Two Cyanidin compound from the Fruits of Acanthopanax divaricatus var. albeofructus

  • Acanthopanax divaricatus var. albeofructus is one of the indigenous medicinal plant and the fruits of Acanthopanax spp. used as a remedial for "wipe out evil wind". Two anthocyanin were isolated from the fruits of Acanthopanax divaricatus var. albeofructus. Their structures were elucidated as cyanidin 3-lathyroside (1) and cyanidin 3-galactoside (2) by chemicophysical and spectroscopic analysis. And also, four chemical, syringin, chlorogenic acid, caffeic acid and acanthoside D were identified. Both anthocyanide were isolated for the first time from Acanthopanax species. cyanidin 3-lathyroside is one of the rare anthocyanin in natural resources.

  • This study was carried out to obtain the basic informations for phenolic compounds by various sections of Acanthopanacis Caulis. The phenolic compounds of Korean Acanthopanacis bark and lignum (Acanthopanax divaricatus var. albeofructus, Acanthopanax koreanum) were measured by the HPLC analysis. The content of total phenolic components of Acanthopanax koreanum bark (1.532%) was about 9.9 times higher than that of Acanthopanax koreanum lignum (0.155%). And also, The content of total phenolic components of Acanthopanax divaricatus var. albeofructus bark (0.420%) was about 2.8 times higher than that of Acanthopanax divaricatus var. albeofructus lignum (0.149%). Eleutheroside E, a functional ingredient of Acanthopanacis Caulis, showed 3.6 times higher contents of Acanthopanax koreanum bark (0.144%) than Acanthopanax koreanum lignum (0.040%). In the case of Acanthopanax divaricatus var. albeofructus bark (0.129%), the content of the eleutheroside E was 1.7 times higher than that of the Acanthopanax divaricatus var. albeofructus lignum (0.074%).

  • Acanthopanax species (Araliaceae) traditionally has been used as analgesics, stimulant of immune system, and replenishment of body functions. Acanthopanax divaricatus var. albeofructus is indigenous plant to Korea. The antioxidant activities of compounds from A divaricatus var. albeofructus were determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) method and thiobarbituric acid reactive substance (TBARS) assay on human plasma low-density lipoprotein (LDL). The triterpenoid and lignan constituents from this plant showed antioxidant activities and the lignan, l-sesamin exhibited the most potent antioxidant activity in Cu$^{2+}$ -induced LDL oxidation.n.

  • Acanthopanax species (Araliaceae) has been traditionally used as analgesics, stimulants of immune system, and replenishment of body functions. Acanthopanax divaricatus var. albeofructus is a indigenous plant in Korea. The anti-lipid peroxidation activities of the loaves and root bark extracts of A divaricatus var. albeofructus were determined. Activities of MeOH extracts were examined in vitro and in vivo with control group intoxicated by carbon tetrachloride. Both extracts showed significant inhibitory effects on the lipid peroxidation.

  • In the present study, water extracts of Acanthopanax divaricatus var, albeofructus and Eleutherococcus divaricatus var. chiisanensis were prepared and their antioxidant and anti-inflammatory effects were compared. The water extract of Acanthopanax divaricatus var, albeofructus showed significantly higher activity in total polyphenol and flavonoid contents, while there were no significant differences in ABTS/DPPH radical scavenging activities and ferric reducing antioxidant power. However, the water extract of Eleutherococcus divaricatus var. chiisanensis at the highest dose showed significantly lower capacity in nitric oxide production than that of Acanthopanax divaricatus var, albeofructus. Taken together, there were differences between Acanthopanax divaricatus var, albeofructus and Eleutherococcus divaricatus var. chiisanensis in total polyphenol/flavonoid contents and anti-inflammatory effect, however, the antioxidant effect was almost similar.

  • We investigated antioxidative activity of the water and ethanol extracts of leaves of Acanthopanax divaricatus var. albeofructus in human dermal fibroblast (HDFs) irradiated by UVA. The irradiation of UVA did not affect the cell viability of HDFs. The antioxidative activity of the extract was investigated by xylenol orange, TBARS (thiobarbituric acid reactive substances) and antioxidant enzyme assay. Both extracts showed H202 scavenging activity and inhibited lipid peroxidation in HDF cells irradiated by UVA. The extracts also recovered enzyme activity in the same cells.

