Synthesis, Characterization and Antimicrobial Activity of New Thiadiazole Derivatives

  • A series of thiadiazole derivatives were synthesized with differently substituted benzoic acids which were cyclized to give differently substituted thiazolidin-4-one. Elemental analysis, IR, $^1H$ NMR, $^{13}C$ NMR and mass spectral data confirmed the structure of the newly synthesized compounds. The derivatives of these moieties were evaluated for antimicrobial activity. Most of the synthesized compounds showed good antimicrobial activity at 200 and $100\;{\mu}g/mL$. Compounds showed most significant antibacterial activity against gram negative test organism Escherichia coli and most significant antifungal activity against test organisms Aspergillus niger and Candida albicans. It was observed that compounds with $OCH_3$ at 3, 4 position of phenyl ring [5(a-l)] were more potent against microbes as compared to compounds having unsubstituted phenyl ring [4(a-l)].

  • 5.7-Dichloro-8-hydroxyquinaldyl-N-ethylcarbamate, one of the carbamate derivative which are generally used as insecticide, was newly synthesized. Its physical properities were determined and chemical structure was indentified by means of I.R., nmr in addition to elemental analysis. The yield of addition, using triethylamine as catalyst, 5.7-dichloro-8-hydroxyquinaldine and isocyanate was better than that of condensation of 5.7-dichloro-8- hydroxyquinaldine with carbamoylchloride. The effect of the cornpond on rabbit's ileum, and antibacterial activity against Staphylococcus aureus, Salmonella typhi, Escherichia coli, and Pseudomonas aeruginosa were examined. It was observed that the dosage over 100 $\mu$g/ml of the compound relaxed rabbit's ileum and the same dosage of the compound inhibited growth of the above strains of bactera.

  • Bioactivity guided fractionation of the cultured filtrates of Aspergillus versicolor, which was derived from a marine sponge Petrosia sp., yielded three polyketides: decumbenones A (1),B (2), and versiol (3). These compounds were identified on the basis of 1D and 2D NMR spectroscopic and MS analysis. The absolute configuration was defined by the modified Mosher's method. The isolated compounds were tested for cytotoxicity against a panel of five human solid tumor cell lines and antibacterial activity against twenty clinically isolated methicillin-resistant strains. This is the first report on the isolation of these compounds from a marine source.

  • Twelve strains of Bifidobacterium spp. were isolated from the feces of healthy Korean 20∼30 years. The identification of genera from isolates were performed by the microscopic observation and fructose-6-phosphate phosphoketolase (F6PPK) activity which is the key enzyme to distinguish the Bifidobacterium spp. from other anaerobic bacteria. To determine the antibacterial resistance patterns, minimum inhibitory concentration (MIC) of several antibiotics (including anti-tuberculosis agents) was determined. Five of the isolate!, showed the high degree of resistance to vancomycin. To investigate the genetic diversity between isolates and type strain of Bifidobacterium spp. from KCTC, we peformed the RAPD-fingerprinting. Using a total set of four primers, it is possible to distinguish the isolates and Bifidobacterium spp. from KCTC. Thus, Bifidobacterium strains isolated from our samples may be a new species or strains of Bifidobacteriurn genera, and have the potential as a probiotics.

  • Enoxacin[1-ethyl-6-fluoro-1,4-dihydro-4-oxo-7-(1-\piperazinyl)-1,8-naphthyridine-3-carboxylic acid, ENX] is a new quinolone antibacterial agent. The method is based on the highly colored charge-transfer complex formation of this drug as a $\pi$-electron donor with 7,7,8,8-tetracyanoquinodimethane(TCNQ) or chloranil(CL) as $\pi$-acceptors. The colored products were measured spectrophotometrically at 842 nm and 552 nm for TCNQ and CL, respectively. The different experimental conditions are optimized. The linearities for TCNQ and CL were $1.6{\;}\mu\textrm{g}/mL~32{\;}\mu\textrm{g}/mL$ and $6.4{\;}\mu\textrm{g}/mL~160{\;}\mu\textrm{g}/mL$, respectively and colors were produced in non-aqueous media. This report describes a simple and ra\pid method for the analysis of enoxacin.

