An Experimental Approach to Secure Freshwater Fish Shelter according to the Water Level Fluctuations in a Shallow Pond (얕은 연못에서 수위변동에 따른 담수 어류 피난처 확보를 위한 실험적 접근)

  • Physical disturbance, which induces a lack of flow rate, frequently occurs in freshwater ecosystem. Due to this, it is required to provide a new fish shelter to resolve. We installed a pilot scale test-bed to scrutinize the relationship between water level and the influence of fish shelter. The proposed ADP (artificial deep pool) is a fish shelter which composed of concrete materials. From the monitoring results in test-bed, it was observed that the population of fish was the highest at the 0.5 m in depth from the water level of experimental pond. But it was more appropriate for shallow water level (<0.3 m) to conserve the total number of fish by increasing the number per unit area despite of lower inner temperature and DO than outer environment. Therefore, inner of ADP was more efficient lentic system for fish to live due to higher WCS, OS, SS, and TS. In addition, there was a relative abundance of WCS fish species such as Acheilognathus koreensis (A. koreensis), Carassius carassius (C. carassius). Considered these results, it is suggested that ADP is appropriate to use for fish shelter and habitat for the fishes in lentic ecosystem.

Poly(ADP-ribose) protects vascular smooth muscle cells from oxidative DNA damage

  • Vascular smooth muscle cells (VSMCs) undergo death during atherosclerosis, a widespread cardiovascular disease. Recent studies suggest that oxidative damage occurs in VSMCs and induces atherosclerosis. Here, we analyzed oxidative damage repair in VSMCs and found that VSMCs are hypersensitive to oxidative damage. Further analysis showed that oxidative damage repair in VSMCs is suppressed by a low level of poly (ADP-ribosyl)ation (PARylation), a key post-translational modification in oxidative damage repair. The low level of PARylation is not caused by the lack of PARP-1, the major poly(ADP-ribose) polymerase activated by oxidative damage. Instead, the expression of poly(ADP-ribose) glycohydrolase, PARG, the enzyme hydrolyzing poly(ADP-ribose), is significantly higher in VSMCs than that in the control cells. Using PARG inhibitor to suppress PARG activity facilitates oxidative damage-induced PARylation as well as DNA damage repair. Thus, our study demonstrates a novel molecular mechanism for oxidative damage-induced VSMCs death. This study also identifies the use of PARG inhibitors as a potential treatment for atherosclerosis.   • The present study was performed to clone and express human poly (ADP-ribose) synthetase (PARS) cDNA in E coli. For these purposes, the CDNA for human poly (ADP-ribose) synthetase, encoding the entire protein, was cloned into pGEM-7Zf(+). The resulting recombinant plasmid pPARS6.1 was restriction enzyme mapped and its identity was confirmed by Southern blot analysis. The pPARS6. 1 contained full-length CDNA of human PARS and the nudeotide sequences were identical with those reported previously. The recombinant protein which migrated as a unique 120 kDa band on 10% SDS-polyacrylamide gels, was identified as PARS by Southwestern blots using nick-translated DNA probes and by activity gels and activity blots using 32 P-NAD as a substrate for poly (ADP-ribose) synthetase (PARS). The signals corresponding to 120 and 98 kDa proteins were obtained following IPTG (0.4 mM) induction of the PARS cDNA cloned into Xba I-digested pGEM-7Zf(+) vector. Nonspecific signals corresponding to 45 and 38 kDa proteins were also shown in both IPTG-induced and noninduced cells. The nonspecific proteins may be products of incomplete translation or proteolytic products of intact PARS.

Distinct Regulation of the sprC Gene Encoding Streptomyces griseus Protease C from Other Chymotrypsin Genes in Streptomyces griseus IFO13350

