소되는 경향이었으며, S. marsecens P76-9는 초기(처리 후 4일)에는 밀도가 감소되다가 처리 후 7일부터 증가되어 처리 후 7일까지도 일정한 수준으로 밀도가 유지되었다.

Monitoring the Ecology of Bacillus During Daqu Incubation, a Fermentation Starter, Using Culture-Dependent and Culture-Independent Methods

  • Daqu, a traditional fermentation starter, has been used to produce attractively flavored foods such as vinegar and Chinese liquor for thousands of years. Although Bacillus spp. are one of the dominant microorganisms in Daqu, more precise information is needed to reveal why and how Bacillus became dominant in Daqu, and next, to assess the impact of Bacillus sp. on Daqu and its derived products. We combined culture-dependent and culture-independent methods to study the ecology of Bacillus during Daqu incubation. Throughout the incubation, 67 presumptive Bacillus spp. isolates were obtained, 52 of which were confirmed by 16S rDNA sequencing. The identified organisms belonged to 8 Bacillus species: B. licheniformis, B. subtilis, B. amyloliquefaciens, B. cereus, B. circulans, B. megaterium, B. pumilus, and B. anthracis. A primer set specific for Bacillus and related genera was used in a selective PCR study, followed by a nested DGGE PCR targeting the V9 region of the 16S rDNA. Species identified from the PCR-DGGE fingerprints were related to B. licheniformis, B. subtilis, B. amyloliquefaciens, B. pumilus, B. benzoevorans, and B. foraminis. The predominant species was found to be B. licheniformis. Certain B. licheniformis strains exhibited potent antimicrobial activities. The greatest species diversity occurred at the Liangmei stage of Daqu incubation. To date, we lack sufficient knowledge of Bacillus distribution in Daqu. Elucidating the ecology of Bacillus during Daqu incubation would enable the impact of Bacillus on Daqu to be accessed, and the quality and stabilization of Daqu-derived products to be optimized.

Growth Performance and Meat Quality of Broiler Chickens Supplemented with Bacillus licheniformis in Drinking Water

  • A feeding trial was conducted to investigate effects of Bacillus licheniformis on growth performance and meat quality of broilers. Nine hundred one-d-old broiler chicks were randomly assigned to 3 experimental groups with three replicate pens of 100 broiler chicks. Three treatments were i) control, ii) basal diets supplemented with 1 ml of B. licheniformis for each in feed water per day iii) basal diets supplemented with 2 ml of B. licheniformis per chick in feed water per day. The supplementation of B. licheniformis significantly increased body weight in grower chickens (p<0.05), and significantly improved the feed conversion in 3 to 6 and 0 to 6 wk feeding period compared with the control group (p<0.05). Additionally, the supplement also resulted in increased protein and free amino acid contents, and decreased fat content in chicken breast fillet (p<0.05). Furthermore, improvement in sensory attributes was observed in broilers fed with the probiotic. In conclusion, B. licheniformis treatments resulted in a significant increase (p<0.05) in broiler productivity based on an index taking into account daily weight gain and feed conversion rate. Meanwhile, the probiotic contributed towards an improvement of the chemical, nutritional and sensorial characteristics of breast fillet. Overall, the study indicates that B. licheniformis can be used as a growth promoter and meat quality enhancer in broiler poultry.
