Study on the Anti-inflammatory Effects of Soyangin-Hyeongbangpaedok-san (소양인 형방패독산의 함염증효과에 대한 실험적 연구)

  • Soyangin-Hyeongbangpaedok-san(SHBPDS) is used for treating upper respiratory infections, In an effort to investigate the anti-inflammatory effects of SHBPDS, we measured production of several cytokines and immunoglobulin in various immune cells. SHBPDS decreased the secretion of TNF-${\alpha}$, but not that of IL-6 in PMA/A23187 stimulated HMC-1 cells. As for mouse B cells, it induced proliferation and caused differential effects in expressions of surface IgE as determined by flow cytometry and secretions of IgE, IgG1, ILA and INF-${\gamma}$as measured by ELISA but showed little change in CD23 or CD69 expression. SHBPDS increased proliferation in anti-CD3/anti-CD28 stimulated CD4 Th cells. Under the Th1/Th2 polarization conditions, SHBPDS at 200 ${\mu}g/m{\ell}$ suppressed the secretion of INF-${\gamma}$ and IL-4. Based on the above results, we conclude that SHBPDS has antiinflammatory activities in mast cells and different immunomodulatroy effects in B cells and Th cells.

Effects of Saposhnikoviae Radix on allergic responses in OVA-induced Allergic rhinitis mice (방풍(防風)의 난알부민 유도 알레르기 비염 마우스에서의 항알레르기 효능 연구)

  • Objectives : In this study, we investigated the effect of Saposhnikoviae Radix on allergic responses in ovalbumin(OVA)-induced Allergic rhinitis(AR) mice. Methods : BALB/c mice were orally administrated with Saposhnikoviae Radix water extract (SRW, 50 mg/kg) or anti-histamine drug, Ketotifen (10 mg/kg) as a reference drug, followed by sensitization and challenge of OVA. Mice were measured clinical symptoms and the serum levels of histamine, IgE, IL-4, and IFN-${\gamma}$, and observed histopathological changes of nasal mucosa H&E staining. Results : SRW significantly decreased rubs and the serum levels of histamine, IgE, and IL-4, and then increased the serum levels of IFN-${\gamma}$ in OVA-induced AR mice, and inhibited histopathological changes of nasal mucosa with inflammation and the eosinophils infilteration. Conclusions : These data suggest that SRW has anti-allergic effect through the inhibitory property of Saposhnikoviae Radix against allergic responses in allergic rhinitis.

Effects of coffee intake on airway hypersensitivity and immunomodulation: an in vivo murine study

  • BACKGROUND/OBJECTIVES: Coffee is a complex chemical mixture, with caffeine being the most well-known bioactive substance. The immunomodulatory and anti-inflammatory properties of coffee and caffeine impact health in various aspects, including the respiratory system. The objective is to investigate the effects of coffee and caffeine on airway hyperresponsiveness and allergic reactions, as well as to analyze and compare associated cytokine profiles. MATERIALS/METHODS: BALB/c mice were intraperitoneally sensitized with ovalbumin (OVA) and given OVA inhalation to induce airway hypersensitivity. Two weeks after sensitization, they were intragastrically gavaged with coffee or caffeine, both containing 0.3125 mg caffeine, daily for 4 weeks. Control mice were fed with double-distilled water. Serum OVA-specific antibody levels were measured beforehand and 5 weeks after the first gavage. Airway hyperresponsiveness was detected by whole body plethysmography after gavage. Cytokine levels of bronchoalveolar lavage and cultured splenocytes were analyzed. RESULTS: Coffee effectively suppressed T helper 2-mediated specific antibody response. Airway responsiveness was reduced in mice treated with either coffee or caffeine. Compared to the control, coffee significantly reduced OVA-specific immunoglobulin (Ig) G, IgG1 and IgE antibody responses (P < 0.05). Caffeine also attenuated specific IgG and IgG1 levels, though IgE level was unaffected. Coffee significantly reduced interleukin (IL)-4 and increased IL-10 concentration in spleen cells and bronchoalveolar lavage fluid (P < 0.05). CONCLUSIONS: Coffee effectively attenuated airway hyperresponsiveness and systemic allergic responses induced by OVA food allergen in mice. As a complex composition of bioactive substances, coffee displayed enhanced immunomodulatory and anti-inflammatory effects than caffeine.

