여주의 amyloid beta 유도 알츠하이머질환 동물 모델에서 인지능력 개선 효과 (Cognitive improvement effects of Momordica charantia in amyloid beta-induced Alzheimer's disease mouse model)

  • 뇌 내 amyloid beta (Aβ) 축적으로 인한 신경독성은 산화적 스트레스를 야기하여 알츠하이머 질환(Alzheimer's disease, AD)을 유도하는 것으로 알려져 있다. 본 연구는 여주(Momordica charantia L.)의 활성분획물인 butanol (BuOH) 분획물의 Aβ25-35 유도 AD 동물모델에서 인지능 개선 효과에 대해 연구하였다. T-미로 실험 및 물체인지실험을 통해서 여주 BuOH 분획물 100 및 200 mg/kg/day 농도 투여군은 AD를 유도한 control군에 비해 유의적으로 새로운 경로와 물체를 탐색하는 비율이 감소되어 공간인지 및 물체인지능력 개선 효과를 확인하였다. 수중미로실험을 통해 학습·기억력에 미치는 효과를 측정한 결과, 여주 BuOH 분획물 투여군은 훈련을 반복할수록 숨겨진 도피대를 찾아가는 시간이 감소함을 통해 학습·기억력 개선 효과를 나타내었다. 여주 BuOH 분획물이 산화적 스트레스 개선 효과에 미치는 효과를 확인하기 위해 뇌, 간, 신장 조직에서 지질과 산화 함량 및 nitric oxideNO 생성량을 측정하였다. 여주 BuOH 분획물을 처리한 군은 Aβ25-35를 주입한 control군에 비해 유의적으로 뇌, 간, 신장 조직에서 지질과산화 함량 및 NO 생성량이 감소되어 산화적 스트레스 개선 효과를 확인하였다. 따라서 본 연구는 여주 BuOH 분획물이 Aβ25-35 유도 AD 동물모델에서 산화적 스트레스 개선을 통해 인지능력 개선 효과를 나타냄을 확인하였으며, 이에 따라 여주는 AD 예방 및 개선용 소재로써의 가능성이 있는 것으로 사료된다.

흰쥐 해마 CA1 영역에서 H2O2에 의한 장기강화 억제에 대한 발효황금 추출물의 효과 (Effects of Fermented Scutellaria Baicalensis Extract on H2O2 - Induced Impairment of Long-term Potentiation in Hippocampal CA1 Area of Rats)

  • Scutellaria baicalensis (SB) has widely used in the treatment for various brain diseases in the field of Oriental medicine. Biofermantation of SB can make major chemical constituents of SB to pass blood-brain barrier easily and to have more potent anti-oxidant ability. There is a little information about the contribution of fermented SB (FSB) to the formation or maintenance of the neural plasticity in the hippocampus. The purpose of this study was to evaluate effects of FSB extract on hydrogen peroxide (H2O2) - induced impairments of the induction and maintenance of long-term potentiation (LTP), an electrophysiological marker for the neural plasticity in the hippocampus. From hippocampal slices of rats, the field excitatory postsynaptic potentials (fEPSPs) were evoked by the electrical stimulation to the Schaffer collaterals - commissural fibers in the CA1 areas and LTP by theta-burst stimulation by using 64 - channels in vitro multi-extracellular recording system. In order to induce oxidative stress to hippocampal slices two different concentrations (200, 400 μM) of H2O2 were given to the perfused aCSF before and after the LTP induction, respectively. The ethanol extract of FBS with concentration of 25 ㎍/ml, 50 ㎍/ml was diluted in perfused aCSF that had 200 μM H2O2, respectively. Oxidative stress by the treatment of H2O2 resulted in decrease of the induction rate of LTP in the CA1 area with a dose - dependent manner. However, the ethanol extract of FSB prevented the reduction of the induction rate of LTP caused by H2O2 - induced oxidative stress with a dose - dependent manner. These results may support a potential application of FSB to ameliorate impairments of hippocampal dependent neural plasticity or memory caused by oxidative stress.

감태 효소 추출물 및 폴리페놀 추출물의 생리활성에 관한 연구 (Biological Potential of Enzymatic and Polyphenol Extracts from Ecklonia cava)

  • 본 연구에서는 감태 효소 추출물과 그것의 폴리페놀 추출물의 화장품 원료로서의 효능을 알아보기 위하여 항산화, 항당화, 미백, 항염 효과와 관련된 실험을 실시하였다. 감태 효소 추출물과 폴리페놀 추출물은 강력한 라디컬 소거능을 가지고 있으며 BSA/Glucose 시스템에서 최종당화생성물의 형성을 저해하는 항당화 활성과 타이로시네이즈 저해를 통한 우수한 미백력을 가지고 있음을 확인하였다. 또한 두 추출물 모두 세포 내에서 $PGE_2$와 NO 생성 저해를 통한 항염 효과를 나타내었다. 이러한 결과를 종합해 볼 때, 감태 효소 추출물과 그 폴리페놀 추출물은 화장품 원료로서의 응용 가능성이 있을 것으로 사료된다.

