Free radical scavenging activity and protective effect from cellular oxidative stress of active compound from eggplant (Solanum melongena L.) (가지(Solanum melongena L.) 활성물질의 라디칼 소거능과 산화적 스트레스에 대한 세포 보호 효과)

  • To investigate the protective effect of eggplant (Solanum melongena L.) and its active compound, delphinidin, we used in vitro and cellular system. The active fraction from eggplant, BuOH fraction, showed protective effect from hydrogen peroxide-induced oxidative stress in WI-38 fibroblast cells. It suggests that eggplant would have the protective activity from radical-induced oxidative damage and its BuOH fraction would play the crucial role with antioxidative activity. In addition, delphinidin, the active compound from eggplant, exerted the strong 1,1-diphenyl-2-picrylhydrazyl scavenging effect with $IC_{50}$ value of 6.59 ${\mu}g/mL$. Furthermore, the cellular oxidative stress was induced by 2,2'-azobis (2-amidinopropane) dihydrochloride (AAPH) in LLC-$PK_1$ cells, while treatment of delphinidin atteunated AAPH-induced oxidative stress as dose-dependent manner. The present study suggests the antioxidative activity of eggplant and delphinidin against free radical-induced oxidative stress.

Effect of Subfractions of Carthamus tinctorius L. Semen on the Lipid Peroxidation and Oxygen Free Radical Scavenging Enzyme Activities in CCl_4$-induced Hepatotoxic Rats (홍화자 분획물의 사염화탄소 유발 간손상 흰쥐에서 지질과산화와 oxygen free radical 제거 효소 활성도에 미치는 영향)

  • Previous studies have shown that methanol extract and its butanol fraction of Carthamus tinctorius L. Semen have the hepatoprotective effect on the CCl4-induced hepatotoxicity. The hepatoprotective effect of the subfractions of butanol fraction has been evaluated by analyzing oxygen free radical scavenging enzyme activities and histopathological examinations. In BS-5 subfraction treated group, the activity of superoxide dismutase has been significantly increased as compared with that of CCl4, treated rats. Antioxidant activity has been evaluated by the examination of the scavenging activity against 1,1-diphenyl-2-picrylhydrazyl radical. BS-5 subfraction has shown strong antioxidant activities. The histopathological examination showed that the treatment of BS-5 subfraction has relieved the ballooning degeneration of hepatocytes which had been generated by CCl4. It appears that the protective effect of BS-5 subfraction would be mediated of the attenuation of lipid peroxidation by acting as a free radical scavenger, which were based on the increase of superoxide dismutase activity.

Stabilization of Quercetin using Organo-hectorite and Its Application in Sunscreen Cosmetics (오가노 헥토라이트를 이용한 쿼세틴 안정화 및 자외선 차단 제품 응용에 관한 연구)

  • In this study, a quercetin benton gel (QUERPLEX) that stabilized quercetin was prepared using organo hectorite, and its efficacy was confirmed. In addition, a comparative study was conducted on the stability and effectiveness of applying this to sunscreen cosmetics. It was confirmed that QUERPLEX remained stable without showing crystal precipitation and growth for 4 weeks. As a result of measuring antioxidant activity using 2,2-diphenyl-1-picrylhydrazyl (DPPH), it showed antioxidant activity depending on the concentration, and showed a radical elimination ability of 70% or more at a concentration of 2,500 ppm or more, confirming a significant effect. Anti-inflammatory activity experiments using RAW 264.7 cells confirmed that NO production decreased in a concentration-dependent manner by reducing NO production by 8% at 25 ㎍/mL, 23% at 50 ㎍/mL, and 84% at 100 ㎍/mL. As a result of confirming the stability of the formulation according to the method of quercetin in the sunscreen formulation, the stability of the formulation was improved when quercetin was added directly to the formulation. It also improved the UV protection index on in vitro and in vivo, which is expected to have the potential as a component that can have a new boosting effect on UV protection. These results suggest that organo hectorite is very effective as a quercetin carrier and that it can be applied in cosmetic formulations by not only expressing the efficacy of quercetin but also bringing about additional effects.

The Anti-oxidative Compounds of Smilax riparia Leaves (Smilax riparia 잎의 항산화 성분)

