The anti-oxidative stress effect of antioxidants in the cell using DCFH-DA (DCFH-DA를 이용한 항산화제의 세포내 oxidative stress 억제 효과에 관한 연구)

  • 본 연구는 널리 알려져 있는 항산화제들의 세포 수준에서의 anti-oxidative stress 효과 및 그 기작을 알아보기 위한 연구이다. 연구에 사용한 항산화제로는 지용성인 retinol, $\alpha$-tocopherol, propyl gallate(PG) 및 butylated hydroxy toluene(BHT)과 수용성인 ascorbic acid, $\alpha$-glucosyl rutin 및 green tea extract를 사용하였으며 이들 항산화제들의 시간별 세포 생존율을 NR assay 로 측정한 후 적정 농도에서 DCFH-DA(2', 7'-dichlorofluorescin-diacetate) 를 이용하여 항산화제들의 anti-oxidative stress 억제 효과를 시간별로 측정하였다. 또한 이들 항산화제의 항산화 기작을 알아보기 위하여 NBT(Nitro-blue-tetrazolium) 및 DPPH(Diphenyl-picry-hydrazl)도 병행하여 실시하였다. Anti-oxidative stress 실험에서 지용성 항산화제들은 전반적으로 수용성 항산화제에 비하여 세포에 대한 독성이 상대적으로 강하여 retinol 의 경우에는 0.01%에서 oxidative stress 억제 효과를 관할할 수 있었으며 1 시간경과 후 측정시 53.1%의 억제 효과를 보여 주었다. PG 의 경우에는 0.1%에서 2 시간 경과 후 측정시 50%의 oxidative stress 억제 효과를 보여주었다. 수용성 항산화제인 green tea extract 및 $\alpha$-glucosyl rutin의 경우에는 1%에서 1시간 경과 후 측정시 각 각 51.6% 및 69.7%의 oxidative stress 억제 효과를 관찰할 수 있었다. 또한 시료처리 후 자외선 조사시 oxidative stress 억제 효과의 경우 수용성 항산화제인 ascorbic acid, $\alpha$-glucosyl rutin 및 green tea extract 와 지용성 항산화제 중에서는 $\delta$-tocopherol 에서만 oxidative stress 억제 효과가 관찰되었으나 자외선을 조사 하지 않았을 때 보다 약 20%-40%까지 억제 효과가 감소되었다. 그리고 PG 및 retinol 의 경우에는 자외선 조사시 독성이 증가하여 oxidative stress 억제 효과를 측정할 수 없었다. NBT실험에서 $\alpha$-glucosyl rutin, $\alpha$-tocopherol 및 PG 1%에서 70%이상의 superoxide anion 생성 억제 효과를 보였으며 DPPH 실험에서는 ascorbic acid 와 PG 1%에서 98%의 hydroxyl radical 생성 억제 효과를 보여 주었다. 본 실험을 통하여 BHT 를 제외하고 전반적으로 세포 수준에서의 oxidative stress 에 대한 억제 효과를 확인해 볼 수 있었으며 특히 수용성 항산화제들에서 두드러진 효과를 보여 주었다.

Buckwheat Extract Increases Resistance to Oxidative Stress and Lifespan in Caenorhabditis elegans (꼬마선충에서 메밀 추출물에 의한 산화성 스트레스 저항성 증가 및 수명 연장 효과)

  • Buckwheat (Fagopyrum esculentum) has been known for having strong anti-oxidant, anti-mutagenic, and anti-carcinogenic activities. The free radical theory of aging, also known as the oxidative stress theory of aging, claims that cellular oxidative damage accumulated with time is a major causal factor of aging. In the present study, we investigated the effect of buckwheat extracts on resistance to oxidative stress and aging using Caenorhabditis elegans as a model system. Survival under an oxidative-stress condition induced by paraquat increased markedly following 500mg/L buckwheat extracts treatment, suggesting lower cellular oxidative damage by buckwheat extracts. A lifespan assay also revealed that treatment of buckwheat extracts significantly extended both the mean and maximum lifespan in C. elegans. Interestingly, this lifespan-extension by buckwheat extracts was not accompanied by reduced fertility. These findings suggest that buckwheat extracts can confer longevity phenotype to C. elegans through its strong anti-oxidant activity and support the aging theory which emphasizes a pivotal role of oxidative stress during aging.

  • [6]-Gingerol, a major polyphenol of ginger (Zingiber officinale), exhibits a variety of biological properties including anti-oxidant, anti-inflammatory and anti-cancer activity. However, the radioprotective effect of [6]-gingerol is still unknown. The aim of this study was to investigate the radioprotective effect of [6]-gingerol against radiation-induced cell cytotoxicity and oxidative stress in HepG2 cells. [6]-Gingerol pretreatment attenuated radiation-induced cell cytotoxicity caused by 5Gy (half lethal dose, $LD_{50}$ of HepG2 cells). The measurements of superoxide dismutase (SOD) and catalase (CAT) activity were also performed. The results showed that [6]-gingerol pretreatment reduced increasing SOD and CAT activity after exposure of IR, indicating that [6]-gingerol protected oxidative stress by regulating cellular antioxidant enzyme (SOD and CAT) activity. These findings suggest that [6]-gingerol acts as a radioprotector by attenuating cell cytotoxicity and oxidative stress.

