Antioxidative Effect and Component Analysis of Niaouli (Melaleuca quinquenervia) Leaf Extracts (니아울리 잎 추출물의 항산화 효과 및 성분 분석)

  • The antioxidative effects and component analysis of the Melaleuca quinquenervia leaf extracts were investigated. All experiments were performed with 50% ethanol extract, ethyl acetate fraction and aglycone fraction obtained from dried M. quinquenervia leaves. The DPPH (1,1-phenyl-2-picrylhydrazyl) scavenging activity ($FSC_{50}$) of ethyl acetate fraction ($10.05{\mu}g/mL$) of M. quinquenervia leaf extracts was similar to (+)-${\alpha}$-tocopherol($8.89{\mu}g/mL$) known as a typical antioxidant. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of the ethyl acetate fraction ($1.61{\mu}g/mL$) and aglycone fraction ($1.07{\mu}g/mL$) of leaf extracts of M. quinquenervia on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescence assay were similar to that of L-ascorbic acid ($1.50{\mu}g/mL$). The cellular protective effect of the extracts on the rose bengal sensitized photohemolysis of human erythrocytes was increased in a concentration dependant manner ($1{\sim}50{\mu}g/mL$). Especially, the cellular protective effects of Aglycone fraction (${\tau}_{50}=158.80min$) and 50% Ethanol extract (${\tau}_{50}=50.1{\pm}0.2min$) on the $^1O_2$-induced cellular damage of human cells were exhibited the higher than (+)-${\alpha}$-tocopherol (${\tau}_{50}=38.0min$). TLC and HPLC were used to analyse active components in the ethylacetate fraction of the extracts. Results showed that avicularin and quercetrin were active components of the extracts. These findings suggest that the M. quinquenervia leaf extracts can be applied to new cosmetics products as an effective antioxidant ingradient.

Water Flow Distribution and Sedimentation Characteristics of Particle Materials in the Sihwa Constructed Wetland (시화호 인공습지의 물흐름 분포 및 입자성물질 퇴적 특성)

  • Flow distribution of water and sedimentation rate were investigated to understand the hydrodynamics and settling characteristics of particulate materials in a constructed wetland for treatment of non-point sources pollutants, the Sihwa constructed wetland, Korea. The Sihwa constructed wetland is divided into three sub-wetlands(the Banwol, the Donghwa and the Samhwa wetlands) to treat the polluted water from three streams, the Banwol stream, the Donghwa stream and the Samhwa stream. From the results of water flow experiment using dye(Rhodamine 50WT Red), it was found that the water flow in the wetland was prevailing at the waterway and open water. Dye was spread slowly in the closed water area planted by plants. The mean hydraulic retention time(HRT) at the upper area of high wetland and lower wetland of Banwol, was found to be 34.1 hr at the upper area and 74.6 hr at the lower area respectively, totaling approximately 108.7 hr(4.5 days). The sedimentation rate was higher at lower area(sites of B, C and D) of the wetland than upper area(site of A which is settling zone). Based on the forecast for 20 years as to the amount of sediment that can be deposited in the open water in the future, the sediment depth of each area would be like this: A: 6.3 cm, B: 8.3 cm, C: 7.0 cm, D: 9.5 cm. The contents of organic materials in the sediment deposited within the sediment trap were found to be higher overly in the first investigation period which had much rainfall, and B, C and D areas were found to have an increased COD accumulation than A area. Also, nitrogen and phosphorus were found to increase in the down-stream of the wetland. The results of this study suggest that a sustainable research and management for the characteristics of water flow pattern and sedimentation changeable as time passes is needs to maintain or improve the efficiency of water treatment in the constructed wetland.

  • The calculation of the resulting fluid motion is an important problem of ship hydrodynamics. For a partially immersed body the condition of constant pressure at the free surface can be linearized. The resulting linear boundary-value problem for the velocity potential is the Neumann-Kelvin problem. The two-dimensional Neumann-Kelvin problem is studied for the half-immersed circular cylinder by Ursell. Maruo introduced a slender body approach to simplify the Neumann-Kelvin problem in such a way that the integral equation which determines the singularity distribution over the hull surface can be solved by a marching procedure of step by step integration starting at bow. In the present pater for the two-dimensional Neumann-Kelvin problem, it has been suggested that any solution of the problem must have singularities in the corners between the body surface and free surface. There can be infinitely many solutions depending on the singularities in the coroners.