  • Rhizoma of Smilax china has been used as anti-inflammatory and analgesic, antiedemic agent in Korean folk medicine. In order to investigate the efficacy of anti-oxidative activity, the activity-guided fractionation and the isolation were performed. Each fractions ($H_2O$ fraction, 20%, 40%, 60%, 100% MeOH fractions and CHCl$_3$ fraction) was examined antioxidant activity by 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging potential. It was revealed that 40%, 20% MeOH fractions and $H_2O$ fractions have significant anti-oxidative activity. From 40% and 20% MeOH fractions two flavonoid glycosides and one procyanidin were isolated and elucidated apigenin-7-Ο-$\alpha$-L-rhamnopyranosyl(1\longrightarrow2)-$\beta$-D-glucopyranoside, apigenin-7-Ο-$\alpha$-L-rhamnopyranosyl(1\longrightarrow6)-$\beta$-D-glucopyranoside and catechin(4$\alpha$\longrightarrow6)epicatechin through their physicochemical data and IR, FAB-MS, $^{13}$ C-NMR, and $^1$H-NMR analysis with authentics, respectively. The isolated compounds were examined by DPPH method. Apigenin-7-Ο-$\alpha$-L-rhamnopyranosyl(1\longrightarrow2)-$\beta$-D-glucopyranoside and catechin (4$\alpha$\longrightarrow6) epicatechin showed powerful radical scavenging activities on DPPH radical among three compounds.

Antioxidant Activity of Gomchi (Ligularia fischeri) Leaves

  • Ligularia fischeri (LF) has been used to treat jaundice, scarlet-fever, rheumatoidal arthritis, and hepatic diseases. This study was carried out to determine the antioxidant activity of the extract from the leaves of LF by using various established in vitro systems with ${\alpha}$-tocopherol as positive control. The results showed that the ethanol extract of LF (0.05-0.5 mg/mL) displayed a strong free radical scavenging activity against 1,1-diphenyl-2-picrylhydrazyl, ${O_2}^{{\cdot}-}$, and $H_2O_2$ radicals. It was observed that LF extract (1 mg/mL) significantly decreased the levels of malondialdehyde (MDA) to 69 and 89% of control, measured by ferric thiocyanate (FTC) and thiobarbituric acid (TBA) test, respectively. LF extract (0.5-2 mg/mL) was also found to inhibit significantly the amount of malondialdehyde formed from liver homogenate to 69-56% of control. Similarly, in the high $Fe^{2+}/ascorbate$ induction system LF extract (1-2 mg/mL) inhibited carbonyl formation measured by 2,4-dinitrophenylhydrazine reaction to 89-79% of control. Like antioxidant activity, the reducing power of LF extract was excellent at concentration of 0.5-2 mg/mL. These results indicate that LF could be used as a potential source of natural antioxidant.

  • Berries and branches essential oil of Juniperus phoenicea were obtained by electromagnetic induction heating assisted extraction and by hydrodistillation with a yield varied from ($1.2{\pm}0.3$ to $2.4{\pm}0.7%$) and from ($0.6{\pm}0.1%$ to $1.1{\pm}0.1%$), respectively. forty eight compounds were identified representing (97.2 - 99.7%) of the oil. ${\alpha}$-Pinene (40.3 - 67.8%) and ${\delta}$-3-carene (13.5 - 26.8%) were the main compounds in berries and branches essential oils. Antioxidant activity was evaluated by three means: inhibition of 2, 2-diphenyl-1-picryl hydrazyl (DPPH) free radical, reducing power and ${\beta}$-Carotene/linoleic acid bleaching. The antioxidant activity of essential oils showed $IC_{50}$ ranging from $67.6{\pm}1.02{\mu}g/mL$ to $131.5{\pm}0.8{\mu}g/mL$ for berries and from $98{\pm}1.25{\mu}g/mL$ to $166.8{\pm}0.29{\mu}g/mL$ for the branches. Berries oil show more potent antioxidant activity compared to branches. This result is supported by the three methods investigated in this work.

Effect of Origanum vulgare Extracts on Hair Regeneration (오레가노 추출물의 모발성장 촉진효과)

  • This study was carried out to investigate the effect of Origanum vulgare extracts on cell proliferation of human hair dermal papilla cell (HHDPC) using sulforhodamine B (SRB) assay, antioxidant activity by 1,1-diphenyl-2-picryl hydrazyl (DPPH) method, expression of insulin-like growth factor-1 (IGF-1) by analyzing reverse transcriptase polymerase chain reaction (RT-PCR) and hair growth in a shaving animal model of C57BL/6 mice topically applying with an amount of 0.1 mL once a day for 3 weeks. The mice were divided into 4 groups including normal group (saline, N), negative control group (dimethyl sulfoxide, NC), positive control group (5 mg/mL minoxidil, PC), and experimental group (Origanum vulgare extracts, OV). Treatment of OV didn't show cytotoxicity in HHDPC up to 10 ${\mu}g/mL$ and exhibited antioxidant activity with $IC_{50}$ of 31.0 ${\mu}g/mL$. IGF-1 expression in the skin was significantly (p<0.05) increased in the PC and OV compared to the N or NC. PC and OV also showed a prominently promoted hair regrowth compared to the N or NC in hair growth observation. The hair regrowth of OV was significantly higher than that of PC (p<0.05). Therefore, these results indicate that O. vulgare extracts effectively stimulated hair growth in an animal model.