Inhibition of MMP-2 and MMP-9 activities by solvent-partitioned Sargassum horneri extracts

  • Background: Matrix metalloproteinases (MMPs) are linked with several complications such as metastasis of cancer progression, oxidative stress, and hepatic fibrosis. Brown seaweeds are being extensively studied for their bioactive molecule content against cancer progression. In this context, Sargassum horneri was reported to possess various bioactivities including antiviral, antimicrobial, and anti-inflammatory partly due to its phenolic compound content. Methods: In this study, potential of S. horneri was evaluated through anti-MMP effect in HT1080 fibrosarcoma cells. S. horneri crude extract was fractionated with organic solvents, namely, water ($H_2O$), n-buthanol (n-BuOH), 85% aqueous methanol (85% aq. MeOH), and n-hexane. The non-toxicity of fraction samples (Sargassum horneri solvent-partitioned extracts (SHEs)) was confirmed by cell-viability assay. SHEs were tested for their ability to inhibit MMP enzymatic activity through gelatin digestion evaluation and cell migration assay. Expressions of MMP-2 and MMP-9 and tissue inhibitors of MMP (TIMPs) were evaluated by reverse transcription and Western blotting. Results: All fractions inhibited the enzymatic activities of MMP-2 and MMP-9 according to gelatin zymography. Except $H_2O$ fraction, fractions hindered the cell migration significantly. All tested fractions suppressed both mRNA and protein levels of MMP-2, MMP-9, TIMP-1, and TIMP-2. Conclusion: Overall, current results suggested that S. horneri has potential to be a good source for anti-MMP agents, and further investigations are underway for better understanding of the action mechanism and isolation and elucidation of the bioactive molecules.

Plant Extracts and Plant-Derived Compounds: Promising Players in Countermeasure Strategy Against Radiological Exposure: A Review

  • Radiation exposure leads to several pathophysiological conditions, including oxidative damage, inflammation and fibrosis, thereby affecting the survival of organisms. This review explores the radiation countermeasure properties of fourteen (14) plant extracts or plant-derived compounds against these cellular manifestations. It was aimed at evaluating the possible role of plants or its constituents in radiation countermeasure strategy. All the 14 plant extracts or compounds derived from it and considered in this review have shown some radioprotection in different in vivo, ex-vivo and or in vitro models of radiological injury. However, few have demonstrated advantages over the others. C. majus possessing antioxidant, anti-inflammatory and immunomodulatory effects appears to be promising in radioprotection. Its crude extracts as well as various alkaloids and flavonoids derived from it, have shown to enhance survival rate in irradiated mice. Similarly, curcumin with its antioxidant and the ability to ameliorate late effect of radiation exposure, combined with improvement in survival in experimental animal following irradiation, makes it another probable candidate against radiological injury. Furthermore, the extracts of P. hexandrum and P. kurroa in combine treatment regime, M. piperita, E. officinalis, A. sinensis, nutmeg, genistein and ginsan warrants further studies on their radioprotective potentials. However, one that has received a lot of attention is the dietary flaxseed. The scavenging ability against radiation-induced free radicals, prevention of radiation-induced lipid peroxidation, reduction in radiation cachexia, level of inflammatory cytokines and fibrosis, are some of the remarkable characteristics of flaxseed in animal models of radiation injury. While countering the harmful effects of radiation exposure, it has shown its ability to enhance survival rate in experimental animals. Further, flaxseed has been tested and found to be equally effective when administered before or after irradiation, and against low doses (${\leq}5Gy$) to the whole body or high doses (12-13.5 Gy) to the whole thorax. This is particularly relevant since apart from the possibility of using it in pre-conditioning regime in radiotherapy, it could also be used during nuclear plant leakage/accidents and radiological terrorism, which are not pre-determined scenarios. However, considering the infancy of the field of plant-based radioprotectors, all the above-mentioned plant extracts/plant-derived compounds deserves further stringent study in different models of radiation injury.