Antioxidant and Cellular Protective Activities of Ecklonia cava Extracts against Reactive Oxyegen Species (감태(Ecklonia cava) 추출물의 항산화 및 세포보호 활성)

  • In this study, we investigated the antioxidative effects of brown seaweed Ecklonia cava extract and its subfractions. All experiments were performed with 50% ethanol extract, ethyl acetate fraction and aglycone fraction of E. cava. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activities ($FSC_{50}$) of ethyl acetate fraction ($FSC_{50}=6.98{\mu}g/mL$) and aglycone fraction ($7.03{\mu}g/mL$) are similar to that of (+)-${\alpha}$-tocopherol ($8.98{\mu}g/mL$) which is a reference control. Reactive oxygen species (ROS) scavenging activity (total antioxidant capacity, $OSC_{50}$) of the aglycone fraction ($OSC_{50}=14.48{\mu}g/mL$) on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescence assay was the strongest among all extract and fractions. However, all samples showed lower antioxidant activities than that of L-ascorbic acid ($6.88{\mu}g/mL$) known as a powerful antioxidant. The protective effect of 50% ethanol extract on the $^1O_2$-induced cellular damage of human erythrocytes was dependent on the concentration from 5 to $50{\mu}g/mL$. Both ethyl acetate fraction and aglycone fraction showed strong cellular protective activities at $10{\mu}g/mL$, where the cellular protective effects (${\tau}_{50}$) of each fraction were recorded 442.0 min and 539.9 min, respectively. Three kinds of extract/fractions of E. cava showed much greater cellular protective activities at $10{\mu}g/mL$ than that of liposoluble antioxidant (+)-${\alpha}$-tocopherol (40.6 min) which is a reference control. These results suggest E. cava extracts and its fractions can be applied as an antioxidant ingredient in a field of cosmetics.

Antioxidant Effect and Component Analysis of Cardiospermum halicacabum Leaf Extracts (풍선덩굴 잎 추출물의 항산화 효과 및 성분 분석)

  • In this study, the antioxidant effect and component analysis for extract and fractions of Cardiospermum halicacabum leaf were investigated. All experiments were performed with 50% ethanol extract, ethyl acetate fraction and aglycone fraction obtained from dried C. halicacabum leaf. The yields of extract and fractions were 16.4, 0.9 and 0.3% per dried powder, respectively. DPPH (1,1-phenyl-2-picrylhydrazyl) radical scavenging activity ($FSC_{50}$) of ethyl acetate fraction ($92.5{\mu}g/mL$) was the greatest radical scavenging activity, but lower than (+)-${\alpha}$-tocopherol ($8.9{\mu}g/mL$). In reactive oxygen species (ROS) scavenging activity (total antioxidant capacity, $OSC_{50}$) on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system, aglycone fraction ($4.2{\mu}g/mL$) was the highest total antioxidant capacity and similar to L-ascorbic acid ($1.5{\mu}g/mL$). The cellular protective effects of C. halicacabum leaf extract and fractions on the $^1O_2$-induced cellular damage of human erythrocytes were exhibited at all concentration-dependent ($5.0-25.0{\mu}g/mL$). Especially, aglycone fraction (${\tau}_{50}$, 76.4 min) in $25.0{\mu}g/mL$ showed the most protective effect among extracts. Components of the ethyl acetate fraction obtained from C. halicacabum extracts were analyzed by TLC, HPLC chromatogram and LC/ESI-MS. Results showed that the ethyl acetate fraction contained some flavonoids, such as apigenin-7-O-glucuronide, apigenin-7-glucosdie and quercitrin hydrate. These results suggest that the extracts and fractions of C. halicacabum leaf may be applied as antioxidant functional cosmetic raw materials.

Inhibitory Effects of Ethanol Extracts from Pine Buds (Pinus densiflora) on Angiotensin Converting Enzyme, Xanthine Oxidase and Nitric Oxide Synthesis (소나무 새순 에탄올 추출물의 angiotensin converting enzyme, xanthine oxidase 및 nitrix oxide synthase 활성)