Study of antimicrobial activity and the mode of action of Anal P5 peptide

  • In a previous study, we showed that Cecropin A (1-8)-Magainin 2 (1-12) hybrid peptide (CA-MA)'s analogue, Anal P5, exhibit broad-spectrum antimicrobial activity. Anal P5, designed by flexible region (positions 9, 10)-substitution, Lys- (positions 4, 8, 14, 15) and Leu- (positions 5, 6, 12, 13, 16, 17, 20) substitutions, showed an enhanced antimicrobial and antitumor activity without hemolysis. The primary objective of the present study was to gain insight into the relevant mechanisms of antimicrobial activities of Anal P5 by using flow cytometric analysis. Anal P5 exhibits strong antifungal activity in a salt concentration independent manner. In addition, Anal P5 causes significant morphological alterations of the bacterial surfaces as shown by scanning electron microscopy, supporting its antibacterial activity. Its potent antibiotic activity suggests that Anal P5 is an excellent candidate as a lead compound for the development of novel antibiotic agents.

Inhibitory Effect of Continentalic Acid from Aralia continentalis on Streptococcus mutans Biofilm

  • In our present study, we investigated the effects of continentalic acid on Streptococcus mutans (S. mutans) biofilm. Methanol extract of Aralia continentalis (A. continentalis) was suspended in water and sequentially partitioned with CHCl3, ethyl acetate (EtOAc), and n-butanol (n-BuOH). The CHCl3 fraction showed the highest activity and an antibacterial compound against S. mutans was isolated from this preparation through various chromatography methods by bioassay guided fractionation. MS, $^1H-NMR$ and $^{13}C-NMR$ analysis showed that the active principle was continentalic acid which was confirmed to show significant inhibitory effects against S. mutans biofilm. These results may provide some scientific rationale for the traditional use these extracts for the treatment of dental diseases.

  • Berberine is one of alkaloids extracted from Phellodendri Cortex or Coptidis Rhizoma and has extensively used as an antibacterial and antidiarrheal drug. In order to increase the bioavailability of berberine preparation, berberine pamoate was synthesized and investigated on its usage in vitro and in vivo. Berberine was more rapidly extracted from herbal plants by hot water extraction method than soxhlet extraction method. Berberine pamoate was easily synthesized from berberine hydrochloride and potassium pamoate solution and identified using the infrared spectrum. Quantitative analysis of berberine was possible in methanol solution by fluorometric determination. The dissolution rate of berberine pamoate was more decreased than that of berberine hydrochloride in simulated gastric fluid and simulated intestinal fluid. The remaining proportion of berberine pamoate in the small intestine of rat was maintained at high concentration for a long time as compared with that of berberine hydrochloride.

The Effect of Well-being Lifestyle and Functional Textile Knowledge on the Perception of High Functional Sports/Leisure Wear Importance (웰빙 라이프스타일과 기능성 섬유에 대한 지식이 고기능성 스포츠레저웨어의 중요도 지각에 미치는 영향)

  • This study analyzes how well-being lifestyle and functional textile knowledge effect the perception of high functional sports/leisure wear importance. A survey was conducted among male and female sports/leisure wear consumers aged 15 and over in the Daegu and Gyeongbuk area from May $12^{th}$ to June $2^{nd}$ 2009. A total of 288 responses were analyzed using descriptive statistics, factor analysis, and regression. As a result, 9 factors were determined as sub-elements of a well-being lifestyle. Knowledge levels for functional textiles were high in elastic, UV blocking, air permeable, and antibacterial properties. The perception of high functional sports/leisure wear importance was positively influenced by functional textile knowledge and two well-being lifestyle constructing factors ('self-confidence/affirmative thinking' and 'health/environment oriented eating habits').

Purification, Structure Determination and Biological Activities of 20(29)-lupen-3-one from Daedaleopsis tricolor(Bull.ex Fr.)Bond.et Sing.

  • The dried fruit-body of Daedaleopsis tricolor was extracted by the petroleum ether. The extracts were purified by liquid-liquid extraction, column chromatography, and recrystallization. The purified compound was a colorless orthorhombic crystal form. Its melting point, molecular weight and molar extinction coefficient $(\varepsilon)$ were estimated $168-170^{\circ}C$, 424 and 3,935 at 208 nm, respectively. Its structure was elucidated to be 20(29)-lupen-3-one by UV-Vis, FT-IR, NMR and X-ray crystallographic analysis. It showed antifungal activities against Saccharomyces cerevisiae and Microsporum gypseum, and antibacterial activities against Escherichia coli, Proteus vulgaris, Pseudomonas pyocyanea, Bacillus subtilis, and Staphylococcus aureus. In addition, this compound showed an antioxidative activity on lipid-peroxidation by 6.4%.
