Objectives : Hyeonggaeyeongyotang Gagambang (HYT) is a herbal medicine prescribed for the treatment of inflammatory diseases, but it is necessary to study the exact therapeutic efficacy. This study aims to investigate the antibacterial and anti-inflmmatory activities of HYT. Methods : Antibacterial activity of HYT was confirmed by staining Escherichia coli, a gram negative strain, and Staphylococcus aureus, a gram positive strain, on solid Lysogeny Broth (LB) medium containing HYT. Antioxidant activity of HYT was confirmed by 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) assay. The phosphorylation of nuclear factor of kappa light polypeptide gene enhancer in B-cells inhibitor, alpha ($I{\kappa}B{\alpha}$) after lipopolysaccharide (LPS) treatment with HYT-treated RAW 264.7 mouse macrophages cells was confirmed by immunoblot analysis and the level of interleukin 1 beta (IL-$1{\beta}$) mRNA expression level was confirmed by quantitative real-time PCR. Results : HYT showed a concentration-dependent antibacterial activity against Escherichia coli and Staphylococcus aureus and also showed excellent antioxidant activity. HYT treatment attenuated the phosphorylation of $I{\kappa}B{\alpha}$induced by LPS treatment in RAW 264.7 mouse macrophages cells. The phosphorylation of $I{\kappa}B{\alpha}$is crucial for the regulation of the expression of various pro-inflammatory cytokines. In addition, IL-$1{\beta}$ mRNA expression level of RAW 264.7 mouse macrophages cells stimulated by LPS treatment was also inhibited by HYT treatment. Conclusions : Through experimental demonstration of the antioxidative, antimicrobial and anti-inflammatory effects of HYT, we demonstrated that HYT is a herbal medicine effective for the treatment of inflammatory diseases caused by various bacterial infections.
한약재로부터 장내의 대표적 유해균인 Clostridium perfringes의 생육을 억제시키는 소재를 탐색한 결과 방기 물추출물에서 강력한 항균활성을 발견하였다. 방기추출물의 항균활성은 pH1부터 pH13까지의 범위에서 안정하였으며 121'C에서 15분간의 열처리 에서도 활성이 유지되었다. 방기 추출물은 Cl. perfringens, Cl.paraputrificum, Cl.ramosum 및 Cl.butyricum 등 실험에 사용한 모든 Clostridium속균의 생육을 강력하게 억제하였다. 그리고 Clostridium 이외의 주요 장내균에 대한 영향을 조사한 결과 Bif.bifidium, Bacteroides fragilis 및 Eubacterium limosum에 대해서는 역시 그 생육을 억제하였으나, Bif. adolescesnits, Bif.infantis, Bif.longum, E.cloi, Enterococcus faecalis 그리고 Staphylococcus aureus에 대해서는 생육억제 효과가 크게 나타나지 않았다. 반면 Lactobacillus acidophilus와 Bif.animalis에 대해서는 생육촉진효과를 보여주었다.
Objectives : We aimed to determine therapeutic effects of probiotics-fermented Magnolia denudata(MD) in the allergic rhinitis model mice. Methods : Polyphenol production, DPPH radical scavenging activity and NO inhibition of fermented MD by different bacterial strains were evaluated to select the one that is most suitable for fermentation. Thirty C57BL/6 mice were divided randomly into 5 groups as follows: normal group, ovalbumin(OVA)-treated plus water fed(CON group), OVA-treated plus unfermented MD fed(UMD group), OVA-treated plus fermented autoclaved MD fed(A-FMD group) and OVA-treated plus fermented unautoclaved MD fed(FMD group). After 9 weeks, we observed changes in the blood cell count, OVA-specific IgE level, nasal rubbing, nasal mucosal tissue and body weight. Results : Extract of MD fermented by Bifidobacterium breve(BB) for 48 hours showed the highest anti-oxidant activity and anti-inflammatory activity out of all the other bacterial strains. The number of eosinophil count in A-FMD, FMD group and platelet count in FMD group showed statistically significant decrease(p<0.05). OVA-specific IgE level decreased in all 3 experimental groups, significantly in UMD and A-FMD group. Nasal symptoms were attenuated in all 3 experimental groups, statistically significant in A-FMD and FMD group (p<0.05). Histologically, infiltration of eosinophils into the nasal mucosa decreased in all 3 experimental groups, especially marked decrease in FMD group. Conclusions : According to the above results, it is considered that probiotics-fermented Magnolia denudata has inhibitory effects on the allergic rhinitis animal models.

