Morphological, Cytological and Molecular Evidence for Intersubgeneric F1 Hybrid between Glycine max x G. tomentella (콩 Glycine max와 G. tomentella의 종간교잡으로부터 얻은 Fl식물체 검증을 위한 형태적 · 세포학적 · 분자유전학적 연구)

  • This study was carried out to demonstrate morphological, cytological and molecular evidence for intersubgeneric $F_1$ hybrid between Glycine tomentella and G. max cv. ‘Baemkong’. Morphological features of $F_1$ plant for pistil and stamen, flower color and growth habit showed intermediate type between G. tomentella and G. max cv. ‘Baemkong’. Chromosome number of $F_1$ plant was 2n=39, which explained the evidence of $F_1$ hybrid between G. tomentella (2n=38) and G. max cv. ‘Baemkong’ (2n=40). Polyacrylamide isoelectric focusing pattern for esterase and peroxidase also showed that the $F_1$ plant was true $F_1$ hybrid between G. tomentella and G. max cv. ‘Baemkong’. From RAPD analysis, we identified that 62 primers showing bands in $F_1$ hybrid had both bands from G. tomentella and G. max cv. ‘Baemkong’, which suggested that this was true $F_1$ hybrid. Based on our results from morphological, cytological and molecular analyses, we suggest that the $F_1$ plant was true intersubgeneric hybrid between G. tomentella and G. max cv. ‘Baemkong’. Our results still remain us further study to recover fertility of $F_1$ hybrids. The occurrence of maternal and/or paternal inheritance in $F_1$ hybrid from intersubgeneric cross between G. tomentella and G. max cv. ‘Baemkong’ need to be explained.

Chemical Composition of Seed from Inbred Lines and Hybrids of Maize Recently Developed in Korea (국내 육성 주요 옥수수 자식계통 및 교잡종의 종실 성분 특성)

  • This study was carried out to evaluate the quality properties, fatty acid composition and amino acid composition of major Korean inbred lines and hybrids of maize for corn breeding. Protein content of $F_2$ seeds was 9.0%, which was slightly lower than those of inbred lines, 11.4%, and $F_1$ seeds, 10.9%. in hybrid maizes. Lipid content of $F_2$ seeds of hybrid maizes was 4.2%, slightly higher than those of inbred lines, 3.3%, and $F_1$ seeds, 3.6%. However, there was no significant difference in ash content. The amount of linoleic acid in inbred lines, $F_1$ and $F_2$ seeds of hybrid maizes was highest among all fatty acids and followed by oleic acid, palmitic acid, stearic acid and linolenic acid. The composition of saturated fatty acid of $F_2$ seeds was 17.6%, similar to $F_1$ seeds but it was lower than that of inbred lines. The composition of unsaturated fatty acid of $F_2$ seeds was 82.4%, similar to $F_1$ seeds but was higher than inbred lines. The content of leucine of $F_2$ seeds was lower than inbred lines and $F_1$ seeds. The content of valine, serine, threonine, cysteine, aspartic acid, lysine, arginine, histidine of $F_2$ seeds was higher than inbred lines and $F_1$ seeds. MMA (monoamino monocarboxylic acid) in inbred lines, $F_1$ and $F_2$ seeds of hybrid maizes was highest among all amino acids and followed by DMA (diamino monocarboxylic acid) and AAA (aromatic amino acid).

Degrees of Heterosis and Inbreeding Depression of Quantitative Characters in Silkworm by Diallel Corsses (누에 이면교잡에 의한 형질발현의 잡종강세와 약세)

  • The purpose of this experiment was to obtain the basic information on heterosis and inbreeding depression of quantitative characters in the silkworm by diallel crosses of seven varieties differing widely in each character. Period of 5th instar cocoon weight, cocoon layer weight, cocoon layer ratio, boiling off ratio, raw silk percent, bave length, bave weight, bave size, weight of first molting larva and fourth molting larva were investigated. The results obtained were summarized as follows : The degrees of positive heterosis of cocoon weight and cocoon layer weight were higher. Period of 5th instar and boiling off ratio were negative heterosis. In F1 generation, cocoon weight and bave weight showed positive heterobeltiosis, while fifth instar-period, cocoon layer ratio and boiling off ratio showed negative heterobeltiosis in the F1 and F2 generation. Inbreeding depression was observed in cocoon layer ratio and boiling off ratio.

