Effect of probiotics Lactobacillus paracasei GKS6, L. plantarum GKM3, and L. rhamnosus GKLC1 on alleviating alcohol-induced alcoholic liver disease in a mouse model

  • BACKGROUND/OBJECTIVES: Heavy alcohol consumption causes the development of alcoholic liver disease (ALD), a neglected but important public health problem. Many studies have pointed out that probiotics could improve gut health, which is also considered to be a cause of ALD. Therefore, this study screened the probiotics, Lactobacillus casei GKC1 (GKC1), L. fermentum GKF3 (GKF3), Bifidobacterium lactis GKK2 (GKK2), L. rhamnosus GKLC1 (GKLC1), L. paracasei GKS6 (GKS6), and L. plantarum GKM3 (GKM3), for their potential benefits in alleviating ALD for applications to disease prevention. SUBJECTS/METHODS: C57BL/6N mice were divided into 8 groups (n = 6 in each): normal control, positive control (alcohol-diet fed), and treatments of feeding probiotics GKC1, GKF3, GKK2, GKLC1, GKS6, and GKM3 under an oral dose 0.82 g/kg B.W. per day by oral gavage. The experiment was conducted for 8 weeks, and the concentrations of alanine aminotransferase (ALT), aspartate aminotransferase, triglyceride (TG), and total cholesterol (TC) in mice were measured. The glutathione (GSH), catalase (CAT), and histology were analyzed after sacrifice. RESULTS: The results showed a decrease in the serum ALT, liver TG, and liver TC levels in the GKS6, GKM3, and GKLC1 groups compared to the positive control. In addition, the decreasing GSH and CAT levels were inhibited in the GKS6 and GKM3 groups. The histopathological results showed that all probiotics could reduce the accumulation of liver fat. Furthermore, there was a significant difference in GKLC1 with lower stomach damage compared to the alcohol-fed mice without any addition of probiotics. CONCLUSIONS: GKLC1, GKS6, and GKM3 can be used as supplements for alleviating the development of ALD.

Bromobenzene의 투여 횟수에 따른 간독성의 차이 (A Study on the Effect of Injection Frequency on the Liver Damage in Rats)

  • 실험동물에 있어서 간조직의 손상 정도에 미치는 xenobiotics의 투여기간에 의한 영향을 검토할 목적으로 hepatotoxin의 일종인 bromobenzene의 투여 횟수에 의한 간손상 정도와 이의 기전을 구명 한 결과는 다음과 같다. 흰쥐에 1일 간격으로 bromobenzene (400 mg/kg)을 복강으로 1회, 3회 및 6회 투여한 실험군을 대상으로 하여 실시한 실험에서 혈청 alanine aminotransferase (ALT)의 활성 및 체중 당 간 무게는 bromobenzene을 1회 투여 한 1일째에는 대조군과 별다른 변동을 볼 수 없었으나 3회 투여 한 3일째에는 유의하게 증가되었으며, 이후 6일째 6회 투여한 실험군에서는 체중 당 간 무게 및 혈청 ALT활성이 3일째 보다 현저히 감소되어 오히려 대조군과 유사하였다. 이와 같이 투여 횟수의 증가에 따른 간손상의 정도가 투여 횟수와 비례하지 않은 것이 어떠한 기전에 의해서 나타나는지를 구명하기 위해 bromobenzene 대사에 관여하는 간조직 중 aniline hydroxylase 및 glutathione S-transferase 활성과 glutathione (GSH) 함량을 측정한 결과, 이들 대사효소 및 GSH 이용률이 bromobenzene 6회 투여군에서 1회 및 3회 투여군 보다 높게 나타났다. 이상 실험 결과를 종합해 볼 때 어떤 독성물질이 생체에 계속 폭로 시 어느 시점에서 중독현상이 경감되는 것은 이 독성물질의 대사율을 증가시켜 해독하려는 생리적응현상이 일어 날 수 있다는 가설을 제시할 수 있다.

한방병원 입원 환자에서 약인성 간 손상 발생율과 약인성 간 손상 환자 치험례 (A Study on the Change of Liver Function Level in Patients Admitted to Korean Medicine Hospital: A Case of One Drug-induced Liver Injury Patient)

  • From January 2014 to December 2016, 38 out of 153 inpatients admitted to the Korean Medicine Hospital for more than one month underwent follow-up blood tests to evaluate their liver function levels, and one drug-induced liver injury (DILI) patient was observed. 1. At the time of admission, six (15.8%) of the 38 patients had abnormal alanine aminotransferase (ALT) levels and five of them recovered after hospitalization. 2. At follow-up, two (5.3%) patients had a Council for International Organizations of Medical Science (CIOMS)-based liver injury. One was identified as a liver injury accompanied by pneumonia, and one was judged as a liver injury caused by the drug. 3. The patient had a suspected DILI and completely recovered after 13 days while discontinuing the herbal medication and by using Sanggangunbi-tang (生肝健脾湯).