Ginseng (the root of Panax ginseng C. A. MEYER, Araliaceae) has been used for traditional medicine in China, Korea, Japan and other Asian countries for the treatment of various diseases including psychiatric and neurologic diseases as well as diabetes mellitus. So far, ginseng saponins (ginsenosides) have been regarded as the principal components responsible for the pharmacological activities of ginseng. Ginsenosides are glycosides containing an aglycone (protopanaxadiol or protopanaxatriol) with a dammarane skeleton and have been shown to possess various biological activities including the enhancement of cholesterol biosynthesis, stimulation of serum protein synthesis, immuno- modulatory effects and anti-inflammatory activity. Several studies using ginsenosides have also reported anti-tumor effects, particularly the inhibition of tumor-induced angiogenesis, tumor invasion and metastasis, and the control of phenotypic expression and differentiation of tumor cells.
The probiotic characteristics of a total of 137 lactic acid bacterial strains isolated from soybean paste were investigated. Among those tested, the D37 strain was selected as a probiotic bacteria due to its acid and bile tolerance, and its strong anti-cancer and antibacterial activities. The D37 strain showed highly stable viability at acidic pH for 2 hr, and was very stable in 10% bovine bile. The viability of human colon cancer HT-29 cells was inhibited more than 60% at a $200\;{\mu}/mL$ concentration of D37 cell-free culture supernatant, and the degree of inhibition was concentration-dependent. The D37 strain showed a wide range of antibacterial activities against food-borne pathogenic bacteria such as Escherichia coli O157, Listeria spp., Vibrio spp., Salmonella spp., and Staphylococcus aureus. According to phenotypic characteristics and the utilization of various sugars, the D37 strain was identified as Lactobacillus cellobiosus.
The purpose of this study was to evaluate the biological activities of extracts of ramie (Boehmeria nivea (L.) Gaud.), hereafter referred to as Bn. Bn extracts from various collecting area were extracted with methanol. Two extracts from our study, Bn-13 and -82, showed significant antioxidant properties, likely due to their ability to scavenge free radicals. In addition, Bn extracts showed stronger anti-bacterial activity against Escherichia coli (Bn-40), Stapylococcus aureus (Bn-33), and Helicobacter pylori (Bn-05). In addition, this study was conducted to evaluate the anti-inflammatory effects of Bn extracts in lipopolyssacharide (LPS)- and interferon-${\gamma}$ (IFN-${\gamma}$)-stimulated RAW 264.7 macrophages cells. Bn-37 significantly inhibited the production LPS/IFN-${\gamma}$-induced nitric oxide. The most noteworthy anti-cancer effect was found in Bn-23. Bn-08 showed inhibition of aldose reductase. This study provides basic information for the development of functional foods.
Staphylococcal enterotoxin B (SEB) is bacterial toxin that induces the activation of immune cells. Because the inhibition of pro-inflammatory effect of SEB can resolve the inflammation, I determined the influence of functional or structural change of SEB on immune cells. The post translational modification of protein occurs through carbamylation. Carbamylation can change the structure of proteins and can modify the biological activity of protein. In the present study, I investigated the effect of carbamylated SEB (CSEB) on the inflammatory response mediated by LPS in HL-60 cells. To determine the anti-inflammatory effect of CSEB, I produced carbamylated SEB using potassium cyanate (KCN) and then examined whether CSEB involved in cytokine releases and apoptosis of LPS-stimulated HL-60 cells. Although CSEB had not any effect on the LPS-stimulated HL-60 cells, the protein levels of IL-8, TNF-${\alpha}$ and IL-$1{\beta}$ were significantly decreased by CSEB without cytotoxicity. CSEB also blocked Akt and NF-${\kappa}B$ activation. These results indicate that the suppressive effect of CSEB in LPS-stimulated cytokine releases is occurred by inhibition of Akt and NF-${\kappa}B$ activity. Through further studies, CSEB may be used as anti-inflammatory molecule that makes the immune system more efficient.
