  • Objectives : Armeniacae semen is the seed of Prunus armenica L. var. ansu MAXIM, and this is classified into Rosaceae. Armeniacae semen has been used for centuries in traditional oriental medicine for the treatment of pain and inflammatory diseases. Amygdalin is the major compound of Armeniacae semen, and it is now being used for the treatment of pain and cancer. Methods : In the present study, we compared the effects of an aqueous extract of Armeniacae semen and a solution of amygdalin extracted from Armeniacae semen on lipopolysaccharide(LPS)-stimulated prostaglandin E2 synthesis and nitric oxide production in mouse BV-2 microglial cells. For this study, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay, reverse transcription-polymerase chain reaction(RT-PCR), prostaglandin E2 immunoassay and nitric oxide detection were performed on mouse BV-2 microglial cells. Results : In the present study, an aqueous extract of Armeniacae semen and an amygdalin solution extracted from Armeniacae semen suppressed prostaglandin E2 synthesis and nitric oxide production by inhibiting the LPS-induced enhancement of cyclooxygenase-2(COX-2) mRNA and the inducible nitric oxide synthase mRNA in mouse BV-2 cells. For the cyclooxygenase-1(COX-1) expression, an aqueous extract of Armeniacae semen showed a more potent suppression effect compared to the amygdalin solution. However, the amygdalin solution more potently suppressed the LPS-induced COX-2 mRNA expression compared to the aqueous extract of Armeniacae semen. Conclusions : As a result, aqueous extract of Armeniacae semen and amygdalin exert anti-inflammatory and analgesic effects.

  • HPLC analysis of three Prunus species, Armeniacae Semen. Persicae Semen, and Mume Semen, showed that 24. 06 mg/g, 5. 79 mg/g and 3/ 10 mg/g of amygdalin in the MeOH extract and 3. 59 mg/g, 5. 41 mg/g and 13. 48 mg/g of benzaldehyde in the MeOH extract hydrolyzed with ${\beta}-glucosidase$ were contained respectively. The MeOH extract of Mume Semen showed strong antibacterial activities against two species of bacteria (E. coli and B. subtilis) while that of Armeniacae Semen showed mild, and that of Persicae Semen showed no effect. Against A. niger, a strong antifungal activity was observed with Armeniacae Semen and only mild activities with Persicae Semen and Mume Semen. None oh the three inhibited the growth of S. cervisiae. The above results may possibly be suggestive of the correlationships between the contents of amygdalin and benzaldehyde in the test extracts with the antimicrobial potencies.

  • Armeniacae Semen is a seed of Prunus armeniaca Linne var. ansu Maximowicz, which belongs to Rosaceae family. It contains amygdalin and fatty oil and is widely used to treat asthma, dysponea and edema. It was reported that D-amygdalin in Armeniacae Semen undergoes hydrolytic reaction by emulsin when using water, and esecially it is almost decomposed when extracting from powder type. we set up a condition where we can achieve the maximun extraction yield through the study of the methods to rstrain emulsin from causing hydrolysis of D-amygdalin in Armeniacae Semen in the aqueous solution and to prevent D-amygdalin from being converted into neoamygdalin. • Background : Nowadays asthma is considered to be the inflammatory disease characterized by airway hyperresponsiveness and pulmonary eosinophilia, and mediated by Th lymphocytes expressing the Th2 cytokine pattern. In many recent studies, molecular biological methods have been used to investigate the role of cytokines in pathogenesis and new therapeutic targets of asthma. Objective : We aimed to identify the effect of Armeniacae Arnarum Semen and Platycodi Radix on the transcriptional activities of cytokine IL-4, IL-5 and IL-6 involved in asthma model. Materials and Methods : RBL-2H3 cell lines were used. Cells were stimulated with calcium inophore for maximal gene expression. After 24 hours of Armeniacae Arnarum Semen and Platycodi Radix-treatment, total cellular RNAs were collected using Trizol solution method. Then transcriptional activities of IL-4, IL-S and IL-6 were measured by RT-PCR with electrophoresis. Results : In IL-4 study, Armeniacae Arnarum Semen treated group showed 48.4% of transcriptional activities compared to the control group and Platycodi Radix treated group showed 45.4% of transcriptional activities compared to the control group. In IL-5 study, Armeniacae Arnarum Semen treated group showed 52.7%of transcriptional activities compared to the control group and Platycodi Radix treated group showed 60.2% of transcriptional activities compared to the control group. In IL-6 study, Armeniacae Amarum Semen treated group showed 42.3% of transcriptional activities compared to the control group and Platycodi Radix treated group showed 69.1% of transcriptional activities compared to the control group. Conclusion : This study shows that Armeniacae Arnarum Semen and Platycodi Radix have the inhibitory effect on the transcription of IL-4, IL-5 and IL-6 gene expression in RBL-2H3 cell lines. Advanced studies are required to investigate the mechanisms of inhibition by herbal medicine in asthma model.
