Study of the Efficacy and Safety of "adenophorae radix extract" in the Obese Patients or Overweight (비만 및 과체중 환자에 대한 사삼추출물의 유효성 및 안전성 평가)

  • Objectives: The purpose of this study is to evaluate the efficacy and biosafety of Adenophorae Radix(AR) extract in obesity or overweight patient. Methods: This study is double-blind, randomized, placebo-controlled intervention Study. 30 patients with BMI $25{\leq}$ and 30> were allotted into two groups at random. In 0, $6^{th}$, $12^{th}$ week, we had checked body weight, waist line, hip line, body fat and abdominal CT scan. In 0, $12^{th}$ week, we also had checked lipid metabolism and biosafety with blood test. Results: AR treatment had a significant effect on suppressing body wight gain (p<0.01) and BMI index(p<0.01). AR treatment reduced plasma TG level but we couldn't find statistical significance. AR treatment had produced no adverse reactions. Conclusions: This study shows that Adenophorae Radix(AR) extract can reduce the weight, BMI. Adenophorae Radix(AR) extract can be used in obesity or overweight patient.

Study on Keratinocyte Differentiation and Skin Barrier Function of Adeonphorae Radix Root Extracts (Adenophorae Radix 뿌리 추출물에 의한 Keratinocyte의 분화 및 피부장벽 기능에 대한 연구)

  • We have studied on the keratinocytes differentiation and skin barrier function using Adenophorae radix (A. radix) root extract, which was known to contain triterpenoid, saponin and starch. A. radix root extracts showed the $PPAR{\alpha}$ expression level of Wy-14,643 $0.5-1.0{\mu}M$ in CV-1 cells. The cornified envelop formation (CE) of human keratinocyte cell line (HaCaT) and normal human keratinocyte (NHK) showed a statistically significant increased compared to the control. When HaCaT cells were treated with A. radix root extract, transglutaminase (TGase-1) was significantly increased. As a result of clinical study of the simple cosmetic formulation containing A. radix root extract for about 2 weeks, TEWL values were significantly decreased and water contents were increased. The ceramides, which were obtained from the inner forearm, were also significantly increased statistically. We suggest that the A. radix root extract can be used as a preventive and therapeutic agent for skin diseases such as dry skin and atopy.

Quantitative and Classification Analyses of Lupenone and ${\beta}$-Sitosterol by GC-FID in Adenophora triphylla var. japonica Hara and Codonopsis lanceolata

  • A simple GC method with a FID detector was developed in order to determine two main compounds (${\beta}$-sitosterol and lupenone) for Adenophorae Radix. ${\beta}$-Sitosterol and lupenone were analyzed by the gradient thermal ramping method. Nitrogen was used as the carrier gas at 108 kPa. The flow rate of gas was 2.0 mL/min; $2{\mu}L$ of filtered sample was injected at a split ratio of 1 : 80. This method was fully validated with respect to linearity, precision, accuracy and robustness. Further, this GC-FID method was applied successfully in order to quantify two compounds in an Adenophorae Radix extract. The GC analytical method for classification analysis was performed by repeated analysis of 59 reference samples in order to differentiate between Adenophora triphylla var. japonica Hara and 14 Codonopsis lanceolata. The results indicate that the GC-FID method is suitable and reliable for the quality evaluation of Adenophorae Radix.

Inhibitory Effect of Water Extract of Adenophorae Radix on the Melanogenesis (소삼 물 추출물의 멜라닌 형성 억제 효과)

  • The aim of this study was to investigate the effect of Adenophorae Radix (AR) on melanogenesis of B16F10 mouse melanoma cells. The cells were treated for 5 days with AR at several concentrations. Treatment with AR suppressed melanin contents as a dose dependent manner without cytotoxicity and morphological change. And the extract of AR also inhibited tyrosinase activity, a key enzyme forming melanin, in a dose-dependent manner, Treatment of the cells with Adenophorae Radix also suppressed the increase of α-MSH (10 nM)-induced melanin contents and tyrosinase activity. These results suggest that AR inhibits melanogenesis and abrogates α-MSH-induced melanogenesis in B16 melanoma cells.