Inhibition of Lung Inflammation by Acanthopanax divaricatus var. Albeofructus and Its Constituents

  • In order to find potential therapeutic agents on lung inflammatory conditions, the extracts of Acanthopanax divaricatus var. albeofructus were prepared and its constituents were isolated. They include lignans such as (+)-syringaresinol (1), acanthoside B (2), salvadoraside (3) and acanthoside D (4), lariciresinol-9-O-${\beta}$-D-glucopyranoside (5) and phenylpropanoids such as 4-[(1E)-3-methoxy-1-propenyl]phenol (6), coniferin (7), and methyl caffeate (8). The extracts and several constituents such as compound 1, 6 and 8 inhibited the production of inflammatory markers, IL-6 and nitric oxide, from IL-$1{\beta}$-treated lung epithelial cells and lipopolysaccharide (LPS)-treated alveolar macrophages. Furthermore, the extracts and compound 4 significantly inhibited lung inflammation in lipolysaccharide-treated acute lung injury in mice by oral administration. Thus it is suggested that A. divaricatus var. albeofructus and its several constituents may be effective against lung inflammation.

  • Acanthopanax divaricatus var. albeofructus (ADA) is commonly ingested as a traditional medicine or as a component of a health drink in Korea. In this phytochemical study, nine phenolics (1 - 9) and three lignans (10 - 12) were isolated from the MeOH extract of the stems of ADA. Chemical structures were elucidated by comparing spectroscopic data with reported values. Nuclear factor kappa B ($NF-{\kappa}B$) inhibitory activity of the isolated compounds was evaluated using an $NF-{\kappa}B$ luciferase assay in HepG2 cells. Among them, compounds 1, 3 - 8, and 11 showed significant inhibitory effects on $TNF{\alpha}$-induced $NF-{\kappa}B$ transcriptional activity in a dosedependent manner, with $IC_{50}$ values ranging from 13.25 to $37.36{\mu}M$. Further studies on potential anti-inflammatory effects and the benefits of phenolic and lignan components from ADA are warranted.

The Comparison of Phytochemical Components from the Berry of Acanthopanax Species (오가피속 식물 열매의 식물화학 성분 비교)

  • This study was carried out to obtain the basic information that can be used to index Acanthopanax in eleven species of China and Korea. The phytochemical components from the berry of Acanthopanax species, were measured by the HPLC analysis. Protocatechuic acid, eleutheroside B, eleutheroside E, scopolin, rutin, hyperoside, chiisanoside, oleanolic acid were found in ethanol extracts from the berry of Acanthopanax species. Total phenolic compound of Acanthopanax sessiliflorum f. chungbuensis berry (0.682%) was about seven times higher than those of Acanthopanax divaricatus f. distigmatis berry (0.091%). As a result, the order of the eleutheroside E content was 1) Acanthopanax sessiliflorum f. chungbuensis (0.554%), 2) A. divaricatus var. albeofructus (0.501%), 3) A. divaricatus f. flavi-flos (0.452%). And also, the order of the chiisanoside content was 1) Acanthopanax senticosus var. subinermis (8.434%), 2) A. seoulense (0.94%), 3) A. divaricatus f. flavi-flos (0.798%).

  • Water extracts of Acanthopanax divaricatus var. albeofructus (ADA) fruits were used to treat hPBMC to determine the mechanisms for the immunomodulatory effects. The secretion level of various cytokines including Th-1 type (IL-2, L-12, $IFN-{\gamma}$ and $TNF-{\alpha}$) and Th-2 type (IL-6, IL-8 and IL-10) were measured using ELISA. A significant increase of Th-1 type cytokine secretion was observed in the presence of extract while Th-2 cytokine, IL-6 was suppressed. Our results suggest that ADA fruit extract may influence the anticancer immune responses towards a predominance of Th-1 cytokines in the immune system.