어류 혈액 성분의 저장 안정성 (Storage Stability of Blood Constituents in Fish)

  • 조피볼락, 잉어 및 무지개송어의 혈청을 상온$(15^{\circ}C)$ 저온$(4^{\circ}C)$, 냉동$(-20{\circ}C)$으로 보존하면서 총단백질(TP), 알부민(ALB), 중성지방(TG), 콜레스테롤(CHOL), 포도당(GLC), 인(P), 나트륨(Na), AST 및 ALT의 활성 변화를 상온과 저온에서는 16일까지, 냉동에서는 30일까지 조사하였다. 어종간에 다소의 차이가 있기는 하지만 대체로 단백질 성분인 TP, ALB과 당 성분인 GLC, 무기질인 P, Na의 농도는 안정하였고, 지방 성분인 TG와 CHOL, AST, ALT의 활성은 불안정하였다 그리고 온도에 따른 혈액 성분의 안정성은 상온에서보다는 저온과 냉동에서 대체로 안정하지만, 성분에 따라서는 냉동하여도 매우 불안정한 것이 있다

Analysis on the Findings of Serum Biochemical Test in Inhabitants Infected with Clonorchis sinensis

  • The findings of serum biochemical tests in the inhabitants residing in some riverside areas of kyongsangnam-do were analized to know the correlation with the Clonorchis sinensis infection. A total of 2,722 fecal samples of inhabitants was examined by Kato's cellophane thick smear and formalin-ether sedimentation methods, and their serums were analized biochemically. Three hundred and thirty-three inhabitants (12.2%) were positive with C. sinensis eggs (egg-positive group), and remaining 2,389 were negative (egg-negative group). In the egg-positive group, the positive .ate of aspartate aminotransferase (AST) was 6.9% (male: 10.3%; female: 3.2%), that of alanine aminotransferase (ALT) was 6.3% (male: 9.1%; female: 3.2%), of $\gamma$-glutamy1 transpeptidase ($\gamma$-GIP) was 9.0% (male: 13.1%; female: 4.4%), and that of $\alpha$-fetoprotein (AEP) was 3.0% (male: 2.9%; female: 3.2%). In the egg-negative group, the positive rate of AST was 5.2% (male: 8.9%; female: 3.2%), that of ALT was 5.1% (male: 8.6%; female: 3.2%), of $\gamma$-GTP was 7.5% (male: 13.0%; female: 4.5%), and that of AFP was 0.3% (male: 0.5%; female: 0.1%). In the egg-positive group, 127 cases (38.1%) were light infection, 108 (32.4%) were moderate infection,64 (19.2%) were heavy infection, and 34 (10.2%) were very heavy infection. The positive rates of all tests were high according to the intensity of infection. From the above results, It was confirmed that prevalence of clonorchiasis is still high in some riverside areas of Kyongsangnam-do, and findings of serum biochemical tests are not different between the egg-positive group and egg-negative group. However, there were some difference in the positive rates of serum AST, ALT, $\gamma$-GTP and AEP by the age, sex and the intensity of infection.

부자(附子) 및 천오(川烏)를 함유한 처방(處方)이 간기능(肝機能)에 미치는 영향(影響)(II) (The Consequences of Long-period Dosages of Herbal Medicine Including Boo-ja and Cheon-o on Liver Injury.(II))

  • Objectives: Boo-ja and Cheon-o are not commonly prescribed, but are necessary for some clinical conditions, dispite the fact that some negative effects have been known to occur with these medicines. Hence, consequences for aspartate aminotransferase (AST), alanine aminotransferase(ALT) due to herbal medicines, including boo-ja and cheon-o are here reported. Methods: From January 2005 to July 2005, iresults were analyzed for 40 inpatients belonging to the sixth internal medicine department of Dongeui Oriental Medical Hospital who took herbal medicine, including boo-ja and cheon-o, over 20 days. The study is about the comparison and the investigation of LFT, and all results were taken upon their hospitalization and upon their departure. The standard of liver injury was used as a standard for their examinations. Results : 1. No Change in normal limit . 34 of the 40 2. Increased AST. ALT from when they were hospitalized : 2 of 40 Normalized AST, ALT from when they were hospitalized with results higher than normal : 6 of 40 3. These results show no side-effects, therefore suggest that these herbal medicines, including boo-ja and cheon-o. do not cause adverse side-effectsfor patients of this kind if administered as was done in this study.

