A Study on Environmental Tolerances of Yeast (효모의 환경내성에 대하여 1)

  • Saccharomyces cerevisiae strain M was cultured in a molasses-containing media with repeated transplantations of the yeasts from one culture to another to adapt to molasses. After that only different amounts of phosphorous and nitrogen sources were added to the media. And then some variations during the culture time and the effects of consituents of cell mass on the functional activity and sensitivity of the cell were investigated. The results obtained were summarized as follows : 1. In the same culture condition of yeasts, the carbohydrates and trehaloses contents were more remarkably increased when small amounts of phosphate and nitrogen sources were added, then when alrge amounts were added, but yield percentage on assimilated sugars was lower. 2. The content of trehalose in yeast cells was reduced remarkably at the early stage in the culture, but this increased remarkably at later stage. When small amounts of nitrogen and phosphate were added to the culture medium, the amount of thehalose in the cells increased greasly. 3. The more protein content was present in the yeast cells, the smaller the carbohydrate and trehalose content, but more amino-N, RNA and moisture content were present in the cells. And in this case fermentability of the cells was stronger, but sugar tolerance was lower. 4. During the preservation period of compressed yeast cells at different temperature, the higher the temperature was, the more rapidly the amount of trehalose in the cells decreased. And in the cell where the amount of trehalose in the cells decreased. And in the cell where the amount of trehalose(carbohydrate) was large and the amount of protein was small, the amount of trehalose decreased at a slower rate during the preservation period.

Authentication of Sesame Oil with Addition of Perilla Oil Using Electronic Nose Based on Mass Spectrometry (전자코-Mass spectrometry를 이용한 들기름이 혼합된 참기름의 판별 분석)

  • Sesame oil was sometimes replaced by mixed oil due to high price in Korean market. To find out authentic sesame oil, electronic nose (E-nose) based on mass spectrometer system was used. Sesame oil was blended with perilla oil at the ratio of 97:3, 94:6, 91:9, 88:12 and 85:15, respectively. Intensities of each fragment from sesame oil by E-nose based on MS were completely different from those of perilla oil. The obtained data was used for discriminant function analysis. For quantitative analysis, the partial least square algorithm was used. The added concentration of perilla oil to sesame oil was correlated with discriminant function first score (DF1) and second score (DF2). From this relationship it could be found out how much perilla oil added. DFA plot indicated a significant separation of pure sesame oil and pure perilla oil. The different geographical origin of sesame oil was used for blending with perilla oil were closed to that of sesame oil. Korean sesame oil mixture and Indian sesame oil one were well separated. And the correlation between mixing ratios and DF1 values was found at the ratio of 97:3, 91:9, and 85:15 (SE vs PE oil), respectively. But the added concentration of perilla oil to sesame oil was correlated with discriminant function first score (DF1). E-nose based on MS system could be used as an efficient method for purity of oil quality.

Feasibility Tests on Struvite Production from Liquid Fertilizer by Utilizing Ferronickel Slag and Sewage Sludge Ash (페로니켈슬래그와 하수슬러지소각재를 이용한 액비로부터 스트루바이트 생산 타당성 연구)