Inhibition of Xanthine Oxidase by Flavonols from Onion Skin (양파껍질에서 분리한 플라보놀의 Xanthine Oxidase 저해기작)

  • The influence of flavonols from onion skin on xanthine oxidase was investigated. Methanol extract was showed 12.8% of yield, 661.3mg% of flavonoids contents and 88.7% of inhibitory effect on xanthine oxidase F1 and F2 fractions were obtained from the methanol extract by ODS and Sephadex LH-20 chromatography. F1 and F2 fractions flavonols(3-OH free) identified by UV/visible spectroscopy. Inhibitory effect of F1 and F2 on xanthine oxidase were increased with increasing concentration. IC50s of F1 and F2 were 0.95$\mu\textrm{g}$ and 0.67$\mu\textrm{g}$, respectively. To confirm the specificity of F1 and F2 against xanthine oxidase, albumin was added to the reaction mixture. The inhibition of F1 and F2 may be due to specific binding to xanthine oxidase. The modes of their inhibitions were of mixed type with respect to xanthine as a substrate.

  • In this paper, we solve the additive ρ-functional inequalities (0.1) ||f(2x-y)+f(y-x)-f(x)|| $\leq$ ||${\rho}(f(x+y)-f(x)-f(y))$||, where ρ is a fixed complex number with |ρ| < 1, and (0.2) ||f(x+y)-f(x)-f(y)|| $\leq$ ||${\rho}(f(2x-y)-f(y-x)-f(x))$||, where ρ is a fixed complex number with |ρ| < $\frac{1}{2}$. Using the direct method, we prove the Hyers-Ulam stability of the additive ρ-functional inequalities (0.1) and (0.2) in β-homogeneous F-spaces.

Evaluation of Reciprocal Cross Design on Detection and Characterization of Mendelian QTL in $F_2$ Outbred Populations

  • A simulation study was conducted to evaluate the effect of reciprocal cross on the detection and characterization of Mendelian QTL in $F_2$ QTL swine populations. Data were simulated under two different mating designs. In the one-way cross design, six $F_0$ grand sires of one breed and 30 $F_0$ grand dams of another breed generated 10 $F_1$ offspring per dam. Sixteen $F_1$ sires and 64 $F_1$ dams were randomly chosen to produce a total of 640 $F_2$ offspring. In the reciprocal design, three $F_0$ grand sires of A breed and 15 $F_0$ grand dams of B breed were mated to generate 10 $F_1$ offspring per dam. Eight $F_1$ sires and 32 $F_1$ dams were randomly chosen to produce 10 $F_2$ offspring per $F_1$ dam, for a total of 320 $F_2$ offspring. Another mating set comprised three $F_0$ grand sires of B breed and 15 $F_0$ grand dams of A breed to produce the same number of $F_1$ and $F_2$ offspring. A chromosome of 100 cM was simulated with large, medium or small QTL with fixed, similar, or different allele frequencies in parental breeds. Tests between Mendelian models allowed QTL to be characterized as fixed (LC QTL), or segregating at similar (HS QTL) or different (CB QTL) frequencies in parental breeds. When alternate breed alleles segregated in parental breeds, a greater proportion of QTL were classified as CB QTL and estimates of QTL effects for the CB QTL were more unbiased and precise in the reciprocal cross than in the one-way cross. This result suggests that reciprocal cross design allows better characterization of Mendelian QTL in terms of allele frequencies in parental breeds.

Fruit Yield and Morphological Characters of Parental Cultivars and Intervarietal Hybrids of Capsicum annuum L. (고추의 친품종 및 품종간 잡종의 과실수량과 형태적 특성)

  • The fruit yield and morpholgical characters of F$_{1}$ hybrids between Capscium annuum L. 'California Wonder', four native Japanese cultivars, and three Korean F$_{1}$ cultuvars were evaluated and the results compared with parental cultivars. The F$_{1}$ hybrids obtained by crossing 'California Wonder' and native Japanese cultivars flowered 3 days earlier on average compared to parental cultivars. The yields of F$_{1}$ hydrids were higher than those of the parental cultivars, especially when one of the parents was 'California Wonder'. Also F$_{1}$ hybrid whose one parent was 'California Wonder' and 'Fushimi Amanaga' gave the highest fruit yield. The number of locules and seeds per fruit from of the 'California Wonder' hybrids whose one parent was 'California Wonder' was more than of the native Japanese cultivars. The plant height in F$_{1}$ hybrids obtained by crossing between 'California Wonder' and four native Japanese cultivars tended to be higher than that of the parental cultivars.
