Production of Bleach-Stable and Halo-Tolerant Alkaline Protease by an Alkalophilic Bacillus pumilus JB05 Isolated from Cement Industry Effluents

  • A new alkalophilic strain of Bacillus pumilus JB¬05 producing bleach-stable and halo-tolerant alkaline protease was isolated from cement industry effluents in Karnataka, India. The effects of carbon and nitrogen sources on protease production by this alkalophilic strain were observed after a 30-h incubation. A high level of alkaline protease activity was obtained in the presence of starch as the carbon and peptone as the nitrogen sources. The partially purified enzyme showed an optimum temperature and pH activity at $58^{\circ}C$ and 10.5, respectively. The enzyme was completely inhibited by PMSF (95.0%) indicating it as a serine protease. It is bleach-stable as it retained 35% original activity in the presence of 10% (v/v) hydrogen peroxide at $30^{\circ}$C after 2 h and is halo-tolerant as it retained 70% original activity in the presence of 2.5 M sodium chloride at $30^{\circ}C$ after 2 h incubation.

  • Objective : The present study designed this study to investigate anti-oxidative effects of EF on HaCaT keratinocyte. Method : The present study measured the amount of polyphenoics and flavonoids, and also measured the levels of catalase, Ascorbate peroxidase (APX), SOD like activities and DPPH free radical scavenging activity. Then the effects of SB on viability and prolferation rates, and protective effects against oxidative stress induced by chemicals such as hydrogen peroxide and rotenone were also investigated. Results and conclusion : EF showed protective effect against cell death of HaCaT keratinocyte induced by rotenone and SNP significantly. In conclusion, these results suggest that EF may have anti-oxidantic action in human skin and also suggest that EF can be used as anti-aging agent.

Antioxidant and α-Amylase Inhibitory Activity of 70% Ethanolic Extract from Morinda citrifolia L. (Noni) (노니가루 70% 에탄올 추출물의 항산화 및 α-Amylase Inhibitory 활성)

  • In this study, polyphenol and flavonoid contents were measured, and DPPH, OH, H2O2 radical scavenging activity, and the α-amylase inhibitory activity were measured to study the antioxidant activity of 70% ethanol extract from Morinda citrifolia L. The polyphenol and flavonoid contents of noni 70% ethanol extract were 29.52 GAE/g and 12.48 CE/g, respectively. Also, the IC50 values of DPPH, hydroxyl radical, and hydrogen peroxide scavenging activity of 70% ethanol extract from noni were 18.70 mg/mL, 26.45 mg/mL, and 35.67 mg/mL, respectively. Measurement of the α-amylase inhibitory activity of 70% ethanol extract from noni showed 45% inhibitory activity at 10 mg/mL.

Diversity in Activities of Peroxidase and Polyphenoloxidase in the Akagare or Helminthosporium-infected Rice Leaves (적고(赤枯) 및 호마엽고(胡麻葉枯) 수도엽중(水稻葉中) Peroxidase와 Polyphenoloxidase의 활성(活性))

  • The activities of peroxidase and polyphenoloxidase were investigated in the rice leaves(the upper halves) diseased with Akagare or Helminthosporium oryzae. The activity of polyphenoloxidase was slightly lower than that of peroxidase in the healthy leaves but it increased 56% in the diseased leaves while peroxidase decreased 35%. It was expected that polyphenoloxidase is dominant in the oxidation of polyphenols, and hydrogen peroxide may accumulate to harmful level due to the decrease of peroxidase activity resulting in non-enzymatic oxidation of polyphenols in the diseased leaves.

Selective Production of Aromatic Aldehydes from Heavy Fraction of Bio-oil via Catalytic Oxidation

  • High value-added aromatic aldehydes (e.g. vanillin and syringaldehyde) were produced from heavy fraction of bio-oil (HFBO) via catalytic oxidation. The concept is based on the use of metalloporphyin as catalyst and hydrogen peroxide ($H_2O_2$) as oxidant under alkaline condition. The biomimetic catalyst cobalt(II)-sulfonated tetraphenylporphyrin ($Co(TPPS_4)$) was prepared and characterized. It exhibited relative high activity in the catalytic oxidation of HFBO. 4.57 wt % vanillin and 1.58 wt % syringaldehyde were obtained from catalytic oxidation of HFBO, compared to 2.6 wt % vanillin and 0.86 wt % syringaldehyde without $Co(TPPS_4)$. Moreover, a possible mechanism of HFBO oxidation using $Co(TPPS_4)/H_2O_2$ was proposed by the research of model compounds. The results showed that this is a promising and environmentally friendly method for production of aromatic aldehydes from HFBO under $Co(TPPS_4)/H_2O_2$ system.