Studies on the Phenolic Compound Content and Antioxidant Efficacy of Rubus idaeus (Raspberry) Cultivar 'Willamette' Fruit Extract Using n-Hexane and Ethyl Acetate (n-Hexane 및 Ethyl Acetate를 이용한 Willamette 품종 라즈베리추출물의 페놀 화합물 함량 및 항산화 효능 연구)

  • In this study, Rubus idaeus (Raspberry) cultivar 'Willamette' fruit extract(RIFE) was prepared from the freeze-dried raspberry powder, n-hexane and ethyl acetate, and then the phenolic compound content, ferric reducing ability, and radical scavenging ability were measured. The raspberry cultivar 'willamette', 'polka', and 'polana' compound fruit extract did not show cytotoxicity up to the concentration of 10%. As a result of conducting an experiment at the concentration, it was confirmed that the total phenolic compound content was 375.3 ppm, and the total flavonoid content was 43.46 ppm, and the ferric reducing ability by ferric reducing antioxidant power (FRAP) reagent was equivalent to FeSO4 0.532 mM. It was confirmed that 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability was 94.5 ± 0.7%, and the 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging ability was 99.4 ± 2.82%, and the nitric oxide (NO) radical scavenging activity was 88.5 ± 0.4%. When compared with the L-ascorbic acid 'standard' solution, DPPH radical scavenging ability was between 25 - 50 ppm / ABTS radical scavenging ability was close to 100 ppm / NO radical scavenging ability was more than 1,000 ppm. These results suggest that the raspberry cultivar 'willamette' fruit extract could be applied as an effective cosmetic material with antioxidant activity.

Antioxidant and inflammation inhibitory effects from fruiting body extracts of Ganoderma applanatum

  • In this study, the antioxidant and anti-inflammatory effects of methanol extract (ME) and hot water extracts (HE) from the fruiting bodies of Ganoderma applanatum were investigated. The 1,1-diphenyl-2-picryl-hydrazy (DPPH) radical scavenging activity of 2.0 mg/mL ME (94.83%) was comparable to that of butylated hydroxytoluene (96.97%), the reference standard. The hydroxyl radical scavenging activities of ME and HE were similar to that of BHT at 2.0 mg/mL, whereas lipid peroxidation activity of the ME and HE were significantly lower than that of BHT. High-performance liquid chromatography analysis showed that the G. applanatum fruiting bodies contained nine phenolic compounds, which might contribute to antioxidant and anti-inflammatory activities. The survival rate of RAW 264.7 macrophages treated with 2.0 mg/mL ME and HE were 65.23 to 68.12% at 2.0 mg/mL, thereby indicating that the extracts were slightly cytotoxic at the concentration tested. The extracts also inhibited the nitric oxide (NO)-mediated expression of inducible nitric oxide synthase (iNOS) protein in lipopolysaccharide-induced RAW 264.7 macrophages and carrageenan-induced paw edema in rats. The study results demonstrated that the fruiting bodies of G. applanatum possessed good antioxidant and anti-inflammatory activities, which might be used to develop novel anti-inflammatory agents.

Effects of Kuseonwangdogo on the Proliferation of Preadipocyte 3T3-L1 Cells, the Anti-Complementary and the Cytotoxic Effects (구선왕도고가 전지방세포(前脂肪細胞) 3T3-L1의 증식(增殖), 항보체활성(抗補體活性) 및 세포독성(細胞毒性)에 미치는 영향)

  • To investigate the anti-complementary and cytotoxic effects of oriental prescription, Kuseonwangdogo, on the proliferation of preadipocyte 3T3- L1 cells, we examined biological effects of Kuseonwangdogo. The results obtained were as follows. 1. After 14 days, the body weight of rats treated with Kuseonwangdogo decreased more than that in the control group (p<0.05). However, the weights of liver, spleen and kidney were unchanged. In serum biochemical test, we examined the level of glucose (GLU) and glutamic pyruvic transaminase (GPT). The levels of GOT and CHOL in serum were decreased remarkably by the administration of Kuseonwangdogo (p<0.05). The haematological examination of the tested group showed significant increment of white blood cells (WBC), hemoglobin concentration (HGB), mean corpuscular hemoglobin (MCH) and monocyte (MO). 2. The effect of Kuseonwangdogo on the proliferation of 3T3-L1 cells was tested by the sulforhodamin B(SRB) assay. The high concentration ($100{\mu}l\;and\;200{\mu}l$) of extracts inhibited the proliferation of 3T3- L1 cells. The p-value was <0.01, respectively. 3. The extract of Kuseonwangdogo showed a potent anti -complementary activity. It was suggested that the active principle may be a kind of polysaccharide molecule. 4. The cytotoxic effects of Kuseonwang dogo and its composing herbs in human liver cells (WRL68) and monkey kidney cells (Vero) were examined by the SRB and 3- (4,5- Dimethylthiazol-2-yl) -2,5 diphenyl-2H- tetrazolium bromide (MTT) assay. Cytotoxic effects were not observed.
