Antioxidative Activity and Component Analysis of Cayratia japonica Extract (오렴매 추출물의 항산화 활성, 성분 분석)

  • In this study, the antioxidative effects, inhibitory effects on elastase, and components of Cayratia japonica extracts were investigated. The free radical(1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activities($FSC_{50}$) of extract/fractions of Cayratia japonica were in the order: 50% ethanol extract(114.3 ${\mu}g/mL$)${\mu}g/mL$)${\mu}g/mL$). Reactive oxygen species(ROS) scavenging activities($OSC_{50}$) of some Cayratia japonica extracts in $Fe^{3+}-EDTA/H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The order of ROS scavenging activities were deglycosylated flavonoid aglycone fraction($OSC_{50},\;3.30{\mu}g/mL$)<50% ethanol extract(1.21 ${\mu}g/mL$)${\mu}g/mL$). Ethyl acetate fraction showed the most prominent scavenging activity. The protective effects of extract/fractions of Cayratia japonica on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The Cayratia japonica extracts suppressed photohemolysis in a concentration dependent manner($1{\sim}25{\mu}g/mL$), particularly deglycosylated flavonoid aglycone fraction exhibited the most prominent celluar protective effect(${\tau}_{50}$, 175.05min at 25 ${\mu}g/mL$). Aglycone fractions obtained from the deglycosylation reaction of ethyl acetate fraction among the Cayratia japonica extracts, showed 2 bands in TLC and 2 peaks in HPLC experiments(360 nm). Two components were identified as luteolin(composition ratio, 47.50%), apigenin(52.50). TLC chromatogram of ethyl acetate fraction of Cayratia japonica extract revealed 3 bands and HPLC chromatogram showed 4 peaks, which were identified as luteolin-7-O-${\beta}$-D-glucopyranoside(composition ratio, 11.14%), apigenin-7-O-${\beta}$-D-glucuronopyranoside(15.38%), luteolin(23.55%) and apigenin(49.92%) in the order of elution time. The inhibitory effect of aglycone fraction on elastase($IC_{50},\;70.5{\mu}g/mL$) was very high. These results indicate that extract/fractions of Cayratia japonica can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. And component analysis of Cayratia japonica extract and antioxidative effects could be applicable to new cosmetics.

Nutrients and Antioxidative Activities of Metasequoia glyptostroboides (메타세콰이어나무 열매와 잎의 영양성분 및 항산화 활성)

  • The nutrients and antioxidative activities of fruits and leaves from Metasequoia glyptostroboides were investigated to provide basic data for the future development of functional foods. The nitrogen-free extract contents of fruit and leaves were 69.04% and 50.47%, respectively. Total phenolic content was higher in leaves (21.75 mg/GAE g) compared to the fruit (19.95 mg/GAE g). The mineral components of fruit and leaves mainly consisted of calcium (2,136.08 and 304.85 mg/100 g, respectively), potassium (1,355.53 and 1,144.04 mg/100 g, respectively), and phosphorous (426.30 and 350.50 mg/100 g, respectively). In terms of amino acid composition, aspartic acid, glutamic acid, proline, and leucine were relatively high, but methionine and cystine were low. The hot water extract from leaves was a more potent free radical-scavenger and had higher reducing activities than extracts from fruit. Thus, phenolics of M. glyptostroboides leaves can be utilized as an effective functional food substance for its natural antioxidant properties.

Effects of gradual loaded exercise on antioxidative enzymes response in normal and obese men (비만인과 정상인간 일회성 점증부하 훈련이 항산화 효소에 미치는 효과)

  • The purpose of this study was to identify the effects of acute exercise on antioxidative enzymes in noramal and obese men. The written informed consent was obtained from 16 participants of eight normally healthy persons and eight obesity ones. The baseline level of obesity was determined at 25 % of the body fat. Any subjects had no experiences in participating in exercise and did not take vitamin or mineral supplement. Blood was sampled at five minutes before exercise, at exhaustion, and after 30 minutes of recovery. All statistical analyses and description methods were computed by SPSS window 14.0. Analysis of Covariance(ANCOVA) was performed to know the differences in values between pre- and post- exercise in each group. We found that after exhaustion, the normal group showed higher erythrocyte superoxide dismutase (SOD) activity compared to the obesity group. At exhaustion, the normal group had lower HSP activity compared to the obesity group. Based on the results, an acute exercise of normally healthy people induced an up-regulation of superoxide dismutase (SOD) activity. This plays an important role in the protection of extra-cellular fluid components against peroxide-mediated damage.