  • Pine trees (Pinus densiflora Sieb. et Zacc.) have been used as a traditional health-promoting medicinal food in Korea. This research was performed to determine the antioxidative and antibacterial activities, tyrosinase, nitric oxide synthesis, angiotensin converting enzyme (ACE), and xanthine oxidase inhibition effects of the pine bud ethanol extract (PBE). Antioxidative activities of PBE were measured by using 2,2-diphenyl-1-picryl-hydrazyl (DPPH) free radical scavenging activity and superoxide dismutase-like activity (SODA). DPPH radical scavenging and SOD-like activities of PBE were remarkably increased in a dose-dependent manner, and were about 88.9% and 47.9% at 1 mg/ml and 10 mg/ml, respectively. The xanthine oxidase and angiotensin converting enzyme activities were inhibited about 71.9% and 60.8% at 1 mg/ml and $100{\mu}g/ml$ of PBE, respectively. The tyrosinase inhibitory activities of PBE were slightly increased in a dose-dependent manner. The PBE showed strong antimicrobial activities on Escherichia coli (E. coli) and Vibrio paraheamolyticus. Stimulation of the macrophages RAW264.7 cells with lipopolysaccharide (LPS) resulted in increased production of nitric oxide (NO) in the medium. However, NO synthesis was reduced up to 54% by addition of PBE at $200{\mu}g/ml$. These results revealed that pine buds have a strong antioxidative and anti-inflammatory activity, and exhibit angiotensin converting enzyme and xanthine oxidase inhibitory activities. This suggests that pine buds have the greatest property as a source for natural health products.

Effects of Radish Leaves Powder on Hepatic Antioxidative System in Rats Fed High-Cholesterol Diet (무청이 고콜레스테롤 식이 흰쥐 간조직의 항산화계에 미치는 영향)

  • The current study examined the effects of radish loaves powder on hepatic antioxidative system in rats fed high-cholesterol diet. Sprague-Dawley male rats weighing 100$\pm$10 g were randomly assigned to normal group (N group), normal diet with 5$\%$ radish leaves powder supplemented group (NR group) and high-cholesterol groups, which were sub-divided into radish leaves powder free diet group (HC group) and 2.5$\%$ (HRL group), 5$\%$ (HRM group), 10$\%$ (HRH group) radish leaves powder supplemented groups. Hepatic super oxide dimutase activity was no significant differences. Hepatic glutathione peroxidase activity was sig-nificantly increased in 5$\%$, 10$\%$ radish leaves powder supplemented groups. Hepatic hydrogen peroxide contents in cytosol were no significantly differences Hepatic hydrogen peroxide contents in mitochondria were sig-nificantly reduced in radish leaves powder supplemented groups. Hepatic superoxide radical contents in mi-crosome were significantly reduced in radish leaves powder supplemented groups. Hepatic superoxide radical contents in mitochondria were significantly reduced in 5$\%$, 10$\%$ radish leaves powder supplemented groups. Hepatic TBARS values were significantly reduced in 5$\%$, 10$\%$ radish leaves powder supplemented groups. Hepatic lipofuscin contents were no significant difference in high-cholesterol groups. Hepatic carbonyl values were significantly reduced in 5$\%$, 10$\%$ radish leaves powder supplemented groups among high-cholesterol groups. The results indicate that radish leaves may reduce oxidative damage by activating antioxidative de-fense system of liver in rats fed high-cholesterol diets.

Antioxidative Effect and Tyrosinase Inhibitory Activity of the Unripened Fruit Extract of Rubus coreanus Miquel (미성숙 복분자 과실의 항산화 효능 및 타이로시네이즈 저해 활성 평가)

  • In this study, the antioxidative effects and inhibitory activities of unripened fruit extract of Rubus coreanus Miquel (R. coreanus Miquel) on tyrosinase were investigated and the potential applicability as a cosmeceutical ingredients was evaluated. All experiments were performed with 50% ethanol extract, ethyl acetate fraction and aglycone fraction of unripened fruit extract of R. coreanus Miquel. The DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging activites ($FSC_{50}$) of 50% ethanol extract (6.56 ${\mu}g/mL$) and ethyl acetate fraction (6.14 ${\mu}g/mL$) of unripened fruit extract of R. coreanus Miquel were higher than (+)-${\alpha}$-tocopherol (8.98 ${\mu}g/mL$), which is known as a typical hydrophobic antioxidant. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of 50% ethanol extract (0.83 ${\mu}g/mL$), ethyl acetate fraction (0.84 ${\mu}g/mL$) and aglycone fraction (1.13 ${\mu}g/mL$) of R. coreanus Miquel on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were higher than L-ascorbic acid (1.5 ${\mu}g/mL$), which is known as s typical hydrophilic antioxidant. The cellular protective effect of extract and fraction of unripened fruit extract of R. coreanus Miquel on the rose bengal sensitized photohemolysis of human erythrocytes was increased in a concentration dependent manner (1 ~ 50 ${\mu}g/mL$). And 50% ethanol extract in 50 ${\mu}g/mL$ showed the most protective effect among extracts (${\tau}_{50}$ = 296.3 min). The inhibitory effects on tyrosinase of ethyl acetate and agylcone fractions were higher than arbutin. These results indicate that unripened fruit extracts of R. coreanus Miquel can be applied to antioxidant scavenging ROS including radical as an alternative whitening agent to replace arbutin.