허브 추출물에 따른 매작과의 품질 특성 (Quality Characteristics of Maejakgwa with Added Herb Extracts)

  • The principal objective of was to assess the effects of herb extracts on the antioxidant activity and quality characteristics of Maejakgwa. The total phenolic and flavonoid contents were found to be the highest by a significant degree in the rosemary, lavender, thyme, lemon balm and olive samples, in order. With regard to electron donating ability, increasing concentrations of added herbs resulted in a more significant rise in the DPPH radical scavenging effect in a range of concentrations of between $100{\sim}1000{\mu}g/m{\ell}$. In particular, the rosemary, lavender, and thyme samples were shown to be lower in antioxidant activity than ascorbic acid, but higher than a-tocopherol, thus the antioxidant activities detected in the rosemary, lavender, and thyme samples were considered to be excellent. With regard to reducing ability, increases in the concentration of added herb extract resulted in significant growth. However, the anti-oxidative activity detected in the rosemary extract was indicated to be superior even to that of $\alpha$-tocopherol when added to concentrations of $500{\sim}1,000{\mu}g/m{\ell}$. In our assessment of external appearance, color, flavor, texture, and overall quality, the highest preference was seen in the control group and in the group with 2% added rosemary extract. The lowest score was earned in the group with a 5% addition of rosemary. The functional ingredients and antioxidant activities of the samples were excellent. Our results appear to indicate that rosemary has some value as a natural antioxidant that can prevent oxidation in food.

강일옥 옥수수의 영양성분, 카로티노이드 함량분석 및 생리활성 평가 (Assessment of Nutritional Components, Carotenoid Content and Physiological Activity of Maize Hybrid for Grain 'Kangilok')

  • 본 연구는 마치종 황색 알곡 품종으로 등록된 강일옥 옥수수 알곡의 처리별 시료 및 속대의 일반성분, 생리활성 검정, 색소 함량을 분석하여 기초자료로 제공하고자 수행되었다. 강일옥 옥수수를 알곡, 거피된 알곡, 알곡 껍질 및 속대로 분리하여 일반성분 분석 결과, 수분함량은 거피된 알곡이 12.04%로 가장 높았고 알곡 껍질의 조회분, 조지방, 조단백질 함량은 각각 3.19%, 8.59%, 12.01%로 강일옥 옥수수 처리별 시료 중 가장 높은 함량을 나타내었다. 강일옥 옥수수 처리별 시료의 무기성분 함량을 분석한 결과, 칼슘, 칼륨, 마그네슘, 철, 망간 및 인의 6종이 검출되었으며 공통적으로 검출된 주요 무기성분은 칼륨과 인이었다. 강일옥 옥수수 속대의 칼륨 함량이 114.33 mg/100 g으로 가장 높았으며 알곡 껍질의 인 함량이 2412.40 mg/100 g으로 가장 높게 측정되었다. 강일옥 옥수수 처리별 시료의 항산화 활성 검정 결과, 강일옥 옥수수 속대 추출물의 DPPH radical 소거능은 93.05%, ABTS radical 소거능은 79.88%로 처리별 시료 중 가장 높게 측정되었으며 알곡, 거피된 알곡 및 알곡껍질 추출물의 DPPH 소거능은 모두 70% 이상으로 우수한 항산화활성을 나타내었다. 강일옥 옥수수 처리별 시료의 항염 활성 검정 결과, 강일옥 옥수수 처리별 시료 추출물은 Raw264.7 대식세포의 생존율에 영향을 미치지 않은 것으로 나타났으며 NO 생성량은 모든 시료 처리군에서 LPS 단독 처리군(con(+))에 비하여 유의적으로 NO 생성을 억제 하는 것으로 나타났다. 강일옥 옥수수 처리별 건조분말시료의 카로티노이드 색소 함량을 분석한 결과, 모든 시료에서 ${\beta}$-carotene은 검출되지 않았고, lutein과 zeaxantin은 알곡과 알곡 껍질에서 검출되었으며 거피된 알곡과 속대에서는 검출되지 않았다. 강일옥 옥수수 알곡의 lutein과 zeaxantin의 함량은 각각 $115.30{\mu}g$/100 g, $50.92{\mu}g$/100 g이었으며 알곡껍질의 lutein과 zeaxantin의 함량은 각각 $37.98{\mu}g$/100 g, $19.27{\mu}g$/100 g이었다.

