T-Cell Death-Associated Gene 51 Is a Novel Negative Regulator of PPARγ That Inhibits PPARγ-RXRα Heterodimer Formation in Adipogenesis

  • The nuclear receptor peroxisome proliferator-activated receptor γ (PPARγ) is the master transcriptional regulator in adipogenesis. PPARγ forms a heterodimer with another nuclear receptor, retinoid X receptor (RXR), to form an active transcriptional complex, and their transcriptional activity is tightly regulated by the association with either coactivators or corepressors. In this study, we identified T-cell death-associated gene 51 (TDAG51) as a novel corepressor of PPARγ-mediated transcriptional regulation. We showed that TDAG51 expression is abundantly maintained in the early stage of adipogenic differentiation. Forced expression of TDAG51 inhibited adipocyte differentiation in 3T3-L1 cells. We found that TDAG51 physically interacts with PPARγ in a ligand-independent manner. In deletion mutant analyses, large portions of the TDAG51 domains, including the pleckstrin homology-like, glutamine repeat and proline-glutamine repeat domains but not the proline-histidine repeat domain, are involved in the interaction with the region between residues 140 and 506, including the DNA binding domain, hinge, ligand binding domain and activation function-2 domain, in PPARγ. The heterodimer formation of PPARγ-RXRα was competitively inhibited in a ligand-independent manner by TDAG51 binding to PPARγ. Thus, our data suggest that TDAG51, which could determine adipogenic cell fate, acts as a novel negative regulator of PPARγ by blocking RXRα recruitment to the PPARγ-RXRα heterodimer complex in adipogenesis.

  • A PEMFC single cell with the active area of $25cm^2$ was used to verify the effect of water management in the anode. Water management is one of the most critical operating variables. In this paper the effect of operating condition change, such as anode humidification and temperature, was investigated under constant current density of $200mA/cm^2$ where possible anode flooding operating area. Also experiments to observe the effect of the anode and cathode stoichiometry change and flow field design on the water management were performed. The water management was effected by the stoichimetry change. The temperature and humidification change also affected the fuel cell performance.

Analysis of the surface sterilization effect of disinfectants (소독제 별 표면소독 효과 분석)

  • The purpose of this study was to compare the number of bacteria before and after the use of surface disinfectants, demonstrate the bactericidal effect of surface disinfectants, and emphasize on the importance of surface disinfectants by recognizing the importance of infection control in dentistry. Chlorhexidine, hydrogen peroxide, ethanol, and chemical disinfectants are commonly used in dentistry. NaOCl was selected as the experimental group, and the bacterium test results obtained by comparing the table without surface sterilization as a control group showed that all disinfectants had an effective bactericidal effect (p<0.05). In the growth inhibition test comparing the experimental and control groups, all results were 100%, proving the effectiveness of surface disinfectants. The results showed that all surface disinfectants preferred and used by medical institutions were effective. Therefore, all surface disinfectants used in the experimental group were effective for surface disinfection for infection control. Dental clinicians should be aware of the necessity of disinfection of surfaces, such as table, chairs, and unit chairs, and make an active effort to ensure that both clinicians and patients are safe from infection.

  • Cancer is an abnormal growth of cells in body which leads to death. These cells are born due to imbalance in cell proliferation mechanism. In 2018, WHO released new statistics on cancer incidence, mortality, and prevalence worldwide i.e., GLOBOCAN 2018 estimates for 28 types of cancer in which more prevalence of cervix and breast cancer. According to survey, in India about 7.8 million cancer deaths and 11.5 million new cases arise in 2018, which will increase to 19.3 million new cases per year by 2025. Though breast cancer as such is not explained anywhere in Ayurvedic compendia, correlations can be done with the Stana Arbuda. Ayurveda, the ancient system of medicine came into existence 1000's of years ago with an objective of maintaining the health of people and treating diseases. Many herbs used in Ayurveda have been screened for activity against cancer and in-vitro and in-vivo studies have given promising leads. The plant, called as "Mother of Medicine", Haritaki has been extensively studied for its various ailments because of its extraordinary healing potency. Haritaki (Terminalia chebula Retz.), Family: Combretaceae have a great therapeutic value and is widely distributed in India. Dried fruit of Terminalia chebula contains high quantities phenolic compounds consist of ellagic acid, gallic acid and chebulic acid. The fruit extract of T. chebula is having different biological properties like anticancer, antioxidant, hepatic and renal protective activities etc. In this study, we focus on the use of CO2 extract of Terminalia chebula, on the breast cancer cell line MDA-MB-231. All tests proved that CO2 extract of Terminalia chebula containing active chemical component, therefore our experiment showed the positive results for CO2 extract of Terminalia chebula against breast cancer cell line cancer MDA-MB-231. The MTT assay results were used to evaluate the anti-cancer activity of the extract. The percentage of cell growth and cell viability were calculated from tabulated result values of MTT assay. Cell viability MTT assay also showed significant growth inhibition, at the same time statistical analysis of MTT assay also proved significant results.

A Study on functional cosmetic ingredients of the invasive plant Spartina anglica (생태교란종 영국갯끈풀의 기능성화장품 원료로서 효능 연구)

  • The Spartina anglica is recognized as a highly invasive plant and active eradication methods are required. In this study, we aimed to determine the physiologic activities of Spartina anglica extracts as a cosmetic ingredient. Antioxidant properties were investigated by different chemical methods including radical quenching (DPPH and ABTS), reducing power(FRAP) assay and aerial part of S. anglica(SAA) extract presented the strongest antioxidant activities. The significant cytotoxicity was not observed up to a concentration of 0.5 mg/mL in RAW264.7 cells and NHDF cells. The anti-inflammatory activity of S. anglica belowground(SAB) extract had strong effects on cell-based systems, including LPS-induced NO and cytokines(TNF-a and IL-6) production in RAW264.7 cells. Collagen synthesis and skin hydration gene expression of S. anglica extract showed the highest anti-wrinkle and moisturizing effect in NHDF cells. Results presented in this study tend to show that the ethanol extracts of S. anglica could be exploited as useful-bio-resource for bioactive substances in functional cosmetics.