  • The sprC gene encodes Streptomyces griseus protease C (SGPC), a bacterial chymotrypsin-like serine protease. Because the published data on sprC was not complete, we cloned and analyzed a new DNA fragment spanning downstream to upstream of the sprC gene from S. griseus IFO13350. The cloned 2.3-kb DNA fragment was placed on a high-copy number plasmid and introduced into Streptomyces lividans TK24. Chymotrypsin activity of the transformant was 8.5 times higher than that of the control after 3 days of cultivation and stably maintained until 9 days of cultivation, which dearly indicated that the cloned 2.3-kb fragment contained the entire sprC gene with its own promoter. When the same construct was introduced in the S. griseus IFO13350 (wild strain) and its two mutant strains in the A-factor regulatory cascade, ${\Delta}adpA$ and HO1, the chymotrypsin activity increased fivefold only in the ${\Delta}adpA$ strain. Transcriptional analysis based on RT-PCR revealed that the sprC gene is normally transcribed in both strains; however, earlier transcription was observed in the wild strain compared with the ${\Delta}adpA$ strain. A gel mobility shift assay showed that the AdpA protein did not bind to the promoter region of sprC. All these data clearly indicate that the expression of sprC is not dependent on the AdpA protein, but is distinctly regulated from other chymotrypsin genes composing an AdpA regulon. Earlier morphological differentiation was observed in S. lividans TK24, and S. griseus IFO13350 and HO1, transformed with the expression vector. The transformant of S. griseus ${\Delta}adpA$ formed markedly larger colonies. Antisense repression of sprC resulted in severe decrease of chymotrypsin activity, down to one-third of the control, and delayed morphological differentiation. All these data suggest that SGPC is related to normal morphogenesis in S. griseus.

  • The purpose of this study was to investigate the antioxidant activities of water soluble browning reaction products (WS-BRPs) isolated 5\ulcorneron korea red ginseng. Antioxidant activities of WS-BRPs were examined with the various systems. All three WS-BRPs (L, S-1 and S-2) were found to have an ability to linoleic acid, Ox-brain autoxidant, Fe$^{2+}$ ADP/NAD system and cumene hydroperoxide system. Especially, S-2 was had the strongest activity of theses three WS-BRPs to scavenge free radicals such as more effective than S-1, L. MDA determination showed the antioxidant effect on linoleic acid oxidation inhibition ratio of 22.5%, 31.7%, 31.9% and 33.5%, respectivity Especially; Ox-brain autoxidant was strong inhibited activity by 49.52%,62,44,97.54% by addition of various concentration. But three WS-BRPs showed weak inhibitory activity on lipid peroxidation in rat hepatic microsomes induced enzymatically and nonenzymaticallyly

Study on Change of Poly ADP Ribose Polymerase in the Rat with Thrombotic Stroke by Full Wave Cockroft Walton method's Transcranial Magnetic Stimulation

  • This study examined the relationships between protein expression and Poly ADP ribose polymerase in brain cell death in brains damaged by thrombotic stroke and treated with the Full Wave- Cockroft Walton (FWCW) method of Transcranial Magnetic Stimulation (TMS). The two-way switching element for TMS drove a half-bridge inverter of the current resonance of direct current voltage (+) and direct current voltage (-), and the experiment was conducted by stimulating the mice with thrombotic stroke through a range of pulses. Thrombotic stroke was caused of ligation of the common carotid artery of male SD mice, and blood reperfusion was conducted five minutes later. Protein expression was examined in immune reaction cells, which reacted to an antibody to Poly ADP ribose polymerase in the cerebrum cells, and western blotting. Observations of the PARP changes after thrombotic stroke showed that the number of Poly ADP ribose polymerase reactions were significantly lower (p < 0.05) in the group treated with TMS of the FWCW than the group with thrombotic stroke 24 hours after its onset. The application of FWCW-TMS helped prevent the necrosis of nerve cells and might prevent the brain damage that occurs as a result of thrombotic stroke, and improve the function recovery and disorder of brain cells.

  • The nascent from of glycosylphosphatidylinositol (GPI)-anchored proteins possesses both amino and carboxy terminal hydrophobic signal sequences to direct processing in the endoplasmic reticulum (ER). Following cleavage of the amino-terminal signal peptide, the carboxy-terminal peptide is processed. Previously, mouse lymphocyte NDA: agrinine ADP-ribosyltransferase (Yac-1) was cloned and the deduced amino acid sequence of the Yac-1 transferase contained hydrophobic amino and carboxy termini, consistent with known signal sequences of GPI-anchored proteins. This tranferase was present on the surface of NMU (rat mammary adenocarcinoma) cells transfected with the wildtype cDNA and was released with phosphatidylinositol-specific phosphilpase C. Expression of the mutant protein, lacking the carboxy terminal hydrophobic sequence, resulted in the peoduction of soluble, secreted from of the transferase. This result shows that carboxy terminal sequence is important for GPI-attachment.