  • Liquid fertilizers made from livestock manure contain high concentrations of nitrogen and phosphorus and thus are used as a fertilizer. However, excessive use of liquid fertilizer causes eutrophication of agricultural land and nonpoint source pollution. In this study, as a means of lowering the nutrient concentrations, struvite ($MgNH_4PO_4{\cdot}6H_2O$) production from the liquid fertilizer was investigated. When liquid fertilizers produced in Gyeonggido were analyzed, its characteristics differed by region and season, but the phosphorus concentration was commonly lower than that of nitrogen. When $K_2HPO_4$ and $MgCl_2$ were added to the liquid fertilizers, the optimal pH for struvite formation was pH 9.5. For environmentally friendly sources of magnesium and phosphate, ferronickel slag (FNS) and sewage sludge ash (SSA) were suspended in deionized water and extracted by sulfuric acid with various mass ratios. The optimum conditions for extracting FNS and SSA were 4.0 M sulfuric acid and 0.35 mass ratio of sulfuric acid to sewage sludge ash, respectively. For forming struvite, 0.233 L of SSA leachate (SSAL) was added into 0.3 L of liquid fertilizer containing 2,586 mg/L of ammonia and 110 mg/L of phosphate, pH was then adjusted to pH 9.5 using 10 M of NaOH. Afterwards 0.333 L of FNS leachate (FNSL) was added to this mixed solution. After a reaction for 1 hr at room temperature, the remaining concentrations of magnesium, ammonium, and phosphate were less than 50 mg/L, 500 mg/L and 150 mg/L, respectively, and 30 g of precipitates were obtained, most of which were struvite.

  • This paper describes the parachute dynamics modeling and simulation results for the development of training simulator of a HALO (High Altitude Low Opening) parachute, which is currently in use for military purposes. The target parachute is a rectangular shaped parafoil and its dynamic model is derived based on the real geometry data as the 9-DOF nonlinear equations of dynamics. The simulation was conducted through the moment of inertia and its aerodynamic derivatives to reflect the real characteristics based on the MATLAB/Simulink. In particular, its modeling includes the typical characteristics of the added mass and moment of inertia, which is shown in the strong effects in Lighter-Than-Air(LTA) flight vehicle. The proposed dynamic modeling was evaluated through the simulation under the spiral turning flight conditions of the asymmetric control inputs and compared with the performance index in the target parachute manual.

  • Added masses of two-dimensional cylinders of curvilinear-element sections with chines, which are similar to marked V character ship sections with either single or double chines, oscillating at high frequency in a free surface of an ideal fluid are calculated for both vertical and horizontal vibration by employing two particular two-parameter families of the conformal transformation. The numerical results are graphically presented in the forms of added mass coefficient curves in terms of the sectional area coefficient and the half beam-draft ratio together with the section contours derived with the employed transformations, and discussed in comparision with those of the Lewis forms and of straightline-element sections with single chine for vertical vibration, and, for horizontal vibration, with those of the Lewis forms.

Effects of Cr and Fe Addition on Microstructure and Tensile Properties of Ti-6Al-4V Prepared by Direct Energy Deposition

  • The effects of Cr and Fe addition on the mechanical properties of Ti-6Al-4V alloys prepared by direct energy deposition were investigated. As the Cr and Fe concentrations were increased from 0 to 2 mass%, the tensile strength increased because of the fine-grained equiaxed prior ${\beta}$ phase and martensite. An excellent combination of strength and ductility was obtained in these alloys. When the Cr and Fe concentrations were increased to 4 mass%, extremely fine-grained martensitic structures with poor ductility were obtained. In addition, Fe-added Ti-6Al-4V resulted in a partially melted Ti-6Al-4V powder because of the large difference between the melting temperatures of the Fe eutectic phase (Ti-33Fe) and the Ti-6Al-4V powder, which induced the formation of a thick liquid layer surrounding Ti-6Al-4V. The ductility of Fe-added Ti-6Al-4V was thus poorer than that of Cr-added Ti-6Al-4V.

  • N-type semi-conducting oxides such as $SnO_{2}$, ZnO, and $ZrO_{2}$ have been known for the detecting materials of inflammable or toxic gases. Of those materials, $SnO_{2}$-based sensors are well known as high sensitive materials to detect toxic gases. And the sensitivity is improved if catalysts are added. Detecting toxic gases, especially DMMP (di-methyl-methyl-phosphonate) and DPGME (Dipropylene glycol methyl ether), was performed by a mixture of Tin oxide ($SnO_{2}$) and Zirconia ($ZrO_{2}$). The films consist of each three different mass% of Zr (from 1 mass% to 5 mass%), and they were tested by XRD, SEM, TEM, BET. Nano-structure, pore and particle size was controlled to verify the sensor's sensing mechanism. The sensors was evaluated at five different degrees (from $200^{\circ}C$ to $400^{\circ}C$) and three different concentrations (from 500 ppb to 1500 ppb). The sensors had good sensitivity of both simulants, and high selectivity of DMMP.