  • Spectroscopies are the most widely used for understanding the crystallographic, chemical, and physical aspects of materials; therefore, numerous commercial and non-commercial software have been introduced to help researchers better handling their spectroscopic data. However, not many researchers, especially early-stage ones, have a proper background knowledge on the choice of fitting functions and a technique for actual fitting, although the essence of such data analysis is peak fitting. In this regard, we present a practical guide for peak fitting for data analysis. We start with a basic-level theoretical background why and how a certain protocol for peak fitting works, followed by a step-by-step visualized demonstration how an actual fitting is performed. We expect that this contribution is sure to help many active researchers in the discipline of materials science better handle their spectroscopic data.

Trapping of Methylglyoxal by Sieboldin from Malus baccata L. and Identification of Sieboldin-Methylglyoxal Adducts Forms

  • The methylglyoxal (MGO) trapping constituents from Malus baccata L. were investigated using incubation of MGO and crude extract under physiological conditions followed by HPLC analysis. The peak areas of MGO trapping compounds decreased, and their chemical structures were identified by HPLC-ESI/MS. Sieboldin was identified as a major active molecule representing MGO-trapping activity of the crude extract. After reaction of sieboldin and MGO, remaining MGO was calculated by microplate assay method using imine (Schiff base) formation of 2,4-dinitrophenylhydrazine (DNPH) and aldehyde group. After 4 h incubation, sieboldin trapped over 43.8% MGO at a concentration of 0.33 mM and showed MGO scavenging activity with an RC50 value of 0.88 mM for the incubation of 30 min under physiological conditions. It was also confirmed that sieboldin inhibited the production of advanced glycation end products (AGE) produced by bovine serum albumins (BSA)/MGO. Additionally, MGO trapping mechanism of sieboldin was more specifically identified by 1H-, 13C-, 2D NMR and, confirm to be attached to the position of C-3' (or 5').

Effect of Vanadium Loading Amount on Pt/V/TiO2 Catalyst on NH3-SCO Reaction (NH3-SCO 반응에서 Vanadium 담지함량이 Pt/V/TiO2 촉매에 미치는 영향)

  • In the study, NH3-SCO (selective catalytic oxidation) reaction activity accodrding to vanadium loading amount were compared when preparing Pt/V/TiO2. Considering both NH3 conversion rate and N2 selectivity, V 2 wt% loading of the catalyst showed the best activity. When the correlation between physical/chemical characteristics and reaction activity was confirmed, it was confirmed that the increase in lattice oxygen and (V3+ + V4+) ratios were active factor. In addition, when the SO2 durability experiment was conducted using the best catalyst, it was confirmed that the influence was insignificant even if the high concentration of SO2 was injected.

Efficiency of PDNR (Polydeoxyribonucleotide) extraction from various plant species and its in vitro wound healing activity (다양한 식물에서의 PDRN(Polydeoxyribonucleotide) 추출 수율 비교 및 상처치유 효능 분석)

  • PDRN (Polydeoxyribonucleotide) is a DNA-derived polymer that promotes self-renewal of damaged cells and tissues as a tissue regeneration active material. PDRN is a DNA fragment cut into small sizes by various physical or chemical methods. When administered to the body, PDRN binds and stimulates the adenosine A2A receptor on the surface of tissue cells to promote cell regeneration, accelerate wound healing, and reduce pain. Although PDRN is prepared from testis or semen of fish in most cass, PDRN extraction from various plants species was performed in the present study. Among 7 tested plant species, the highest DNA yield and purity was obtained form mugwort (Chrysanthemum coronarium, C.c), followed by broccoli (Brassica oleracea, B.o). Then, we evaluated the in vitro wound healing capacity of PDRNs prepared from these two selected plants. PDRN from C.c and B.o. significantly stimulated the wound healing process at ㎍/ml range. The present study suggests that PDRN from plant species can be an effective alternative to PDRN from marine organism.

Preparation and Electrochemical Characterization of Porous Carbon Foam from Waste Floral Foam for Supercapacitors (폐 플로랄 폼을 이용한 슈퍼커패시터용 다공성 탄소 폼 제조 및 전기화학 성능 평가)

  • The recycling of solid waste materials to fabricate carbon-based electrode materials is of great interest for low-cost green supercapacitors. In this study, porous carbon foam (PCF) was prepared from waste floral foam (WFF) as an electrode material for supercapacitors. WFF was directly carbonized at various temperatures of 600, 800, and 1,000 ℃ under an inert atmosphere. The WFF-derived PCF (C-WFF) was found to have a specific surface area of 458.99 m2/g with multi-modal pore structures. The supercapacitive behavior of the prepared C-WFF was evaluated using a three-electrode system in a 6 M KOH aqueous electrolyte. As a result, the prepared C-WFF as an active material showed a high specific capacitance of 206 F/g at 1 A/g, a rate capability of 36.4 % at 20 A/g, a specific power density of 2,500 W/kg at an energy density of 2.68 Wh/kg, and a cycle stability of 99.96 % at 20 A/g after 10,000 cycles. These results indicate that the C-WFF prepared from WFF could be a promising candidate as an electrode material for high-performance green supercapacitors.