Development of a Thermoplastic Oral Compensator for Improving Dose Uniformity in Radiation Therapy for Head and Neck Cancer (두경부암 방사선치료 시 선량 균일도 향상을 위한 Thermoplastic 구강 보상체의 개발)

  • Aquaplast Thermoplastic (AT) is a tissue-equivalent oral compensator that has been developed to improve dose uniformity at the common boundary and around the treated area during radiotherapy in patients with head and neck cancer. In order to assess the usefulness of AT, the degree of improvement in dose distribution and physical properties were compared to those of oral compensators made using paraffin, alginate, and putty, which are materials conventionally used in dental imprinting. To assess the physical properties, strength evaluations (compression and drop evaluations) and natural deformation evaluations (volume change over time) were performed; a Gafchromic EBT2 film and a glass dosimeter inserted into a developed phantom for dose verification were used to measure the common boundary dose and the beam profile to assess the dose delivery. When the natural deformation of the oral compensators was assessed over a two-month period, alginate exhibited a maximum of 80% change in volume from moisture evaporation, while the remaining tissue-equivalent properties, including those of AT, showed a change in volume that was less than 3%. In a free-fall test at a height of 1.5 m (repeated 5 times as a strength evaluation), paraffin was easily damaged by the impact, but AT exhibited no damage from the fall. In compressive strength testing, AT was not destroyed even at 8 times the force needed for paraffin. In dose verification using a glass dosimeter, the results showed that in a single test, the tissue-equivalent (about 80 Hounsfield Units [HU]) AT delivered about 4.9% lower surface dose in terms of delivery of an output coefficient (monitor unit), which was 4% lower than putty and exhibited a value of about 1,000 HU or higher during a dose delivery of the same formulation. In addition, when the incident direction of the beam was used as a reference, the uniformity of the dose, as assessed from the beam profile at the boundary after passing through the oral compensators, was 11.41, 3.98, and 4.30 for air, AT, and putty, respectively. The AT oral compensator had a higher strength and lower probability of material transformation than the oral compensators conventionally used as a tissue-equivalent material, and a uniform dose distribution was successfully formed at the boundary and surrounding area including the mouth. It was also possible to deliver a uniformly formulated dose and reduce the skin dose delivery.

Antioxidant and Cellular Protective Effects of Parthenocissus tricuspidata Stem Extracts Fermented by Lactobacillus pentosus (Lactobacillus pentosus 발효에 의한 담쟁이덩굴 줄기 추출물의 항산화 및 세포보호 효과)

  • In this study, the antioxidant activities, cellular protective effects, and inhibitory effects on elastase of non-fermented and fermented extracts of Parthenocissus tricuspidata (P. tricuspidata) stem using Lactobacillus pentosus were investigated. The free radical scavenging activities ($FSC_{50}$) of non-fermented and fermented extracts were 42.3 and $34.5{\mu}g/mL$, respectively, in which the activity after fermentation was approximately 18.4% higher. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) in $Fe^{3+}-EDTA/H_2O_2$ system of non-fermented and fermented extracts were 2.6 and $2.5{\mu}g/mL$, respectively. The activity after fermentation was approximately 4.2% higher. In the $^1O_2$-induced cellular damage of erythrocytes, the cellular protective effects (${\tau}_{50}$) of non-fermented and fermented extracts were 126.4 and 173.0 min at $50{\mu}g/mL$, respectively. The activity after fermentation was approximately 34.0% higher. The effect of fermented extract was 3.9 times higher than $(+)-{\alpha}$-tocopherol (${\tau}_{50}=43.4min$), known as a lipophilic antioxidant at $50{\mu}g/mL$. The inhibitory effect of elastase was investigated to predict the anti-wrinkle efficacy using Hs68 human fibroblasts cells. The elastase inhibitory activities ($IC_{50}$) of non-fermented and fermented extracts were 873.6 and $687.8{\mu}g/mL$, respectively, and the activity after fermentation was approximately 21.3% higher. These results indicated that fermented extract of P. tricuspidata stem has potentials as natural cosmetic ingredients with antioxidant and anti-wrinkle effect.
