Protective Effect of Tongyuhwalhyeol-tang on Liver Injury in Thioacetamide-induced Rat (Thioacetamide 유발 간손상모델에서 통규활혈탕의 간보호효과)

  • Objectives : Liver disease is an inflammatory reaction caused by oxidative stress, viral, alcohol, and drug properties. Inflammatory reaction causes hepatitis and chronic hepatitis is persistent, it progresses to liver fibrogenesis and liver cancer. The aim of this study was to confirm the hepatoprotective effect of Tongyuhwalhyeol-tang(Tongqiaohuoxue Decoction) (TH) and Gamtongyuhawlhyeol-tang(GTH) in TAA-induced liver injury animal model. Methods : The antioxidant activities were evaluated through in vitro experiments, such as 1, 1-diphenyl-2-picrylhydrazyl (DPPH) and 2, 2'-azinobis-3-ethyl-benzothiazoline-6-sulfonic acid (ABTS) radical scavenging assays, total polyphenol and total flavonoid content measurement. To confirm the liver protective effect, induced by Thioacetamide (TAA) for 3 days injection at 200 mg/kg rats. TH and GTH were treated 3 days at 200 mg/kg/day. The changes of reactive oxygen species (ROS), peroxynitrite ($ONOO^-$), alanine aminotransferanse (ALT) and aspartate aminotransferase (AST) in serum were analyzed after experiment. Also, expression of anti-inflammation, anti-oxidant related proteins were investigated by western blot analysis. Results : TH was inhibited the antioxidant activities. In the TAA-induced rat, TH decreased ROS, $ONOO^-$, ALT, AST level in serum. Inflammation related protein expressions increased in TAA-induced rat compared to normal rat. However, TH group inhibited the down expression of these proteins. Also, anti-oxidant related protein expressions increased in TH group compared TAA-induced rat. Conclusion : Therefor, these results suggested that TH provided hepatoprotective effects on the hepatic injury leading to the reduction of inflammatory response. In addition, the effect of TH was superior to that of GTH.

The Anti-oxidant and Anti-microbial Activities of Purified Syringin from Cortex Fraxini (진피로부터 정제한 Syringin의 항산화 및 항균 활성)

  • The purpose of this study is to evaluate the anti-oxidant and anti-microbial activity of syringin isolated from Cortex Fraxini to investigate their potential for use as safe natural compounds. Purified syringin was dissolved in distilled water for each concentration and used in each experiment. Syringin showed higher 2,2-Diphenyl-1-picrylhydrazyl radical scavenging than butylated hydroxytoluene (BHT) at a concentration of 50 ㎍/ml. In 2,2'-Azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging, activity was similar to that of BHT at all concentrations. In antioxidant protection factor measurement, activity of syringin slightly increased as the concentration increased, as did the inhibitory effect of thiobarbutric acid reactive substances. In evaluating anti-microbial activity, the clear zones of Listeria monocytogenes KCTC 13064, Staphylococcus aureus KCTC 1916, Escherichia coli KCTC 2571, and Helicobacter pylori HPKCTC B0150 at a concentration of 200 ㎍/ml were found to be 17.8 mm, 20.45 mm, 17.05 mm, and 16.8 mm, respectively, but no clear zone was observed in the case of Candida albicans ATCC 10231. The activity against water-soluble antioxidants was therefore superior to that against lipid-soluble antioxidants. Anti-microbial activity was examined by inhibiting growth against gram-positive and -negative strains, and anti-fungal activity was not observed. Based on the results of this study, syringin has possible applications as a natural anti-oxidant and anti-microbial material.

Anti-oxidant and Anti-inflammatory Effects of Ethanol Extracts from Aerial Part of Coriandrum sativum L. (고수(Coriandrum sativum L.) 지상부 추출물의 항산화 및 항염증 활성 효과)

  • Coriandrum sativum L., an annual herbaceous plant of Apiaceae family. The present study evaluated the anti-oxidant activities and anti-inflammatory effects of ethanol extracts of C. sativum. The anti-oxidant activities of C. sativum were measured by total contents of polyphenol, flavonoid, DPPH and ABTS radical scavenging and reducing power activity. And anti-inflammatory effects of C. sativum were measured by LPS-induced RAW 264.7 cells. The results showed that the contents of total polyphenol and flavonoid were 76.03 ± 1.36 mg of gallic acid equivalents/g and 182.23 ± 4.32 mg of rutin equivalents/g at concentration 1 mg/mL of C. sativum. The DPPH radical scavenging activity was found to be 52.8% at 500 ㎍/mL. The ABTS radical scavenging activity was shown in 58.3% after exposure to 1,000 ㎍/mL. Reducing power activity was found to be 66.8% at 2,000 ㎍/mL. The inhibitory effect of NO production was found to be 65% concentration 500 ㎍/mL. In the generation quantity of inflammatory cytokines such as TNF-α and IL-1β in cell culture medium, the expression levels of inflammatory proteins in cells were showed decrease with the increase of concentration. Therefore, we suggest that the C. sativum should be a potential source of alternative anti-inflammatory drug with good anti-inflammatory effects.