Inhibitory Effect of Methanolic Extract from Adenophorae Radix on Melanogenesis (사삼 메탄올 추출물의 멜라닌생성 억제효과)

  • Effects of methanolic extract from Adenophorae Radix (AR) on melanogenesis were investigated in mouse melanoma B16F10 cells. The methanol extract of AR was partitioned into Hexane, Ethyl acetate (EA), Butanol, H₂O, and exogenously added to the culture medium for 72 hours at the concentration of 10, 50, 100 and 200 ㎍/㎖. Of the four partitions, Hexane and EA partion of AR reduced tyrosinase activity, which is the key enzyme for a melanogenesis, as well as melanin contents. But the EA partition was less toxic for B16F10 cells and has more efficient melanin-reducing effect than the former. In addition, the EA partition dramatically lightened the color of cell pellet and significantly decreased the level of tyrosinase protein expression. In these results, EA partition of AR reduced melanin synthesis of B16F10 mouse melanoma cells by down regulating the tyrosinase activity and tyrosinase protein expression. Therefore, it is anticipated that AR is a candidate for an efficient whitening agent which supresses melanogenesis.

  • HAE-06 extract is a mixture of four medicinal plants, namely Lonicerae Folium et Caulis (Lonicera japonica), Scutellariae Radix (Scutellaria baicalensis), Adenophorae Radix (Adenophora triphylla var. japonica), and Polygonati Oddorati Rhizoma (Polygonatum odoratum var. pluriflorum). The HAE-06 extract demonstrated a concentration-dependent relaxing effect and enhanced cAMP production in bronchial smooth muscle that had been stimulated to contract with acetylcholine. Using a blocker, it was confirmed that the effect was through the β2-adrenergic receptor/cAMP/PKA pathway. In addition, it is thought that the HAE-06 extract has a bronchial smooth muscle relaxation effect by reducing the inflow of Ca2+ through the K+ and Ca2+ channels present in the sarcoplasmic membrane. If research continues in the future, it is believed that it will be possible to use it as a material for pharmaceuticals and functional foods.

Inhibitory Effect of Water Extract of Adenophorae Radix on the Melanogenesis (사삼 물 추출액의 멜라닌 형성 억제 효과)

  • Recently many efforts were focused to understanding the mechanical insights of melanogenesis to develop the agents for hyper-pigmentation and hypo-pigmentation. In the melanin biosynthetic pathway, tyrosinase is the rate limiting enzyme, and ${\alpha}$-melanocyte stimulating hormone(MSH) or cAMP-elevating agents stimulate melanogenesis and enhance the melanin synthesis and the tyrosinase activity. The author has analyzed the effects of Radix Trichosanthis on the basal Melanogenic activities of Bl6/F10 mouse melanoma cells, and on the ${\alpha}$-MSH or forskolin-induced melanogenesis. Radix Trichosanthis alone markedly suppressed melanin content and tyrosinase activity in a dose-dependent manner. Pretreatment of the cells with Radix Trichosanthis also suppressed the increase of ${\alpha}$-MSH(10 nM) or forskolin(20 ${\mu}$M)-induced melanin content and tyrosinase activity. The decrease in the tyrosinase activity was paralled by a decrease in the abundance of tyrosinase protein and tyrosinase promoter activity. Pretreatment of the cells with Radix Trichosanthis also inhibited the increase of forskolin(20 ${\mu}$M) induced the amount of tyrosinase protein and tyrosinase promoter activity. The results of DOPA staining revealed that pretreatment of the cells with Radix Trichosanthis showed less intensity than B16 melanoma cells stimulated with ${\alpha}$-MSH or forskolin. These results suggest that Radix Trichosanthis inhibits melanogenesis and abrogates ${\alpha}$-MSH and cAMP-induced melanogenesis in B16 melanoma cells.

