This study was carried out to investigate the effect of the bleeding times(30sec., 90sec., 150sec.) at slaughtering process on meat quality and storage properties of broiler. The redness$(a^{\ast}\;value)$ of skin, wing, leg muscle decreased at high bleeding time(150sec.). However, there was no significant difference in breast muscle. WHC(water holding capacity) of breast muscle decreased from $63.64\%$ at low bleeding time(30sec.) to $61.06\%$ at high bleeding time. TBARS(thiobarbituric acid-reactive substance) values were 0.18 mgMA/kg at the low bleeding time, 0.16 mgMA/kg at the middle bleeding time(90sec.) and 0.21mgMA/kg high bleeding time on 3 days of storage. Total aerobic plate counts(TPC) were $6.25logCFU/cm^2$ at the low bleeding time, $6.25logCFU/cm^2$ at the middle bleeding time and $6.53logCFU/cm^2$ at the high bleeding time. The TPC was increased as the bleeding time increased. In conclusion, meat color of chicken were acceptable when the carcasses were slaughtered at the high bleeding time.

The aim of this study was to shorten the time required for subculture and bacterial identification and obtain a simple and rapid identification method for new test methods for bloodstream infections. The following results were obtained using a mass spectrometer. In Vitek 2, 208 (81.8%) cases were well-identified and 45 isolates were not identified in blood cultures. Among 208 cases, 146 (57.5%) were Gram positive bacteria and 108 (42.5%) were Gram negative bacteria. In total, 233 were identified to the species level and 21 were identified to the genus level. The identification error was found to be Propionibacterium acnes as Clostridium bifermentans. The accuracy of Enterobacteriaceae, glucose non-fermentative bacilli (GNFB), and staphylococci were 81/83 (97.6%), 12/15 (80.0%), and 72/85 (84.7%), respectively. The concordance rate of Vitek 2 and Vitek MS by the direct method was 81.8% and 45 isolates were not identified. Most of the unidentified bacteria were Gram positive bacteria (N=37). The Gram positive bacteria were streptococci (14), coagulase-negative staphylococci (CNS) (11), enterococci (3), Staphylococcus aureus (2), Micrococcus spp. (2), Bacillus spp. (2) and Actinomyces odontolyticus, Finegoldia magna, and Peptostreptococcus spp. The results reporting time was reduced to 24~72 hours compared to the conventional method. The rate of identification of the aerobic and anaerobic cultures was similar, but the use of an anaerobic culture did not require a dissolution process, which could shorten the sample preparation time. These results suggest that the method of direct identification in blood cultures is very useful for the treatment of patients. In further studies, it might be necessary to further improve the method for identifying streptococci and CNS, which were lacking in accuracy in this study.