Protectvie effects of Lonicerae Japonicae Flos against hydrogen peroxidase-induced oxidative stress on Human keratinocyte, HaCaT cells (Hydrogen peroxide로 산화적 스트레스가 유도된 HaCaT keratinocyte에서 금은화의 세포 보호 효과)

  • Objectives : Lonicerae Japonicae Flos (LJF) has been shown anti-oxidant, anti-inflammatory, anti-viral, anti-rheumatoid properties. However, it is still largely unknown whether LJF inhibits skin injury against oxidative stress in human keratinocyte, HaCaT cells. The purpose of this study was to evaluate the protective effects of LJF against hydrogen peroxide($H_2O_2$)-induced oxidative stress in human keratinocytes, HaCaT cells. Methods : To evaluate out the protective effects of LJF on oxidative injury in HaCaT cells, an oxidative stress model of HaCaT cells was established under a suitable concentration (500 ${\mu}M$) hydrogen peroxide. HaCaT keratinocyte cells were pre-treated with LJF (0.1, 0.25 or 0.5 mg/ml), and then stimulated with $H_2O_2$. Then, the cells were harvested to measure the cell viability, DNA damage, and release of reactive oxygen species (ROS). Results : LJF (0.1, 0.25 or 0.5 mg/ml) itself did not show any significant toxicity in HaCaT cells. The treatment of $H_2O_2$ caused the oxidative stress, leading to the cell death, and DNA injury. However, pretreatment with LJF reduced cell death, and DNA injury. The stimulation of $H_2O_2$ on HaCaT cells resulted in excessive release of ROS, which is the main factor of oxidative stress. The excessive release of ROS was inhibited by LJF treatment significantly. Conclusions : These results could suggest that LJF exhibited the protective effects of HaCaT cells against $H_2O_2$-induced oxidative stress by inhibiting ROS release. It could be explained that LJF inhibit skin damages against oxidative stress. Thus, LJF would be useful for the development of drug or cosmetics treating skin troubles.

An anti-clastogenic Role of Selenium in Arsenic- and Chromium-induced Oxidative Stress Causing Chromosomal Damages (비소와 크롬에 의한 산화적 스트레스와 염색체 상해에 대한 셀레늄의 방어 효과)

  • This experiment was carried out to examine the roles of selenium in arsenic- and chromium-induced oxidative stress, which results in chromosomal damage, such as sister chromatid exchange (SCE) and chromosomal aberration (CA). For this purpose, the frequency of CA and SCE related to the level of 0xidative stress were analyzed. Selenium decreased the frequency of CA induced by As. In order to evaluate the effect of selenium on clastogenic factors, media from As- and Cr-treated cells were ultrafiltered and added again to cells in the presence or absence of selenium. Selenium decreased the frequency of SCE by As and Cr. This observation indicates the possibility of presence of clastogenic factor. In addition, the clastogenic factor would be involed in oxidative stress since selenium decreased the level of oxidative stress. Thus, it is suggested that selenium may play a role as an anti-clastogenic effector by preventing the oxidative stress, thereby decreasing the frequency of Asand Cr-induced chromosomal damage.

  • Colorectal cancer (CRC) is a major worldwide health problem owing to its high prevalence and mortality rates. Carcinogenesis in the colon is a multistage and multifactorial process. An imbalance between free radical exposure and anti-oxidant defense systems may leads to oxidative stress and attack of macromolecules which can alter signal transduction pathways and gene expression. Consequently, oxidative damage can lead to cellular dysfunction and contribute to pathophysiological processes in a variety of diseases including CRC. One factor tightly associated with CRC is chronic inflammation, which can be present from the earliest stage of tumor onset. Unpolished rice is an attractive chemoprevention in CRC due to their anti-oxidant and anti-inflammatory activities. The aim of this paper is to review evidence linking oxidative stress and inflammation to CRC and to provide essential background information for understanding future research on oxidative stress and inflammation on CRC. Mechanisms of action of unpolished rice in CRC carcinogenesis are also discussed.

Anti-oxidative Effect of Some Plant Extracts Against Nitric Oxide-induced Oxidative Stress on Neuronal Cell (Nitric oxide에 의해 산화적 스트레스를 받은 Neuronal cell에 항산화 효과를 가지는 수종 생약추출물의 검색)

  • The objective of this study is screening the anti-oxidative effects of several plant MeOH extracts against oxidative stress in Neuroblastoma cell. Oxidative stress has been implicated in the pathogenesis of many neurotoxicity, neurodegenerative disorders and cell death. This oxidative stress is generated by ROS (Reactive Oxygen Species) such as nitric oxide, nitrogen dioxide, peroxyl, superoxide ($O_2^-$), hydroxyl, alkoxyl. So, in the present study, we induced oxidative stress by treatment of sodium nitroprusside (2.5 mM) in human neuroblastoma SH-SY5Y cell which was treated samples before 24hr, and cell viability was measured by MTT reduction assay. Of those tested, the extracts of Paeonia japonica (roots), Eucommia ulmoides (炒)(barks), Paeonia japonica (曝乾)(roots), Phyllostachys bambusoides (stems), Polygala tenuifolia (去心, 炒)(roots), Paeonia japonica (roots), Polygala tenuifolia (roots), Machilus thunbergii (barks), Mallotus japonicus (leaves), Poria cocos (whole), Sophora flavescens (roots), Angelica tenuissima (roots), Angelica gigas (當歸尾)(roots) showed anti-oxidative effects[$EC_{50}$<15.20 ${\mu}g$/ml(Carnosine:Positive control)]in dose dependent manner.

