In this study, the NABH558 gene expression system was constructed to efficiently produce neoagarobiose hydrolase (NABH) in Saccharomyces cerevisiae strain. The ADH1 and GAL10 promoters of the pAMFα-NABH and pGMFα-NABH plasmids were examined to determine the suitable promoter for the NABH558 gene expression, respectively. The effect of promoter and carbon sources on NABH558 gene expression was investigated by transforming each plasmid into the S. cerevisiae 2805 strain. The NABH activity in the 2805/pAMFα-NABH strain was 0.069 unit/ml/DCW in YPD medium, whereas that in the 2805/pGMFα-NABH strain was similar (0.02-0.027 unit/ml/DCW) irrespective of the medium composition. The higher NABH activity in the YPD medium was due to the increased NABH558 gene transcription. NABH produced in the recombinant strains could degrade agarose to galactose and AHG. This indicated that ADH1 promoter was a more optimal promoter for the expression of NABH558 gene than the GAL10 promoter. The NABH activity induced by the ADH1 promoter was about 3-fold higher than that induced by the GAL10 promoter.

Wet-injury often occurs in upland cereals growing in the paddy field due to oxygen deficiency in the rhizosphere caused by excessive water in the soil. Under hypoxia, energy metabolism is diminished causing nonreversible damage to root cells. This study was conducted to investigate effects of hypoxia on root growth and enzymes involved in the fermentative energy metabolism in upland cereals including barley, wheat, rye and triticale. Young seedlings were subject to hypoxia for up to 7 days. Root fresh weight and dry weight were decreased significantly by hypoxia for 5 to 7 days in all cereal seedlings. Root growth retardation under hypoxia was lowest in barley. Hypoxia-induced increases in activity and isozyme expression of alcohol dehydrogenase (ADH) and lactate dehydrogenase (LDH) were commonly observed in roots of all cereal seedlings. The inherent ADH activity levels were higher in barley but the hypoxia-induced increases in ADH activities were lowest in barley than other cereals. The inherent LDH activity levels were lower in barley and the hypoxia-induced increases in LDH activities were lower in barley than other cereals. The results suggest the importance of the rapid enhancement of fermentative enzyme systems for increased tolerance to hypoxia.

This study is to analyze the effects of the growth temperature of mungbean sprouts ($15{\sim}30{\pm}1^{\circ}C$) on the yield ratio, content of phenolic compounds and DPPH (1,1-diphenyl-2-picrylhydrazyl), ADH (alcohol dehydrogenase), ALDH (aldehyde dehydrogenase) activities of the sprouts. When the growth temperature of mungbean sprouts was higher, the yield ratio of the sprouts was higher while the hard seed rate was lower, but $25{\pm}1^{\circ}C$ and $30{\pm}1^{\circ}C$ showed no regular tendency. The content of the total phenol from the ethanol extract of the sprouts was higher in the growth temperature of $15{\pm}1$, $20{\pm}1$, and $25{\pm}1^{\circ}C$, while the content of total flavonoid was higher in the growth temperature of $15{\pm}1$, and $20{\pm}1^{\circ}C$. The DPPH radical scavenging activity of the ethanol extract of the sprouts was higher when the growth temperature was lower, while the activity of ADH and ALDH showed no regular tendency according to the growth temperature. Considering the yield ratio, content of phenolic compounds, biological activities of mungbean sprouts, the optimum cultivation temperature of mungbean sprouts may be $20{\sim}25^{\circ}C$.
To develop an effective anti-hangover product, hot-water extracts of 25 medicinal herbs were screened for inhibition or activation of alcohol dehydrogenase(ADH) and acetaldehyde dehydrogenase(ALDH), and 12 herbs were selected for further study. Chosen medicinal herb extracts(CMHEs) were fermented by Lactobacillus delbruechii subspecies lactis for 10 days at $35^{\circ}C$ after saccharification with nuruk(malt inoculated by 5 types of microbs) for 72 hours at $35^{\circ}C$ and both CMHEs and fermented CMHEs(FCMHEs) were explored for anti-hangover effects in vitro. We found significant ADH inhibition by hot-water extracts of Pueraria thunbergiana, Hovenia dulcis Thunb, Lycium chinense, Glycyrrhiza uralensis, Acanthopanax sessiliflorus, Liriope platyphylla, and Ixeris dentata, and significant ALDH activation by extracts of Acanthopanax sessiliflorus, Lycium chinense, Ixeris dentata, and Polypori umbellati of the Polyporaceae. The ADH effects on CMHE and FCMHE were -20.22% and -62.63% of control values, and the ALDH effects 173.20% and 280.17%, respectively. In rats given 20%(v/v) alcohol(15 mL/kg), FCMHEs significantly decreased blood acetaldehyde concentrations on 3 hours after ethanol administration, in a dose-dependent manner(p<0.05). Notably, blood acetaldehyde concentrations were markedly reduced in animals given FCMHEs(400 mg/kg) compared to levels seen in rats receiving CADB(commercial alcohol detoxification beverage). Thus, anti-hangover effects were promoted by fermentation of certain medicinal herb extracts.

Alcohol dehydrogenase (ADH) and Aldehyde dehydroganase (ALDH) activities of turnip extracts with ultrasonification at $60,\;100^{\circ}C$ temperature were investigated. Ultrasonification extracts showed a increase as compared with the normal extracts. ALDH activities were high than ADH activities. So, turnip extracts was shown efficient plants in alcohol oxidation. Appraisers liked powder extracts better than liquid extracts in sensory score.