Serum Liver Enzyme Pattern in Birth Asphyxia Associated Liver Injury

  • Purpose: To study temporal pattern of serum liver enzymes levels in newborns with hepatic injury associated with birth asphyxia (BA). Methods: Singleton term newborns with BA and ${\leq}72$ hours of age admitted to neonatal intensive care unit were prospectively enrolled. Term newborns with physiological jaundice and without BA were studied as controls. Serum liver enzymes were measured at <24 hours, 24-72 hours, and at 6-12 days of age for cases and at 1-6 days of age for controls. BA was defined by 1 minute Apgar score <7 or delayed or absent cry with hypoxic ischemic encephalopathy. BA-associated liver injury was defined as serum alanine aminotransferase (ALT) elevation beyond +2 standard deviation (ALT > +2 SD) above the mean of control subjects at any of the three time points. Results: Sixty controls and 62 cases were enrolled. Thirty-five cases (56%) developed BA-associated liver injury (ALT>81 IU/L). They had higher serum levels of ALT, aspartate aminotransferase, lactate dehydrogenase than the control infants, with peak at 24-72 hours. In controls, serum liver enzyme levels were significantly higher in appropriate-for-date (AFD) babies than small-for-date (SFD) babies. Serum enzyme pattern and extent of elevation were comparable between SFD and AFD babies. Degree of serum liver enzyme elevation had no relationship with severity of hypoxic encephalopathy. Conclusion: Serum liver enzyme elevation is common in BA; it peaks at 24-72 hours followed by a sharp decline by 6-12 days of age. Pattern and extent of enzyme elevation are comparable between SFD and AFD babies.

흰쥐의 일차배양 간세포에서 Daidzin, Daidzein, Genistein 및 Puerarin의 간 보호 활성 평가 (Hepatoprotective Activities of Daidzin, Daidzein, Genistein and Puerarin in Primary Cultured Rat Hepatocytes)

  • The aim of this study was to investigate the protective activities of daidzin, daidzein, genistein or puerarin, active isoflavonoids of Puerariae Radix, on the hepatocyte injury induced by carbon tetrachloride (CCl$_4$, 10 mM), tert-butyl hydroperoxide (TBH, 0.5 mM) and D-galactosamine (GalN, 30 mM). Primary cultures of rat hepatocytes (18 hr cultured) were treated with CCl$_4$, TBH or GalN and various concentrations (0.1, 1, 10 and 100 ${\mu}$M) of daidzin, daidzein, genistein or puerarin. CCl$_4$ significantly increased the levels of lactate dehydrogenase (LDH), alanine aminotransferase (ALT) and aspartate aminotransferase (AST). The increase in LDH level was attenuated by daidzein, genistein and puerarin. Puerarin also inhibited the increase in AST level induced by CCl$_4$. The increases in LDH and ALT levels induced by TBH were significantly attenuated by daidzin and genistein treatments. GalN markedly increased the levels of LDH, ALT and AST These increases were significantly attenuated by daidzein. Daidzin also inhibited the increases in LDH and AST levels induced by GalN. The increases in LDH and ALT levels were attenuated by genistein and puerarin, respectively. These results suggest that daidzin and daidzein possess hepatoprotective activities.

  • The aim of this study was to investigate the protective effects of glycynhizin, active glycosides of Glycyrrhizae Radix, and baicalin, bioactive flavonoid isolated from Scutellariae Radix, on hepatocyte injury induced by carbon tetrachloride(CCl$_4$, 10 mM), tert-butyl hydroperoxide (TBH, 0.5 mM), and D-galactosamine (GaIN, 30 mM). Primary cultures of rat hepatocyte (18 hr cultured) were treated with CCl$_4$, TBH, or GaIN and various concentrations (0.1, 1, 10, and 100 ${\mu}$M) of glycyrrhizin or baicalin. Activity was accessed by determining the release of lactate dehydrogenase (LDH) and aminotransferses. CCl$_4$ significantly increased the levels of LDH, alanine aminotransferase (ALT), and aspartate aminotransferase(AST) and these increases were prevented by baicalin concentrations of 0.1,1, and 100 ${\mu}$M. The increases in ALT and AST levels were reduced by glycyrrhizin concentration of 100 ${\mu}$M. The level of LDH was markedly increased by TBH, and this increase was reduced by both glycyrrhizin and baicalin. ALT and AST levels were increased by TBH, which were prevented by glycynhizin and bacalin, respectively: GaIN markedly increased the levels of LDH, ALT and AST These increases was significantly reduced by both glycyrrhizin and baicalin. These results suggest that glycynhizin and baicalin possess the hepatoprotective activity.

Effect of Elsholtzia splendens Extracts on the Blood Lipid Profile and Hepatotoxicity of the Mice

  • Effects of extracts obtained from the flowers of Elsholtzia splendens on the serum lipid profile and hepatotoxicity in mice were investigated. Female ICR mice were given E. splendens ethanolic extract (ESEs) orally at a dose of 10 or 50 mg/kg BW for 50 days. Significant dose-dependent decreases in triglyceride and low-density lipoprotein (LDL)-cholesterol of serum were observed. In addition, ESEs prolonged the lag-time of LDL oxidation in vitro. In the serum of ICE mice given ESEs orally at 10 and 50 mg/kg BW, the serum levels of aspartate aminotransferase (AST) and lactic dehydrogenase (LDH) increased significantly, while total protein, albumin, creatinine, alanine aminotransferase (ALT), and total bilirubin did not change. Therefore, ESEs may be beneficial to human health, although it has some hepatotoxicity.