The Convergence Study on the Effects of White Ginseng Complex Extracts on OVA-induced Allergic Asthma in Mice (백삼복합물이 난알부민으로 유도된 천식 마우스에서의 천식개선에 대한 융복합 연구)

  • The aim of the convergence study is to evaluate anti-asthma effects of white ginseng complex extract(WGCE) on OVA-induced allergic asthma in mice. WGCE was administered at 100 mg/kg and 300 mg/kg to mice, where asthma was induced by OVA. Th2 cytokine including IL-4, IL-5 and IL-13 were measured by Luminex. Also, OVA-IgE and eosinophil were measured by haemocytometer and BALF total cells were measured microscope. Production of IL-4, IL-5, IL-13 and OVA-IgE in serum was decreased, respectively, in comparison with control. The eosinophil in whole blood decreased significantly. In addition, WGCE groups showed a decrease in the BALF total cells. These results demonstrated that WGCE decreases the Th2 cytokine and asthma factors. Therefore, we strongly suggest that WGCE could be effectively used as a therapeutic drug based on its anti asthma factors.

Anti-acne and Anti-atopic Dermatitis Effect of Plant Extracts Including Eucommia ulmoides Oliv and Phellodendron amurense (두충나무, 황벽나무 등을 포함하는 수목추출물의 항여드름 및 항아토피 효과)

  • In this study, plant extracts including Eucommia ulmoides Oliv. and Phellodendron amurense were studied to test possible application for cosmetics and skin related medicine. Anti-oxidation effect of plant extracts was measured by DPPH free radical scavenging activity and it was insignificant at low concentration, however, it was as good as vitamin C, excellent anti-oxidation agent, at $1000{\mu}g/ml$. Anti-bacterial effect was tested by disc diffusion method, and plant extracts showed mild anti-bacterial effect for normal skin flora, Staphylococcus epidermidis while it indicated strong anti-bacterial effect for acne inducing Propionibacterium acne. Therefore it had powerful potential for anti-acne material because of selectivity. Anti-atopic dermatitis effect was tested by hairless mouse and plant extracts recovered damaged skin to near normal condition after 14 days of treatment. IgE concentration in treated mouse was decreased 16% compared with control. From the research, plant extracts indicated strong anti-acne and anti-atopic dermatitis effect, and showed strong potential for cosmetics and skin related medicine.

The Experimental Stydy on the Anti-Allergic Rhinitis Effects of the Sochungryong-tang (小靑龍湯이 알레르기 鼻炎에 미치는 效果에 대한 實驗的 硏究)

  • Allergic rhinitis is an inflammation of the nasal mucosa which is triggered by an allergic reaction. When exposed to certain allergens, the IgE covered mast cells degranulate releasing inflammatory mediators and cytokines which results in a local inflammatory reaction. In many recent studies, molecular biological methods have been used to investigate the role of cytokines in pathogenesis and new therapeutic targets of allergic rhinitis. This experimental study was done to research effects of Sochungryong-tang. We have studied effect of mice on OVA-induced production of IL-4, IL-5, IFN-${\gamma}$ by murine splenocytes and effect of OVA-induced total IgE and OVA-specific IgE. The results were as follows; 1. In IL-4 study, Sochungryong-tang treated group was proved significant inhibitory effect(p〈0.005). 2. In IL-5 study, Sochungryong-tang treated group was proved significant inhibitory effect(p〈0.05). 3. In JFN-${\gamma}$ study, Sochungryong-tang treated group was proved significant inhibitory effect(p〈0.000001). 4. In Total IgE, Sochungryong-tang treated group was proved significant inhibitory effect(p〈0.000001). 5. In OVA-specific IgE, Sochungryong-tang treated group didn't showed significant inhibitory effect. Depending upon above results, it is considered that Sochungryong-tang has the inhibitory effect on the allergic rhinitis of mice and suggested that it could be used in relieving patients of the symptoms which are caused by allergic rhinitis.

Comparison on anti-allergic activities of leaves from Domestic and Chinese species of Cudrania tricuspidata (토종과 중국도입종간 꾸지뽕잎의 항알레르기작용 비교)