Comparative study of the biological activities effect of Mentha arvensis L. extracts from water and 80% ethanol (열수 및 80% 에탄올로 추출한 박하의 생리활성 비교연구)

  • The purpose of this study was to investigated the biological activities such as cytotoxic, anti-oxidant and anti-inflammatory using Mentha arvensis L. extracts. Mentha arvensis L. was prepared by extracting with DW and 80% ethanol, after cell viability was assessed by MTT assay using RAW 264.7 cells. Antioxidant activities, and Anti-inflammatory activities measured through changes in the levels of reactive oxygen species (ROS), nitric oxide (NO), leukotrien B4 (LTB4), and anti- or in-flammatory cytokines($IL-1{\beta}$, IL-6, tumor necrosis factor $(TNF)-{\alpha}$, and IL-10) on LPS-induced RAW 264.7 cells. All test measured by ELISA reader and Luminex. Mentha arvensis L. was no cytotoxic in water and 80% ethanol extracts, and Concentration of 100 ug/ml of 80% ethanol extract was suppressed the productions of ROS in LPS-induced RAW 264.7 cells. Also, Productions of NO, LTB4, inflammatory or anti-inflammatory cytokines showed efficacy change that dose-independent of all extracts. In other words, Mentha arvensis L. showed significant biological activities showing anti-inflammatory, and anti-oxidant. These results may be developed as a raw material for new health food and therapeutics to ease the symptoms mentioned above.

Effects of Salvia plebeia Herb Extracts on Anti-oxidant Activity and Whitening action (곰보배추 추출물이 항산화 및 피부미백효과에 미치는 영향)

  • Whitening and anti-oxidant effects were observed in order to investigate the biological activations of Salvia plebeia herb ethanol extracts. No toxicity was found in both B16F10 melanoma cells and Raw 264.7 cells exposed to Salvia plebeia herb ethanol extracts for 48 hour. The extracts showed significant antioxidant activity in cell-free and cell-cultured system. In the DPPH radical assay, it removed dose-dependently DPPH radicals and showed 77.6% at $100{\mu}g/mL$. In the cells, it also significantly removed silica-induced ROS generation and LPS-induced NO production in a dose dependent manner. Using L-DOPA and L-tyrosine as a substrate, tyrosinase activity was inhibited using Salvia plebeia herb ethanol extracts in a dose-dependent manner. The supression occurred to be in the B16F10 melanoma cells, where dose-dependently inhibited Salvia plebeia herb ethanol extracts of $1{\mu}g/M$ ${\alpha}$-melanocyte stimulated hormone-induced melanin production and the inhibitory effect was 30.7% at a concentration of $100{\mu}g/mL$. This suggests that the Salvia plebeia herb ethanol extracts are usable for cosmetic product developments for anti-oxidant and whitening effects.

  • Effect of dietary 2.0 % brown seaweed(Undaria pinnatifida) with bean extract on anti-oxidant system and TNF-$\alpha$ levels were evaluated in blood of 2 week-old broiler chicks activated innate immune response. Dietary brown seaweed and activation of innate immune response decreased MnSOD activities. while activation of innate immune reponse only increased CuZnSOD activities in erythrocyte cytosol. Activation of innate immune response lowered plasma SOD activity in birds fed seaweed with bean extract, increased peroxide levels, and decreased peroxidase activity in plasma. Brown seaweed with bean extract reduced TNF-$\alpha$ levels and increased ovotransferrins concentrations in plasma. The result indicated that dietary 2.0 % brown seaweed with bean extract affect innate immune response changing anti-oxidant system and TNF-$\alpha$ levels in broiler chicks.

  • The anti-oxidant components in Panax ginseng, which were known to have antiaging effect, were fractionated by various solvents and then isolated by the preparative thin layer chromatography. A developing system with silicagel G using toluene-chloroform-actene (20 : 13 : 13) as mobile phase was applied. The components were investigated the anti-oxidant activity by ${\alpha}$, ${\alpha}'-diphenyl-{\beta}-picrylhydrazyl$ (DPPH) at 517nm. Two effective components were obtained from red ginseng and were indophenol reducing substances.

