Phytochemical Constituents of Acanthopanax senticosus (Rupr. & Maxim.) Harms Stem

  • Five constituents were isolated from the stem of Acanthopanax senticosus. Their structures were elucidated as (-)-sesamin (1), iso-fraxidin (2), 5-hydroxymethylfurfural (3), syringin (4) and acanthoside D (5) by spectral analysis. Among these compounds, 5-hydroxymethylfurfural (3) was isolated for the first time from this plant.

  • Three-dimensional (3D) culture system is useful technique for study of in vivo environment and it was used various experiments. This study was investigated to establish of embryo co-culture system and changes of PAs activity in 3D cultured endometrial cells of pigs. In results, growth of stromal cells into gel matrix were detected only with endometrial and myometrial cells. The most rapid growth of stromal cells were confirmed in $2.5{\times}10^5cells/ml$ and gel matrix containing 15% FBS. Expression of urokinase-PA (uPA) after treatment of hCG (0.5, 1.0, 1.5 and 2.0 IU/ml) were higher than without hCG, but, there are not significant difference among the treatment. On the other hand, expression of uPA after treatment of $IL-1{\beta}$ (0.1, 1, 10 and 100 ng/ml) were higher than without $IL-1{\beta}$, but, there are not significant difference. Expression of uPA after treatment of estrogen (0.2, 2, 20 and 200 ng/ml) were not difference, but PA activity was significantly decreased (p<0.05). Blastocyst was producing in PZM-3 medium containing FBS and endometrial cells were grown in PZM-3 medium. When embryos development with cultured endometrial cells, cleavage rates were not significant difference and blastocyst were not produced in co-culture with stromal cells and 3D culture system. 3D culture system had similar activity to in vivo tissue and these features are very useful for study of in vivo physiology. Nevertheless 3D culture system was not proper in embryo co-culture system. Therefore, we suggest that 3D culture system with embryo co-culture need continuous research.

Optimization of Culture Conditions for the Bioconversion of Vitamin $D_3\;to\;1{\alpha}$,25-Dihydroxyvitamin $D_3$ Using Pseudonocardia autotrophica ID9302

  • We assessed the ability of a Pseudonocardia sp. from soil samples to bioconvert vitamin $D_3$. The optimal culture conditions for the bioconversion of vitamin $D_3$ to active $1{\alpha}$,25-dihydroxyvitamin $D_3$ were investigated by varying the carbon and nitrogen sources, the metal salt concentrations, the initial pH, and the temperature. Microbial transformations were carried out with the addition of vitamin $D_3$ dissolved in ethanol. They were sampled by extraction with methanol-dichloromethane and the samples were examined by HPLC. Optimum culture conditions were found to be 0.4% yeast extract, 1% glucose, 3% starch, 1% fish meal, 0.2% NaCl, 0.01% $K_2HPO_4$, 0.2% $CaCO_3$, 0.01% NaF, and pH 7.0 at $28^{\circ}C$. The optimal timing of the addition of vitamin $D_3$ for the production of calcitriol by Pseudonocardia autotrophica ID9302 was concurrent with the inoculation of seed culture broth. Maximum calcitriol productivity and the yield of bioconversion reached a value of 10.4mg/L and 10.4% respectively on the 7th day in a 75L fementer jar under the above conditions.

  • Seven compounds were isolated from the stem of Acanthopanax senticosus by repeated col-umn chromatography. Their structures were elucidated as isovanillin (1), (-)-sesamin (2), iso-fraxidin (3), (+)-syringaresinol (4), 5-hydroxymethylfurfural (5), eleutheroside B (6), and eleuth-eroside E (7) by spectral analysis. Among them, isovanillin (1) was isolated for the first time from the family Araliaceae.