  • A peroxopolyoxotungsten-based ionic hybrid was synthesized by anion-change of peroxopolyoxometalate (POM) $PW_4O{_{24}}^{3-}$ with dicationic long-chain alkyl imidazolium ionic liquids. The characterization was conducted by FT-IR, TGA, $^1H$-NMR and CHN Elemental analyses. Its catalytic performance was evaluated by the epoxidation of soybean oil with $H_2O_2$ under solvent-free condition, including testing of organic cations influence, catalytic reusability and reaction conditions. The catalyst was proved to be a highly efficient recyclable catalyst for epoxidation of various vegetable oils with $H_2O_2$, showing high $H_2O_2$ utilization efficiency, high catalytic activity, convenient recovery and good reuse ability.

Template Synthesis of Nitrogen-Doped Short Tubular Carbons with Big Inner Diameter and their Application in Electrochemical Sensing

  • Nitrogen-doped short tubular carbons (N-STCs) with big inner diameter have been successfully synthesized via carbonization of polydopamine (PDA) wrapped halloysite nanotubes (HNTs). The obtained N-STCs have average length of $0.3{\mu}m$ with big inner diameter (50 nm), thin wall (2-3 nm) and large surface area ($776m^2g^{-1}$), and show excellent electrochemical properties. As an example in electrochemical applications, N-STCs were used to electrochemically detect hydrogen peroxide ($H_2O_2$) and glucose. The results showed that the N-STCs modified glassy carbon (N-STCs/GC) electrode had much better analytical performance (lower detection limit and wider linear range) compared to the acid-treated carbon nanotubes (AO-CNTs) based GC electrode. The unique structure endows N-STCs the enhanced electrochemical performance and promising applications in electrochemical sensing.

  • A disposable electrochemical immunosensor system has been developed for the detection of herbicide in aqueous samples. Disposable screen printed carbon electrodes(SPCE) were used as basic electrodes and an enzyme, horseradish peroxidase (HRP), and anti-herbicide antibodies was immobilised on to the working electrode of SPCE by using avidin-biotin coupling reactions. An herbicide-glucose oxidase conjugates have been used for the competitive immunoreaction with sample herbicides. The enzymatic reaction between the conjugated glucose oxidase and glucose added generates hydrogen peroxide, which was reduced by the peroxidase immobilised. The latter process caused an electrical current change, due to direct re-reduction of peroxidase by a direct electron transfer mechanism, which was measured to determine the herbicides in the sample. The optimal operational condition was found to be: $20\;{\mu}gl-1$ deglycosylated avidin loading to the working electrode and working potential +50 mV vs. Ag/AgCl. The total assay time was 15 min after sample addition. The detection limits for herbicides, atrazine and simazine, were found to be 3 ppb and 10 ppb, respectively.

Protective Effects of Ramie (Boehmeria nivea) against Oxidative Stress in C6 Glial Cells

  • β amyloid protein (Aβ) plays a critical role in the pathogenesis of Alzheimer's disease (AD) and possibly in Aβ-induced mitochondrial dysfunction and oxidative stress. Aβ can directly cause reactive oxygen species (ROS) production. Overproduction of ROS is considered to be involved in the pathogenesis of neurodegeneration of AD. Here, we investigated 9 kinds of ramie (Boehmeria nivea, (L.) Gaud., BN; hereafter denoted as BN) for their protective action against oxidative stress in a cellular system using C6 glial cells. We observed loss of cell viability and high levels of ROS generation after treatment with hydrogen peroxide (H2O2) and Aβ25-35. However, treatments with BN extracts led to an increase in cell viability and decrease in ROS production induced by H2O2 and Aβ25-35. In particular, the extracts of BN-01 (seobang variety from Seocheon) and BN-09 (local variety from Yeonggwang) showed excellent anti-oxidative properties. This indicates that BN extracts could prevent neurodegeneration by reducing oxidative stress in cells.

The Radioprotective Effect of Ginseng Extracts on the liver in Mice that was irradiated by radiation (방사선이 조사된 생쥐 간에서 인삼추출물이 방사선 방어효과에 미치는 영향)

  • Radioprotective effects of ginseng extracts on liver damage induced by high energy x-ray were studied. To one group of ICR male mice were given white(50 mg/kg/day for 7days, orally) and fermenta ginseng extracts(500 mg/kg/day for 7days, orally) before irrdiation. To another group were irradiated by 5 Gy dose of high energy x-ray. Contrast group were given with saline(0.1 ml). This study also investigated the radioprotective effect between SOD, CAT, hydrogen peroxide and ginseng extracts on hepatic damage. This study measured the level of superoxide dismutase(SOD), catalase(CAT), hydrogen peroxide($H_2O_2$) in liver tissue. Administrating orally white (50 mg/kg/day for 7days, orally) and fermenta ginseng extracts(500 mg/kg/day), the activity of SOD, CAT were generally increased and the hydrogen peroxide($H_2O_2$) was decreased. After irradiation, the activity of SOD, CAT were generally decreased and the hydrogen peroxide($H_2O_2$) was increased. Therefore, ginseng extracts increased antioxidative enzyme activity. And We know that the antioxidatant effect of extracts from white and fermenta ginseng protect radiation damage by direct antioxidant effect involving SOD, CAT. It was included that ginseng can protect against radiation damage through its antioxidatant properties.