  • Critical parameters affecting sensitivity and specificity of an enzyme-linked immunosorbent assay(ELISA) for detection of antibodies to avian infections bronchitis virus(IBV) were standardized. By adopting the optimized conditions an equation calculating ELISA antibody titers from the observations at single serum dilution was formulated. The purified antigen of IBV Mass-41 strain was dispensed into polystyrene microplate wells at a concentration of 300ng per well($100{\mu}l$) and the plates were coated by completey drying at $37^{\circ}C$. Diluted chicken serum and horseradish peroxidase conjugated goat anti-chicken IgG were added in order in $100{\mu}l$ volumes per well and allowed to react for 30 minutes each at room temperature. Just before use and after each reaction the plates were washed three times with distilled water. Finally o-phenylenediamine solution was added as an enzyme substrate. After incubation for another 15 minutes at room temperature absorbances were read at 492nm. Hyperimmune serum against Mass-41 strain was used as internal reference positive(IRP) serum. After repeated titration of IRP and negative serum, a constant titer of IRP was determined. Serum titrations were carried out for various sample sera together with IRP and negative sera and the observed titers of sample sera were corrected by reflecting the ratio between observed and constant titers of IRP serum. These corrected titers of the sample sera were plotted against sample/positive(S/P) OD ratios. All the OD's measured in the serum titrations were also corrected by substracting negative serum OD. The following equation was formulated from the above data; $Log_{10}$ ELISA titer=$5.568({\log}_{10}S/P)+4.161$ Thus it was possible to calculate ELISA titer by measuring absorbance at 1/400 single serum dilution. Titer measured by cross ELISA tests employing Mass-41 strain and three local IBV isolates were similar. These results suggest that the ELISA tests standardized in this study can be used for evaluating not only vaccinal immunity but also for infection status against fields IBV's.

  • This study explored the analytical conditions for 21 veterinary antibiotics which have been popularly sold in South Korea in 2014 but have not yet been targeted in EPA method 1694. Most of the selected antibiotics were separated by a reverse-phase C18 column with a combination of (buffered) water and organic polar solvent, which was commonly methanol and acetonitrile in the gradient elution mode. Volatile additives such as formic acid, ammonium acetate and ammonium formate were usually added to the mobile phases to minimize asymmetrical and tailing of antibiotics' peaks and to increase their ionization in mass spectrometry. The analytical methods of aminoglycoside antibiotics were distinct from those of the other antibiotics in terms of adoption of ion-pair chromatography (IPC) and hydrophilic interaction liquid chromatography (HILIC) capable of retaining and separating extremely polar compounds due to their hydrophilicity. Trifluoroacetic acid or heptafluorobutyric acid was frequently added to the mobile phase as an ion-pair reagent for the IPC. Tandem mass spectrometry was numerously applied to the detection of antibiotics using positive electrospray ionization (ESI) and the selected reaction monitoring (SRM) mode. All reviewed analytical methods had been/were validated by evaluating recovery, limits of detection and quantification, decision limit or detection capability of the methods.

  • Melamine has been used to increase the protein contents of powdered foods by adding a nitrogen source. Samples were prepared by blending melamine with powdered products such as powdered milk and coffee whitener. Melamine was added at ratios of 100:0, 80:20, 50:50, 30:70, and 0:100 (w/w), respectively. A Mass Spectrometry-based Electronic Nose and DFA (discriminant function analysis) were used to determine the amount of melamine. The electronic nose results were completely separated by the mixing ratios of melamine.