The Effects of Anti-Oxidant and Phototoxic Inhibitory Activity of Rubus coreanus Fruits (복분자의 항산화 및 광독성 억제활성에 미치는 영향)

  • Objectives : Oxidation by active oxygen in the body, phototoxicity and photoallergic interest has grown, antioxidant and phototoxicity inhibiting substances for research progress. The purpose of this study was to examine the effects on antioxidant and phototoxicity of Rubus coreanus Fruits. Meterial and Methods : Hs68 cell lines using the DPPH radical scavenging activity, cytotoxic, phototoxic inhibitory activity and apoptosis were measured. Results : 1. In MTT assay, the concentrations of Rubus coreanus Fruits that were used on the test had no cytotoxicity. 2. In DPPH radical scavenging activity, the concentration of $50{\mu}g/ml$, $100{\mu}g/ml$ anti-oxidant effect of Rubus coreanus Fruits was statistically significant increased than control group in dose-dependantly. 3. In phototoxic inhibitory activity, Rubus coreanus Fruits dose-dependently increased the cell viability of Hs68 cell lines. 4. The concentration of $50{\mu}g/ml$ Rubus coreanus Fruits inhibited the enrichment of nucleus in the Hs68 cell stimulated with UVB. Conclusions : These results indicate that Rubus coreanus Fruits has anti-oxidant effects and Phototoxic Inhibitory Activity. If further study is performed, the use of Rubus coreanus Fruits will be valuable and beneficial in the therapy of skin aging and damage.

Effect of methyl donors supplementation on performance, immune responses and anti-oxidant variables in broiler chicken fed diet without supplemental methionine

  • Objective: Methionine (Met) is involved in methyl group transfer besides protein synthesis. As the availability is limited and cost is high for synthetic Met, reductions in its inclusion in broiler diet may be possible by supplementing the low Met diets with methyl donors (MD) like betaine (Bet), folic acid (FA), vitamin B12 (B12), and biotin (Bio). An experiment was conducted to study the effects of supplementing the MD on performance (average daily gain [ADG], daily feed intake, feed efficiency [FE]), anti-oxidant variables, immune responses and serum protein concentration in broilers fed sub-optimal concentrations of dietary Met. Methods: Maize-soybean meal diet was used as control (CD). Different MD like Bet (0.2%), B12 (0.1 mg), FA (4 mg), or Bio (1.5 mg/kg) were supplemented to basal diet (BD) having no supplemental Met. The BD without MD was kept for comparison. Each diet was fed ad libitum to 10 replicates of 25 chicks in each from 1 to 42 d of age. Results: At the end of experiment, the ADG in MD group was higher than BD and lower than CD. The FE improved with FA or Bet compared to the BD. Breast meat weight was higher in Bet compared to the BD, while it was intermediate between BD and CD in other groups. The lipid peroxidation reduced with Bio, B12, or Bet, while the glutathione peroxidase activity improved with Bio or B12 compared to the BD. Lymphocyte proliferation improved with Bet compared to the BD. The serum protein concentrations increased with FA, Bio, or Bet compared to those fed BD. Conclusion: It can be concluded that the ADG can be improved partially with supplementation of MD while the FE improved with FA or Bet. Some MD also reduced the stress indices and improved immune responses compared to the BD fed broilers.

Effect of Anti-oxidant, Anti-inflammatory and Anti-invasive of PMA-induced Matrix Metalloproteinase (MMP-2) and MMP-9 Activities of Water Extract and Solvent Fractions of Saururus Chinensis (삼백초 물 추출물과 유기용매 분획물의 항산화, 항염증 및 PMA에 의해 유도된 MMP-2 및 MMP-9활성 침윤 억제 효과)

  • Saururus chinensis is a perennial plants, its flavonoid compound is known to exhibit anti-oxidative activity. This study was aimed to investigate the effect of Water Extract and Solvent Fractions of Saururus chinensis on antioxidant, anti-inflammatory and anti-invasive of Phorbol 12-myristate 13-acetate (PMA)-induced matrix metalloproteinase (MMP-2) and MMP-9 activities. Plant samples were fractionated into hexane, CHCl3, ethyl acetate, butanol, and water fractions, and each of these was assayed individually. The water fraction showed the highest extraction yield at 9.25%(w/w). Anti-oxidative activity was analyzed by DPPH assay. Cell viability was detected by the MTS assay. Anti-inflammatory activity was assayed by the nitric oxide (NO) production in mouse macrophage Raw 264.7 cells. The activity and mRNA expression of MMP-2 and MMP-9 in human oral squamous carcinoma YD-10B cells were examined by zymography and RT-PCR. As results, MMP-2/-9 activation was increased in PMA induced YD-10B cells. In PMA-treated YD-10B cells, the increased mRNA expression and protein activation of MMP-2/-9 were significantly inhibited in the ethyl acetate fraction. The ethyl acetate fraction showed the highest anti-oxidative activity at 73.38%. The ethyl acetate fraction at non-cytotoxic concentrations significantly exhibited the anti-inflammatory activity of Raw 264.7 cells in dose-dependent manner. In conclusion, these findings demonstrate that the ethyl acetate fraction obtained from a chinensis water extract potentiates a promising therapeutic anti-invasive agent and, therefore, as an anti-cancer drug for cancer prevention and therapy in oral cancer.