Effects of Red Ginseng Component on the Antioxidative Enzymes Activities and Lipid Peroxidation in the Liver of Mice (홍삼 추출물 투여가 생쥐간에서 항산화 효소 활성과 지질과산화에 미치는 효과)

  • The effects of each component (water extracts, alcohol extracts, lipophillic extracts, total saponin, panaxadiol, panaxatriol) of red ginseng on the antioxidative enzyme activities were investigated in the liver in order to screen antioxidative components of red ginseng. 20∼25g ICR mouse which were pretreated with 50 mg/kg body weight of red ginseng component for 15 days. The ability of red ginseng component to protect against oxidative damage to the mouse liver was examined by determining the level of lipid peroxidation (MDA), hydroperoxide (H$_2$O$_2$) and the activities of superoxide dismutase (SOD), catalase. The hepatic total-SOD activity was highest in lipophilic extracts group and panaxadiol group next (p<0.01). The content of hepatic hydroperoxide was lowest in the order of panaxatriol group and alcohol extracts group (p < 0.01). The hepatic catalase activity in the liver was highest in order of lipophillic extracts group (p <0.01) and total saponin group (p<0.05). Finally the lipid peroxidation (MDA) level was lowest in lipophillic extracts group, alcohol extracts group and panaxadiol next (p <0.01). In conclusion, the order of effectiveness of antioxidants was to be lipophillic extracts>panaxadiol >total saponins.

Chemical Compositions and Antioxidative Activities of Sweet Potato Foliages Harvested by the Cultivation Period and Tips Location (재배기간 및 마디별 고구마 끝순의 성분과 항산화 활성)

  • The foliage of sweet potato [Impomoea batatas (L.) Lam] is an excellent source of food material due to the functional components of polyphenol and carotenoid. In this study, the lightness (L-value), polyphenol contents, and DPPH radical scavenging activity of the foliage of Shinwhangmi, Hayanmi and Suioh cultivars harvested after different cultivation periods (60, 90 and 120 days after planting) and according to tip locations (1st~5th, 6th~10th, 11th~15th tips) were investigated. As a result, Shinwhangmi showed a lower lightness value (60 days after planting and 1st~5th tips). The longer the cultivation periods and the closer the tips, the higher the contents of polyphenol and DPPH radical scavenging activity were in lyophilized foliage of Shinwhangmi, Hayanmi and Suioh. In the 1st~10th tip of Shinwhangmi, Hayanmi and Suioh cultivated for 90 days and dried by 2 step hot-air (1st: $70^{\circ}C$ for 8 hr, 2nd: $80^{\circ}C$ for 4 hr), the polyphenol and ${\beta}$-carotenoid contents were 5.0, 3.9 and 4.0 mg/g and 75.6, 71.6 and 63.1 mg/g, respectively. The DPPH radical scavenging activities and ABTS radical scavenging activities ($EC_{50}$) were 0.29, 0.36 and 0.33 mg and 0.12, 0.15 and 0.11 mg, respectively. These antioxidative activities were over two times stronger than spinach.

Changes of Contents of ${\beta}-Carotene$ and Vitamin C and Antioxidative Activities of Juice of Angelica keiskei stored at different conditions (저장조건에 따른 신선초 생즙의 베타카로틴과 비타민 C의 함량 및 항산화능의 변화)

  • Contents of ${\beta}-carotene$ and vitamin C in the vegetable(Angelica keiskei) juice were measured as a function of storing temperature$(-18^{\circ}C{\sim}35^{\circ}C)$ and period$(6{\sim}72\;hours)$. Content of ${\beta}-carotene$ was highest in the fresh juice and the degree of destruction of the ${\beta}-carotene$ was significantly increased as the storage temperature increased. Vitamin C in the juice stored at $4^{\circ}C$ decreased less than that of ${\beta}-carotene$. There was no significant difference in fatty acid compositions among in the fresh, freeze-dried juice and juice stored for 72 hours. Antioxidative activities of components extracted from the juice(fresh and stored for 24 hours) followed by incubating for 1 day were higher than that of ${\alpha}-tocopherol$ and then significantly decreased as incubation prolonged.

Antioxidative Activity of Extract from Bangah Herb (방아 추출물의 항산화 효과)

  • Bangah, one of the herbs grown in Korea, was investigated for its antioxidant activity. The ether extracts of bangah herb was separated into neutral, phenolic, acidic and basic fractions and further separated into subfractions. Antioxidative activities were measured by hydrogen donating activity (HDA), peroxide value (POV), thiobarbituric acid (TBA) value and inhibition activity against lipid peroxidation of rat liver microsomes, The subfraction components were identified by GC/MS and NMR. Phenolic, though being very small in quantity, showed higher antioxidant activity at all assay system by hydrogen donating activity. POV, TBA value and inhibition activity against lipid peroxidation of rat liver microsomes. Five subfractions(P-1, P-2, P-3, P-4 and P-5) were fractionated from phenolic fraction of bangah herbs, and subfraction P-2 among them showed strong antioxidant activity on a level with BHT or gallic acid at each assay system. Four compounds (peak I, peak II, peak III and peak IV) were isolated by gas chromatogram of TMS derivatives of subfraction P-2 and thes compounds were confirmed to be phenolic substance having -OH and COOH group. There subfractions (N-1, N-2 and N-3) were fractionated from neutral fraction of bangah herbs, and subfraction N-2 among them showed highest antioxidant activity and inhibition activity against lipid peroxidation of rat liver microsomes. Subfraction N-2 was indentified to be estragole by H-NMR spectroscopy.