This study was conducted to evaluate the microbiological contamination on commonly used hand towels in the child care centers and to investigate the toxin gene and toxin production ability of food-borne pathogens. A total of 22 commonly used hand towels including 7 for before use and 15 for during use were tested. The average number of total aerobic bacteria and fungi were 6.2 log CFU/100 $cm^2$ and 4.1 log CFU/100 $cm^2$. Coliform bacteria were detected in 4 out of 7 before used towels (57.1%) and all of during used towels (100%). These results showed that the sanitary conditions of hand towels in the child care centers should be improved promptly. Among the pathogenic bacteria, Staph. aureus and B. cereus without Salmonella spp. were detected in 5 (22.7%) and 11 (50.0%) out of 22 hand towels. All of Staphy. aureus isolated in this study did not possess any toxin genes and did not produce enterotoxin. The detection rate of hblC, hblD, and hblA toxin genes in B. cereus was 72.7, 72.7, and 54.5% respectively. The possession rate of nheA, nheB, and nheC toxin genes showed 81.8, 72.7, and 54.5% respectively. The cytK and entFM toxin genes were presented at 45.5 and 90.0% in B. cereus. The HBL was detected in 8 out of 11 B. cereus isolates (72.7%) and 5 B. cereus isolates produced NHE (45.5%). Ten out of eleven B. cereus isolates (90.9%) produced one or more enterotoxin such as HBL and NHE. From the results, using a private hand towel or paper towel is required to prevent the cross-contamination between commonly used hand towel and children's hands in the child care center.
To study the productivity of single cell protein from the petroleum hydrocarbon utilizing yeasts, 242 soil samples, such as oil soaked soil of gas stations and garage, coal, farm soil, and sewage, from 135 places in Korea were collected. From these samples 468 yeast strains which utilize petroleum hydrocarbon as a sole organic carbon source were isolated and identified by observing the growth rates. For the identified strains optimum culture conditions were determined and analysis of cell components were performed. 1. 90.8% of petroleum hydrocarbon utilizing yeast strains were found from oil soaked soil and about 10% from coal, farm soil and sewage etc. 2. The yeast strain of the highest cell productivity was isolated from oil soaked soil and was identified as Candida curvata HY-69-19. 3. The optimum culture conditions for the selected yeast strain were found to be pH 5.0, $28^{\circ}C$ and affluent aerated state. 4. Candida curvata HY-69-19 was found to utilize favorably the heavy gas oil fractionated at above $268.9^{\circ}C$ as carbon source and urea as inorganic nitrogen source. 5. The growth curve of this strain on heavy gas oil medium showed that the yeast has a lag phase up to 18 hours and logarithmic growth phase between 24 to 42 hours. Generation time was found to be between 3.8 and 4.5 hours during the logarithmic growth phase. 6. About 300 mg dried cells per heavy gas oil was harvested under the culture conditions of adjusted pH to 5.0 at time intervals of 6 hours for 54 hours and heavy gas oil urea for shaking culture medium. 7. Chemical composition of the yeast cell was found to be 40.25%, 14.81%, 24.32% and 10.63% for crude protein, crude lipid, carbohydrate and ashes, respectively.
An antimicrobial bacterium to pathogenic microorganisms, strain $W5-1^T$ was isolated from Korean fermented-food Kimchi. The isolate was Gram-staining-variable, strictly aerobic, rod-shaped, endospore-forming, and motile with peritrichous flagella. It grew at $15-40^{\circ}C$, at pH 6.0-10.0, and in the presence of 0-4% NaCl. Strain $W5-1^T$ could hydrolyze esculin and xylan, and assimilate $\small{D}$-mannose, but not $\small{D}$-mannitol. Strain $W5-1^T$ showed antimicrobial activity against Listeria monocytogens, Pseudomonas aeruginosa, Staphylococcus aureus, and Salmonella typhi. The G+C content of the DNA of strains $W5-1^T$ was 52.6 mol%. The predominant respiratory quinone was menaquinone-7 (MK-7) and the major cellular fatty acids were $C_{16:0}$, antieiso-$C_{15:0}$, $C_{18:0}$, and $C_{12:0}$. The strain contained meso-diaminopimelic acid in cell-wall peptidoglycan. On the basis of 16S rRNA gene sequence and phylogenetic analysis, the strain W5-1 was shown to belong to the family Paenibacillaceae and was most closely related to Paenibacillus pinihumi $S23^T$ (98.4% similarity) and Paenibacillus tarimensis $SA-7-6^T$ (96.4%). The DNA-DNA relatedness between the isolate and Paenibacillus pinihumi $S23^T$ was 8.5%, indicating that strain $W5-1^T$ represented a species in the genus Paenibacillus. On the basis of the evidence from this polyphasic study, it is proposed that strain $W5-1^T$ is considered to represent a novel species of the genus Paenibacillus, for which the name Paenibacillus kimchicus sp. nov. is proposed. The type strain is $W5-1^T$ (=KACC $15046^T$ = $LMG 25970^T$).
A marine bacterium, designated as strain 50C-3, was isolated from a seawater sample collected from the East Sea of South Korea. The strain is a Gram-negative, aerobic, yellow colored polar-flagellated bacterium that grows at $20-50^{\circ}C$ and pH 5.5-8.5. Optimal growth occurred at $40-50^{\circ}C$, at pH 6.5-7.5, and in the presence of 2% (w/v) NaCl. Based on 16S rRNA gene sequence similarity, the isolate was considered to represent a member of the genus Ruegeria. The result of this analysis showed that strain 50C-3 shared 99.4% and 96.98% sequence similarity with Ruegeria intermedia CC-GIMAT-$2^T$ and Ruegeria lacuscaerulensis ITI-$1157^T$, respectively. Furthermore, strain 50C-3 showed clear differences from related strains in terms of several characteristics such as motility, carbon utilization, enzyme production, etc. The DNA G+C content was 66.7 mol%. Chemotaxonomic analysis indicated ubiquinone-10 (Q-10) as the predominant respiratory quinone. Based on phenotypic, chemotaxonomic, and phylogenetic characteristics, the isolate represents a novel variant of the Ruegeria intermedia CC-GIMAT-$2^T$, for which we named Ruegeria sp. 50C-3 (KCTC23890=DSM25519). Strain 50C-3 did not produce cellulase and agarase, but produced alkaline phosphatase, ${\alpha}$-galactosidase, and ${\beta}$-galactosidase. The three enzymes showed stable activities even at $50^{\circ}C$ and thus regarded as thermostable enzymes. Especially, the ${\beta}$-galactosidase activity enhanced by 1.9 times at $50^{\circ}C$ than that at $37^{\circ}C$, which may be very useful for industrial application.
In order to obtain basic information on the production of single cell protein from petroleum, more than 400 yeast strains were isolated from various soil samples in Korea utilizing petroleum hydrocarbon as the sole carbon source. A yeast strain showing the highest cell yield among the isolated strains was selected and identified. The optimal culture condition was searched in the flasks shaken throughout the procedure. And the growing characteristics for the selected yeast strain and chemical analysis of the yeast cell component were carried out. The results obtained were as follows: 1. The selected yeast strain was identified as Candida curvata and we named it Candida curvata-SNU 70. 2. The composition of the medium proposed for the present yeast strain is: Light Gas Oil 30ml, Urea 400mg, Ammonium sulfate 100mg, Potasium phosphate (monobasic) 670mg, Sodium phosphate (dibasic) 330mg, Magnesium sulfate 500mg, Calcium carbonate 3g, Yeast extract 50mg, Tween 20 0.05ml, Tap water 1,000ml. 3. Other culture conditions employed for the yeast were pH 5.5-7.0, temp. $30^{\circ}C$ under an affluent aerobic state. 4. Addition of light gas oil in portions to the culture media as the growth proceeded was more effective, especially in the cultivation on the higher oil concentration media. 5. Studies on the propagation of the yeast cells in the light gas oil medium revealed that the yeast has the lag phase lasted 16 hours and the logarithmic growth phase covered 16 to 28 hours. The specific growth rate was about $0.22\;hr^{-1}$ and doubling time was 3.2 hrs. during the logarithmic growth phase. 6. Under the cultural condition employed, the cell yield against the amount of light gas oil (wt%) was 16.1% and the protein content of the dried yeast cells was 48.4%.

