Inhibitory Constituents of LPS-induced Nitric Oxide Production from Arctium lappa

  • The methanolic extract from the seeds of Arctium lappa was found to inhibit the LPS-induced nitric oxide (NO) production in murine macrophage RAW264.7 cells. Bioassay-guided fractionation of a methylene chloride soluble fraction led to the isolation of three lignan compounds, arctiin (1), arctigenin (2), and lappaol B (3). Their structures were elucidated by UV, IR, MS, and NMR data, as well as by comparison with those of the literatures. Arctigenin (2) and lappaol B (3) had an iNOS inhibitory activity with $IC_{50}$ values of 12.5 and $25.9\;{\mu}M$, respectively.

우엉 뿌리 추출물의 항산화 활성 및 세포 독성 평가 (Antioxidant Activity and Cytotoxicity on cell of Arctium lappa L. root extract)

  • 본 연구는 우엉 뿌리 추출물의 항산화 활성 및 피부에 대한 안전성을 평가하기 위하여 총 폴리페놀 함량, 총 플라보노이드 함량, DPPH radical 소거 활성을 통하여 항산화 활성을 살펴보고, B16F10 melanoma 세포에 대한 세포 독성 및 자외선 A에 대한 피부 세포 보호 효과를 확인하였다. 또한 화장품 소재로서의 활용을 검증하기 위하여 1차 피부 첩포 테스트를 실시하였다. 본 실험 결과 우엉뿌리 추출물의 함량이 증가됨에 따라 높은 폴리페놀과 플라보노이드의 함량이 확인되었으며, DPPH radical 소거 활성을 확인하였다. B16F10세포에 대한 세포 독성을 확인한 결과 B16F10 melanoma 세포에 대해 독성이 낮고, 자외선 A에 대해 80% 이상의 세포 보호 효과를 확인하였다. 또한 1차 피부 첩포 테스트를 통해 우엉 뿌리 추출물이 피부에 자극이 거의 없음을 확인하였다. 이러한 결과를 통하여 우엉뿌리 추출물이 항산화 활성과 자외선에 대한 피부 보호 효과가 뛰어나고 피부 세포에 대한 독성이 낮으며, 피부에 대한 안전성이 확인됨에 따라 화장품 소재로서의 가능성을 확인하였다.

  • A simple quantitative HPLC method has been developed for differentiating two plants of Asteraceae family viz. S. costus and A. lappa by using a pharmacologically active constituent chlorogenic acid and symmetry C18 column clubbed with a binary gradient using acetonitrile: 0.1% phosphoric acid and detected using a PDA at 327 nm. Quantitatively chlorogenic acid was found to be more in A. lappa (0.140%) than in S. costus (0.087%).

  • As a continuation of the previous studies, a third group of thirty five hot aqueous extracts from natural products were screened for platelet activating factor(PAF) receptor binding antagonistic acitivities using rabbit platelet. The results demonstrate that Arctium lappa is potential source of PAF antagonist.

Yield and Free Sugar Contents of Burdock (Arctium lappa L.) depending on Nitrogen Levels

  • Excessive nitrogen fertilization influences crop yields and quality as well as environmental pollution. In this study, yields, nitrogen use efficiency and free sugar contents of burdock (Arctium lappa L.) were evaluated at different levels of nitrogen fertilization. Nitrogen fertilizer was applied at 5 levels (0, 50, 100, 150, 200%) based on the conventional fertilization ($N=230kg\;ha^{-1}$), and phosphate and potassium fertilizer were treated by conventional P and K fertilization ($P_2O_5-K_2O=140-210kg\;ha^{-1}$) in all plots. The root yields of burdock were the highest in N 100~150% treatment plots. Nitrogen use efficiency and nitrogen recovery decreased from over N 150% treatment. Nitrogen uptake of root was greater than that of shoot in N 50~200% treatments. Fructose contents in root were inversely proportional to the level of nitrogen fertilization. As considering nitrogen recovery and root quality, economical burdock yield was obtained in N $230kg\;ha^{-1}$.

혈관내피세포에서 우방자(牛蒡子) 에탄올 추출물의 항염증 효과 (Anti-Inflammatory Effect of Ethanol Extract from the Seeds of Arctium Lappa L. in Vascular Endothelial Cells)

  • Objectives: The seeds from Arctium lappa have been considered for its various pharmacological properties, which include anti-carcinogenic, anti-inflammatory, anti-diabetic, and anti-viral activities. Methods: In the present study, we investigated the anti-inflammatory effect of the ethanol extract from the seeds of Arctium lappa L (EAL) on cytokine-induced vascular inflammation in human umbilical vein endothelial cells (HUVEC). Results: Pretreatment with EAL significantly decreased tumor necrosis factor alpha ($TNF-{\alpha}$)-induced cell adhesion molecules expression such as intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and endothelial-selectin (E-selectin) in a dose-dependent manner. Cell adhesion assay showed that pretreatment with EAL suppressed HUVEC-monocyte adhesion by $TNF-{\alpha}$ over $1{\mu}g/ml$ concentration. We investigated the involvement of nuclear transcription factor kappa-B ($NF-{\kappa}B$) in $TNF-{\alpha}$-induced vascular inflammation. $NF-{\kappa}B$ p65 nuclear expression was induced by $TNF-{\alpha}$, however, pretreatment with EAL was attenuated that nuclear translocation. In cytoplasm, EAL was also attenuated $TNF-{\alpha}$-induced decrease of inhibitor of ${\kappa}B-{\alpha}$ ($I{\kappa}B-{\alpha}$) expression. Moreover, EAL significantly decreased $TNF-{\alpha}$-induced production of intracellular reactive oxygen species (ROS). Conclusions: Taken together, our findings suggest that seeds of Arctium lappa L could be a therapeutic herb for prevention of cardiovascular diseases throughout the inhibition of vascular endothelial inflammation.