  • Objectives : This study was performed to compare the effect of two Cudrania tricuspidata cultivars; Sancheong native (CT) and varieties in china(SCT) on immediate hypersensitivity of the anaphylactic type and Ova-induced allergic asthma mouse model by calculating serum cytokines and IgE. Methods : We investigated the free radical scavanging effect and quantify total phenol contents and total flavonoids of two Cudrania tricuspidata cultivars; Sancheong native(CT) and varieties in china(SCT). The sample was extracted by 80% EtOH. To induce the allergic asthma, in the control group and the CT group, mice of each group were sensitized intraperitoneally with ovalbumin (OVA) solution at the 1st, the 7th and the 14th day. After then, sensitization was performed by aerosol allergen challenges with 1% OVA solution intratracheally at the 21th, the 23th, 25th and the 27th day. At the 29th day, the mice were killed and the changes of interferon-${\gamma}$, interleukin-4, 5 and 10, total IgE and OVA-specific IgE in serum were checked. Results : CT inhibited compound 48/80-induced systemic anaphylaxis 90% with a dose of 100 mg/kg body weight at 1 hr before injection of compound 48/80. In the allergic asthma mouse model, IFN-${\gamma}$ was did not increased in the CT and SCT group than that in the control group. IL-4, IL-5, the total IgE and OVA-specific IgE were decreased in the CT group as compared with the control group and these results were statistically significant. Conclusions : Considering the above experimental results, this study showed that Sancheong native cultivar could reduce the allergic reaction.

Anti-allergic Effect of Ethanolic Extract of Flos Sophora japonica L. on Ca++ Ionophore Stimulated Murine RBL-2H3 Cells (괴화(槐花) 에탄올 추출물이 RBL-2H3 비만세포에서 Ca++ Ionophore에 의한 알레르기 반응 조절에 미치는 효과)

  • Elevation of intracellular calcium ($Ca^{{+}{+}}$) triggers degranulation of mast cells by bypassing receptor activation. Flos Sophora japonica L. has been used as a natural dying source and has been reported to have biological activities such as anti-inflammatory and anti-allergic effects through $Fc{\varepsilon}RI$ and IgE crosslinking. In the present investigation, we report the regulatory effect of ethanolic extract of Flos Sophora japonica L. (S.F) on allergic mediators produced by $Ca^{{+}{+}}$ ionophore activation in mast cells. S.F significantly inhibited calcium ionophore (A23187)-induced interleukin (IL)-4 and tumor necrosis factor (TNF)-${\alpha}$ production as well as mast cell degranulation. Furthermore, administration of S.F suppressed allergic reactions in a 2,4-dinitrofluorobenzene (DNFB)-induced allergic dermatitis mouse model. Both oral administration and ear painting using 50 mg/kg of S.F significantly reduced levels of cytokines such as IL-4, TNF, and interferon-${\gamma}$ in ear tissues compared to the DNFB alone-treated group. Serum IgE level in the S.F-treated group also decreased compared to the DNFB alone-treated group. Weights of spleens and lymph nodes in the S.F-treated groups also decreased compared to the control group. Considering the data, we conclude that S.F mediates its anti-allergic effects not only through $Fc{\varepsilon}RI$ stimulation but also $Ca^{{+}{+}}$ influx in mast cells.

Suppressive Effects of Korean Indigenous Acanthopanax divaritacus on the Allergic Inflammation

  • The water extracts of root, stem, and leaf from Korean indigenous Acanthopanax divaritacus were examined for their suppressive effects against allergic inflammations such as lipoxygenase activity, ${\beta}-hexosaminidase$ release, inflammatory cytokine production, and serum IgE level. The root extract inhibited the release of ${\beta}-hexosaminidase$, a degranulation marker, from rat basophilic leukemia cells (RBL-2H3) much more potently than the stem and leaf extracts. The root extract also significantly reduced the expression of $TNF-{\alpha}\;and\;IL-1{\beta}$ in the RBL-2H3 cells challenged with antigen. Moreover, there was a significant fall in the serum IgE level by the treatment of the root extract. Taken together, the root extract could be the most potent inhibitor of allergic inflammation, suppressing ${\beta}-hexosaminidase$ release and inflammatory cytokine expression, as well as reducing the rise of serum IgE level.

Inhibitory Effect of Lycopus lucidus on Mast Cell-Mediated Immediate-Type Allergic Reactions (택란의 비만세포 매개 즉시형 알레르기 반응의 억제 효과)

  • The effect of aqueous extract of Lycopus lucidus Turcz. (Labiatae)(LLAE) on mast cell-mediated immediate-type allergic reactions was investigated. LLAE (0.01 to 1 mg/g) dose-dependently inhibited systemic anaphylaxis induced by compound 48/80. LLAE (0.001 to 1 mg/g) also dose-dependently inhibited local anaphylaxis activated by anti-dinitrophenyl (DNP) IgE. When LLAE was pretreated at the same concentration with systemic anaphylaxis, serum histamine levels were reduced in a dose-dependent manner. LLAE (0.001 to 1 mg/mι) dose-dependently inhibited histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80. The level of cAMP in human mast cell line (HMC-1) cells, when LLAE (1 mg/mι) was added, significantly was increased, compared with that of normal control. These results provide evidences that LLAE may be beneficial in the treatment of allergic diseases.