  • 본 연구에서는 전통 발효 식품에서 분리한 B. subtilis Y3-7 균주의 배양액이 $\alpha$-glucosidase의 활성에 미치는 영향을 살펴보았다. B. subtilis Y3-7 균주를 TSB 배지에 30시간 이상 배양한 배양액은 1.62 mg/mL의 농도로 yeast $\alpha$-glucosidase의 활성을 50%까지 저해할 수 있었다. 또한 효소학적 분석을 통해 이것이 배양액 내 물질이 효소의 기질과 경쟁적으로 작용하여 억제효과가 나타남을 확인하였다. B. subtilis Y3-7 균주의 배양액은 yeast 뿐만 아니라 실험동물의 장내 $\alpha$-glucosidase의 활성에도 영향을 미쳐 당부하에 따른 급격한 혈당상승을 억제할 수 있었다. 이와 같은 결과로 보아 대량배양이 가능한 B. subtilis Y3-7을 통해 탄수화물 흡수에 중요한 역할을 하는 $\alpha$-glucosidase 활성억제 물질을 얻음으로써 항당뇨 물질의 안정적인 생산이 기대되는 바이다.

  • Organic acid (OA) profiling analysis was performed in culture media from Lactobacillus pentosus K34 (L. pentosus K34) and Pediococcus lolli PL24 (P. lolli PL24) by gas chromatography-mass spectrometry (GC-MS) following methoxime/tert-butyldimethylsilyl derivatives. 12 OAs were positively identified in culture media. Most of OA levels from L. pentosus K34 of hetero lactic fermentation were found to be higher when compared with those from P. lolli PL24 of homo lactic fermentation, which may explain different OA metabolism in each strain. In addition, the distorted dodecagonal star patterns were readily distinguishable, and the characteristics of each strain were well represented. The present study demonstrates that the OA metabolic profiling method by GC-MS combined with star pattern recognition is useful for the monitoring study of characteristic OA metabolism in various microorganisms.

Hyperosmotic Stimulus Down-regulates $1{\alpha}$, 25-dihydroxyvitamin $D_3$-induced Osteoclastogenesis by Suppressing the RANKL Expression in a Co-culture System

  • The hyperosmotic stimulus is regarded as a mechanical factor for bone remodeling. However, whether the hyperosmotic stimulus affects $1{\alpha}$, 25-dihydroxyvitamin $D_3$ ($1{\alpha},25(OH)_2D_3$)-induced osteoclastogenesis is not clear. In the present study, the effect of the hyperosmotic stimulus on $1{\alpha},25(OH)_2D_3$-induced osteoclastogenesis was investigated in an osteoblast-preosteoclast co-culture system. Serial doses of sucrose were applied as a mechanical force. These hyperosmotic stimuli significantly evoked a reduced number of $1{\alpha},25(OH)_2D_3$-induced tartrate-resistant acid phosphatase-positive multinucleated cells and $1{\alpha},25(OH)_2D_3$-induced bone-resorbing pit area in a co-culture system. In osteoblastic cells, receptor activator of nuclear factor ${\kappa}B$ ligand (RANKL) and Runx2 expressions were down-regulated in response to $1{\alpha},25(OH)_2D_3$. Knockdown of Runx2 inhibited $1{\alpha},25(OH)_2D_3$-induced RANKL expression in osteoblastic cells. Finally, the hyperosmotic stimulus induced the overexpression of TonEBP in osteoblastic cells. These results suggest that hyperosmolarity leads to the down-regulation of $1{\alpha},25(OH)_2D_3$-induced osteoclastogenesis, suppressing Runx2 and RANKL expression due to the TonEBP overexpression in osteoblastic cells.