Pseudomonas aeruginosa ATCC 27853에 대한 약용식물 추출물들의 활성 (Activity of Nature Plants Extracts against Pseudomonas aeruginosa ATCC 27853)

  • Pseudomonas aeruginosa에 대한 항균물질을 찾기 위하여 한의학에서 전통적으로 사용되어 오던 34종류 약용식물로부터 에탄올 추출물들을 분리하여 항균활성을 조사하였다. Gardenia jasminoides, Arctium lappa, Citrus unshiu 및 Phellodendron amurense 등의 에탄올 추출물들이 P. aeruginosa ATCC 27853에 대해서 항균활성을 나타냈다. 이들 중 Gardenia jasminoides의 에탄올 추출물은 P. aeruginosa에 대해 강한 항균활성을 나타냈다. 앞으로 이들 약용식물들의 추출물들이 P. aeruginosa에 대한 천연항균물질로서 사용될 수 있을 것으로 기대된다.

Agrobacterium tumefaciens에 대한 약용식물의 탐색 I (The Screening of Medicinal Plant Extracts against Agrobacterium tumefaciens I)

  • 본 연구는 약용식물로부터 항균활성 물질을 찾기 위하여 Glycyrrhiza uralensis, Dryopteris crassirhizoma, Dictamnus albus, Paeonia lactiflora, Angelica dahurica, Spirodela polyrhiza, Cimicifuga heracleifolia, Bupleurum falcatum, Magnolia kobus, Artemisia princeps, Arctium lappa, Aster tataricus, Hovenia dulcis, Citrus unshiu, Asparagus cochinchinensis, Gardenia jasminoides, Smilax china, Hovenia dulcis, Prunus sargentii, Scutellaria baicalensis등의 약용식물을 사용하였다. 총 20종류의 약용식물로부터 에탄올 추출물을 분리하여 Agrobacterium tumefaciens에 대한 항균활성을 조사하였다. Glycyrrhiza uralensis, Dryopteris crassirhizoma, Cimicifuga heracleifolia, Bupleurum falcatum,의 추출물이 Agrobacterium tumefaciens에 대한 항균활성을 나타냈다.

우엉 뿌리 에탄올 추출물이 Streptozotocin으로 유발된 당뇨 흰쥐의 항산화에 미치는 영향 (Antioxidative Effect of Ethanol Extract on Arctium lappa root in Streptozotocin Induced Diabetic Rats)

  • Streptozotocin(STZ)을 45mg/kg, body weight(b.w)의 용량으로 흰쥐의 미정맥에 투여 한 후 유발된 당뇨 흰쥐에게 우엉 뿌리 에탄올 추출물을 1일 1회 7일간 1,000mg/kg,bw의 용량으로 투여하고 항산화작용에 관여하는 glutathione-s-transferase(GST), catalase(CAT), glutathioneperoxidase (GSH-Px)활성과 malondialdehyde(MDA)와 glutathione(GSH) 함량을 측정한 결과 우엉 뿌리 에탄올 추추물 투여군에서 MDA 함량, CAT와 GSH-Px 활성 등의 유의적인 감소(p<0.05)를, GSH 함량과 GST 활성은 유의적인 증가(p<0.05)를 나타내었다. 이와 같이 우엉 뿌리 에탄올 추출물이 항산화 개선효과를 갖는 유효성분을 함유하고 있음을 알 수 있었다.

채취시기에 따른 우엉 잎의 휘발성 향기성분 분석 (Analyses of the Volatile Flavor Composition of Burdock (Arctium lappa L.) Leaves according to Harvesting Season)

  • This study investigated the chemical composition of burdock (Arctium lappa L.) leaves essential oil, and the quantitative changes of the major terpene compounds according to the specific harvesting season. The essential oils obtained by the hydrodistillation extraction (HDE) method from the aerial parts of the burdock leaves were analyzed by gas chromatography (GC) and GC-mass spectrometry (GC-MS). The essential oil composition of this plant was characterized by the higher content of phytol and 6,10,14-trimethyl-2-pentadecanone. Seventy seven (98.28%) volatile flavor compounds were identified in the essential oil from the burdock leaves harvested during the spring season of 2012, and phytol (33.47%) and 6,10,14-trimethyl-2-pentadecanone (32.47%) were the most abundant compounds. Eighty eight (99.08%) compounds were identified in the essential oil from the leaves harvested during the autumn season of 2012, and in this case, phytol (37.35%) and 6,10,14-trimethyl-2-pentadecanone (34.67%) were also the most abundant compounds. These two volatile components were confirmed as the major oil components of the burdock leaves during the time of any harvest. The ratio between the two components contained in the burdock essential oils did not differ significantly by harvesting season. But overall, the essential oil harvested during the spring season contained 65.94% of the two major components, while for the essential oil harvested during the autumn season, the total amount of these two major components was 72.02%. While the main ingredients of the essential oils were found to be unchanged from one harvest time to the next, it was found to differ in content. For the burdock leaves, the quality index of the volatile constituents according to the harvest time would be more useful for utilizing the total quantity other than the proportion between phytol and 6,10,4-trimethyl-2-pentadecone.