Ginsenosides are major constituents of ginseng and are known to be responsible for its pharmacological properties. This study aimed to investigate the detoxification effect of a mixture containing black red ginseng powder, red ginseng extract, Puerariae radix extract, and Hovenia dulcis extract, on SD (Sprague Dawley) rats treated with 30% ethanol. Thirty minutes before treatment, the animals were orally administered different concentrations of the mixture or water. Results revealed that the concentration of ethanol in blood serum was significantly decreased in the black red ginseng mixture treated group in a dose-dependent manner, as compared with that of the control group. The blood level of acetaldehyde increased until 1 hr after alcohol administration, but the levels rapidly decreased later. Furthermore, ADH and ALDH activities in the hepatic tissue were also increased in the black red ginseng mixture administered group, than in the control group. These results indicate that the black red ginseng mixture has the ability of decomposing alcohol by increasing the ADH and ALDH activities responsible for alcohol metabolism.
$Microbulbifer$ sp. AJ-3 was used to acquire the degrading products from $Capsosiphon$$fulvescens$ (DPCF), which were investigated to determine its physiological activities. A crude enzyme extract from $Microbulbifer$ sp. AJ-3 hydrolyzes polysaccharide substrates such as agar, agarose, alginic acid, fucoidan, laminaran, starch, and chitin. Among them, agarose, laminaran, and alginic acid showed higher activities, especially alginic acid. The antioxidant activity of DPCF was measured by using 2,2-diphenyl-1-picryl-hydrazyl (DPPH) and superoxide dismutase (SOD)-like activities and were about 32% and 93% at 2 mg/ml, respectively. In addition, the nitrite-scavenging activity of DPCF was about 82%, 53%, and 12% at pH levels of 1.2, 3.0, and 6.0, respectively. To determine the influence of DPCF on alcohol metabolism, the generating activity of reduced-nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) was measured. The facilitating rate of ADH activity by DPCF was 130% at 2 mg/ml. The tyrosinase inhibitory activity of DPCF was slightly increased in a dose-dependent manner and was about 28% at 2 mg/ml. These results indicated that the enzymatic products from DPCF have a strong antioxidant, nitrite scavenging, and alcohol metabolizing activity.
In this study, increased alcohol decomposition efficacy (ADH) of Hoveina dulcis extract by Carbohydrate-Hydrolyzing Enzymes was investigated. Carbohydrate decomposition enzymes such as Maxinvert (Invertase), Optidex L-400 (Glucoamylase) and Rohament CL (Cellulase & Pectinase) were added to Hoveina dulcis extract at different concentrations (0.01, 0.05, 0.1, 0.5 and 1%) for 48 hrs, after which samples were taken every 6 hrs for determination of ADH activity. As the enzyme concentration became higher, ADH activity also increased. Especially, the addition of 1% Rohament CL increased enzyme activity to 76% at 30 hrs incubation, after which the increase in activity stopped. In the rat and human body experiment, enzymatic decomposition of Hovenia dulcis extract by addition of 1% Rohament CL was also effective in decreasing serum alcohol concentration and respiration. Especially, in the early stage after alcohol consumption, the efficacy of enzyme treatment of Hovenia dulcis extract was more effective. These results show that if the glycoside forms of active compounds such as flavonols in Hovenia dulcis extract are converted into aglycone forms, alcohol decomposition capability can be enhanced.

식이성 아연이 에탄올의 생체대사율에 미치는 영향을 검토코저 흰쥐에 식이중 아연의 함량(100ppm, 5ppm)을 달리하여 성장시키면서 에탄올을 4주 및 7주간 급여한 다음, 체중증가량과 에탄올의 대사에 관여한다고 알려져 있는 alcohol dehydrogenase, microsomal ethanol oxidizing system, catalase 및 aldehyde dehydrogenase의 활성변동과 혈중 에탄올 제거율을 측정한 결과는 다음과 같다. 실험기간 중 체중증가량은 대조군에 비해 Zn이 부족한 실험군에서 감소되었으며, 에탄올을 투여한 CE군과 ZnDE군은 control에 비해 현저히 감소하였다. Zn 부족한 군(ZnD)에서의 간 ADH, MEOS의 활성 및 혈액중 에탄올 제거율이 아연이 충분히 함유된 대조군에 비해 감소하였으나, catalase와 AIDH의 활성은 별다른 차이를 관찰할 수 없었다. 한편 에탄올을 투여한 CE 및 ZnDE군에서는 대조군(C 및 ZnD)에 비하여 ADH, MEOS 및 혈액중 에탄올 제거율이 증가하였으며 그 증가율은 아연을 충분히 급여한 CE군에서 아연이 부족한 ZnDE 군에 비하여 높게 나타났다. AIDH의 활성은 에탄올의 투여에 의해 CE군에서는 증가하였으나 ZnDE군에서는 별다른 변동을 관찰할 수 없었으며 catalase의 활성은 전실험군에 있어서 차이를 발견할 수 없었다.

This study is purposed to check up the natural 12 kinds of herbal extracts suitable for hangover and based on the results of contents of phenolic compounds, ABTS radical scavenging activity, alcohol dehydrogenase (ADH), and acetaldehyde dehydrogenase (ALDH). Selected 8 kinds of herbal extracts are blended according to the efficacy and the pearson's correlation between each content and activity. C. sinensis var. sinensis, P. densiflora Gnarl and P. lobata Ohwi showed excellent ADH activity, P. lobata Ohwi had a strong correlation between the content and efficacy, and C. sinensis var. sinensis, P. densiflora Gnarl had a negative correlation. Through the ADH and ALDH activity test of F.1 to F.7, the F.7 showed the highest synergic effect and selected as an optimal formulation. F.7 intake-group, the breath alcohol concentration was significantly reduced to 58% after 30 minutes and 27% after 120 minutes, compared to right after alcohol consumption. After alcohol consumption, there was a significant improvement effect (p<0.05) in tired and thirst in the intake group compared to the non-intake group.