The objectives of this study were to investigate the microbial contamination levels on food service in university and to provide the information of microbial contamination to improve food safety. A total of 288 samples were collected during summer and winter season between 2006 and 2008 from 4 food services located in the university in Western Gyeongnam and were used to detect sanitary indicator bacteria [aerobic plate count (APC), coliform, and Escherichia coli] and pathogenic bacteria (Staphylococcus aureus, Salmonella spp.). As a result, APC and coliform for hand and kitchen utensils which are used often by the employee were detected at high levels of 1.1~5.5 and 1.3~5.3 log CFU/($100\;cm^2$, hand), respectively. The contamination levels of APC and coliform in cooked foods and drinking water were 0.8~6.4 and 1.3~5.0 log CFU/(g, mL), respectively. Especially, the cooked foods showed the highest contamination for APC (2.1~6.4 log CFU/g) and coliform (1.0~5.0 log CFU/g). We think the reason that the cooked foods may be contaminated with APC and coliform on cooking process by using employee's hand and kitchen utensils. Moreover, S. aureus for hand and kitchen utensils was detected at levels of 2.8~3.0 and 2.0~2.3 log CFU/(g, hand), but Salmonella spp. was not detected. According to the above results, contamination levels of the samples were mostly decreased irrespective of summer and winter season. The results obtained indicated that it is necessary to periodic monitoring for microorganism contamination and education about personal and environmental hygiene to employee for ensuring food safety of food service in university.

This study investigated the effects of silkworm droppings(SWD) added diet feeding on chemical composition of pork loin meat. Pigs were divided into 7 treatment groups(10 pigs/group) and subjected to one of seven treatment diets(O, 1 0/0, 2 0/0, 3% SWD for 4 weeks, 1 0/0, 2 0/0, 3% SWD for 8 weeks, SWD diets; total fed diets) before slaughter. Pork loin were collected from the animals(1l0 kg body weight) slaughtered at a commercial slaughter house. Pork loin meat were aerobic packaged and then stored at$4^{\circ}C$ for 2, 5, 8, or 12 days. Samples were analyzed for TBARS, sensory evaluation, myoglobin content, fatty acid composition, amino acid and mineral content. TBARS value was not significantly different between the control and SWD treatment groups. Myoglobin content of SWD fed group pork was significantly increased than that of control group(P<0.05). Marbling score and acceptability were increased in SWD fed pork compared to those in control pork. Na, Mg, Ca, Mn and Fe content of SWD fed group pork was significantly decreased than those of control group(P<0.05). P content was significantly increased than that of control group(P< 0.05). In the change of amino acid composition, the contents of aspartic acid, serine and tyrosine were decreased by dietary SWD-supplementation, whereas the SWD-supplementation resulted in the higher glutamic acid, alanine and isoleucine. As dietary SWD was increased in feed, the content of oleic acid was significantly increased, but stearic acid was significantly decreased(P<0.05).

BACKGROUND: To design and develop a constructed wetland for effective livestock wastewater treatment, it is necessary to understand the removal mechanisms of various types of pollutants in constructed wetlands. The objective of this study was to evaluate the treatment efficiency of pollutants under different types of fraction in constructed wetland system for treating piggery wastewater. METHODS AND RESULTS: The piggery wastewater treatment plant that consisted of activated sludge tank, aerobic and anaerobic beds was constructed. The concentration of COD(Chemical oxygen demand) in effluent by fraction was 71.5 mg/L for soluble COD, 142 mg/L insoluble COD. The concentration of SS(Suspended solid) in effluent by existing form was 102 mg/L for volatile SS, 15.5 mg/L for fixed SS. The concentration of T-N(Total nitrogen) and T-P(Total phosphorus) in effluent by existing form were 12.8 mg/L and 3.05 mg/L for dissolved form, 35.0 mg/L and 1.93 mg/L for suspended form. The removal efficiencies of COD, SS, T-N and T-P in hot season(summer and autumn) were higher than those in cold season(spring and winter). The removal efficiencies of COD, SS, T-N and T-P in effluent were 98.0, 99.0, 98.2 and 99.2% for all seasons, respectively. CONCLUSION(S): The results indicated that removal types of pollutants were insoluble COD(ICOD), volatile SS(VSS), dissolved T-N(DTN) and dissolved T-P(DTP) in constructed wetlands for treating piggery wastewater.