Protective Effects of Cellular Membrane and Component Analysis of Polygonum aviculare Extracts (마디풀 추출물의 세포 보호 효과 및 주성분 분석)

  • In this study, the antioxidative effects and component analysis of Polygonum aviculare (P. aviculare) extracts were investigated. The ethyl acetate and the aglycone fraction from P. aviculare extracts were more active than (+)-${\alpha}$-tocopherol and $\small{L}$-ascorbic acid, which are known as strong antioxidants for their antioxidative activity by the DPPH method and chemiluminescence assay. The cellular protective effects of fractions of P. aviculare on the rose-bengal sensitized photohemolysis of human erythrocytes, increased in a concentration dependent manner ($1-10{\mu}l$). In particular, the ethyl acetate fraction at a concentration of $10{\mu}l$ showed the most prominent protective effect among all the extracts (${\tau}_{50}$, 314.70 min). TLC and HPLC chromatogram of the ethyl acetate fraction of P. aviculare extracts revealed 3 main bands (PA8, PA5, PA6) and peaks (peak 1, peak 2, peak 3), which were identified as myricetin-3-O-rhamnoside (myricitrin, PA8, peak 1), quercetin-3-${\alpha}$-rhamnoside (quercitrin, PA6, peak 3) by LC/ESI-MS/MS and $^1H$-NMR respectively. These results indicate that fractions from P. aviculare could be applicable to new functional cosmetics as antioxidants.

Changes in Free Sugar, Coixol Contents and Antioxidant Activities of Adlay Sprout (Coix lacryma-jobi L. var. ma-yuen Stapf.) according to Different Growth Stage (생육시기 별 새싹율무의 유리당, 기능성 성분 및 항산화활성 변화)

  • Background: Adlay (Coix lacryma-jobi L. var. ma-yuen Stapf.) is an important medicinal and cereal crop that contains high levels of protein, fatty acids and crude fiber, as well as calcium, phosphorus, zinc, minerals, and essential amino acids. Despite its economic significance, little is known about biological activity in adlay sprouts. This study investigated the total free sugar, polyphenol and flavonoid content, and antioxidative activity in adlay sprouts at different growth stages. Methods and Results: The total free sugar and coixol content of germinated adlay sprouts were analyzed by high performance liquid chromatography. The total phenolic content was measured by the Folin and Denis, and flavonoid content by the Davis method. Antioxidative activity was tested by the 2,2-diphenyl-1-picryl hydrazyl method. All measured component, including total phenolic and coixol content ($2.71{\pm}0.02mg/g$ and $59.70{\pm}0.01mg/g$), and antioxidant capacity ($IC_{50}$; $453.93{\mu}g/m{\ell}$) were highest in 3 day old adlay sprouts. In contrast, total free sugar and flavonoid content (3.02% and $0.60{\pm}0.02mg/g$) were highest in sprouts at 5 days after sowing. In addition, there was a strong positive correlation between antioxidative activity and total polyphenol content (r = 0.902, p < 0.001). Conclusions: Adlay sprouts between 3 days and 5 days following germination contain high levels of functional components and free sugar. This study therefore suggests that adlay sprouts can be developed as a valuable health-promoting food and a good source of natural antioxidants.

Alleviating Effects of Euphorbiae humifusae L. Extract on the Neurotoxicity Induced by Lead (납의 신경독성에 대한 지금초 추출물의 독성경감 효과)

  • This study examined the neurotoxicity induced by lead acetate (LA) on cultured C6 glioma cells and the protective effects of Euphorbiae humifusae L. (EH) extract against LA-induced cytotoxicity. In this study, LA exhibited neurotoxicity in a dose-dependent manner compared to the control, and was determined to be highly-toxic according to the toxic criteria. The $XTT_{50}$ value of LA was calculated at a concentration of $38.6{\mu}M$ after C6 glioma cells were incubated for 72 hours in the media containing $30{\sim}50{\mu}M$ of LA, respectively. In addition, LA-induced neurotoxicity was suggested to correlate with the level of oxidative stress because vitamin E, an antioxidant, increased the cell viability damaged by LA-induced cytotoxicity. The EH extract effectively prevented cell injury from LA-induced cytotoxicity via its antioxidative effects, such as inhibitory ability of lipid peroxidation, superoxide dismutase-like activity and 1,1-diphenyl-2-picrylhydrazyl-radical scavenging activity. These antioxidative effects may result in components, such as polyphenol or flavonoids including gallic acid or quercetin. In conclusion, natural resources, such as EH extracts, may be a useful putative agent for the treatment of diseases associated with oxidative stress, such as lead neurotoxicity.