Poly(ADP-ribosyl)ation of p53 Contributes to TPEN-Induced Neuronal Apoptosis

  • Depletion of intracellular zinc by N,N,N,N-tetrakis(2-pyridylmethyl) ethylenediamine (TPEN) induces p53-mediated protein synthesis-dependent apoptosis of mouse cortical neurons. Here, we examined the requirement for poly(ADP-ribose) polymerase (PARP)-1 as an upstream regulator of p53 in zinc depletion-induced neuronal apoptosis. First, we found that chemical inhibition or genetic deletion of PARP-1 markedly attenuated TPEN-induced apoptosis of cultured mouse cortical neurons. Poly(ADP-ribosyl)ation of p53 occurred starting 1 h after TPEN treatment. Suggesting the critical role of PARP-1, the TPEN-induced increase of stability and activity of p53 as well as poly(ADP-ribosyl)ation of p53 was almost completely blocked by PARP inhibition. Consistent with this, the induction of downstream pro-apoptotic proteins PUMA and NOXA was noticeably reduced by chemical inhibitors or genetic deletion of PARP-1. TPEN-induced cytochrome C release into the cytosol and caspase-3 activation were also blocked by inhibition of PARP-1. Taken together, these findings indicate that PARP-1 is essential for TPEN-induced neuronal apoptosis.

Effects of ATP and ADP on iron uptake in rat heart mitochondria

  • Iron uptake in mitochondria and fractionated mitochondria compartments was studied to understand iron transport in heart mitochondria. The inner membrane is most active in iron uptake. Mitochondrial uptake was dependent on iron concentration and the amount of mitochondria. Iron transport was inversely proportional to pH in the range of 6.0 to 8.0. Iron transport reached a maximum after 30 min of incubation at $37^{\circ}C$. Iron uptake was inhibited by 1 mM ATP and stimulated by 1 mM ADP. The oxidative phosphorylation inhibitor oligomycin inhibited iron uptake, but rotenone and antimycin A did not. The divalent ions $Mg^{2+}$, $Cu^{2+}$, $Mn^{2+}$, and $Zn^{2+}$ suppressed iron uptake at $10\;{\mu}M$ and stimulated it at 1 mM. The divalent ion $Ca^{2+}$ stimulated iron uptake at $10\;{\mu}M$ and suppressed it at 1 mM, competing with iron. The uptake of calcium was stimulated by 10 to $1000\;{\mu}M$ ATP, while iron uptake was stimulated reciprocally by 10 to $1000\;{\mu}M$ ADP, suggesting that these ions have movements similar to those of ATP and ADP.

A Study on Development of Freshwater Fish Shelter and Evaluation of Water Quality for the Reduction of Thermal Stress in Shallow Pond (얕은 연못에서 담수 어류의 열성 스트레스 저감을 위한 피난처 개발 및 수질환경 조사 연구)

  • The purpose of this study evaluates the water quality of artificial deep pool (ADP) during the late spring and summer. we monitored the water quality, as pH, dissolved oxygen (DO), temperature and conductivity at two stations (St. 1. open water, St. 2. in the ADP). The water quality in the ADP is very stable, and temperature and DO are lower than the open water (average value; temperature $1.4{\sim}3.2^{\circ}C$, DO 2.4~3.6 mg/L). In particular, daily variation of temperature in the open water showed above $4^{\circ}C$, but ADP remained stable. The water quality was analyzed using two-way analysis of variance (ANOVA). The results of the analysis showed difference about temperature, pH, DO, conductivity (two-way ANOVA, p<0.05). The ADP has created an aqua environment in thermal and DO gradients by depth. About 1.2 m, Temperature and DO were sharply decreased. The Rhodeus uyekii is dominant species in pond of this study. The ADP offers optimum water temperature ($16.5{\sim}18.5^{\circ}C$) to the Rhodeus uyekii about spawning. Consequently, the ADP offers stable habitat than open water for fish and aquatic organisms during the summer. It is also a good shelter for fish from a thermal stress.