Protective effect of Samultang and its four herbal plants against reactive oxygen species in vitro and cellular system

  • Oxidative stress and overproduction of free radicals have been reported to be a major pathological hallmark of neurodegenerative diseases. Samultang has been known as a beneficial agent to treat liver disease and cardiovascular diseases. However, the anti-oxidant activities and neuro-protective effects of Samultang against oxidative stress still have not been evaluated yet. The aim of the present study was to investigate the anti-oxidant and protective effects of Samultang and its four herbal plants, Paeonia lactiflora (PL), Ligusticum striatum (LS), Rehmannia glutinosa (RG), and Angelica gigas (AG), in vitro system and in SH-SY5Y neuronal cells. The extracts of Samultang strongly increased the radical scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical (${\cdot}OH$), and nitric oxide (NO) in a concentration-dependent manner. Furthermore, we investigated the protective effects of Samultang on cellular damage against oxidative stress induced by hydrogen peroxide ($H_2O_2$) in SH-SY5Y cells. Treatment with Samultang alleviated the oxidative stress from $H_2O_2$ by increasing the cell viability and decreasing the intracellular reactive oxygen species levels. Based on these results, we further investigated the radical scavenging effects of PL, LS, RG, and AG. In our results, PL had the highest DPPH, ${\cdot}OH$, and NO radical scavenging activities. Thus, PL has a crucial role in Samultang, which has anti-oxidative and neuro-protective effects. The present research suggests that Samultang and PL have protective roles against oxidative stress from $H_2O_2$-induced neuronal cell death.

Four active monomers from Moutan Cortex exert inhibitory effects against oxidative stress by activating Nrf2/Keap1 signaling pathway

  • Paeonol, quercetin, β-sitosterol, and gallic acid extracted from Moutan Cortex had been reported to possess anti-oxidative, anti-inflammatory, and anti-tumor activities. This work aimed to illustrate the potential anti-oxidative mechanism of monomers in human liver hepatocellular carcinoma (HepG2) cells-induced by hydrogen peroxide (H2O2) and to evaluate whether the hepatoprotective effect of monomers was independence or synergy in mice stimulated by carbon tetrachloride (CCl4). Monomers protected against oxidative stress in HepG2 cells in a dose-response manner by inhibiting the generation of reactive oxygen species, increasing total antioxidant capacity, catalase and superoxide dismutase (SOD) activities, and activating the antioxidative pathway of nuclear factor E2-related factor 2/Kelch-like ECH-associated protein 1 (Nrf2/Keap1) signaling pathway. We found that the in vitro antioxidant capacities of paeonol and quercetin were better than those of β-sitosterol and gallic acid. Furthermore, paeonol apparently diminished the levels of alanine transaminase and aspartate aminotransferase, augmented the contents of glutathione and SOD, promoted the expressions of Nrf2 and heme oxygenase-1 proteins in mice stimulated by CCl4. In HepG2 cells, paeonol, quercetin, β-sitosterol, and gallic acid play a defensive role against H2O2-induced oxidative stress through activating Nrf2/Keap1 pathway, indicating that these monomers have anti-oxidative properties. Totally, paeonol and quercetin exerted anti-oxidative and hepatoprotective effects, which is independent rather than synergy.

Antioxidant and Protective Effects of Palmul-tang on Ultraviolet B (UVB)-induced Damage in Human Keratinocytes (팔물탕의 항산화 효과와 자외선으로 유도된 각질형성세포 손상에 대한 보호효과)

  • Objective : In this paper, we investigated the anti-oxidative capacities and protective effects of water extract of palmul-tang (PMT) against Ultraviolet B(UVB)-induced oxidative damage in human keratinocytes(HaCaT). Method : To evaluate the anti-oxidative activities of PMT, we measured scavenging activities on 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation and reducing power of PMT. To give an oxidative stress to HaCaT cells, UVB was irradiated with $40mJ/cm^2$ to HaCaT cells. To detect the protective effects of PMT against UVB, we measured cell viability, apoptotic bodies and reactive oxygen species(ROS) generation. Results : PMT showed the anti-oxidative activities by scavenging DPPH radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation. Also PMT showed high reducing values. The UVB-induced oxidative conditions led to the cell apoptosis. However, treatment with PMT reduced oxidative stress conditions, including inhibition of cell apoptosis and expression of ROS. Conclusion : PMT had anti-oxidative activities and exhibited protective effects against UVB on HaCaT cells. PMT would be useful for the development of cosmetics treating UVB-induced skin aging.