Water Extract of Fermented New Korean Medicinal Mixture (F-MAPC) Controls Intracellula Adipogenesis and Glut-4 dependent Glucose Uptake in 3T3-L1 Adipocytes and L6 Myoblasts (세포 내 지방생성과 Glut-4 의존성 포도당 운반에 미치는 발효복합한약 물추출물(F-MAPC)의 영향)

  • Objectives : The aim of this study was to investigate the effects water extract of fermented new korean medicinal mixture, combinations of Mori Folium, Adenophorae Radix, Phllostachyos Folium and Citri Pericarpium (F-MAPC), on adipocyte differentiation, adipogenesis and glucose uptake using undiffernentiated 3T3-L1 adipocytes and L6 myoblasts. Methods : Each herb and those mixture were respectively fermented and then extracted with water. We carried on MTT assay for check-up on cell toxicity, Oil Red O staining for determination of cell differentiation and intracelluar adipogenesis. Western blot analysis for measurement of pAMPK and pACC, $C/EBP{\alpha}$, $PPAR{\gamma}$ and Glut-4 protein expressions were performed. Results : F-MAPC showed significant inhibitory activity on adipocyte differentiation in 3T3-L1 preadipocytes without affecting cell toxicity as assessed by measuring fat accumulation, and this effect was 2 fold higher in 0.2 mg/ml F-MAPC than that of the same dose of each fermented herbal extract alone. In addition, these effects were associated with modulation of adipogenic transcription factors, such as $C/EBP{\alpha}$, $PPAR{\gamma}$, as well as stimulated phosphorylations of AMPK and ACC. Translocation of Glut-4 was significantly increased by 10.2% in L6 cells treated with 0.2 mg/ml F-MAPC compared with that of control. Conclusions : These results demonstrate that F-MAPC may be an ideal candidate for therapy of obesity and diabetes by disturbing the differentiation into adipocytes, as well as the inducement of intramuscular glucose uptake from blood.

The Inhibitory Effects of Sabaek-san and Sabaeksan plus Sasam on the IL-6, IL-8 and GM-CSF mRNA Levels in Human Epithelial Cells (사백산(瀉白散)과 사백산가사삼(瀉白散加沙蔘)이 인간 기관지상피세포의 IL-6, IL-8 및 GM-CSF 발현에 미치는 영향)

  • Objective : We aimed to identify the dose-dependent inhibitory effects of Sabaek-san(瀉白散) and Sabaeksan plus Sasam(Adenophorae Radix) 瀉白散加沙蔘) on the mRNA expressions of Interleukin(IL)-6, IL-8 and granulocyte macrophage colony stimulating factor(GM-CSF) involved in the asthma model. Materials and Methods : Through this study, BEAS-2B cell lines, human epithelial cells were used. These cells were stimulated by tumor necrosis factor(TNF)-${\alpha}$, IL-1${\beta}$ and histamine for artificial inflammatory expression. ${\beta}$-action messenger RNA(mRNA) was used for standards. After each 24hours of Sabaeksan and Sabaeksan plus Sasam treatment, total cellular RNAs were collected by treating RNA zol directly on living cells, Then the transcriptional activities of IL-6, 8 and GM-CSF were measured by RT-PCR with electrophoresis, Results : The mRNA expressions of IL-6 are significantly inhibited compared to those of controlled group at 40 and 100ug/ml of Sabaeksan extract and $100{\mu}g/ml$ of Sabaeksan plus Sasam extract (p<0.05). The mRNA expressions of IL-8 are significantly inhibited compared to that of controlled group at 2.40 and 100 ug/ml of Sabaeksan extract and $40.100{\mu}g/ml$ of Sabaeksan plus Sasam extract(p<0.05) THe mRNA expressions of GM-CSF are significantly inhibited compared to those of the controlled group at $100{\mu}g/ml$ of Sabaeksan extract adn $40.100{\mu}g/ml$ of Sabaeksan plus Sasam extract.(p<0.05) Conclusions : This study shows that Sabaeksan and Sabaeksan plus Sasam have dose-dependent inhibitory effects on the mRNA expressions of IL-6, IL-8 and GM-CSF in human epithelial cells. Therefore, these types of herb medicine may inhibit the inflammatory process of asthma. Advanced studies are required to investigate the mechanisms of inhibition by herb medicine in the asthma model.