천연물 유래 난치성 피부질환 기능성 소재를 탐색하기 위해 느릅나무 가지를 초임계 유체 기술을 활용하여 추출을 수행하였고, 확보된 느릅나무 초임계 추출물을 수종의 피부상재균에 대하여 항균력 실험을 실시하였고, 모든 균종에 대해서 항균력이 확인이 되었으며, ABTS 라디컬 소거능과 NO 소거능 실험을 통해서 느릅나무 초임계 추출물의 항산화 활성과 항염증 활성의 우수한 활성이 확인이 되어서 산화적 스트레스 질환과 염증성 피부질환에 대해서 예방 및 치료 보완 기능성 소재로 개발이 가능성이 높다는 것을 알 수 있었다. 결론적으로 느릅나무 초임계 추출물은 피부상재균에 대한 항균활성, 라디컬 소거능에 의한 항산화 활성, 과 생성된 NO 소거능에 의한 항염증활성이 각각 확인이 되었다. 이 상의 결과로 느릅나무 초임계 추출물은 항균효능, 항산화효능, 항염증 효능을 갖는 기능성 원료로서의 가능성을 확인할 수 있었고, 향후 만성 염증성 피부면역 질환 개선 및 치료 보조제 관련 의약품 또는 기능성 향장품 개발을 위한 기초 연구 자료가 될 것으로 사료된다.
본 연구의 목적은 황백을 포함한 총 14가지의 한약재를 증류 추출하여 얻은 시료(A.C.C. 추출물)의 항염증 활성을 확인하고 임상적인 효과를 평가하는 것이다. 이를 위해, Lipopolysaccharide(LPS)로 자극한 RAW 264.7세포에서 세포가 방출하는 nitric oxide(NO) 생성량과 염증성 사이토카인인 tumor necrosis factor(TNF)-${\alpha}$, interleukin(IL)-$1{\beta}$ 및 IL-6 생성량의 변화를 확인하였다. 그 결과, A.C.C. 추출물은 세포 독성없이 LPS가 증가시킨 NO와 염증성 사이토카인들의 생성을 강력하게 억제하였다. 또한, A.C.C. 추출물은 Pseudomonas aeruginosa, Staphylococcus aureus, MRSA(Methicillin-resistant Staphylococcus aureus), Candida albicans and Streptococcus mutans 등의 세균들에서 99.9%의 강력한 세균 감소율을 보여 주었다. 이는 A.C.C. 추출물이 항염증 효과와 함께 강력한 항균 효과를 가진 유효한 성분인 것을 의미한다. 또한, 기저귀 발진, 가려움증, 땀띠 증상을 겪고 있는 영 유아에게 A.C.C. 추출물을 도포하였을 때, 발진, 아토피, 짓무름, 가려움증, 땀띠 등의 증상 개선 효과를 나타내었으며, 영 유아 피부의 땀띠가 2주일 경과 후 상당히 완화된 것을 육안으로 확인할 수 있었다. 이는 A.C.C. 추출물이 임상적으로도 항염증 및 항균작용을 통하여 기저귀 발진 및 땀띠 등의 증상을 완화 시키는 물질임을 확실히 한 것이다. 그러므로, A.C.C. 추출물이 염증성 질환에 대한 효과적인 대안이 될 수 있을 것으로 사료된다.
The MeOH extract of seeds of Cerbera odollam Gaertn. (Apocynaceae) was screened for its antibacterial, cytotoxic and neuropharmacological activities. The extract showed moderate anti-bacterial activity against Salmonella typhi, Streptococcus saprophyticus, and Streptococcus pyogenes. It exhibited high level cytotoxicity against brine shrimp ($LC_{50}$: $3\;{\mu}g/ml$). The extract potentiated pentobarbital induced sleeping time in mice which was further supported by the exploratory behavior test at dose of 25 mg/kg. The overall results tend to suggest the antibacterial, cytotoxic and CNS depressant activities of the extract.