Intramuscular Administration of Zinc Metallothionein to Preslaughter Stressed Pigs Improves Anti-oxidative Status and Pork Quality

  • This study was conducted to determine the effects of exogenous zinc-metallothionein (Zn-MT) on anti-oxidative function and pork quality. After feeding a corn-soybean meal-based diet for two weeks, 48 pigs ($Duroc{\times}Landrace{\times}Chinese\;Black Pig$) were assigned randomly to four groups. Pigs in Group 1 were maintained under non-stress conditions, whereas pigs in Groups 2, 3 and 4 were aggressively handled for 25 min to produce stress. Pigs in Groups 1, 2, 3, and 4 received intramuscular administration of saline (control group; CON), 0 (negative control group; NCON), 0.8 (low dose group; LOW), and 1.6 (high dose group; HIGH) mg rabbit liver Zn-MT per kg body weight, respectively. Pigs were slaughtered at 3 and 6 h post-injection. Zn-MT treatment increased (p<0.05) the activities of superoxide dismutase (SOD) and glutathione-peroxidase (GSH-PX) while decreasing the concentration of malondialdehyde (MDA) in liver. These responses were greater (p<0.05) at 6 h than at 3 h post Zn-MT injection. Zn-MT treatment increased (p<0.05) hepatic SOD mRNA levels in a time and dose-dependent manner and decreased (p<0.05) serum glutamate-pyruvate transaminase and lactate dehydrogenase activities (indicators of tissue integrity). Zn-MT administration decreased (p<0.05) lactate concentration and increased (p<0.05) pH and water-holding capacity in the longissimus thorasis meat. Collectively, our results indicate that intramuscular administration of Zn-MT to pre-slaughter stressed pigs improved tissue anti-oxidative ability and meat quality.

포도씨의 총페놀 성분, 항산화능 및 아질산염소거능에 대한 마이크로웨이브 추출조건 예측 (Prediction of the Optimum Conditions for Microwave-Assisted Extraction of the Total Phenolic Content and Antioxidative and Nitrite-scavenging Abilities of Grape Seed)

  • 포도씨에 함유된 항산화 및 아질산염소거성분에 대한 마이크로웨이브 최적 추출조건을 예측하고자, 추출조건(microwave power 0~120 W, 에탄올 농도 0~100%, 추출시간 1~5 min)을 독립변수로 하고 조건별 추출물의 총페놀 함량, 항산화능 및 아질산염소거능을 종속변수로 하여 회귀 분석함으로써 최적 추출조건을 예측하였다. 도출된 회귀식의 $R^2$는 0.8024-0.9498 범위로 나타났다. 추출물 기능성분들의 최대 예측값과 최적 추출조건은 총페놀 함량의 경우 3.19%(microwave power 142.32W, 에탄올농도 44.30%, 추출시간 4.23 min)와 항산화능의 경우 1.22(84.44W, 56.60%, 3.28 min)로 나타났다. 그리고 pH 1.2~3.0에서 99.5% 이상의 높은 아질산염 소거능을 보인 추출물의 최적 추출조건 범위는 microwave power 30.80~106.58W, 에탄올 농도 49.32~55.18%, 추출시간 3.72-4.58 min으로 예측되었다. 이상의 결과에서 총페놀 함량과 항산화능은 높은 상관성을 보이면서 microwave power에, 아질산염소거능은 에탄올 농도에 영향을 많이 받는 것으로 나타났다.

Effects of cranberry powder on serum lipid profiles and biomarkers of oxidative stress in rats fed an atherogenic diet

  • This study investigated that the antioxidative effect of freeze-dried cranberry powder against protein and lipid oxidation and ameliorative effect of serum lipid profile in rat fed atherogenic diet. Six weeks old male Sprague-Dawley rats were divided into the following four groups: normal diet group with 5% com oil(control), atherogenic diet group with 5% com oil, 10% lard, 1% cholesterol, and 0.5% sodium cholate(HFC), atherogenic plus 2% cranberry powder diet group(HFC+C2), and atherogenic plus 5% cranberry powder diet group(HFC+C5), and respective diet and water were fed daily for 6 weeks. After the experimental period, the serum lipid profile, such as total cholesterol, HDL-cholesterol, LDL-cholesterol, and triglyceride, ferric reducing ability of plasma(FRAP), plasma phenolics content, superoxide dismutase(SOD) activity, serum protein carbonyl and thiobarbituric acid reactive substances(TBARS) levels were examined. Total phenolic compound and total flavonoid levels in freeze-dried cranberry powder were 9.94 mg/g and 8.12 mg/g, respectively. Serum total cholesterol and LDL-cholesterol levels were not significantly different for cranberry powder treatment, but serum HDL-cholesterol level was significantly increased in HFC+C5 group compared with HFC group. Plasma FRAP value tended to be increased by cranberry powder treatment though there was no significant difference. Plasma total phenol concentrations and SOD activities were not significantly different among all groups. Serum protein carbonyl and TBARS levels were significantly decreased in HFC+C5 group compared with HFC group. Overall results suggested that freeze-dried cranberry powder might have the serum lipid improving effect, as well as anti oxidative effect demonstrated by its protective effect against protein and lipid oxidation.