3차원 Co-culture 시스템을 통한 BMSC의 NP-like Cell로의 분화 (The Differentiation of bone Marrow Stromal Cells into NP-like Cells through 3-Dimensional Co-culture System)

  • The goal of this study is to investigate the effect and potential of three-dimensional Co-culture of BMSCs (bone marrow stromal Cells) and NP (nucleus pulposus) Cells on the differentiation of BMSCs into NP-like Cells. The NP Cells and BMSCs were isolated and cultured from New Zealand White rabbits. The isolated NP Cells and BMSCs were prepared in different alginate beads. Those two types of beads were separated by a track-etched membrane of $3\;{\mu}m$ pore in a 6-well culture plate. No growth factors were used. In addition to these, NP and BMSC were cultured in the beads independently for control. The number of Cells in Co-culturing system was half of those in two control groups. Proliferation and production of glycosaminoglycan (GAG) were evaluated along with histological observation. The GAG production rate(GAG contents/Cell) of Co-cultured BMSCs were much higher than that of BMSCs cultured alone. The total amounts of GAG produced by BMSCs in Co-culturing system were larger than those produced by BMSCs in control group and were comparable with those produced by NP alone even the number of each Cell was half of BMSCs in Co-culturing system. This study showed the potential of differentiation of BMSCs into NP-like Cells through three-dimensional Co-culture system even without any chemical agents.

두 종 미세 녹조류의 연속배양을 통한 바이오매스 생산성 비교 (Comparison of Biomass Productivity of Two Green Microalgae through Continuous Cultivation)

  • In the present study, the biomass productivity of two green microalgae (Chlorella sp. and Dunaliella salina DCCBC2) were assessed in a 12 L tubular photobioreactor under optimum culture conditions. In the batch culture optimization process, the Chlorella sp. biomass was obtained as 1.2 g/L under atmospheric air as a sole $CO_2$ source and other culture conditions as follows: light intensity, temperature, pH, $NH_4Cl$ and $K_2HPO_4$ were 100 ${\mu}E/m^2/s$, $27^{\circ}C$, 7.0, 20.0 mM and 2.0 mM, respectively. On the other hand, 2.9 g/L of D. salina DCCBC2 biomass production was observed under the following conditions: light intensity, temperature, pH, $KNO_3$ and $K_2HPO_4$were 80 ${\mu}E/m^2/s$, $27^{\circ}C$, 8.0, 3.0 mM and 0.025 mM, respectively. At 1% $CO_2$ supply to the reactor, the Chlorella sp. production was reached 1.53 g/L with 25% increment under the same operating conditions. In addition, the maximum D. salina DCCBC2 biomass was observed as 3.40 g/L at 3% $CO_2$ concentration. Based on the aforementioned optimized conditions, the dilution rate and maximal biomass productivity of Chlorella sp. and D. salina DCCBC2 in the continuous cultivation were 0.4/d and 0.6 g/L/d and 0.6/d and 1.5 g/L/d, respectively.

Effects of a Soaking-Fermentation-Drying Process on the Isoflavone and ${\gamma}$-Aminobutyric acid Contents of Soybean

  • In our study, lactic acid bacteria (LAB)-fermented whey solutions were applied in the soybean soaking process to minimize bacterial contamination and to enrich the biologically functional components of isoflavone and $\gamma$-aminobutyric acid (GABA). Among the 11 LAB tested, Bifidobacteria infantis and a mixed culture (Lactobacillus acidophilus, Bifidobacteria lactis, and Streptococcus thermophilus; ABT-3) displaying the greatest $\beta$-glucosidase activity were selected to produce improved biologically functional soybean preparations. In the soybean soaking processing (without water spraying), the LAB-cultured 10% whey solution was used to soak and to ferment the soybeans and the fermented soybeans were finally dried by heat-blowing at $55^{\circ}C$. The processing conditions used in this study demonstrated that the final soybean product had a reduced contamination by aerobic and coliform bacteria, compared to raw soybeans, likely due to the decrease in pH during LAB fermentation. The aglycone content of the isoflavone increased up to 44.6 mg per 100 g of dried soybean by the processing method, or approximately 8-9 times as much as their initial content. The GABA contents in the processed samples increased as the processing time of soaking-fermentation proceeded as well. The soybean sample that fermented by ABT-3 culture for 24 hr showed the greatest increase in GABA content (23.95 to 97.79 mg/100 g), probably as a result of the activity of glutamate decarboxylases (GAD) released from the soybean or produced by LAB during the soaking process.
